Study of the novel non-xanthine heterocyclic compound GU 285 as a potent non-selective adenosine receptor antagonist in the rat

The novel pyrazolo[3,4-d]pyrimidine compound GU285 (4-amino-6-alpha-carbamoylethylthio-1- phenylpyrazolo[3,4-d]pyrimidine, CAS 134896-40-5) was examined for its ability (1) to inhibit binding of adenosine (ADO) receptor ligands in rat brain membranes, (2) to antagonise functional responses to ADO agonists in rat right and left atria and coronary resistance vessels, and (3) to reduce the fall in heart rate and arterial blood pressure produced by the ADO A1 agonist N6-cyclopentyladenosine (CPA) in the intact, anaesthetized rat. GU285 competitively inhibited binding of the ADO A1 agonist [3H]-R-N6-phenylisopropyladenosine (R-PIA) yielding a Ki value of 11 (7-18) nmol.l-1 (geometric mean +/- 95% Cl). When assayed against the ADO A2A selective agonist [3H]-2-[p-(2-carboxyethyl)- phenethylamino]-5'-N-ethylcarboxamidoadenosine, (CGS21680), a Ki of 15 (10-24) nmol.l-1 was obtained. In spontaneously beating right atria, GU285 competitively antagonized negative chronotropic effects of R-PIA with a pA2 of 8.7 +/- 0.3 and in electrically paced left atria, GU285 competitively antagonized negative inotropic effects of R-PIA with a pA2 of 9.0 +/- 0.1. In the potassium-arrested, perfused rat heart GU285 (1 mumol.l-1) antagonized only the high sensitivity, ADO A2B mediated component of the biphasic relaxation of the coronary vasculature produced by NECA. The low sensitivity component was unchanged. GU285 (1 mumol.kg-1) antagonized the negative chronotropic and hypotensive effects of the adenosine A1 agonist CPA in anaesthetized rats, producing a 10-fold rightward shift in the dose-response relationship. These data demonstrate that in the rat, GU285 is a potent, non-selective adenosine receptor antagonist that maintains its activity in vivo.

O Deputado Fábio Feldmann (PMDB-SP) informa que as estações ecológicas e as áreas de proteção ambiental existem somente no papel, sendo necessária uma efetiva implantação para a preservação dos recursos naturais. O Deputado César Cals Neto (PDS-CE) declara que a audiência da Constituinte, no Estado do Ceará, fortalece a luta para preservar o meio ambiente no estado e em todo o país. A Constiutinte irá criar o princípio geral, depois haverá o detalhamento e a operacionalização de leis que sejam realmente válidas, viáveis e cumpridas. Em Fortaleza (CE) a Frente Nacional de Ação Ecológica na Constituinte visitou o aterro do lixo, uma das zonas mais insalubres da capital. O Deputado Raimundo Bezerra (PMDB-CE) ressalta que o Brasil deveria ter aterros sanitários, industrialização do lixo com proteção ao homem e ao meio ambiente. No encontro com o governador do Ceará, Tasso Jeiressati (PMDB-CE), constituintes abordaram suas preocupações com a defesa ecológica. O governador informou que o governo do Estado está criando uma superintendência do meio ambiente como forma de acompanhar todos esses problemas. Jeiressati também afirma que a visita dessa Comissão ajuda a trazer novas atitudes aos problemas não atentados. Constituintes lembram que no substitutivo do relator Bernardo Cabral (PMDB-AM), existe todo um capítulo destinado ao meio ambiente. Este capítulo afirma que a Floresta Amazônica, Mata Atlântica, Serra do Mar, Pantanal e Zona Costeira são patrimônio nacional. O Deputado Ruy Nedel (PMDB-RS) declara que está estarrecido com o processo imobiliário em grande parte do litoral nacional que tem eliminado os valores naturais da região.

Se transcribe la Ley de fomento del empleo juvenil de Italia, de 1 de junio de 1977, n. 285. Consta de treinta artículos en total, agrupados en cinco títulos. Los principales objetivos que perseguía esta Ley eran: fomentar el empleo de los jóvenes en actividades agrícolas, artesanales, comerciales, industriales y de servicios, desarrolladas por empresas individuales o asociadas, cooperativas y sus consorcios y entidades económicas y públicas; financiar programas regionales de trabajo productivo para obras y servicios de utilidad social, con referencia especial al sector agrícola, y programas de servicios y trabajos propuestos por las administraciones centrales; animar al acceso de los jóvenes al cultivo de la tierra y realizar planes de formación profesional para generar desarrollo durante el año 1977.

Commission v. Gazprom: The antitrust clash of the decade? CEPS Policy Brief No. 285, 31 October 2012

This new CEPS Policy Brief boldly asserts that the antitrust case launched by DG Competition against Gazprom on September 4th will turn out to be the landmark antitrust case of this decade, in much the same way that Microsoft v. Commission was the defining antitrust lawsuit of the last decade. The paper argues that, for a host of political and economic reasons, this case is likely to be hard fought by both sides to a final prohibition decision and then onwards into the EU courts. In the process, the European gas market and the powers of DG Competition in the energy field are likely to be transformed.

Quality in emergency departments: a study on 3,285,440 admissions

INTRODUCTION: A multi-centre study has been conducted, during 2005, by means of a questionnaire posted on the Italian Society of Emergency Medicine (SIMEU) web page. Our intention was to carry out an organisational and functional analysis of Italian Emergency Departments (ED) in order to pick out some macro-indicators of the activities performed. Participation was good, in that 69 ED (3,285,440 admissions to emergency services) responded to the questionnaire. METHODS: The study was based on 18 questions: 3 regarding the personnel of the ED, 2 regarding organisational and functional aspects, 5 on the activity of the ED, 7 on triage and 1 on the assessment of the quality perceived by the users of the ED. RESULTS AND CONCLUSION: The replies revealed that 91.30% of the ED were equipped with data-processing software, which, in 96.83% of cases, tracked the entire itinerary of the patient. About 48,000 patients/year used the ED: 76.72% were discharged and 18.31% were hospitalised. Observation Units were active in 81.16% of the ED examined. Triage programmes were in place in 92.75% of ED: in 75.81% of these, triage was performed throughout the entire itinerary of the patient; in 16.13% it was performed only symptom-based, and in 8.06% only on-call. Of the patients arriving at the ED, 24.19% were assigned a non-urgent triage code, 60.01% a urgent code, 14.30% a emergent code and 1.49% a life-threatening code. Waiting times were: 52.39 min for non-urgent patients, 40.26 min for urgent, 12.08 for emergent, and 1.19 for life-threatening patients.
