This work aimed at an evaluation of the classical iodine method for quantification of vitamin C (L-ascorbic acid) in fruit juices, as well as at a search into the stability of this so popular vitamin under different conditions of pH, temperature and light exposition, in addition to a proposal of a new quantification method. Our results point to the persistent reversibility of the blue color of the starch-triiodide complex at the end point when using the classical iodine titration, and the overestimation of the true vitamin concentration in fruit juices. A new quantification method is proposed in order to overcome this problem. Surprising conclusions were obtained regarding the controversial stability of L-ascorbic acid toward atmospheric oxygen, at low pH, even in fruit juice and at room temperature, showing that the major problem concerned with aging of fruit juices is proliferation of microorganisms rather than expontaneous oxidation of L-ascorbic acid.

ABSTRACT Functional genomic analyses require intact RNA; however, Passiflora edulis leaves are rich in secondary metabolites that interfere with RNA extraction primarily by promoting oxidative processes and by precipitating with nucleic acids. This study aimed to analyse three RNA extraction methods, Concert™ Plant RNA Reagent (Invitrogen, Carlsbad, CA, USA), TRIzol® Reagent (Invitrogen) and TRIzol® Reagent (Invitrogen)/ice -commercial products specifically designed to extract RNA, and to determine which method is the most effective for extracting RNA from the leaves of passion fruit plants. In contrast to the RNA extracted using the other 2 methods, the RNA extracted using TRIzol® Reagent (Invitrogen) did not have acceptable A260/A280 and A260/A230 ratios and did not have ideal concentrations. Agarose gel electrophoresis showed a strong DNA band for all of the Concert™ method extractions but not for the TRIzol® and TRIzol®/ice methods. The TRIzol® method resulted in smears during electrophoresis. Due to its low levels of DNA contamination, ideal A260/A280 and A260/A230 ratios and superior sample integrity, RNA from the TRIzol®/ice method was used for reverse transcription-polymerase chain reaction (RT-PCR), and the resulting amplicons were highly similar. We conclude that TRIzol®/ice is the preferred method for RNA extraction for P. edulis leaves.

A confiança no médico e o sucesso terapêutico dependem, além de outros fatores, de uma boa comunicação entre profissional de saúde e paciente. O presente estudo buscou conhecer a percepção dos pacientes sobre aspectos da comunicação não verbal que influenciam a consolidação da confiança no médico. Trata-se de um estudo transversal, descritivo e analítico, com amostra aleatória de pacientes alocada em locais públicos da cidade. Foram realizadas 182 entrevistas com pessoas de idades entre 18 e 88 anos, registrando-se preferência pelo perfil mais tradicionalista do profissional, com roupas brancas, cabelos aparados e sem acessórios. O uso de maquiagem e acessórios (brincos, pulseiras, etc.) para mulheres é aceito com moderação. Existe restrição ao uso de piercings, tatuagens e brincos em homens, especialmente pela população idosa. Confirmando a importância da aparência na comunicação médico-paciente, o estudo destaca a necessidade da inserção do tema nos currículos médicos, uma vez que esses aspectos são pouco discutidos na formação do profissional médico, o que permitirá reflexões sobre aspectos não verbais da comunicação na relação médico-paciente e poderá influenciar positivamente as atitudes dos novos profissionais.

A extensão universitária é um processo educativo, cultural e científico, que articula o ensino e a pesquisa de forma indissociável, possibilitando uma relação transformadora entre Universidade e sociedade. Nesse contexto, o projeto Unimontes Solidária tem como objetivo promover ação solidária articulada, por meio dos diversos cursos da Universidade Estadual de Montes Claros (Unimontes), nos municípios carentes das regiões norte e noroeste de Minas Gerais, Vales do Jequitinhonha e do Mucuri, de forma a equacionar os problemas sociais, reduzir a exclusão e as disparidades regionais, colocando a serviço da sociedade o conhecimento que é produzido na Universidade. As ações do projeto no município de Indaiabira (MG) proporcionaram benefícios tanto aos acadêmicos do curso de Medicina como à comunidade local, a partir das atividades extracurriculares desenvolvidas, ampliando o espaço de aprendizado oferecido pela instituição, complementando a formação acadêmica, bem como amenizando as demandas inerentes ao município relacionadas à saúde. Pesquisar, aprender sobre temas relacionados e estender o conhecimento adquirido à comunidade é a tríade que sustenta a formação acadêmica nas mais modernas metodologias de ensino.

O presente trabalho teve por objetivo isolar e identificar fungos de solo de sistemas agroflorestais e avaliar a atividade enzimática destes fungos. Estes foram isolados pela técnica de diluição sucessiva e avaliados quanto à capacidade de degradar amido, celulose e caseína. Foram identificadas 11 espécies de fungos anamorfos (Hyphomycetes) distribuídos em cinco gêneros: Aspergillus, Cladosporium, Curvularia, Fusarium e Penicillium. As espécies estudadas não apresentaram atividade amilolítica satisfatória. Cladosporium cladosporioides (Fres.) De Vries e Penicillium chrysogenum Thom apresentaram maior índice de relação enzimática (IRE) para protease e celulase. Entre as espécies estudadas seis apresentaram maior Índice de Relação Enzimática (IRE) para celulase em comparação as demais enzimas.

Contexte et problématique. Selon l’Association Canadienne d’Équitation Thérapeutique (ACET), l’équitation thérapeutique comprend toutes les activités équestres s’adressant à une clientèle présentant des déficiences. L’équitation thérapeutique compte plusieurs approches, notamment l’hippothérapie, une stratégie de réadaptation offerte par des ergothérapeutes, physiothérapeutes et orthophonistes. L’hippothérapie se base sur le mouvement tridimensionnel induit par le cheval lequel favorise l’amélioration de diverses fonctions neuromotrices notamment le tonus du tronc et de la tête, la posture debout et les ajustements posturaux. Bien que les approches d’équitation thérapeutique prennent de l’ampleur au Québec, il n’y a toujours aucune règlementation officielle. Il existe donc une confusion importante entre les différentes approches d’équitation thérapeutique et l’hippothérapie actuellement reconnue comme la seule approche médicale de réadaptation utilisant le cheval. Les clientèles présentant de lourdes déficiences neuro-musculo-squelettiques se voient donc régulièrement référées dans des centres qui n’offrent pas d’hippothérapie et sont confrontées à des risques importants. Objectifs. Modéliser les interventions d’équitation thérapeutique afin de rendre explicites les composantes de ces interventions et les liens qui les unissent ainsi qu’analyser la plausibilité des interventions à atteindre les résultats escomptés. Méthodologie. Les interventions d’équitation thérapeutique ont été modélisées par des entrevues réalisées auprès des principales personnes offrant des services d’équitation thérapeutique au Québec. Une revue de la littérature a été conduite sur les principes de réadaptation qui sous-tendent les interventions. L’ensemble des données recueillies ont été analysé selon une procédure habituelle d’analyse de contenu qualitatif. Résultats. Les modèles créés permettent d’améliorer les connaissances des pratiques d’équitation thérapeutiques et d’hippothérapie au Québec. Conséquences. Les modèles permettent d’entamer une réflexion sur la règlementation de ces pratiques au Québec et au Canada ainsi que de soutenir les processus de références dans les différents centres québécois.

We developed 15 new polymorphic microsatellites for the plethodontid salamander Ensatina eschscholtzii. Loci were isolated from a genomic library from Ensatina eschscholtzii xanthoptica enriched for (AAAG)(n) repetitive elements. The number of alleles per locus ranged from 4 to 20 (mean 9) in the sampled population. Observed heterozygosity ranged from 0.37 to 1. None of the loci deviated from Hardy-Weinberg equilibrium or showed significant linkage disequilibrium after a Bonferroni correction for multiple comparisons. All loci amplified in the six other subspecies of the Ensatina eschscholtzii complex. These new markers will prove useful in measuring gene flow and population structure as well as patterns of mating and sperm use in Ensatina.

We isolated 13 polymorphic microsatellite markers from the satin bowerbird, Ptilonorhynchus violaceus from a genomic library enriched in (AAGG)(n) repetitive elements and characterized them in 20 individuals. The number of alleles ranged from two to 18 per locus with the observed heterozygosity ranging from 0.15 to 1.00. These markers will be useful for analysing questions concerning parentage, population genetic structure and models of speciation.

Abstract : 5-Methylcytosine is an epigenetic mark, which can be oxidized to 5-hydroxymethylcytosine (5hmC) in DNA by ten-eleven translocation (TET) oxygenases. It is an initial step in the demethylation of 5mC. Levels of 5hmC is relatively high in the brain compared to other organs, but these levels are known to be significantly reduced during the development of a brain tumor, especially in glioblastoma multiforme (GBM). However, no known mechanisms may fully explain this abnormality. The objectives of my project were to (1) understand the implications of the demethylation pathway mediated by TET, and (2) gain a deeper insight in the epigenetic make-up of brain tumors. (1) U87 cells were incubated with 5mC, 5hmC, 5-formylcytosine (5fC) or co-incubated of 5hmC with 3,4,5,6-tetrahydro-2’-deoxyuridine (dTHU) over a timeline of 0, 24, 48 and 96 hours. (2) 130 brain tumors (GBM= 79; grade II/III= 51) were obtained directly from surgery and immediately suspended in DNA extraction buffer. Both cell samples and tumor tissues underwent DNA extraction and DNA digestion protocols. The percent per cytosine (%/C) was obtained by quantification of 5mC, 5hmC, 5fC, 5-hydroxymethyluracil (5hmU) and 5formyluracil (5fU) using LC-MS/MS. (1) Cellular incubations showed that it is possible to increase levels of 5hmC in DNA, but also a slight increase in 5mC levels throughout the experiment. 5HmC levels dramatically increased by 1.9-fold after 96h. On the other hand, no increase was observed in 5fC levels. Both 5hmC and 5fC incubations were accompanied by high increases in 5hmU and 5fU levels respectively. The addition of dTHU to the 5hmC incubation decreased 5hmU incorporation by 65%. (2) The average levels of 5mC, 5hmC and 5fC, in brain tumors, were 4.0, 0.15 and 0.021 %/C respectively. 5HmU and 5fU levels were present at comparable levels of 5hmC and 5fC. Levels of 5hmC, 5hmU and 5fU were significantly lower in the DNA of GBM specimens. There was a strong correlation between 5mC with 5hmC and 5fC in GBM, but this was absent in low grade tumors. The presence of 5hmU and 5fU in brain tumor and the increase in their levels during cell incubations indicate a deamination activity in these cancerous cells, which may impinge on the cellular levels of 5hmC, in particular. Furthermore, upon the incubations with 5hmC, downstream levels of 5fC did not increase suggesting a TET malfunction. TET activity is maintained in GBMs, but impaired in low grade tumors due to isocitrate dehydrogenase-1 (IDH1) mutations. Therefore, in brain tumors, a strong deamination activity and TET impairment may lead to epigenetic reduction of 5hmC.
