Se ha realizado un estudio sobre la microbiota fúngica asociada a la fumagina del madroño, determinándose las especies de pulgones recogidas sobre madroños muestreados en la Comunidad de Madrid (España). Las especies de pulgones del madroño encontradas en los muestreos han sido dos: Aphis arbuti Ferrari y Wahlgreniella nervata (Gillette). La especie más frecuente ha sido W. nervata cuya presencia se observó en el 80% de las muestras, mientras que A. arbuti estuvo presente en el 35% de ellas. Parecen ser especies bastante especÃficas del madroño, de las que no se habÃan recogido citas en la Comunidad de Madrid. Los análisis microbiológicos realizados sobre las hojas de madroño muestran que no existen diferencias apreciables entre las que visualmente tienen negrilla y pulgones de aquellas que están aparentemente normales. La microbiota fúngica total de las ramillas es muy semejante a la de las hojas. Sobresalen entre los géneros y/o especies Alternaria, Aspergillus, Niger, Aureobasidium, Cladosporium y Fusarium. Este último género representado por dos especies (F. dinerum y F. solari) no ha incrementado considerablemente su presencia cuando se ha utilizado un medio especÃfico para el análisis.

The common Mediterranean ornamental strawberry-tree (Arbutus unedo L.) produces an edible reddish sweet berry that is found to be bland and tasteless unless it is consumed overripe, otherwise it is discarded or used as basic agricultural sub residue. The bioactive properties of this fruit have been reported and related with phenolic compounds, mainly flavan-3-ols, such as catechin and procyanidins, which has opened the opportunity to exploit their extraction from alternative sources.The common Mediterranean ornamental strawberry-tree (Arbutus unedo L.) produces an edible reddish sweet berry that is found to be bland and tasteless unless it is consumed overripe, otherwise it is discarded or used as basic agricultural sub residue. This study compares and optimizes the maceration, microwave and ultrasound extraction techniques in the recovery of a catechin extract from Arbutus unedo L. fruits and evaluate the stability of flavan-3-ols during storage and application processes. To obtain conditions that maximize the catechin extraction yield, a response surface methodology was used. Maceration and microwave extractions were found to be the most effective methods, capable of yielding 1.38±0.1 and 1.70±0.3 mg of catechin/g dry weight (dw) in the corresponding optimal extraction conditions. The optimal conditions for maceration were 93.2±3.7 min, 79.6±5.2 ºC and 23.1±3.7 % of ethanol, while for the microwave extraction were 42.2±4.1 min, 137.1±8.1 ºC and 12.1±1.1 % of ethanol. The microwave system was a quicker solution, conducting to slightly higher yields of catechin than maceration, but this one needed lower temperatures to reach similar yields. The ultrasound method was the least effective solution in terms of catechin yield extraction (0.71±0.1 mg/g at 42.4±3.6 min, 314.9±21.2 W and 40.3±3.8 %. ethanol). The stability was tested with of the catechin-enriched extract (60% flavan-3-ols and 22% catechin), obtained under the best maceration conditions, was tested. Therefore, catechin-enriched extracts were submitted to physical and chemical stability studies, considering the main affecting variables (time, temperature and pH): i) a stability study of the extracts during storage as powder system; and ii) a stability study of the extracts in simulated food environment (aqueous solution system). The measured responses were the flavan-3-ols and catechin contents, determined by HPLC-DAD, and the antioxidant activity of the extracts evaluated by hydrophilic assays. Mechanistic and phenomenological equations were used to describe the responses, and the optimal conditions for flavan-3-ols (including catechin) stability as powder extract during a month were pH= 5.4 and T= -20ºC; while its stability in aqueous solution remained during the 24 h of application at pH<4 and T<30ºC. Gastrointestinal stromal tumors (GIST) are mesenchymal neoplasms frequently caused by a gain of function mutation in KIT or PDGFRα, two tyrosine kinase receptors (TKR). For this reason, they are successfully treated with imatinib, a tyrosine kinase inhibitor (TKI). However, the therapy is typically long-term ineffective due to imatinib resistance, which represents the main issue in the clinic of GISTs. Although numerous efforts have been made in the last two decades to develop novel therapies for imatinib-resistant GISTs, the approvals of multi-target TKIs have only improved the clinical outcomes modestly. Emblematic is the recent failure of ripretinib in the phase III INTRIGUE trial, decisively marking the end of the paradigm only based on the central role of KIT secondary mutations in imatinib resistance, and the consequent seeking of multi-target TKIs as the solution. Consistent with this clinical result, preclinical studies have revealed numerous mechanisms of resistance that are not targetable with multi-target TKIs, indicating that imatinib resistance is more multifaceted than initially hypothesized and explaining the modest efficacy of these latter. In this scenario, the absence of drugs capable of long-term counteracting the rise of imatinib-resistant subclones unavoidably leads to progressive disease and metastasis. In particular, the onset of metastases remarkably impacts the median overall survival and determines the most GIST-related deaths. Therefore, new therapy proposals are needed. Here, we present two project lines investigating novel strategies to counteract imatinib-resistant GISTs.

O medronheiro é um arbusto da região mediterrânica que pode ser encontrada por todo o paÃs. Ao contrário do que verifica na região sul do paÃs, no concelho de Castelo de Paiva é atribuÃda uma reduzida importância económica a esta espécie. Com o intuito de preservar e potenciar a produção desta espécie e contribuir para a dinamização da economia do concelho, procedeu-se à caracterização morfológica e genética de uma amostra da população de medronheiros de Castelo de Paiva. A caracterização morfológica e genética foi realizada para um total de 10 genótipos. Para tal recolheram-se 70 folhas aleatoriamente em cada árvore. Em 40 folhas mediu-se o comprimento, largura, comprimento do pedúnculo, peso fresco, peso seco e determinou-se a área foliar. Dos caracteres morfológicos analisados, aqueles que se revelaram mais úteis na distinção dos vários genótipos foram: comprimento do pedúnculo, peso fresco e peso seco. As restantes 30 folhas foram utilizadas para a caracterização genética. Esta caracterização foi realizada recorrendo a um marcador de DNA, ISSR. Os 5 primeiros exemplaresutilizados na técnica de ISSR demonstraram-se polimórficos. Os resultados da caracterização genética sugerem que a variabilidade genética na população é média a alta.

Els incendis forestals són una pertorbació amb un paper decisiu en l’estructura i dinà mica dels ecosistemes mediterranis. La majoria de les seves espècies vegetals presenten mecanismes de resposta al foc, com la germinació de llavors i la rebrotada d’individus cremats. Les masses forestals regenerades a partir de rebrots assoleixen densitats massa altes i una baixa producció, i, per tant, és fonamental dur a terme una gestió mitjançant tractaments silvÃcoles. El principal objectiu d’aquest projecte és quantificar l’efecte de la selecció de rebrots i la selecció de rebrots més la desbrossada sobre el creixement de l’Arbutus unedo. S’han estudiat 12 parcel—les en regeneració després dels incendis de 1985, 1986 i 1994 al terme municipal d’Esparreguera. Els resultats mostren que els dos tractaments afavoreixen de la mateixa manera el creixement dels peus d’Arbutus unedo, a causa de la disminució de la competència intraespecÃfica i interespecÃfica. La desbrossada (a nivell de parcel—la, no d’individu), no obstant, provoca un increment probablement perjudicial de l’alçada dels rebrots, per la major disponibilitat de llum. Per tal de proposar un model de gestió forestal, s’ha realitzat una anà lisi multicriterial dels diferents escenaris, on s’han considerat altres criteris, com són el model de combustible, la possibilitat de pastura i el cost econòmic. L’alternativa preferida en els boscos d’Arbutus unedo és la selecció de rebrots i la desbrossada.

Photosynthetic organisms have sought out the delicate balance between efficient light harvesting under limited irradiance and regulated energy dissipation under excess irradiance. One of the protective mechanisms is the thermal energy dissipation through the xanthophyll cycle that may transform harmlessly the excitation energy into heat and thereby prevent the formation of damaging active oxygen species (AOS). Violaxanthin deepoxidase (VDE) converts violaxanthin (V) to antheraxanthin (A) and zeaxanthin (Z) defending the photosynthetic apparatus from excess of light. Another important biological pathway is the chloroplast water-water cycle, which is referred to the electrons from water generated in PSII reducing atmospheric O2 to water in PSI. This mechanism is active in the scavenging of AOS, when electron transport is slowed down by the over-reduction of NADPH pool. The control of the VDE gene and the variations of a set of physiological parameters, such as chlorophyll florescence and AOS content, have been investigated in response to excess of light and drought condition using Arabidopsis thaliana and Arbutus unedo.. Pigment analysis showed an unambiguous relationship between xanthophyll de-epoxidation state ((A+Z)/(V+A+Z)) and VDE mRNA amount in not-irrigated plants. Unexpectedly, gene expression is higher during the night when xanthophylls are mostly epoxidated and VDE activity is supposed to be very low than during the day. The importance of the water-water cycle in protecting the chloroplasts from light stress has been examined through Arabidopsis plant with a suppressed expression of the key enzyme of the cycle: the thylakoid-attached copper/zinc superoxide dismutase. The analysis revealed changes in transcript expression during leaf development consistent with a signalling role of AOS in plant defence responses but no difference was found any in photosynthesis efficiency or in AOS concentration after short-term exposure to excess of light. Environmental stresses such as drought may render previously optimal light levels excessive. In these circumstances the intrinsic regulations of photosynthetic electron transport like xanthophyll and water-water cycles might modify metabolism and gene expression in order to deal with increasing AOS.

As lapas do género Patella são moluscos gastrópodes em crescente utilização na alimentação humana, sendo um petisco muito apreciado em várias regiões costeiras de Portugal. De acordo com a fileira do pescado, o consumo de produtos da pesca ricos em ácidos gordos polinsaturados têm vindo a substituir os produtos de origem animal terrestre através dos produtos filetados, reestruturados e pré-cozinhados. A oxidação dos alimentos é a causa mais importante na deterioração da qualidade nutricional e sensorial dos produtos da pesca. Assim nos últimos anos, tem-se verificado um interesse crescente na utilização de antioxidantes naturais de origem vegetal (frutas e vegetais) em detrimento de antioxidantes sintéticos, aquando da elaboração de produtos reestruturados de origem animal (hambúrgueres, salsichas e patés). Para além deste efeito antioxidante, existe uma relação benéfica entre o consumo de frutas e vegetais, ricos em compostos fenólicos e a prevenção de certas doenças. Este trabalho pretende produzir e caracterizar ao nÃvel fÃsico-quÃmico, sensorial e microbiológico um paté elaborado a partir de lapas, com adição do fruto silvestre medronho, bem como a valorização destas matérias-primas ao proporcionar uma mais-valia nutricional/funcional e económica com o desenvolvimento de um produto inovador (paté). Por outro lado, avaliou-se a estabilidade oxidativa do paté em armazenamento refrigerado durante 90 dias bem como a aceitabilidade do mesmo. Sendo assim, produziram-se três tipos de patés inovadores: i) paté de lapa com BHT (PCON), ii) paté de lapa com 3% de medronho (PAU3) e iii) paté de lapa com 6% de medronho (PAU6). Os resultados da avaliação da estabilidade oxidativa (PAU6>PAU3>PCON) confirmam a elevada capacidade do extrato de medronho rico em compostos fenólicos como inibidor eficaz na oxidação lipÃdica do paté de lapa e com boa aceitação sensorial.

Evergreen trees in the Mediterranean region must cope with a wide range of environmental stresses from summer drought to winter cold. The mildness of Mediterranean winters can periodically lead to favourable environmental conditions above the threshold for a positive carbon balance, benefitting evergreen woody species more than deciduous ones. The comparatively lower solar energy input in winter decreases the foliar light saturation point. This leads to a higher susceptibility to photoinhibitory stress especially when chilly (< 12 C) or freezing temperatures (< 0 C) coincide with clear skies and relatively high solar irradiances. Nonetheless, the advantage of evergreen species that are able to photosynthesize all year round where a significant fraction can be attributed to winter months, compensates for the lower carbon uptake during spring and summer in comparison to deciduous species. We investigated the ecophysiological behaviour of three co-occurring mature evergreen tree species (Quercus ilex L., Pinus halepensis Mill., and Arbutus unedo L.). Therefore, we collected twigs from the field during a period of mild winter conditions and after a sudden cold period. After both periods, the state of the photosynthetic machinery was tested in the laboratory by estimating the foliar photosynthetic potential with CO2 response curves in parallel with chlorophyll fluorescence measurements. The studied evergreen tree species benefited strongly from mild winter conditions by exhibiting extraordinarily high photosynthetic potentials. A sudden period of frost, however, negatively affected the photosynthetic apparatus, leading to significant decreases in key physiological parameters such as the maximum carboxylation velocity (Vc,max), the maximum photosynthetic electron transport rate (Jmax), and the optimal fluorometric quantum yield of photosystem II (Fv/Fm). The responses of Vc,max and Jmax were highly species specific, with Q. ilex exhibiting the highest and P. halepensis the lowest reductions. In contrast, the optimal fluorometric quantum yield of photosystem II (Fv/Fm) was significantly lower in A. unedo after the cold period. The leaf position played an important role in Q. ilex showing a stronger winter effect on sunlit leaves in comparison to shaded leaves. Our results generally agreed with the previous classifications of photoinhibition-tolerant (P. halepensis) and photoinhibitionavoiding (Q. ilex) species on the basis of their susceptibility to dynamic photoinhibition, whereas A. unedo was the least tolerant to photoinhibition, which was chronic in this species. Q. ilex and P. halepensis seem to follow contrasting photoprotective strategies. However, they seemed equally successful under the prevailing conditions exhibiting an adaptive advantage over A. unedo. These results show that our understanding of the dynamics of interspecific competition in Mediterranean ecosystems requires consideration of the physiological behaviour during winter which may have important implications for long-term carbon budgets and growth trends.
