Basement membranes are specialized sheets of extracellular matrix found in contact with epithelia, endothelia, and certain isolated cells. They support tissue architecture and regulate cell behaviour. Laminins are among the main constituents of basement membranes. Due to differences between laminin isoforms, laminins confer structural and functional diversity to basement membranes. The first aim of this study was to gain insights into the potential functions of the then least characterized laminins, alpha4 chain laminins, by evaluating their distribution in human tissues. We thus created a monoclonal antibody specific for laminin alpha4 chain. By immunohistochemistry, alpha4 chain laminins were primarily localized to basement membranes of blood vessel endothelia, skeletal, heart, and smooth muscle cells, nerves, and adipocytes. In addition, alpha4 chain laminins were found in the region of certain epithelial basement membranes in the epidermis, salivary gland, pancreas, esophagus, stomach, intestine, and kidney. Because of the consistent presence of alpha4 chain laminins in endothelial basement membranes of blood vessels, we evaluated the potential roles of endothelial laminins in blood vessels, lymphatic vessels, and carcinomas. Human endothelial cells produced alpha4 and alpha5 chain laminins. In quantitative and morphological adhesion assays, human endothelial cells barely adhered to alpha4 chain-containing laminin-411. The weak interaction of endothelial cells with laminin-411 appeared to be mediated by alpha6beta1 integrin. The alpha5 chain-containing laminin-511 promoted endothelial cell adhesion better than laminin-411, but it did not promote the formation of cell-extracellular matrix adhesion complexes. The adhesion of endothelial cells to laminin-511 appeared to be mediated by Lutheran glycoprotein together with beta1 and alphavbeta3 integrins. The results suggest that these laminins may induce a migratory phenotype in endothelial cells. In lymphatic capillaries, endothelial basement membranes showed immunoreactivity for laminin alpha4, beta1, beta2, and gamma1 chains, type IV and XVIII collagens, and nidogen-1. Considering the assumed inability of alpha4 chain laminins to polymerize and to promote basement membrane assembly, the findings may in part explain the incomplete basement membrane formation in these vessels. Lymphatic capillaries of ovarian carcinomas showed immunoreactivity also for laminin alpha5 chain and its receptor Lutheran glycoprotein, emphasizing a difference between normal and ovarian carcinoma lymphatic capillaries. In renal cell carcinomas, immunoreactivity for laminin alpha4 chain was found in stroma and basement membranes of blood vessels. In most tumours, immunoreactivity for laminin alpha4 chain was also observed in the basement membrane region of tumour cell islets. Renal carcinoma cells produced alpha4 chain laminins. Laminin-411 did not promote adhesion of renal carcinoma cells, but inhibited their adhesion to fibronectin. Renal carcinoma cells migrated more on laminin-411 than on fibronectin. The results suggest that alpha4 chain laminins have a counteradhesive function, and may thus have a role in detachment and invasion of renal carcinoma cells.

Serum butyrylcholinesterase (BCHE) activity is associated with obesity, blood pressure and biomarkers of cardiovascular and diabetes risk. We have conducted a genome-wide association scan to discover genetic variants affecting BCHE activity, and to clarify whether the associations between BCHE activity and cardiometabolic risk factors are caused by variation in BCHE or whether BCHE variation is secondary to the metabolic abnormalities. We measured serum BCHE in adolescents and adults from three cohorts of Australian twin and family studies. The genotypes from approximately 2.4 million single-nucleotide polymorphisms (SNPs) were available in 8791 participants with BCHE measurements. We detected significant associations with BCHE activity at three independent groups of SNPs at the BCHE locus (P = 5.8 x 10(-262), 7.8 x 10(-47), 2.9 x 10(-12)) and at four other loci: RNPEP (P = 9.4 x 10(-16)), RAPH1-ABI2 (P = 4.1 x 10(-18)), UGT1A1 (P = 4.0 x 10(-8)) and an intergenic region on chromosome 8 (P = 1.4 x 10(-8)). These loci affecting BCHE activity were not associated with metabolic risk factors. On the other hand, SNPs in genes previously associated with metabolic risk had effects on BCHE activity more often than can be explained by chance. In particular, SNPs within FTO and GCKR were associated with BCHE activity, but their effects were partly mediated by body mass index and triglycerides, respectively. We conclude that variation in BCHE activity is due to multiple variants across the spectrum from uncommon/large effect to common/small effect, and partly results from (rather than causes) metabolic abnormalities.

In recent years, there has been intense interest in the potential health benefits of dietary derived plant polyphenols and antioxidants. A new variety of Prunus salicina, Queen Garnet plum (QGP), was developed as a high anthocyanin, high antioxidant plum, in a Queensland Government breeding program. Following consumption of 400 mL QGP juice (QGPJ; 1,117 mg anthocyanins) by two healthy male subjects, QGP anthocyanins (cyanidin-3-glucoside and cyanidin-3-rutinoside) were excreted mainly as methylated and glucuronidated metabolites in urine (0.5% of the ingested dose within 24 h). Furthermore, QGPJ intake resulted in a threefold increase in hippuric acid excretion (potential biomarker for total polyphenols intake and metabolite), an increased urinary antioxidant capacity and a decreased malondialdehyde excretion (biomarker for oxidative stress) within 24 h as compared with the polyphenol-/antioxidant-free control. Results from this pilot study suggest that metabolites, and not the native QGP anthocyanins/polyphenols, are most likely the bioactive compounds in vivo.

Background Increased disease resistance is a key target of cereal breeding programs, with disease outbreaks continuing to threaten global food production, particularly in Africa. Of the disease resistance gene families, the nucleotide-binding site plus leucine-rich repeat (NBS-LRR) family is the most prevalent and ancient and is also one of the largest gene families known in plants. The sequence diversity in NBS-encoding genes was explored in sorghum, a critical food staple in Africa, with comparisons to rice and maize and with comparisons to fungal pathogen resistance QTL. Results In sorghum, NBS-encoding genes had significantly higher diversity in comparison to non NBS-encoding genes and were significantly enriched in regions of the genome under purifying and balancing selection, both through domestication and improvement. Ancestral genes, pre-dating species divergence, were more abundant in regions with signatures of selection than in regions not under selection. Sorghum NBS-encoding genes were also significantly enriched in the regions of the genome containing fungal pathogen disease resistance QTL; with the diversity of the NBS-encoding genes influenced by the type of co-locating biotic stress resistance QTL. Conclusions NBS-encoding genes are under strong selection pressure in sorghum, through the contrasting evolutionary processes of purifying and balancing selection. Such contrasting evolutionary processes have impacted ancestral genes more than species-specific genes. Fungal disease resistance hot-spots in the genome, with resistance against multiple pathogens, provides further insight into the mechanisms that cereals use in the “arms race” with rapidly evolving pathogens in addition to providing plant breeders with selection targets for fast-tracking the development of high performing varieties with more durable pathogen resistance.

Key features of the synthesis include the utility of a hitherto unexplored beta-ketophosphonate derived from lactic acid and Yamaguchi lactonization leading to the title compounds. El autoconcepto es una pieza esencial para el desarrollo adecuado del individuo, por lo que en este trabajo se elabora y aplica una intervención en relación al logro de un autoconcepto ajustado. Para ello, se llevan a cabo una serie de dinámicas en un grupo-clase y periodo de tiempo específicos. Asimismo, se utilizan una serie de instrumentos de medida previa y posteriormente a la intervención, con el objetivo de evaluar las posibles variaciones en lo que al autoconcepto de los alumnos respecta. Finalmente, se concluyen ciertas variaciones que más que reflejar grandes cambios en el autoconcepto del alumnado, indican la importancia y posible eficacia de programas que dirigen su atención a dicho constructo

Thirty-nine participants from 18 countries, including women fishworkers, representatives of fishworker organizations and NGOs, activists and researchers, met at Mahabalipuram, India, during 7-10 July 2010, to discuss the theme “Recasting the Net: Defining a Gender Agenda for Sustaining Life and Livelihoods in Fishing Communities”. The workshop was meant to reflect on what needs to be done to develop a ‘gender agenda’ for sustaining life and livelihoods in fisheries. It was also organized against the backdrop of the growing international recognition of small-scale fisheries and efforts to enhance their contribution to food security and poverty alleviation, as evidenced in the deliberations of the the Committee on Fisheries (COFI) of the Food and Agriculture Organization of the United Nations (FAO). This publication—the proceedings of the Mahabalipuram workshop—will be useful for fishworker organizations, gender activists, researchers, policymakers, fish farmers, members of civil society and anyone interested in gender, fisheries and livelihoods. Hoje, a literatura é bastante limitada no que se refere a estudos sobre a influência da mídia no consumo de alimentos, especialmente se pretendemos articular cultura e saúde. Neste estudo, buscamos compreender a prática discursiva presente na publicidade televisiva de um refrigerante a partir do olhar de diversos segmentos da sociedade. Trata-se de um estudo exploratório, que parte de um filme publicitário de um refrigerante muito consumido. Realizamos dez grupos focais envolvendo profissionais e estudantes do campo da Nutrição e da Comunicação e prestadores de serviços de limpeza de uma universidade, num total de 74 participantes. Entre os temas que surgiram a partir da análise do material oriundo dos grupos focais e tomando por referência teórica autores como Bourdieu, Bauman e Ayres, destacamos a recontextualização e a fragmentação da vida humana e, em seu interior, da alimentação dita saudável na prática discursiva publicitária, neste momento em que, no Brasil, a prevalência de obesidade e de outras doenças crônico-degenerativas a ela associada encontra-se em elevação e onde há movimentos na sociedade militando pela regulamentação da publicidade de alimentos industrializados. Parece-nos que, ao reduzir o ser humano à condição de consumidor tentando levá-lo para o mundo idealizado dos sonhos, colocam-se os objetivos de aumento de vendas e de lucros acima daqueles relativos à promoção de saúde, à boa alimentação e ao bem viver. A indústria de alimentos juntamente com as agências de publicidade operam fenômenos inerentes a esse modelo de financiamento da televisão através da publicidade que reforçam perspectivas fragmentárias, recontextualizadas e conflituosas de pensar a saúde, a alimentação e a vida. Não contribuem, ou mesmo, dificultam a construção de projetos de felicidade num mundo mais justo e humano.

We investigate the influence of vacuum organic contaminations on laser-induced damage threshold (LIDT) of optical coatings. Anti-reflective (AR) coatings at 1064 nm made by Ta2O5/SiO2 are deposited by the ion beam sputtering method. The LIDTs of AR coatings are measured in vacuum and in atmosphere, respectively. It is exhibited that contaminations in vacuum are easily to be absorbed onto optical surface because of lower pressure, and they become origins of damage, resulting in the decrease of LIDT from 24.5 J/cm(2) in air to 15.7 J/cm(2) in vacuum. The LIDT of coatings in vacuum has is slightly changed compared with the value in atmosphere after the organic contaminations are wiped off. These results indicate that organic contaminations are the main reason of the LIDT decrease in vacuum. Additionally, damage morphologies have distinct changes from vacuum to atmosphere because of the differences between the residual stress and thermal decomposability of filmy materials.

经整理我国缨鱼属鱼类仅有1种Crossocheilusreticulatus(Fowler);原置于此属的种CrossocheilusbamaensisFang和CrosscheilusliuchengensisLiang则归并于另一新属拟缨鱼鹰Pseudocrossocheilusgen.nov,其上唇消失,吻皮与下唇相连,上颌与下唇相连以及口角须发达等特征与缨鱼属不同;同时其口角须发达、下唇两侧有纵行的额沟以及下唇前缘具有显著的乳实状角质突起等特征与直口鲮属Rectoris相异。
