Recent deep-ocean exploration has revealed unexpectedly widespread and diverse coral ecosystems in deep water on continental shelves, slopes, seamounts, and ridge systems around the world. Origin and growth history of these cold-water coral mounds are poorly known, owing to a lack of complete stratigraphic sections through them. Here we show high-resolution oxygen isotope records of planktic foraminifers from the base to the top of Challenger Mound, southwest of Ireland, which was drilled during Integrated Ocean Drilling Program Expedition 307. Challenger Mound began to grow during isotope stage 92 (2.24 million years ago (Ma)), immediately after the onset of Northern Hemisphere glaciation and the initiation of modern stratification in the northeast Atlantic. Mound initiation was likely due to reintroduction of Mediterranean Outflow Water (MOW) and ensuing development of a density gradient with overlying northeastern Atlantic water (NEAW), where organic matter was prone to be stagnated and fueled the coral ecosystem. Coral growth continued for 11 glacial-interglacial cycles until isotopic stage 72 (1.82 Ma) with glacial siliciclastic input from the continental margin. After a long hiatus that separates the lower mound and the upper mound, coral growth restored for a short time in isotope stages 19-18 (0.8-0.7 Ma) in which sediments were either eroded or not deposited during a full glacial stage. The development pattern of the water mass interface between MOW and NEAW might have changed, because of the fluctuations of the MOW production which is responsible for the amplitude in ice volume oscillations from the low-amplitude 41 ka cycles for the lower mound to the high-amplitude 100 ka cycles for the upper mound. The average sedimentation and CaCO3 production rates of the lower mound were evaluated 27 cm/ka and 31.1 g/cm2/ka, respectively.

The Plio-Pleistocene intensification of Northern Hemisphere continental ice-sheet development is known to have profoundly affected the global climate system. Evidence for early continental glaciation is preserved in sediments throughout the North Atlantic Ocean, where ice-rafted detritus (IRD) layers attest to the calving of sediment-loaded icebergs from circum-Atlantic ice sheets. So far, Early-Pleistocene IRD deposition has been attributed to the presence of high-latitudinal ice sheets, whereas the existence and extent of ice accumulation in more temperate, mid-latitudinal regions remains enigmatic. Here we present results from the multiproxy provenance analysis of a unique, Pleistocene-Holocene IRD sequence from the Irish NE Atlantic continental margin. There, the Challenger coral carbonate mound (IODP Expedition 307 site U1317) preserved an Early-Pleistocene record of 16 distinctive IRD events, deposited between ca 2.6 and 1.7 Ma. Strong and complex IRD signals are also identified during the mid-Pleistocene climate transition (ca 1.2 to 0.65 Ma) and throughout the Middle-Late Pleistocene interval. Radiogenic isotope source-fingerprinting, in combination with coarse lithic component analysis, indicates a dominant sediment source in the nearby British-Irish Isles, even for the oldest, Early-Pleistocene IRD deposits. Hence, our findings demonstrate, for the first time, repeated and substantial (i.e. marine-terminating) ice accumulation on the British-Irish Isles since the beginning of the Pleistocene. Contemporaneous expansion of both high- and mid-latitudinal ice sheets in the North Atlantic region is therefore implied at the onset of the Pleistocene. Moreover, it suggests the recurrent establishment of (climatically) favourable conditions for ice sheet inception, growth and instability in mid-latitudinal regions, even in the earliest stages of Northern Hemisphere glacial expansion and in an obliquity-driven climate system.

Sr isotope stratigraphy provides a new age model for the first complete section drilled through a deep-water coral mound. The 155-m-long section from Challenger Mound in the Porcupine Sea-bight, southwest of Ireland, is on Miocene siliciclastics and consists entirely of sediments bearing well-preserved cold-water coral Lophelia pertusa. The 87Sr/86Sr values of 28 coral specimens from the mound show an upward-increasing trend, correspond to ages from 2.6 to 0.5 Ma, and identify a significant hiatus from ca. 1.7 to 1.0 Ma at 23.6 m below seafloor. The age of the basal mound sediments coincides with the intensification of Northern Hemisphere glaciations that set up the modern stratification of the northeast Atlantic and enabled coral growth. Mound growth persisted throughout glacial-interglacial fluctuations, reached a maximum rate (24 cm/k.y.) ca. 2.0 Ma, and ceased at 1.7 Ma. Unlike other buried mounds in Porcupine Seabight, Challenger Mound was only partly covered during its growth interruption, and growth restarted ca. 1.0 Ma.

Microcantilever-based biosensors have been found increasing applications in physical, chemical, and biological fields in recent years. When biosensors are used in those fields, surface stress and mass variations due to bio-molecular binding can cause the microcantilever deform or the shift of frequency. These simple biosensors allow biologists to study surface biochemistry on a micro or nano scale and offer new opportunities in developing microscopic biomedical analysis with unique characteristics. To compare and illustrate the influence of the surface stress on the frequency and avoid unnecessary and complicated numerical solution of the resonance frequency, some dimensionless numbers are derived in this paper by making governing equations dimensionless. Meanwhile, in order to analyze the influence of the general surface stress on the frequency, a new model is put forward, and the frequency of the microcantilever is calculated by using the subspace iteration method and the Rayleigh method. The sensitivity of microcantilever is also discussed. (19 refs.)

The geneswere cloned for the two apoprotein subunits, alpha and beta, of phycocyanin from the cyanobacterium Spirulina maxima (=Arthrospira maxima) strain F3. The alpha- and beta-subunit gene-coding regions contain 489 bp and 519 bp, respectively. The beta-subunit gene is upstream from the alpha-subunit gene, with a 111-bp segment separating them. Similarities between the alpha-subunits of S. maxima and nine other cyanobacteria were between 58% and 99%, as were those between the beta-subunits. The maximum similarity between the alpha- and beta-subunits from S. maxima was 27%.

We introduced a targeted single base deletion at codon 307 of the rds-peripherin gene in mice, similar mutations being known to cause autosomal dominant retinitis pigmentosa (RP) in man. Histopathological and electroretinographic analysis indicate that the retinopathy in mice homozygous for the codon 307 mutation appears more rapid than that in the naturally occurring null mutant, the rds(-/-) mouse, suggesting that the rds-307 mutation displays a dominant negative phenotype in combination with that due to haplosufficiency. RP is the most prevalent cause of registered visual handicap in those of working age in developed countries, the 50 or so mutations so far identified within the RDS-peripherin gene accounting for up to 10% of dominant cases of the disease. Given the sequence homologies that exist between the murine rds-peripherin and the human RDS-peripherin gene, this disease model, the first to be generated for peripherin-based RP using gene targeting techniques, should in principle be of value in the work-up in mice of therapeutics capable of targeting transcripts derived from the human gene.
