Insertion of a malE B-Galactosidase fusion protein into the envelope of Escherichia coli disrupts biogenesis of outer membrane proteins and processing of inner membrane proteins

The synthesis of a membrane-bound MalE ,B-galactosidase hybrid protein, when induced by growth of Escherichia coli on maltose, leads to inhibition of cell division and eventually a reduced rate of mass increase. In addition, the relative rate of synthesis of outer membrane proteins, but not that of inner membrane proteins, was reduced by about 50%o. Kinetic experiments demonstrated that this reduction coincided with the period of maximum synthesis of the hybrid protein (and another maltose-inducible protein, LamB). The accumulation of this abnormal protein in the envelope therefore appeared specifically to inhibit the synthesis, the assembly of outer membrane proteins, or both, indicating that the hybrid protein blocks some export site or causes the sequestration of some limiting factor(s) involved in the export process. Since the MalE protein is normally located in the periplasm, the results also suggest that the synthesis of periplasmic and outer membrane proteins may involve some steps in common. The reduced rate of synthesis of outer membrane proteins was also accompanied by the accumulation in the envelope of at least one outer membrane protein and at least two inner membrane proteins as higher-molecular-weight forms, indicating that processing (removal of the N-terminal signal sequence) was also disrupted by the presence of the hybrid protein. These results may indicate that the assembly of these membrane proteins is blocked at a relatively late step rather than at the level of primary recognition of some site by the signal sequence. In addition, the results suggest that some step common to the biogenesis of quite different kinds of envelope protein is blocked by the presence of the hybrid protein.

Nuori Suomi : kirjallistaiteellinen joulualbumi

Nuoren Suomen ryhmä eli nuorsuomalaiset oli 1800-luvulla Suomen politiikkaan ja kulttuurielämään syntynyt radikaali liike, joka ajoi voimakkaasti yhteiskunnallisia uudistuksia ja edusti kulttuurielämän uusia virtauksia. Nuoren Suomen ryhmän keskeisin foorumi oli vuonna 1889 perustettu Päivälehti, jonka joulukirjana vuodesta 1891 vuoteen 1903 ilmestynyt kirjallistaiteellinen albumi oli nimeltään Nuori Suomi. Päivälehden lakkauttamisen jälkeen Nuori Suomi joulualbumi jatkoi ilmestymistään Santeri Ivalon toimittamana 1904 - 1923, siirtyi Suomen Kirjailijaliitolle vuonna 1924 ja lopulta Eljas Erkolle vuonna 1930. Albumi ilmestyi vuoteen 1940 saakka kerran vuodessa. Sen 50 vuosikerrassa on yhteensä yli 1000 artikkelia tai kaunokirjallista teosta, sekä saman verran kuvia aikakauden merkittävimpien kuvataiteilijoiden töistä.

Jugamos con lo que tiramos : una propuesta para impartir Educación Física en las escuelas rurales de Castilla y León.

La experiencia se realiza en varios centros rurales de la provincia de Valladolid. Todos los profesores y profesoras implicadas trabajan en dichos centros y además pertenecen a la Asociación de Educación para la Paz 'La Comba'. Los objetivos propuestos con esta experiencia son los siguientes: 1) Planificar y desarrollar una propuesta de programación para el área de Educación Física en Educación Primaria, orientada a la escuela rural partiendo de la legislación vigente. 2) Ofrecer al profesorado de Educación Física de los Centros Rurales agrupados, alternativas orientadas a solventar la falta de materiales y de recursos en dichos centros. 3) Elaborar una serie de sesiones de clase, adaptadas a la escuela rural, orientadas a concretar en la práctica los objetivos y contenidos considerados como básicos en el área de Educación Física. El desarrollo de la experiencia se concreta en cuatro fases: 1) Análisis de la situación inicial. 2) Elaboración de estrategias orientadas a solventar en lo posible los problemas observados. 3) Puesta en práctica, recogida de datos y análisis de los mismos. 4) Conclusiones y redacción definitiva de la propuesta de programa para escuelas rurales. Entre los materiales elaborados se destaca un dossier memoria donde se recoge la experiencia. El trabajo no ha sido publicado.

Lo audiovisual y la educación en el tiempo libre.

La televisión, el video, el cino y, actualmente, la navegación en internet tienen una irrefutable presencia en las pautas de comportamiento de los adolescentes. Ocupan buena parte de su tiempo de ocio e influyen en sus procesos educativos. Este artículo ofrece diversas perspectivas sobre dicha influencia así como indicaciones útiles para poder emplear los medios audiovisuales en distintas actividades educativas.

Wasserabpressende Fördereinrichtung zur energieeffizienten Entwässerung von Schüttgut

Für die Optimierung und somit Energieeinsparung beim Trocknen von Grüngut wurde eine mechanische Vorentwässerung entwickelt. Dieser Schritt wurde notwendig, da sich die politischen und ökonomischen Rahmenbedingungen, durch Subventionskürzungen und steigenden Energiepreisen, zu Ungunsten der Trocknungsbetriebe verändert haben und eine rein thermische Trocknung kaum noch rentabel ist. In einem gemeinsamen Kooperationsprojekt zwischen der TU Chemnitz und der fömat GmbH entstand eine Schneckenpresse speziell für Grüngut ohne dabei wichtige Nährstoffe beim Entwässern mit auszuspülen. Seit 2010 ist diese Fördereinrichtung in den Trocknungsprozess vollständig integriert und es konnten während einer vollständigen Trocknungssaison in ca. 1.800 Betriebsstunden mehr als 10.500 m³ Wasser aus 47.200 t Grüngut abpressen werden. Dies hatte zur Folge, dass eine Energieeinsparung von über 25 % erreicht wird. Die Qualität des Grünfutters wurde dabei nicht beeinträchtigt und die wichtigen Nährstoffe blieben im Grüngut nahezu vollständig erhalten.
