This work aims to study the effects of adding antioxidants, such as, α- tocopherol and BHT on the thermal and oxidative stability of biodiesel from cottonseed (B100). The Biodiesel was obtained through the methylical and ethylical routes. The main physical and chemical properties of cotton seed oil and the B100 were determined and characterized by FTIR and GC. The study of the efficiency of antioxidants, mentioned above, in concentrations of 200, 500, 1000, 1500, 2000ppm, to thermal and oxidative stability, was achieved by Thermogravimetry (TG), Differential Thermal Analysis (DTA), Differential Scanning Calorimetry (DSC), Differential Scanning Calorimetry - Hi-Pressure (P-DSC) and Rancimat. The Biodiesel obtained are within the specifications laid down by Resolution of ANP No7/2008. The results of TG curves show that the addition of both antioxidants, even in the lowest concentration, increases the thermal stability of Biodieseis. Through the DTA and DSC it was possible to study the physical and chemical transitions occurred in the process of volatilization and decomposition of the material under study. The initial time (OT) and temperature (Tp) of oxidation were determined through the P-DSC curve and they showed that the α-tocopherol has a pro-oxidant behavior for some high concentrations. The BHT showed better results than the α-tocopherol, with regard to the resistance to oxidation

Recent lines of evidences indicate that several pathological conditions, as cardiovascular diseases, are associated with oxidative stress. In order to validate a butylated hydroxytoluene (BHT)-induced experimental model of oxidative stress in the cardiac tissue and serum lipids, 12 Wistar rats were divided into two groups, a control group and the BHT group, Which received BHT i.p. twice a week (1500 mg/kg body Weight) during 30 days. BHT group presented lower body weight gain and heart weight. BHT induced toxic effects on serum through increased triacylglycerols (TG), VLDL and LDL-cholesterol concentrations. The heart of BHT animals showed alteration of antioxidant defenses and increased concentrations of lipid hydroperoxides, indicating elevated lipoperoxidation. TG concentrations and lactate dehydrogenase activities were elevated in the cardiac Muscle of BHT animals. Thus, long-term administration of BHT is capable to induce oxidative and metabolic alterations similarly to some pathological disorders, constituting an efficient experimental model to health scientific research. The use of antioxidants either to prevent or retard food's lipids oxidation was approved after inquires that verified their security within a daily intake limit. In this study, the methodology was developed and validated for the analysis of synthetic antioxidants: propylgallate (PG), tert-butylhydroquinone (TBHQ), butylhydroxyanisole (BHA), octylgallate (OG) and butylhydroxytoluene (BHT) in vegetables oils, margarine and hydrogenated fats by high performance liquid chromatographic. The methodology revealed itself efficient, with recovery rates above 90% for all antioxidant substances, besides good linearity in concentration range of 40-240 mg kg-1 (r = 0,999), repeatability with CV < 3,7% and limit of quantification 16.55, 10.32, 1.40, 3.76 and 9.30 mg/kg for BHT, BHA, PG, OG and TBHQ, respectively.

Total phenolic contents, antioxidant activity and chemical composition of propolis samples from three localities of Minas Gerais state (southeast Brazil) were determined. Total phenolic contents were determined by the Folin-Ciocalteau method, antioxidant activity was evaluated by DPPH, using BHT as reference, and chemical composition was analyzed by GC/MS. Propolis from Itapecerica and Paula Candido municipalities were found to have high phenolic contents and pronounced antioxidant activity. From these extracts, 40 substances were identified, among them were simple phenylpropanoids, prenylated phenylpropanoids, sesqui- and diterpenoids. Quantitatively, the main constituent of both samples was allyl-3-prenylcinnamic acid. A sample from Virginopolis municipality had no detectable phenolic substances and contained mainly triterpenoids, the main constituents being alpha-and beta-amyrins. Methanolic extracts from Itapecerica and Paula Candido exhibited pronounced scavenging activity towards DPPH, indistinguishable from BHT activity. However, extracts from Virginopolis sample exhibited no antioxidant activity. Total phenolic substances, GC/MS analyses and antioxidant activity of samples from Itapecerica collected monthly over a period of 1 year revealed considerable variation. No correlation was observed between antioxidant activity and either total phenolic contents or contents of artepillin C and other phenolic substances, as assayed by CG/MS analysis.

The effect of butylated hydroxytoluene/butylated hydroxyanisole blend (BHT/BHA), and rosemary and oregano extracts, added individually or in combination, on lipid oxidation and fatty acid composition was investigated on irradiated frozen beef burgers. Irradiation treatment was carried out using a (60)CO semi-industrial irradiator at doses of 6, 7 and 8 kGy, and then the treated meat samples were stored at -20 degrees C for 90 days. Lipid oxidation and fatty acid composition of beef samples were evaluated by measurement of TBARS and gas chromatography, respectively. The results of the experiment showed that rosemary extract, applied alone and in combination with either BHT/BHA or oregano extracts was more effective in maintaining a low oxidation level in the samples compared to oregano extract used individually or in combination with BHT/BHA. Results also showed no significant differences (p > 0.05) in fatty acid composition in all analyzed samples, although some changes in terms of decreased PUFA and MUFA, beside of slight increase of SFA content were observed. However, these differences do not correlate positively neither with the irradiation dose nor the type of antioxidant. Thus, there is a potential application of these spices as natural antioxidants in irradiated meats. (C) 2009 Elsevier Ltd. All rights reserved.

Several epidemiological and research studies suggest that a high intake of foods rich in natural antioxidants increases the antioxidant capacity of the plasma and reduces the risk of some kinds of cancers, heart diseases, and stroke. These health benefits are attributed to a variety of constituents, including vitamins, minerals, fiber, and numerous phytochemicals, such as flavonoids. Thus, in addition to measuring the composition of the usual macronutrients and micronutrients, it seems important to also measure the antioxidant capacity of foods. For this purpose, 28 foods including fruits, vegetables and commercially-frozen fruit pulps were analyzed for antioxidant capacity. The antioxidant capacity of the foods varied from 0.73 to 19.8 mu mol BHT equiv/g. The highest values were observed for wild mulberries (19.8 mu mol BHT equiv/g), acai fruit pulp (18.2 mu mol BHT equiv/g) and watercress (9.6 mu mol BHT equiv/g). The antioxidant capacities are only indicative of the potential of the bioactive compounds; however, these data are important to explore and understand the role of fruit, vegetables and other foods in health promotion. The effect of addition of rosemary and oregano extracts on the sensory quality of irradiated beef burger was investigated. Batches of beef burgers were prepared with 400 ppm of rosemary or oregano extract and a group prepared with 200 ppm of synthetic butyl-hydroxytoluene (BHT)/butyl-hydroxy-anisol (BHA) was used as a control. Half of each formulation was irradiated at the maximum dose allowed for frozen meat (7 kGy). Samples were kept under frozen conditions (-20 degrees C) during the whole storage period, including during irradiation. Two analyses were performed after 20 and 90 days to verify the influence of the addition of the different types of antioxidants and the effect of irradiation and storage time on the acceptance of the product. Thirty-three and thirty-four untrained panelists were invited to participate in the first and second test, respectively. A structured hedonic scale ranging from 1 to 9 points was used in both analyses. BHT/BHA formulation obtained the highest score (6.73) and regarding the natural antioxidants, oregano received better acceptance (6.36). Irradiated samples formulated with oregano received a lower score, 6.03 in the first test and 5.06 in the second one, compared to the non-irradiated sample (6.36 and 5.79). In the second test (90 days), the sample formulated with BHT/BHA and which was irradiated received a higher score (6.59) when compared to the non-irradiated one (5.85). In both tests, the irradiated samples formulated with rosemary extract obtained a better score compared to the non-irradiated one, the scores being 5.00-3.82 and 5.00-3.76 in the first and second test, respectively. Our results allowed us to conclude that the natural antioxidants, rosemary and oregano extracts, present a good alternative for replacing synthetic additives in food industries, and that the irradiation process, in some cases, may help to enhance the sensory quality of food. The antioxidant activity of natural and synthetic compounds was evaluated using five in vitro methods: ferric reducing/antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydradzyl (DPPH), oxygen radical absorption capacity (ORAL), oxidation of an aqueous dispersion of linoleic acid accelerated by azo-initiators (LAOX), and oxidation of a meat homogenate submitted to a thermal treatment (TBARS). All results were expressed as Trolox equivalents. The application of multivariate statistical techniques suggested that the phenolic compounds (caffeic acid, carnosic acid, genistein and resveratrol), beyond their high antioxidant activity measured by the DPPH, FRAP and TBARS methods, showed the highest ability to react with the radicals in the ORAC methodology, compared to the other compounds evaluated in this study (ascorbic acid, erythorbate, tocopherol, BHT, Trolox, tryptophan, citric acid, EDTA, glutathione, lecithin, methionine and tyrosine). This property was significantly correlated with the number of phenolic rings and catecholic structure present in the molecule. Based on a multivariate analysis, it is possible to select compounds from different clusters and explore their antioxidant activity interactions in food products.

We report on the cardiovascular effects of noradrenaline (NA) microinjection into the hypothalamic supraoptic nucleus (SON) as well as the central and peripheral mechanisms involved in their mediation. Microinjections of NA 1, 3, 10, 30 or 45 nmol/100 nL into the SON caused dose-related pressor and bradycardiac response in unanesthetized rats. The response to NA 10 nmol was blocked by SON pretreatment with 15 nmol of the alpha(2)-adrenoceptor antagonist RX821002 and not affected by pretreatment with equimolar dose of the selective alpha(1)-adrenoceptor antagonist WB4101, suggesting that local alpha(2)adrenoceptors mediate these responses. Pretreatment of the SON with the nonselective beta-adrenoceptor antagonist propranolol 15 nmol did not affect the pressor response to NA microinjection of into the SON. Moreover, the microinjection of the 100 nmol of the selective alpha(1)-adrenoceptor agonist methoxamine (MET) into the SON did not cause cardiovascular response while the microinjection of the selective alpha(2)adrenoceptor agonists BHT920 (BHT, 100 nmol) or clonidine (CLO, 5 nmol) caused pressor and bradycardiac responses, similar to that observed after the microinjection of NA. The pressor response to NA was potentiated by intravenous pretreatment with the ganglion blocker pentolinium and was blocked by intravenous pretreatment with the V(1)-vasopressin receptor antagonist dTyr(CH2)5(Me)AVP, suggesting an involvement of circulating vasopressin in this response. In conclusion, our results suggest that pressor responses caused by microinjections of NA into the SON involve activation of local alpha(2)-adrenoceptor receptors and are mediated by vasopressin release into circulation. This study was undertaken to test whether the structural remodelling of pulmonary parenchyma can be sequentially altered in a model and method that demonstrate the progression of the disease and result in remodelling within the lungs that is typical of idiopathic pulmonary fibrosis. Three groups of mice were studied: (i) animals that received 3-5-di-tert-butyl-4-hydroxytoluene (BHT) and were killed after 2 weeks (early BHT = 9); (ii) animals that received BHT and were killed after 4 weeks (late BHT = 11); (iii) animals that received corn oil solution (control = 10). The mice were placed in a ventilated Plexiglas chamber with a mixture of pure humidified oxygen and compressed air. Lung histological sections underwent haematoxylin-eosin, immunohistochemistry (epithelial, endothelial and immune cells) and specific staining (collagen/elastic fibres) methods for morphometric analysis. When compared with the control group, early BHT and late BHT groups showed significant decrease of type II pneumocytes, lower vascular density in both and higher endothelial activity. CD4 was increased in late BHT compared with early and control groups, while CD8, macrophage and neutrophil cells were more prominent only in early BHT. The collagenous fibre density were significantly higher only in late BHT, whereas elastic fibre content in late BHT was lower than that in control group. We conclude that the BHT experimental model is pathologically very similar to human usual interstitial pneumonia. This feature is important in the identification of animal models of idiopathic pulmonary fibrosis that can accurately reflect the pathogenesis and progression of the human disease.

O uso do biodiesel como um substituto total ou parcial do diesel de petróleo é significativo, entretanto a sua baixa estabilidade oxidativa é um ponto desfavorável. Portanto o uso de antioxidantes é viável desde que diminuam a velocidade de degradação promovido pela oxidação. Mas a ação de um antioxidante está relacionada com a composição química do biodiesel ao qual é adicionado. Considerando a importância e o crescimento da indústria de biodiesel, a necessidade de utilização de antioxidantes, e a busca por novas fontes como matérias primas, faz-se necessário estudar a eficiência de antioxidantes para o aumento da vida útil do biodiesel, além de avaliar novas culturas com potencial de produção como o pinhão manso (Jatropha curcas L.) e nabo forrageiro (Raphanus sativus L.). O objetivo deste trabalho foi avaliar os antioxidantes BHT (butilhidroxitolueno), PA (ácido pirogálico) e PG (galato de propila), em concentrações de 50, 200, 500, 1000 e 2000 mg/kg, nos biodiesel de pinhão manso, nabo forrageiro e soja, empregando a metodologia PetroOXY, ensaio acelerado de oxidação, para a determinação do seu período de indução. Os biodieseis foram obtidos através de reações de transesterificação, utilizando rota metílica, com rendimentos superiores a 80% e foram caracterizados através de alguns parâmetros físico-químicos. Foi observada a eficiência do uso de antioxidantes no aumento da vida útil do biodiesel. E com o aumento da concentração houve um aumento do período de indução. A eficiência do BHT foi sempre crescente com o aumento da concentração para todos os biodieseis enquanto que para o PG e PA houve uma diminuição da eficiência na maior concentração. A metodologia usada para avaliação da estabilidade oxidativa, PetroOXY, mostrou-se eficaz, de boa repetibilidade e reprodutibilidade, além de ser mais rápida e completa que o método Rancimat. Na concentração mais elevada, 2000 mg/kg, o melhor desempenho foi para o BHT, nas concentrações inferiores a 2000 mg/kg, a melhor eficiência foi para o PA ~ PG, nos três biodieseis estudados, soja, nabo forrageiro e pinhão manso.

Natural and synthetic xanthone derivatives are well-known for their ability to act as antioxidants and/or enzyme inhibitors. This paper aims to present a successful synthetic methodology towards xanthenedione derivatives and the study of their aromatization to xanthones. Additionally their ability to reduce Fe(III), to scavenge DPPH radicals and to inhibit AChE was evaluated. The results demonstrated that xanthenedione derivative 5e, bearing a catechol unit, showed higher reduction capacity than BHT and similar to quercetin, strong DPPH scavenging activity (EC50 = 3.79 ± 0.06 μM) and it was also showed to be a potent AChEI (IC50 = 31.0 ± 0.09 μM) when compared to galantamine (IC50 = 211.8 ± 9.5 μM).

El principal objetivo de este proyecto es consolidar una línea de investigación en el campo de la Química Analítica, área que por el momento está poco desarrollada en nuestro país y particularmente en la Universidad Nacional de Córdoba. Para ello se tiene previsto continuar con el estudio de inmovilización de reactivos de interés práctico para su uso en la modificación de electrodos con el fin de emplearlos como sensores electroquímicos. Se comenzará con el estudio de electrodos químicamente modificados con partículas de polímeros preparados a partir de una mezcla de prepolímero resol de fenol o fenoles sustituidos y formaldehído. Estos polímeros tienen una muy buena respuesta para la cuantificación de fenoles y compuestos etílicos en general, los que no sólo son contaminantes industriales muy importantes, sino que además están relacionados con la industria de la alimentación debido a que algunos compuestos de base fenólica se usan como antioxidantes. Se tiene previsto analizar la síntesis y caracterización de prepolímero, la preparación del electrodo de trabajo y la optimización del sistema en un sistema de flujo continuo. También se considerará la preparación de la muestra y la evaluación del método. Finalmente se compararán los resultados obtenidos con aquellos logrados mediante la técnica patrón que recomienda la Association of Official Analytical Chemist (AOAC). El estudio se llevará a cabo con el análisis del antioxidante 2,6-di-tert-butilo-4-cresol (BHT) en diferentes muestras de aceites vegetales e industriales. También se analizarán en muestras de agua de la ciudad de Córdoba y del río Suquía el contenido total de fenoles.

O objetivo deste trabalho foi avaliar o efeito da inclusão de extratos etanólicos na ração, obtidos do caroço e da casca da manga, sobre o desempenho de frangos e a oxidação lipídica da carne. Foram utilizados 360 pintos machos da linhagem Ross 308, de um dia de idade, distribuídos em delineamento inteiramente casualizado, com seis tratamentos e seis repetições de dez aves. Os tratamentos consistiram de: ração sem adição de antioxidante (controle); ração com adição de 200 ppm do antioxidante butilato de hidroxitolueno (BHT); ração com 200 ou 400 ppm de extrato da casca da manga (ECAS); e ração com 200 ou 400 ppm de extrato do caroço da manga (ECAR). A adição de BHT ou dos extratos da manga não influenciou significativamente o consumo de ração, o ganho de peso e a conversão alimentar. A adição de BHT e a de 400 ppm de ECAR proporcionaram maior estabilidade lipídica da carne fresca, mensurada pelas substâncias reativas ao ácido tiobarbitúrico antes do armazenamento. O extrato etanólico do caroço da manga, na dosagem de 200 e 400 ppm, retarda a oxidação lipídica da carne de frangos armazenada por 15 dias.

O objetivo deste trabalho foi avaliar o efeito de extratos etanólicos do caroço e da casca de manga, sobre o desempenho de poedeiras e sobre a qualidade e estabilidade lipídica dos ovos. Um total de 180 poedeiras comerciais Hisex White foi distribuído ao acaso em seis tratamentos, com cinco repetições de seis aves. Os tratamentos consistiram de: ração sem adição de antioxidante; ração com 200 ppm do antioxidante butilato de hidroxitolueno (BHT); ração com 200 ou 400 ppm de extrato da casca de manga (Ecas); ração com 200 ou 400 ppm de extrato de caroço de manga (Ecar). Foram avaliados: o consumo de ração, a produção de ovos, o peso do ovo, a massa de ovo produzida (grama por ave por dia), a conversão alimentar e características de qualidade dos ovos. A oxidação lipídica da gema durante o armazenamento foi determinada pela quantificação das substâncias reativas ao ácido tiobarbitúrico. As aves alimentadas com a ração sem adição de antioxidantes produziram ovos com os piores valores de unidade Haugh e maior oxidação lipídica da gema. Os teores de 400 ppm de Ecas e 200 ou 400 ppm de Ecar foram efetivos na prevenção de danos oxidativos aos ovos durante o armazenamento e podem ser utilizados na alimentação das poedeiras como substituto ao antioxidante sintético.
