Diffusion tensor imaging (DTI) studies have identified changes in white matter tracts in schizophrenia patients and those at high risk of transition. Schizotypal samples represent a group on the schizophrenia continuum that share some aetiological risk factors but without the confounds of illness. The aim of the current study was to compare tract microstructural coherence as measured by
fractional anisotropy (FA) between 12 psychometrically defined schizotypes and controls. We investigated bilaterally the uncinate and arcuate fasciculi (UF and AF) via a probabilistic tractography algorithm (PICo), with FA values compared between groups. Partial correlations were also examined between measures of subclinical hallucinatory/delusional experiences and FA values. High schizotypes
were found to have significantly higher FA values in bilateral UF only, but failed to reach significance in each hemisphere. In the whole sample there was a positive correlation between increasing FA values and measures of hallucinatory experience in the right AF. These findings suggest subtle changes in microstructural coherence are present in schizotypes. Correlations between mild hallucinatory experience and increasing FA values could indicate increasing coherence could be associated with symptom formation.

The arcuate fasciculus (AF), a white matter tract linking temporal and inferior frontal language cortices, can be disrupted in stroke patients suffering from aphasia. Using diffusion tensor imaging (DTI) tractography it is possible to track AF connections to neural regions associated with either phonological or semantic linguistic processing. The aim of the current study is to investigate the relationship between integrity of white matter microstructure and specific linguistic deficits.

Il cervello umano è composto da una rete complessa, formata da fasci di assoni, che connettono le diverse aree cerebrali. Il fascio arcuato collega l’area imputata alla com- prensione del linguaggio con quella dedicata alla sua produzione. Il fascio arcuato è presente in entrambi gli emisferi cerebrali, anche se spesso è utilizzato prevalente- mente il sinistro. In questa tesi sono state valutate, in un campione di soggetti sani, le differenze tra fascio arcuato destro e sinistro, utilizzando la trattografia, metodica avanzata e non invasiva che permette la ricostruzione della traiettoria delle fibre con immagini RM (Risonanza Magnetica) pesate in diffusione. A questo scopo ho utilizzato un algoritmo probabilistico, che permette la stima di probabilità di connessione della fibra in oggetto con le diverse aree cerebrali, anche nelle sedi di incrocio con fibre di fasci diversi. Grazie all’implementazione di questo metodo, è stato possibile ottenere una ricostruzione accurata del fascio arcuato, an- che nell’emisfero destro dove è spesso critica, tanto da non essere possibile con altri algoritmi trattografici. Parametrizzando poi la geometria del tratto ho diviso il fascio arcuato in venti seg- menti e ho confrontato i parametri delle misure di diffusione, valutate nell’emisfero destro e sinistro. Da queste analisi emerge un’ampia variabilità nella geometria dell’arcuato, sia tra diversi soggetti che diversi emisferi. Nell’emisfero destro l’arcuato incrocia maggiormente fibre appartenenti ad altri fasci. Nell’emisfero sinistro le fibre dell’arcuato sono più compatte e si misura anche una maggiore connettività con altre aree del cervello coinvolte nelle funzioni linguistiche. Nella seconda fase dello studio ho applicato la stessa metodica in due pazienti con lesioni cerebrali, con l’obiettivo di testare il danno del fascio arcuato ipsilaterale alla lesione e stimare se nell’emisfero controlaterale si innescassero meccanismi di plastic- ità strutturale. Questa metodica può essere implementata, in un gruppo di pazienti omogenei, per identificare marcatori RM diagnostici nella fase di pianificazione pre- chirurgica e marcatori RM prognostici di recupero funzionale del linguaggio.

In the Iberian Variscides several first order arcuate structures have been considered. In spite of being highly studied their characterization, formation mechanisms and even existence is still debatable. Themain Ibero-Armorican Arc (IAA) is essentially defined by a predominantNW–SE trend in the Iberian branch and an E–Wtrend in the Brittany one. However, in northern Spain it presents a 180° rotation, sometimes known as the Cantabrian Arc (CA). The relation between both arcs is controversial, being considered either as a single arc due to one tectonic event, or as the result of a polyphasic process. According to the last assumption, there is a later arcuate structure (CA), overlapping a previousmajor one (IAA). Whatever themodels, they must be able to explain the presence of a Variscan sinistral transpression in Iberia and a dextral one in Armorica, and a deformation spanning from the Devonian to the Upper Carboniferous. Another arcuate structure, in continuity with the CA, the Central-Iberian Arc (CIA) was recently proposed mainly based upon on magnetic anomalies, geometry of major folds and Ordovician paleocurrents. The critical review of the structural, stratigraphic and geophysical data supports both the IAA and the CA, but as independent structures. However, the presence of a CIA is highly questionable and could not be supported. The complex strain pattern of the IAA and the CA could be explained by a Devonian — Carboniferous polyphasic indentation of a Gondwana promontory. In thismodel the CA is essentially a thin-skinned arc,while the IAA has a more complex and longer evolution that has led to a thick-skinned first order structure. Nevertheless, both arcs are essentially the result of a lithospheric bending process during the Iberian Variscides.

Background. The majority of studies investigating the neural mechanisms underlying treatment-induced recovery in aphasia have focused on the cortical regions associated with language processing. However, the integrity of the white matter connecting these regions may also be crucial to understanding treatment mechanisms. Objective. This study investigated the integrity of the arcuate fasciculus (AF) and uncinate fasciculus (UF) before and after treatment for anomia in people with aphasia. Method. Eight people with aphasia received 12 treatment sessions to improve naming; alternating between phonologically-based and semantic-based tasks, with high angular resolution diffusion imaging conducted pre and post treatment. The mean generalized fractional anisotropy (GFA), a measure of fiber integrity, and number of fibers in the AF and UF were compared pre and post treatment, as well as with a group of 14 healthy older controls. Results. Pre treatment, participants with aphasia had significantly fewer fibers and lower mean GFA in the left AF compared with controls. Post treatment, mean GFA increased in the left AF to be statistically equivalent to controls. Additionally, mean GFA in the left AF pre and post treatment positively correlated with maintenance of the phonologically based treatment. No differences were found in the right AF, or the UF in either hemisphere, between participants with aphasia and controls, and no changes were observed in these tracts following treatment. Conclusions. Anomia treatments may improve the integrity of the white matter connecting cortical language regions. These preliminary results add to the understanding of the mechanisms underlying treatment outcomes in people with aphasia post stroke.

Purpose To highlight the finding of occult areas of poor epithelial adhesion in the superior perilimbal cornea in a minority of patients with recalcitrant recurrent corneal erosion syndrome presenting with corneal erosion elsewhere on the corneal surface. Patient population 31 eyes of 31 consecutive patients with corneal erosion undergoing mechanical debridement of the epithelium prior to diamond burr keratectomy for recurrent corneal erosion. Methods Determine the location and incidence of poor epithelial adhesion distant from the initial erosion by use of mechanical debridement with a dry microsponge. Results During debridement, 8 of 31 eyes (25.8%) displayed a large arcuate area of occult dysfunction of adhesion in the superior perilimbal area. None of these patients showed recurrence over a mean of 18 month after diamond burr keratectomy (95% confidence interval 0-36.9%). Conclusion Mechanical debridement with a microsponge identified a significant minority of patients with poor adhesion in the superior perilimbal cornea away from the area of obvious erosion and increased the target area for diamond burr keratectomy. This two pronged approach allowed successful management of this group.

A new evaluation of the elastic thickness (Te) structure of the Indian Shield, derived from isotropic fan wavelet methodology, documents spatial variations of lithospheric deformation in different tectonic provinces correlated with episodic tectono-thermal events. The Te variations corroborated by shear velocity, crustal thickness, and seismogenic thickness reveal the heterogeneous rheology of the Indian lithosphere. The thinned, attenuated lithosphere beneath Peninsular India is considered to be the reason for its mechanically weak strength (<30 km), where a decoupled crust-mantle rheology under different surface/subsurface loading structures may explain the prominent low Te patterns. The arcuate Te structure of the Western Dharwar province and a NNE-trending band of low Te anomaly in the Southern Granulite Terrane are intriguing patterns. The average Te values (40-50 km) of the Central Indian Tectonic Zone, the Bastar Craton, and the northern Eastern Ghats Mobile Belt are suggestive of old, stable, Indian lithosphere, which was not affected by any major tectono-thermal events after cratonic stabilization. We propose that the anomalously high Te (60-85 km) and high S-wave velocity zone to the north of the Narmada-Son Lineament, mainly in NW Himalaya, and the northern Aravalli and Bundelkhand Cratons, suggest that Archean lithosphere characterized by a high velocity mantle keel supports the orogenic topographic loads in/near the Himalaya. The Te map clearly segments the volcanic provinces of the Indian Shield, where the signatures of the Reunion, Marion, and Kerguelen hotspots are indicated by significantly low Te patterns that correlate with plume- and rift-related thermal and mechanical rejuvenation, magmatic underplating, and crustal necking. The correlations between Te variations and the occurrence of seismicity over seismically active zones reveal different causal relationships, which led to the current seismogenic zonation of the Indian Shield. Fluvial systems form landscapes and sedimentary deposits with a rich hierarchy of structures that extend from grain- to valley scale. Large-scale pattern formation in fluvial systems is commonly attributed to forcing by external factors, including climate change, tectonic uplift, and sea-level change. Yet over geologic timescales, rivers may also develop large-scale erosional and depositional patterns that do not bear on environmental history. This dissertation uses a combination of numerical modeling and topographic analysis to identify and quantify patterns in river valleys that form as a consequence of river meandering alone, under constant external forcing. Chapter 2 identifies a numerical artifact in existing, grid-based models that represent the co-evolution of river channel migration and bank strength over geologic timescales. A new, vector-based technique for bank-material tracking is shown to improve predictions for the evolution of meander belts, floodplains, sedimentary deposits formed by aggrading channels, and bedrock river valleys, particularly when spatial contrasts in bank strength are strong. Chapters 3 and 4 apply this numerical technique to establishing valley topography formed by a vertically incising, meandering river subject to constant external forcing—which should serve as the null hypothesis for valley evolution. In Chapter 3, this scenario is shown to explain a variety of common bedrock river valley types and smaller-scale features within them—including entrenched channels, long-wavelength, arcuate scars in valley walls, and bedrock-cored river terraces. Chapter 4 describes the age and geometric statistics of river terraces formed by meandering with constant external forcing, and compares them to terraces in natural river valleys. The frequency of intrinsic terrace formation by meandering is shown to reflect a characteristic relief-generation timescale, and terrace length is identified as a key criterion for distinguishing these terraces from terraces formed by externally forced pulses of vertical incision. In a separate study, Chapter 5 utilizes image and topographic data from the Mars Reconnaissance Orbiter to quantitatively identify spatial structures in the polar layered deposits of Mars, and identifies sequences of beds, consistently 1-2 meters thick, that have accumulated hundreds of kilometers apart in the north polar layered deposits.

Crianças de mães fumantes são mais suscetíveis a se tornarem adultos obesos e se viciarem em drogas ou alimentos palatáveis. Drogas e alimentos ativam a via mesolímbica de recompensa, causando sensação de prazer que induz ainda mais o consumo. Assim, avaliamos a relação entre a exposição apenas à nicotina ou à fumaça do cigarro durante a lactação com a preferência alimentar e sistema dopaminérgico de recompensa cerebral das proles, em dois modelos de programação: Modelo I: no 2o dia pós-natal (PN), lactantes receberam implante de minibombas osmóticas que liberam nicotina (NIC) ou salina (C), durante 14 dias. Em PN150 e novamente em PN160, as proles foram divididas em 4 grupos para um desafio alimentar: N-SC e C-SC que receberam ração padrão; N-SSD e C-SSD que podiam escolher livremente entre as dietas hiperlipídica e hiperglicídica. A ingestão alimentar foi avaliada após 12 h. As mães foram sacrificadas apenas na 21 da lactação (desmame) e as proles em PN15 (com nicotina), PN21 e PN170 (ausência da NIC). Ao desmame, as ratas lactantes NIC apresentaram menor conteúdo de tirosina hidroxilase (TH), maior OBRb e SOCS3 na area tegmentar ventral (VTA); menor TH, maior receptor de dopamina 1 (D1R), receptor de dopamina 2 (D2R) e transportador de dopamina (DAT) no núcleo accumbens (NAc); maior conteúdo de TH no estriado dorsal (DS); e maior D2R e SOCS3 no núcleo arqueado (ARC). Em PN15, os filhotes NIC apresentaram maior conteúdo de D1R, D2R e menor DAT no NAc, enquanto em PN21, apresentaram apenas menor DAT no DS, e menor conteúdo de pSTAT3 em ARC. Aos 170 dias, as proles SSD demonstraram maior preferência para a ração hiperlipídica. No entanto, os animais N-SSD consumiram mais ração hiperglicidica do que as proles C-SSD. A prole N apresentou menor conteúdo de D2R e DAT no NAc e menor D2R no ARC. Modelo II: as mães e suas proles foram divididas em: expostos à fumaça do cigarro (grupo S: 4 vezes / dia, do 3 ao 21 dia de lactação), e expostos ao ar filtrado (grupo C). Em PN175, as proles foram divididas em 4 grupos para o desafio alimentar S-SC, C-SC, S-SSD e C-SSD. A ingestão alimentar foi avaliada após 30 min e 12 h. Em PN180, as proles foram sacrificadas. O grupo S-SSD ingeriu mais das rações palatáveis do que o grupo C-SSD em 30 min e 12 h. Ambos os grupos preferiram a ração hiperlipídica. No entanto, os animais S-SSD consumiram mais ração hiperlipídica do que C-SSD em 30 min. A prole S apresentou menor conteúdo de TH no VTA, menor conteúdo de TH, D2R e maior conteúdo de D1R no NAc e menor OBRb no ARC. Demonstramos que tanto a nicotina isolada como a exposição à fumaça do cigarro durante a lactação resultaram em mudanças no sistema dopaminérgico das proles, programando o comportamento alimentar devido à diminuição da dopamina no NAc.

This study was undertaken to investigate the effect of emotional stress on humoral immunoactivity and to examine whether the sympathetic nervous system was involved in the immunomodulation. In the present study, two types of emotional stressors were used. One was footshock apparatus used to cause the rats which were given footshock before, emotional stressed; the other was an empty water bottle used to cause the rats which were trained to drink water at two set times each day, emotional stressed. The effect of emotional stress on the primary immune function (anti-ovallum antibody level and spleen index), the endocrine response (corticosterone level, epinephrine and norepinephrine level), the behavioral changes (freezing, defecation, grooming and attacking behavior) were investigated. The main results were: 1. Two types of emotional stress significantly increased the level of plasma corticosterone, norepinephrine and epinephrine, as well as freezing, defecation and attacking behavior. 2. Two types of emotional stress significantly decreased the level of anti-ovallum antibody. A negative correlation between catecholamine level (epinephrine and norepinephrine) and antibody level or spleen index was found. 3. β-adrenergic receptor antagonist propranolol could reverse the immunomodulation induced by emotional stress. 4. After two types of emotional stress, c-fos expression was observed in the following brain areas or nucleus; arcuate nucleus, anterior commissure nucleus, diffuse part of dorsalmedial nucleus hypothalamus, lateral dorsal nucleus thalamus, medial nucleus amygdala, solitary nucleus, frontal cortex and cingulum. These brain areas and nucleus are involved in the central modulation of the autonomic nervous system. Taken together, these findings demonstrate that emotional stress can suppress humoral immunity and the activation of the sympathetic nervous system is involved in the humoral immunomodulation induced by emotional stress.
