Two new epimeric chlorinated withaphysalins, rel-(4 beta,5 beta,6 alpha,18S,22R)- and rel-(4 beta,5 beta,6 alpha,18R,22R)-6-chloro-18,20-epoxy-18-ethoxy-4,5-dihydroxy-1- oxowitha-2,24-diene-26,22-lactone (1 and 2 resp.), together with the new rel-(4 beta,5 beta,6a,18R,22R)-6-chloro-18,20-epoxy-4,5-dihydroxy-18-methoxy-1-oxowitha-2,24-diene-26,22-lactone (3) and rel-(3 beta,4 beta,5 beta,6 beta,18R,22R)-5,6:18,20-diepoxy-3,18-diethoxy-4-hydroxy-1-oxowith-24-ene-26,22-lactone (4) were isolated from the leaves of Acnistus arborescens and named withaphysalins TW, respectively. The final structures and the complete 1H- and 13C-NMR assignments of the three chlorowithaphysalins 13 were performed by means of HR-ESI-MS and 1D- and 2D-NMR experiments, including COSY, HSQC, and HMBC, beside comparison with spectral data of analogous compounds from the literature. The structure of 4 was also confirmed by means of a single-crystal X-ray diffraction analysis.

In the first phytochemical study of the Aureliana genus (Solanaceae), two new withanolides, 1 and 2, together with two known sterols, were isolated from the MeOH extract of the leaves of Aureliana fasciculata var. fasciculata. The structures were established as (4S,22R)-16 alpha-acetoxy-5 beta,6 beta-epoxy-4 beta,17 alpha-dihydroxy-1-oxowitha-2,24-dienolide (aurelianolide A) and (4S,22R)-16 alpha-acetoxy-4 beta,17 alpha-dihydroxy-1-oxowitha-2,5,24-trienolide (aurelianolide B). The new compounds possessed the unusual 16 alpha,17 alpha-dioxygenated group and were fully characterized by spectroscopic techniques, including (1)H- and (13)C-NMR (DEPT), as well as 2D-NMR (HMBC, HMQC, (1)H, (1)H-COSY, NOESY) experiments, and HR-MS.

Lipoma arborescens é uma condição rara de moléstia intra-articular, usualmente monoarticular, caracterizada por extensa proliferação dos vilos sinoviais e hiperplasia da gordura subsinovial. O tecido sinovial é progressivamente substituído por células maduras de gordura na membrana sinovial. O presente trabalho é o relato de caso de uma condição rara de lipoma arborescens tanto intra-articular (glenoumeral) como da bursa subacromial-subdeltoide além de ruptura do tendão do supraespinhoso. As apresentações clínicas, histológicas e radiográficas assim como o tratamento são discutidos no presente estudo. A apresentação do caso contempla também a avaliação radiográfica, ressonância magnética e exame patológico. Apesar do lipoma arborescens ser uma condição rara, tal hipótese deve ser considerada frente a um caso com hiperproliferação sinovial e lipossubstituição da sinovial.

La presente investigación se planteó reemplazar el uso de insecticidas sintéticos, formulando un champú bioinsecticida de aplicación canina mediante la acción biocida del aceite esencial deAmbrosia arborescens Mill (Altamisa). La planta se recolectó en las laderas del rio Tomebamba, cercanas al Campus Balzay de la Universidad de Cuenca Parroquia San Joaquín. La recolección se realizó durante los meses de Enero a Marzo del 2016. El desarrollo y formulación del producto se realizó en el Laboratorio de Biotecnología, Facultad de Ciencias Químicas de la Universidad de Cuenca. La obtención del aceite esencial de A. arborescens se realizó mediante hidrodestilación por el método Clevenger, con un rendimiento del 0,14%. La actividad biocida se estableció en un ensayo “in vitro” ante el nematodo Panagrellus redivirus, determinándose la dosis letal (DL50) de 250 uL/mL. Debido a la moderada DL50y bajo rendimiento, se planteó como estrategia, determinar el DL50 del extracto orgánico de A. arborescens, el cual se obtuvo mediante una extracción con metanol, consiguiendo un rendimiento del 2 % y DL50de 31,25 uL/mL. De acuerdo estos resultados se procedió a realizar pruebas en pulgas de perros(Ctenocephalides canis) con el extracto de A. arborescens, estableciendo una efectividad del 100 % a la concentración de 46,875 mg/mL en el periodo de tiempo más corto, siendo esta la dosis aplicada para la formulación del champú. El extracto metanólico de A. arborescens presentó elevada actividad biocida, comparado con el aceite esencial. Esta sustancia activa es promisoria en la formulación de bioinsecticidas para mascotas.

Four Alternaria species groups (A. longipes, A. arborescens, A. alternata/A. tenuissima and A. tenuissima/A. mali) are associated with leaf blotch and fruit spot of apple in Australia. There is no information on the variability of pathogenicity among the species and isolates within each species causing leaf blotch or fruit spot. We used a detached leaf assay and an in planta fruit inoculation assay to determine the pathogenicity and virulence of the four Alternaria species. Our results showed that isolates within the same species were not specific to either leaf or fruit tissue and showed great variability in pathogenicity and virulence, indicating cross-pathogenicity, which may be isolate dependent rather than species dependent. Generally, virulence of A. tenuissima and A. alternata isolates on leaf and fruit was higher than other species. Isolates of all species groups were pathogenic on leaves of different cultivars, but pathogenicity on fruit of different cultivars varied among isolates and species. Implications of our findings on prevalence of the diseases in different apple-producing regions in Australia and the development of targeted disease management of the diseases are discussed

以锦鸡儿属中柠条锦鸡儿(Caraganakorshinskii)、中间锦鸡儿(C.intermedia)和树锦鸡儿(C.arborescens)种子为外植体,通过对增殖阶段和生根阶段培养基的筛选,探讨了细胞分裂素、生长素、矿质营养和活性炭对这3个树种不同发育阶段的影响。结果表明:(1)继代增殖阶段:柠条锦鸡儿、中间锦鸡儿和树锦鸡儿最佳的基本培养基为MS,最适合的激素浓度配比中,柠条锦鸡儿用BA2mg·L-1+GA2mg·L-1分生率最高;中间锦鸡儿用BA2mg·L-1+GA1mg·L-1分生率最高;树锦鸡儿用BA2mg·L-1+NAA0.1mg·L-1分生率最高。蔗糖是最好的碳源。(2)生根阶段:柠条锦鸡儿诱导生根的适合培养基为1/4MS+NAA0.1mg·L-1和MS+NAA0.1mg·L-1,中间锦鸡儿为1/4MS+NAA0.1mg·L-1,树锦鸡儿为1/2MS+NAA0.1mg·L-1和1/4MS+NAA0.1mg·L-1。活性炭对生根率无显著影响,但普遍增加了根的条数和根的长度。气培生根以MS+IBA50mg·L-1+3%白糖处理较好。

The present paper reports on 22 species collected by the Brazilian Program of Living Resources in the Exclusive Economic Zone (REVIZEE). A new genus and species of Cribrilinidae, Corbuliporina crepida n. gen. et sp., is described, along with seventeen other new species: Chaperia brasiliensis n. sp., Amastigia aviculifera n. sp., Isosecuriflustra pinniformis n. sp., Cellaria subtropicalis n. sp., Melicerita brasiliensis n. sp., Arachnopusia haywardi n. sp., Smittina migottoi n. sp., Hippomenella amaralae n. sp., Rogicka joannae n. sp., Malakosaria atlantica n. sp., Turbicellepora winstonae n. sp., Rhynchozoon coalitum n. sp., Stephanollona angusta n. sp., Stephanollona arborescens n. sp., Aulopocella americana n. sp., Conescharellina cookae n. sp. and Conescharellina bocki n. sp. Chorizopora brongniartii (Audouin, 1826) is recorded for the first time in Brazilian waters and a new combination for Rhynchozoon arborescens Canu & Bassler, 1928 is established. New illustrations and taxonomic remarks are included for two little-known species from Brazil, Rogicka scopae (Canu & Bassler, 1928) and Fenestrulina ampla Canu & Bassler, 1928. A compilation of species recorded from deeper waters of the Brazilian coast is included.

Barbaloin is a bioactive glycosilated 1,8-dihydroxyanthraquinone present in several exudates from plants, Such as Aloe vera, which are used for cosmetic or food purposes. It has been shown that barbaloin interacts with DMPG (dimyristoylphosphatidylglycerol) model membranes, altering the bilayer structure (Alves, D. S.; Perez-Fons, L.; Estepa, A.; Micol, V. Biochem. Pharm. 2004, 68, 549). Considering that ESR (electron spin resonance) of spin labels is one of the best techniques to monitor structural properties at the molecular level, the alterations caused by the anthraquinone barbaloin on phospholipid bilayers will be discussed here via the ESR signal of phospholipid spin probes intercalated into the membranes. In DMPG at high ionic strength (10 mM Hepes pH 7.4 + 100 mM NaCl), a system that presents a gel-fluid transition around 23 degrees C, 20 mol % barbaloin turns the gel phase more rigid, does not alter much the fluid phase packing, but makes the lipid thermal transition less sharp. However, in a low-salt DMPG dispersion (10 mM Hepes pH 7.4 + 2 mM NaCl), which presents a rather complex gel-fluid thermal transition (Lamy-Freund, M. T.; Riske, K. A. Chem. Phys. Lipids 2003, 122, 19), barbaloin strongly affects bilayer structural properties, both in the gel and fluid phases, extending the transition region to much higher temperature values. The position of barbaloin in DMPG bilayers will be discussed on the basis of ESR results, in parallel with data from sample viscosity, DSC (differential scanning calorimetry), and SAXS (small-angle X-ray scattering).

Nas últimas décadas o conhecimento florístico e taxonômico sobre a família Solanaceae no Rio Grande do Sul tem recebido incremento significativo de informações. Dos gêneros com espécies nativas, Bouchetia, Calibrachoa, Nicotiana, Nierembergia, Petunia e Solanum já foram alvo de revisões. Visando complementar o estudo desta família no Estado, foram estudados os gêneros Acnistus, Athenaea, Aureliana, Brunfelsia, Capsicum, Cestrum, Dyssochroma, Grabowskia, Lycianthes, Solandra e Vassobia, além das espécies do gênero Solanum seção Pachyphylla (Cyphomandra sensu lato). Foram catalogadas 22 espécies nativas, sendo três destas, novas citações de ocorrência. Para os onze primeiros gêneros foram identificados 19 espécies, dos quais cinco são de ocorrência restrita (Athenaea picta, Dyssochroma longipes, Grabowskia duplicata, Lycianthes rantonnei e Solandra grandiflora). A seção Pachyphylla do gênero Solanum está representada por três espécies. Dessa forma, foram atualizados os dados quali e quantitativos sobre a participação da família Solanaceae na flora sul-riograndense. Para cada gênero e para cada táxon específico foram elaboradas descrições, e acrescentados dados sobre fenologia, hábitat, comentários pertinentes e mapas de ocorrência. Chaves analíticas para identificação das espécies, quando mais de uma, também foram elaboradas. A revisão de doze herbários regionais possibilitou a elaboração de uma sinopse taxonômica e de uma chave analítica para identificação dos gêneros presentes no Estado. Dos 27 gêneros confirmados, 22 têm espécies nativas. O número de espécies nativas é de 114 e o de introduzidas, cultivadas ou assilvestradas, é até o momento 26.

It was decided to carry out a morphological and molecular characterization of the Italian Alternaria isolatescollected from apple , and evaluate their pathogenicity and subsequently combining the data collected. The strain collection (174 isolates) was constructed by collecting material (received from extension service personnel) between June and August of 2007, 2008, and 2009. A Preliminary bioassays were performed on detached plant materials (fruit and leaf wounded and unwounded), belonging to the Golden cultivar, with two different kind of inoculation (conidial suspension and conidial filtrate). Symptoms were monitored daily and a value of pathogenicity score (P.S.) was assigned on the basis of the diameter of the necrotic area that developed. On the basis of the bioassays, the number of isolates to undergo further molecular analysis was restricted to a representative set of single spore strains (44 strains). Morphological characteristics of the colony and sporulation pattern were determined according to previous systematic work on small-spored Alternaria spp. (Pryor and Michaelides, 2002 and Hong et al., 2006). Reference strains (Alternaria alternata, Alternaria tenuissima, Alternaria arborescens and four Japanese strains of Alternaria alternata mali pathotype), used in the study were kindly provided by Prof. Barry Pryor, who allows a open access to his own fungal collection. Molecular characterization was performed combining and comparing different data sets obtained from distinct molecular approach: 1) investigation of specific loci and 2) fingerprinting based on diverse randomly selected polymorphic sites of the genome. As concern the single locus analysis, it was chosen to sequence the EndoPG partial gene and three anonymous region (OPA1-3, OPA2- and OPa10-2). These markers has revealed a powerful tool in the latter systematic works on small-spored Alternaria spp. In fact, as reported in literature small-spored Alternaria taxonomy is complicated due to the inability to resolve evolutionary relationships among the taxa because of the lack of variability in the markers commonly used in fungi systematic. The three data set together provided the necessary variation to establish the phylogenetic relationships among the Italian isolates of Alternaria spp. On Italian strains these markers showed a variable number of informative sites (ranging from 7 for EndoPg to 85 for OPA1-3) and the parsimony analysis produced different tree topologies all concordant to define A. arborescens as a mophyletic clade. Fingerprinting analysis (nine ISSR primers and eight AFLP primers combination) led to the same result: a monophyleic A. arborescens clade and one clade containing both A. tenuissima and the A. alternata strains. This first attempt to characterize Italian Alternaria species recovered from apple produced concordant results with what was already described in a similar phylogenetic study on pistachio (Pryor and Michaelides, 2002), on walnut and hazelnut (Hong et al., 2006), apple (Kang et al., 2002) and citurus (Peever et al., 2004). Together with these studies, this research demonstrates that the three morphological groups are widely distributed and occupy similar ecological niches. Furthermore, this research suggest that these Alternaria species exhibit a similar infection pattern despite the taxonomic and pathogenic differences. The molecular characterization of the pathogens is a fundamental step to understanding the disease that is spreading in the apple orchards of the north Italy. At the beginning the causal agent was considered as Alteraria alternata (Marshall and Bertagnoll, 2006). Their preliminary studies purposed a pathogenic system related to the synthesis of toxins. Experimental data of our bioassays suggest an analogous hypothesis, considering that symptoms could be induced after inoculating plant material with solely the filtrate from pathogenic strains. Moreover, positive PCR reactions using AM-toxin gene specific primers, designed for identification of apple infecting Alternaria pathovar, led to a hypothesis that a host specific toxin (toxins) were involved. It remains an intriguing challenge to discover or not if the agent of the “Italian disease” is the same of the one previously typified as Alternaria mali, casual agent of the apple blotch disease.
