Aflatoxin B1 (AFB1), ochratoxin A (OTA) and fumonisin B1 (FB1) are contaminants which have been shown to regularly co-occur in a range of foods. However, only a small number of studies have evaluated the interactive effect of binary and tertiary mycotoxins. The present study evaluated the effects of low levels of each mycotoxin in combination at their EU regulatory limits. Toxic effect with respect to cell viability was measured by MTT and neutral red assays, assessing mitochondria and lysosome integrities respectively. Individual toxicity showed that OTA (10 μg/ml) was the most cytotoxic mycotoxin in all three cell lines studied (caco-2, MDBK and raw 264.7). Binary combinations were cytotoxic to the MDBK cell line in the order [OTA/FB1] > [AFB1/FB1] > [AFB1/OTA], whilst all effects observed were classified as being additive. Tertiary combinations of AFB1, FB1 and OTA at the EU regulatory limits were tested and not found to exhibit measurable cytotoxicity in MDBK, caco-2 or raw 264.7 cells. However by increasing these concentrations above the legal limits to OTA (3 μg/ml), FB1 (8 μg/ml) and AFB1 (1.28 μg/ml), cytotoxicity was observed with up to 26% reduction in cell viability and synergistic effects were evident with regard to mitochondrial integrity. © 2014 Elsevier Ltd. Fungi isolated from marine organisms have been shown to produce several interesting secondary metabolites with important biological activities. Such chemical diversity may be associated to environmental stress conditions and may represent an important source of NCE for bioprospection. Quinolactins belong to a rare fungi-alkaloid class with a unique N-methyl-quinolone moiety fused to a lactam ring and present several bioactivities1. Fungi strain Dm1 was isolated from red alga Dichotomaria marginata, collected from Brazil SE coast, and was grown in sterile rice solid media at 26oC 2, which was then extracted with MeOH. The MeCN fr. from the MeOH extract was chromatographed over Sephadex LH-20 and fr. 4 afforded quinolactin (QL) alkaloids B1, B2 and A, whereas fr. 5 afforded quinolactin D1 after purification by HPLC-DAD. Structural determination of pure compounds was based on HRMS, UV, and NMR spectral analyses, in addition to comparison with literature data and Antimarin® databank. UV data indicated the presence of similar chromophores with λmax at ca. 247 and 320nm. HRMS and tandem MS analyses using both negative and positive ion modes for the isolated compounds indicated their molecular formula and structural features, as for QL B1: C15H16O2N2 [M+H 257], which showed one fragment at m/z 214 [-CHNO]; QL B2: C15H16O3N2 [M+H 273], with product ions at m/z 230 [-CHNO.] and m/z 186 [-C4H9NO.]; for QL A: C16H18N2O2 [M+H 271], which presented one ion at m/z 214, due to loss of fragment (-C4H9) from the molecular ion; and for QL D1: C16H18N2O3 [M+H 287], with product ions at m/z 186 [-CHNO] and m/z 230 [-C4H9]. Such data suggested fragmentation proposals, e.g. for Quinolactin B1 (Fig. 1), which confirmed the structures of the isolated quinolactins, and may represent an important contribution for the sustainable exploration of marine biodiversity.

Phytochromes are red/far-red light photoreceptors that mediate a variety of photomorphogenic processes in plants, from germination to flowering. In addition, there is evidence that phytochromes are also part of the stress signalling response, especially in response to water deficit stress, which is the major abiotic factor limiting plant growth and crop productivity worldwide. In this study, we used the phyA (far red-insensitive; fri), phyB1 (temporary red-insensitive; tri) and phyB2 mutants of tomato (Solanum lycopersicum L.) to study the roles of these three phytochromes in drought stress responses. Compared to wild type (WT) plants grown under water-deficit stress conditions, the fri, tri, and phyB2 mutants did not exhibit altered dry weights, leaf areas, stomatal densities, or stomatal opening. The stomatal conductance of all three mutants was severely reduced under both fully-hydrated and water-deficit conditions. Although relative water contents did change after drought stress in each mutant, the most significant reduction in water potential during water stress was observed in the fri mutant. However, this mutant returned its water status to WT levels during rehydration. Although the phyB2 mutant lost more water from detached leaves during abscisic acid (ABA) treatment, phyB2 behaved like WT plants, indicating that this mutant was not insensitive to ABA. Overall, these results indicate that the phytochromes phyA, phyB1, and phyB2 modulate drought stress responses in tomato.

Barite crusts were formed by an intermittent hydrothermal vent with output temperature from 85 to 465°C. Principal sources of supply of sulfate sulfur are sea water, evaporites, and tholeiitic basalts of the Red Sea rift. Sulfides and sulfates were formed in conditions of isotope disequilibrium with respect to sulfur because rate of precipitation of sulfur compounds from hydrothermal solution was high compared with rate of isotope exchange.

Barite ores with admixture of sphalerite that occur in the Atlantis-II Deep are indicated by their geological position, structure, and mineral and chemical characteristics and by isotope composition of sulfur and oxygen in them to be of hydrothermal origin. They were produced close to an orifice of an intermittent hydrothermal vent by chemogenic precipitation of barite onto cooled basalt.

Barley (Hordeum vulgare L.) is a long-day plant whose flowering is enhanced when the photoperiod is supplemented with far-red light, and this promotion is mediated by phytochrome. A chemically mutagenized dwarf cultivar of barley was selected for early flowering time (barley maturity daylength response [BMDR]-1) and was made isogenic with the cultivar Shabet (BMDR-8) by backcrossing. BMDR-1 was found to contain higher levels of both phytochrome A and phytochrome B in the dark on immunoblots with monoclonal antibodies from oat (Avena sativa L.) that are specific to different members of the phytochrome gene family. Phytochrome A was light labile in both BMDR-1 and BMDR-8, decreasing to very low levels after 4 d of growth in the light. Phytochrome B was light stable in BMDR-8, being equal in both light and darkness. However, phytochrome B became light labile in BMDR-1 and this destabilization of phytochrome B appeared to make BMDR-1 insensitive to photoperiod. In addition, both the mutant and the wild type lacked any significant promotion of flowering in response to a pulse of far-red light given at the end of day, and the end-of-day, far-red inhibition of tillering is normal in both, suggesting that phytochrome B is not involved with these responses in barley.

As aflatoxinas são metabólitos secundários produzidos por fungos toxigênicos das espécies Aspergillus flavus, A. parasiticus e A. nomius. São amplamente encontradas em matérias-primas de rações animais, em especial o milho, e têm a capacidade de levar a quadros clínicos agudos ou crônicos de aflatoxicose, caracterizados por, desde a morte por hepatite aguda até a diminuição do desempenho zootécnico por diminuição de peso ou consumo de ração. A aflatoxina B1 tem sido considerada o metabólito mais perigoso, uma vez que possui alto poder hepatotóxico, além de ser mutagênica e carcinogênica. Atualmente a ciência trabalha rumo à descoberta de substâncias que sejam indicadoras confiáveis de contaminação por componentes tóxicos em homens e em animais, os chamados biomarcadores, que medem uma mudança celular, biológica ou molecular em um meio biológico (tecidos humanos, células ou fluídos) que fornecem informação a respeito de uma doença ou exposição a uma determinada substância. Sua detecção pode auxiliar na identificação, no diagnóstico e no tratamento de indivíduos afetados que podem estar sob risco, mas ainda não exibem os sintomas. Sendo assim, com o auxílio de análises que confirmem a patogenicidade da aflatoxina B1 (determinação da atividade de enzimas hepáticas, da avaliação da função renal, de hematologia, da dosagem de minerais séricos e da avaliação de desempenho zootécnico), o objetivo deste trabalho foi avaliar a aplicabilidade da determinação de resíduos hepáticos de aflatoxinas e do aduto sérico AFB1-lisina na avaliação da eficiência de adsorventes em frangos de corte. Utilizou-se 240 pintos de 1 dia, machos, de linhagem Cobb 500®, distribuídos aleatoriamente em 4 dietas experimentais: Controle Negativo: Ração Basal (RB); RB + 0,5% de adsorvente ((aluminosilicato de cálcio e sódio hidratado/HSCAS); RB + 0,5% de adsorvente + 500 µg de AFB1/kg de ração e; RB + 500 µg de AFB1/kg de ração.Os resultados experimentais mostram que o efeito deletério da AFB1, na concentração utilizada, é mais pronunciado que os efeitos protetores do HSCAS sobre os parâmetros de saúde dos animais. Não houve ação efetiva do adsorvente utilizado sobre quase nenhuma variável estudada, apenas para a redução das lesões histopatológicas em fígado, na redução da concentração de gama-glutamiltransferase (GGT), fósforo e aumento da contagem de hemáceas aos 21 dias de idade. Porém, influenciou positivamente a redução de resíduos hepáticos de aflatoxina G1 aos 21 dias e as concentrações de AFB1-lisina sérica aos 21 e aos 42 dias de idade. Estes dados são importantes porque permite concluir que, embora sintomatologicamente o HSCAS não tenha exercido função efetiva, molecularmente foi capaz de mostrar de eficácia sobre os alguns biomarcadores de aflatoxinas no organismo das aves

Context. Several clusters of red supergiants have been discovered in a small region of the Milky Way close to the base of the Scutum-Crux Arm and the tip of the Long Bar. Population synthesis models indicate that they must be very massive to harbour so many supergiants. Amongst these clusters, Stephenson 2, with a core grouping of 26 red supergiants, is a strong candidate to be the most massive young cluster in the Galaxy. Aims. Stephenson 2 is located close to a region where a strong over-density of red supergiants had been found. We explore the actual cluster size and its possible connection to this over-density. Methods. Taking advantage of Virtual Observatory tools, we have performed a cross-match between the DENIS, USNO-B1 and 2MASS catalogues to identify candidate obscured luminous red stars around Stephenson 2, and in a control nearby region. More than 600 infrared bright stars fulfill our colour criteria, with the vast majority having a counterpart in the I band and >400 being sufficiently bright in I to allow observation with a 4-m class telescope. We observed a subsample of ~250 stars, using the multi-object, wide-field, fibre spectrograph AF2 on the WHT telescope in La Palma, obtaining intermediate-resolution spectroscopy in the 7500–9000 Å range. We derived spectral types and luminosity classes for all these objects and measured their radial velocities. Results. Our targets turned out to be G and K supergiants, late (≥ M4) M giants, and M-type bright giants (luminosity class II) and supergiants. We found ~35 red supergiants with radial velocities similar to Stephenson 2 members, spread over the two areas surveyed. In addition, we found ~40 red supergiants with radial velocities incompatible in principle with a physical association. Conclusions. Our results show that Stephenson 2 is not an isolated cluster, but part of a huge structure likely containing hundreds of red supergiants, with radial velocities compatible with the terminal velocity at this Galactic longitude (and a distance ~6 kpc). In addition, we found evidence of several populations of massive stars at different distances along this line of sight.

Context. The open cluster NGC 7419 is known to contain five red supergiants and a very high number of Be stars. However, there are conflicting reports about its age and distance that prevent a useful comparison with other clusters. Aims. We intend to obtain more accurate parameters for NGC 7419, using techniques different from those of previous authors, so that it may be used as a calibrator for more obscured clusters. Methods. We obtained Strömgren photometry of the open cluster NGC 7419, as well as classification spectroscopy of ~20 stars in the area. We then applied standard analysis and classification techniques. Results. We find a distance of 4 ± 0.4 kpc and an age of 14 ± 2 Myr for NGC 7419. The main-sequence turn-off is found at spectral type B1, in excellent agreement. We identify 179 B-type members, implying that there are more than 1200 M⊙ in B stars at present. Extrapolating this to lower masses indicates an initial cluster mass of between 7000 and 10 000 M⊙, depending on the shape of the initial mass function. We find a very high fraction (≈40%) of Be stars around the turn-off, but very few Be stars at lower masses. We also report for the first time a strong variability in the emission characteristics of Be stars. We verified that the parameters of the red supergiant members can be used to obtain accurate cluster parameters. Conclusions. NGC 7419 is sufficiently massive to serve as a testbed for theoretical predictions and as a template to compare more obscured clusters. The distribution of stars above the main-sequence turn-off is difficult to accommodate with current evolutionary tracks. Though the presence of five red supergiants is marginally consistent with theoretical expectations, the high number of Be stars and very low number of luminous evolved B stars hint at some unknown physical factor that is not considered in current synthesis models.

This paper presents a new method of eye localisation and face segmentation for use in a face recognition system. By using two near infrared light sources, we have shown that the face can be coarsely segmented, and the eyes can be accurately located, increasing the accuracy of the face localisation and improving the overall speed of the system. The system is able to locate both eyes within 25% of the eye-to-eye distance in over 96% of test cases.
