A 10-fold BAC library for giant panda was constructed and nine BACs were selected to generate finish sequences. These BACs could be used as a validation resource for the de novo assembly accuracy of the whole genome shotgun sequencing reads of giant panda newly generated by the Illumina GA sequencing technology. Complete sanger sequencing, assembly, annotation and comparative analysis were carried out on the selected BACs of a joint length 878 kb. Homologue search and de novo prediction methods were used to annotate genes and repeats. Twelve protein coding genes were predicted, seven of which could be functionally annotated. The seven genes have an average gene size of about 41 kb, an average coding size of about 1.2 kb and an average exon number of 6 per gene. Besides, seven tRNA genes were found. About 27 percent of the BAC sequence is composed of repeats. A phylogenetic tree was constructed using neighbor-join algorithm across five species, including giant panda, human, dog, cat and mouse, which reconfirms dog as the most related species to giant panda. Our results provide detailed sequence and structure information for new genes and repeats of giant panda, which will be helpful for further studies on the giant panda.

Sugarcane has an importance in Brazil due to sugar and biofuel production. Considering this aspect, there is basic research being done in order to understand its physiology to improve production. The aim of this research is the Base Excision Repair pathway, in special the enzyme MUTM DNA-glycosylase (formamidopyrimidine) which recognizes oxidized guanine in DNA. The sugarcane scMUTM genes were analyzed using four BACs (Bacterial Artificial Chromosome) from a sugarcane genomic library from R570 cultivar. The resulted showed the presence in the region that had homology to scMUTM the presence of transposable elements. Comparing the similarity, it was observed a highest similarity to Sorghum bicolor sequence, both nucleotide and peptide sequences. Furthermore, promoter regions from MUTM genes in some grass showed different cis-regulatory elements, among which, most were related to oxidative stress, suggesting a gene regulation by oxidative stress

El Centro Histórico de Cuenca tiene como actividades predominantes el comercio, la administración, gestión de servicios, entre otros. Sin embargo, el uso de vivienda se ha ido perdiendo a través de los años. Este abandono perjudica a toda la ciudad, puesto que las personas buscan establecer sus nuevos domicilios en zonas periféricas, ocasionando disgregación y dispersión espacial de actividades, alterando el correcto crecimiento de la ciudad. Así mismo, uno de los mayores problemas que enfrenta el Centro Histórico es el no poder desarrollarse como un organismo vivo, teniendo un uso combinado de vivienda y comercio, que permite que la zona esté activa todo el tiempo. La habitabilidad es fundamental para el mantenimiento y buena salud de los Centros Históricos. Cuenca no es la excepción y es necesario devolverle a esta zona de la ciudad una función residencial más intensa. Por estas razones y a manera de estudio, se ha decidido hacer un proyecto de diseño de modelos arquitectónicos de vivienda multifamiliar en predios vacíos del área de Primer Orden del Centro Histórico de Cuenca, no solo respetando la normativa municipal, sino también aplicando la normativa BACS ensamble!, desarrollada por el Departamento de Investigación Espacio y Población a través del grupo LlactaLab Ciudades Sustentables de la Universidad de Cuenca, que defiende la posibilidad de generar barrios compactos sustentables, en zonas consolidadas de la ciudad, que ayuden a construir un modelo de ciudad diferente a la actual, donde el ser humano sea el centro de reflexión.

Crash statistics that include the blood alcohol concentration (BAC) of vehicle operators reveal that crash involved motorcyclists are over represented at low BACs (e.g., ≤0.05%). This riding simulator study compared riding performance and hazard response under three low dose alcohol conditions (sober, 0.02% BAC, 0.05% BAC). Forty participants (20 novice, 20 experienced) completed simulated rides in urban and rural scenarios while responding to a safety-critical peripheral detection task (PDT). Results showed a significant increase in the standard deviation of lateral position in the urban scenario and PDT reaction time in the rural scenario under 0.05% BAC compared with zero alcohol. Participants were most likely to collide with an unexpected pedestrian in the urban scenario at 0.02% BAC, with novice participants at a greater relative risk than experienced riders. Novices chose to ride faster than experienced participants in the rural scenario regardless of BAC. Not all results were significant, emphasising the complex situation of the effects of low dose BAC on riding performance, which needs further research. The results of this simulator study provide some support for a legal BAC for motorcyclists below 0.05%.

Introduction: Within the context of road safety it is important that workload (the portion of a driver’s resources expended to perform a task) remains at a manageable level, preventing overloading and consequently performance decrements. Motorcyclists are over represented in crash statistics where the vehicle operator has a positive, low blood alcohol concentration (BAC) (e.g., 0.05%). The NASA task load index (NASA-TLX) comprises sub-scales that purportedly assess different aspects of subjective workload. It was hypothesized that, compared to a zero BAC condition, low BACs would be associated with increases in workload ratings, and decrements in riding performance. Method: Forty participants (20 novice, 20 experienced) completed simulated motorcycle rides in urban and rural scenarios under low dose BAC conditions (0.00%, 0.02%, 0.05% BAC), while completing a safety relevant peripheral detection task (PDT). Six sub-scales of the NASA-TLX were completed after each ride. Riding performance was assessed using standard deviation of lateral position (SDLP). Hazard perception was assessed by response time to the PDT. Results: Riding performance and hazard perception were affected by alcohol. There was a significant increase in SDLP in the urban scenario and of PDT reaction time in the rural scenario under 0.05% BAC compared to 0.00% BAC. Overall NASA-TLX score increased at 0.02% and 0.05% BAC in the urban environment only, with a trend for novices to rate workload higher than experienced riders. There was a significant main effect of sub-scale on workload ratings in both the urban and rural scenarios. Discussion: 0.05% BAC was associated with decrements in riding performance in the urban environment, decrements in hazard perception in the rural environment, and increases in overall ratings of subjective workload in the urban environment. The workload sub-scales of the NASA-TLX appear to be measuring distinct aspects of motorcycle riding-related workload. Issues of workload and alcohol impaired riding performance are discussed.

The tomato I-3 gene introgressed from the Lycopersicon pennellii accession LA716 confers resistance to race 3 of the fusarium wilt pathogen Fusarium oxysporum f. sp. lycopersici. We have improved the high-resolution map of the I-3 region of tomato chromosome 7 with the development and mapping of 31 new PCR-based markers. Recombinants recovered from L. esculentum cv. M82 × IL7-2 F2 and (IL7-2 × IL7-4) × M82 TC1F2 mapping populations, together with recombinants recovered from a previous M82 × IL7-3 F2 mapping population, were used to position these markers. A significantly higher recombination frequency was observed in the (IL7-2 × IL7-4) × M82 TC1F2 mapping population based on a reconstituted L. pennellii chromosome 7 compared to the other two mapping populations based on smaller segments of L. pennellii chromosome 7. A BAC contig consisting of L. esculentum cv. Heinz 1706 BACs covering the I-3 region has also been established. The new high-resolution map places the I-3 gene within a 0.38 cM interval between the molecular markers RGA332 and bP23/gPT with an estimated physical size of 50-60 kb. The I-3 region was found to display almost continuous microsynteny with grape chromosome 12 but interspersed microsynteny with Arabidopsis thaliana chromosomes 1, 2 and 3. An S-receptor-like kinase gene family present in the I-3 region of tomato chromosome 7 was found to be present in the microsyntenous region of grape chromosome 12 but was absent altogether from the A. thaliana genome.

• In December 1986 funds were approved to double the intensity of random breath testing (RBT) and provide publicity support for police efforts. These changes were considered necessary to make RBT effective. • RBT methods were changed in the metropolitan area to enable block testing (pulling over a block of traffic rather than one or two cars), deployment of police to cut off escape routes, and testing by traffic patrols in all police subdivisions. Additional operators were trained for country RBT. • A publicity campaign was developed, aimed mainly at male drivers aged 18-50. The campaign consisted of the “cardsharp” television commercials, radio commercials, newspaper articles, posters and pamphlets. • Increased testing and the publicity campaigns were launched on 10 April 1987. • Police tests increased by 92.5% in May – December 1987, compared with the same period in the previous four years. • The detection rate for drinking drivers picked up by police who were cutting off escape routes was comparatively high, indicating that drivers were attempting to avoid RBT, and that this police method was effective at detecting these drivers. • A telephone survey indicated that drivers were aware of the messages of the publicity campaign. • The telephone survey also indicated that the target group had been exposed to high levels of RBT, as planned, and that fear of apprehension was the major factor deterring them from drink driving. • A roadside survey of driver blood alcohol concentrations (BACs) by the University of Adelaide’s Road Accident Research Unit (RARU) showed that, between 10p.m. and 3a.m., the proportion of drivers in Adelaide with a BAC greater than or equal to 0/08 decreased by 42%. • Drivers under 21 were identified as a possible problem area. • Fatalities in the twelve month period commencing May 1987 decreased by 18% in comparison with the previous twelve month period, and by 13% in comparison with the average of the previous two twelve month periods (commencing May 1985 and May 1986). There are indications that this trend is continuing. • It is concluded that the increase in RBT, plus publicity, was successful in achieving its aims of reductions in drink driving and accidents.

Nicastrin (NCSTN) is a component of the ?-secretase complex and therefore potentially a candidate risk gene for Alzheimer's disease. Here, we have developed a novel functional genomics methodology to express common locus haplotypes to assess functional differences. DNA recombination was used to engineer 5 bacterial artificial chromosomes (BACs) to each express a different haplotype of the NCSTN locus. Each NCSTN-BAC was delivered to knockout nicastrin (Ncstn(-/-)) cells and clonal NCSTN-BAC(+)/Ncstn(-/-) cell lines were created for functional analyses. We showed that all NCSTN-BAC haplotypes expressed nicastrin protein and rescued ?-secretase activity and amyloid beta (Aß) production in NCSTN-BAC(+)/Ncstn(-/-) lines. We then showed that genetic variation at the NCSTN locus affected alternative splicing in human postmortem brain tissue. However, there was no robust functional difference between clonal cell lines rescued by each of the 5 different haplotypes. Finally, there was no statistically significant association of NCSTN with disease risk in the 4 cohorts. We therefore conclude that it is unlikely that common variation at the NCSTN locus is a risk factor for Alzheimer's disease.

Il y a des problemes qui semblent impossible a resoudre sans l'utilisation d'un tiers parti honnete. Comment est-ce que deux millionnaires peuvent savoir qui est le plus riche sans dire a l'autre la valeur de ses biens ? Que peut-on faire pour prevenir les collisions de satellites quand les trajectoires sont secretes ? Comment est-ce que les chercheurs peuvent apprendre les liens entre des medicaments et des maladies sans compromettre les droits prives du patient ? Comment est-ce qu'une organisation peut ecmpecher le gouvernement d'abuser de l'information dont il dispose en sachant que l'organisation doit n'avoir aucun acces a cette information ? Le Calcul multiparti, une branche de la cryptographie, etudie comment creer des protocoles pour realiser de telles taches sans l'utilisation d'un tiers parti honnete. Les protocoles doivent etre prives, corrects, efficaces et robustes. Un protocole est prive si un adversaire n'apprend rien de plus que ce que lui donnerait un tiers parti honnete. Un protocole est correct si un joueur honnete recoit ce que lui donnerait un tiers parti honnete. Un protocole devrait bien sur etre efficace. Etre robuste correspond au fait qu'un protocole marche meme si un petit ensemble des joueurs triche. On demontre que sous l'hypothese d'un canal de diusion simultane on peut echanger la robustesse pour la validite et le fait d'etre prive contre certains ensembles d'adversaires. Le calcul multiparti a quatre outils de base : le transfert inconscient, la mise en gage, le partage de secret et le brouillage de circuit. Les protocoles du calcul multiparti peuvent etre construits avec uniquements ces outils. On peut aussi construire les protocoles a partir d'hypoth eses calculatoires. Les protocoles construits a partir de ces outils sont souples et peuvent resister aux changements technologiques et a des ameliorations algorithmiques. Nous nous demandons si l'efficacite necessite des hypotheses de calcul. Nous demontrons que ce n'est pas le cas en construisant des protocoles efficaces a partir de ces outils de base. Cette these est constitue de quatre articles rediges en collaboration avec d'autres chercheurs. Ceci constitue la partie mature de ma recherche et sont mes contributions principales au cours de cette periode de temps. Dans le premier ouvrage presente dans cette these, nous etudions la capacite de mise en gage des canaux bruites. Nous demontrons tout d'abord une limite inferieure stricte qui implique que contrairement au transfert inconscient, il n'existe aucun protocole de taux constant pour les mises en gage de bit. Nous demontrons ensuite que, en limitant la facon dont les engagements peuvent etre ouverts, nous pouvons faire mieux et meme un taux constant dans certains cas. Ceci est fait en exploitant la notion de cover-free families . Dans le second article, nous demontrons que pour certains problemes, il existe un echange entre robustesse, la validite et le prive. Il s'effectue en utilisant le partage de secret veriable, une preuve a divulgation nulle, le concept de fantomes et une technique que nous appelons les balles et les bacs. Dans notre troisieme contribution, nous demontrons qu'un grand nombre de protocoles dans la litterature basee sur des hypotheses de calcul peuvent etre instancies a partir d'une primitive appelee Transfert Inconscient Veriable, via le concept de Transfert Inconscient Generalise. Le protocole utilise le partage de secret comme outils de base. Dans la derniere publication, nous counstruisons un protocole efficace avec un nombre constant de rondes pour le calcul a deux parties. L'efficacite du protocole derive du fait qu'on remplace le coeur d'un protocole standard par une primitive qui fonctionne plus ou moins bien mais qui est tres peu couteux. On protege le protocole contre les defauts en utilisant le concept de privacy amplication .

Bachman’s Sparrow (Peucaea aestivalis), an endemic North American passerine, requires frequent (≤ 3 yr) prescribed fires to maintain preferred habitat conditions. Prescribed fires that coincide with the sparrow’s nesting season are increasingly used to manage sparrow habitat, but concerns exist regarding the effects that nesting-season fires may pose to this understory-dwelling species. Previous studies suggested that threats posed by fires might be lessened by reducing the extent of prescribed fires, thereby providing unburned areas close to the areas where fires eliminate ground-cover vegetation. To assess this hypothesis, we monitored color-marked male Bachman’s Sparrows on 2 sites where the extent of nesting-season fires differed 5-fold (> 70 ha vs. < 15 ha). Monthly survival for males did not differ between the large- and small-extent treatments, and survival rates exceeded 90% for all months except one during the second year of our study when fires were applied later in the season. Male densities also did not differ between treatments, but treatment-by-year interactions pointed to effects relating to the specific time that fires were applied. The distances separating observations of marked males before and after burns were smaller on small-extent treatments in the first year of study but larger on the small-extent treatments in the second year of study. Burn extents also had no consistent effect on postburn reproductive status. The largest extent we examined could have been too small to affect sparrow populations, but responses may also reflect sustainable metapopulation dynamics in a setting where a large sparrow population is maintained at a regional scale (> 100,000 ha) using frequent prescribed fire (≤ 2-yr return intervals). Additional research is needed regarding the effects that nesting-season fires may have on small, isolated populations as well as sites where much larger burn extents (> 100 ha) or longer burn intervals (> 2 yr) are used.

Introgression in Festulolium is a potentially powerful tool to isolate genes for a large number of traits which differ between Festuca pratensis Huds. and Lolium perenne L. Not only are hybrids between the two species fertile, but the two genomes can be distinguished by genomic in situ hybridisation and a high frequency of recombination occurs between homoeologous chromosomes and chromosome segments. By a programme of introgression and a series of backcrosses, L. perenne lines have been produced which contain small F. pratensis substitutions. This material is a rich source of polymorphic markers targeted towards any trait carried on the F. pratensis substitution not observed in the L. perenne background. We describe here the construction of an F. pratensis BAC library, which establishes the basis of a map-based cloning strategy in L. perenne. The library contains 49,152 clones, with an average insert size of 112 kbp, providing coverage of 2.5 haploid genome equivalents. We have screened the library for eight amplified fragment length polymorphism (AFLP) derived markers known to be linked to an F. pratensis gene introgressed into L. perenne and conferring a staygreen phenotype as a consequence of a mutation in primary chlorophyll catabolism. While for four of the markers it was possible to identify bacterial artificial chromosome (BAC) clones, the other four AFLPs were too repetitive to enable reliable identification of locus-specific BACs. Moreover, when the four BACs were partially sequenced, no obvious coding regions could be identified. This contrasted to BACs identified using cDNA sequences, when multiple genes were identified on the same BAC.

Repetitive DNAs have been extensively applied as physical chromosome markers on comparative studies, identification of chromosome rearrangements and sex chromosomes, chromosome evolution analysis, and applied genetics. Here we report the characterization of repetitive DNA sequences from the Nile tilapia (Oreochromis niloticus) genome by construction and screening of plasmid library enriched with repetitive DNAs, analysis of a BAC-based physical map, and hybridization to chromosomes. The physical mapping of BACs enriched with repetitive sequences and C(o)t-1 DNA (DNA enriched for highly and moderately repetitive DNA sequences) to chromosomes using FISH showed a predominant distribution of repetitive elements in the centromeric and telomeric regions and along the entire length of the largest chromosome pair (X and Y sex chromosomes) of the species. The distribution of repetitive DNAs differed significantly between the p arm of X and Y chromosomes. These findings suggest that repetitive DNAs have had an important role in the differentiation of sex chromosomes. (c) 2007 Elsevier Ltd. All rights reserved.
