The role of catecholamines in the control of the GnRH pulse generator is unclear as studies have relied on the use of peripheral or intracerebroventricular injections, which lack specificity in relation to the anatomical site of action. Direct brain site infusions have been used, however, these are limited by the ability to accurately target small brain regions. One such area of interest in the control of GnRH is the median eminence and arcuate nucleus within the medial basal hypothalamus. Here we describe a method of stereotaxically targeting this area in a large animal (sheep) and an infusion system to deliver drugs into unrestrained conscious animals. To test our technique we infused the dopamine agonist, quinpirole or vehicle into the medial basal hypothalamus of ovariectomised ewes. Quinpirole significantly suppressed LH pulsatility only in animals with injectors located close to the lateral median eminence. This in vivo result supports the hypothesis that dopamine inhibits GnRH secretion by presynaptic inhibition in the lateral median eminence. Also infusion of quinpirole into the medial basal hypothalamus suppressed prolactin secretion providing in vivo evidence that is consistent with the hypothesis that there are stimulatory autoreceptors on tubero-infundibular dopamine neurons. RT-PCR followed by 5'- and 3'- rapid amplification of cDNA ends was used to clone and sequence ovine prolactin-releasing peptide (PrRP). The cDNA was characterised by short 5'- and 3'-untranslated regions and a GC-rich (71%) coding region. The nucleotide and deduced amino acid sequences for the coding region showed 95.6 and 94.9% identity with bovine PrRP but the amino acid sequence of PrRP31 was conserved between these species. Northern blot analysis and RT-PCR showed that, as in the rat, the peptide was more abundantly expressed in the brainstem than the hypothalamus. However, in the ovine hypothalamus, PrRP mRNA expression was more widespread than in the rat, with expression detected in both rostral and caudal parts of the mediobasal hypothalamus. Portugal has a strong tradition of cheesemaking from raw ewe's milk; most of these cheeses are still made on a traditional farmhouse scale. Their production is protected by Protected Designation of Origin (PDO) but the specific biochemical aspects of the majority still need to be characterised. Two different cheesemaking procedures, traditional and semi-industrial, were compared technologically, biochemically and microbiologically. It was observed that, despite the highly significant difference between artisanal and semi-industrial cheeses (P < 0.001), both products were within the limits of national regulations for most parameters except maturation temperature, humidity and the value for the maturation index. Although the present study was not fully representative of the region, the results obtained suggest that the specific regulations for Serpa cheese should be revised and that other parameters, such as moisture and salt-in-moisture content, which are very much dependent on the cheesemaking process, should be included in order to characterise better this traditional cheese.

To study the annual pattern of plasma melatonin and progesterone concentrations in hair [Santa Ines (SI)] and wool [Romney Marsh (RM) and Suffolk (SU)] ewe lambs kept under natural photoperiods at 21 degrees 59'S, 12 ewe lambs (four/breed) were used. For melatonin, blood samples were collected monthly throughout the year at the onset (17:00, 19:00 and 21:00 hr) and end (04:00, 06:00 and 08:00 hr) of the night, and for progesterone the samples were collected in the morning, two to three times a week throughout the year. Plasma melatonin concentrations at different times of the day changed according to the season. In diurnal periods (17:00 and 8:00 hr) no seasonal differences were observed but they became evident in the nocturnal intervals (21:00 and 4:00 hr) and transitional night-day (6:00 hr) times. The patterns of melatonin secretion were higher in winter and autumn than in spring and summer. The patterns of plasma progesterone secretion were affected by interaction between breed and season. There was no seasonal variation in plasma progesterone concentrations for SI females. The progesterone pattern for RM and SU females varied with season. The plasma levels were higher in autumn and winter than in spring and summer. At 21 degrees 59'S hair and wool ewe lambs showed the same annual pattern of plasma melatonin concentration while the annual progesterone profiles were quite different. For SI females this pattern was constant along all seasons and for RM and SU females this pattern was higher during autumn and winter than spring and summer.

The objective of this experiment was to evaluate effects of treatments with intravaginal progesterone (N) device and/or 48 h temporary weaning (TW) on reproductive performance of postpartum anestrous crossbred Angus x Nelore cows, throughout the breeding season (BS). Anestrous cows (n = 296; 53 5 DPP, body condition score between 2.5 and 3.5, on a 1-5 scale) were assigned randomly to four treatments (2 x 2 factorial design): (1) Control Group (no treatment; n = 73); (2) TW Group (48 h TW beginning on day 0 of BS; n = 70); (3) P4 Group (Intravaginal P4 device between days -7 and 0 of BS; n = 73); and (4) P4+TW Group (Intravaginal P4 device between days -7 and 0 of BS followed by 48h TW; n=70). on day 0 of BS (when P4 devices were removed and TW was initiated), cows had their ovaries evaluated by ultrasonography aiming to evaluate size of the largest follicle. Signs of behavioral estrus were observed twice a day (morning and evening), between days 0 and 25 of BS, and cows observed in estrus were inseminated 12 h later. Between days 26 and 80 of BS, cows were submitted to natural service. Pregnancy diagnosis was conducted by transrectal ultrasonography on days 32 and 53 (pregnancies by AI) and 114 (pregnancies by natural service). Rates of detection of estrus during the first 3 days and within 25 days of BS were greater in cows receiving TW (45.0% compared with 9.6% and 50.0% compared with 29.5%, respectively; P < 0.01). Conception rate in cows inseminated in the first 3 days of BS was greater in cows treated with P4 (54.8% compared with 11.1 %; P < 0.01). Combining P4+TW increased pregnancy rate in the first 3 days (Control: 1.4%, TW: 4.3%, P4: 8.2% and P4+TW: 24.3%; P < 0.01) and in 25 days of BS (Control: 10.9%, TW: 7.1%, P4: 13.7% and P4+TW: 28.6%; P < 0.05). Pregnancy rate at the end of BS did not differ among treatment groups. In conclusion, TW alone increased estrous behavior whereas P4 alone benefited conception. Combining P4 with TW improved pregnancy rate with direct benefits to behavioral estrus and conception. (c) 2008 Elsevier B.V. All rights reserved.
