miR-219-5p inhibits receptor tyrosine kinase pathway by targeting EGFR in glioblastoma

Glioblastoma is one of the common types of primary brain tumors with a median survival of 12-15 months. The receptor tyrosine kinase (RTK) pathway is known to be deregulated in 88% of the patients with glioblastoma. 45% of GBM patients show amplifications and activating mutations in EGFR gene leading to the upregulation of the pathway. In the present study, we demonstrate that a brain specific miRNA, miR-219-5p, repressed EGFR by directly binding to its 3'-UTR. The expression of miR-219-5p was downregulated in glioblastoma and the overexpression of miR-219-5p in glioma cell lines inhibited the proliferation, anchorage independent growth and migration. In addition, miR-219-5p inhibited MAPK and PI3K pathways in glioma cell lines in concordance with its ability to target EGFR. The inhibitory effect of miR-219-5p on MAPK and PI3K pathways and glioma cell migration could be rescued by the overexpression of wild type EGFR and vIII mutant of EGFR (both lacking 3'-UTR and thus being insensitive to miR-219-5p) suggesting that the inhibitory effects of miR-219-5p were indeed because of its ability to target EGFR. We also found significant negative correlation between miR-219-5p levels and total as well as phosphorylated forms of EGFR in glioblastoma patient samples. This indicated that the downregulation of miR-219-5p in glioblastoma patients contribute to the increased activity of the RTK pathway by the upregulation of EGFR. Thus, we have identified and characterized miR-219-5p as the RTK regulating novel tumor suppressor miRNA in glioblastoma.

O Senador José Fogaça (PMDB-RS) informa que essa é uma decisão de competência do Presidente da Comissão de Sistematização, que deverá tomá-la em conjunto com o Presidente da Assembleia Nacional Constituinte (ANC), Deputado Ulysses Guimarães. O Deputado Miro Teixeira (PMDB-RJ) declara que a proposta é o envio do novo substitutivo do Relator, com o parecer e as emendas, para a discussão no âmbito da Comissão. Sessão da Assembleia Nacional Constituinte presta homenagem a Marcos Freire, que morreu em um acidente aéreo no Pará. O Líder do PMDB, Senador Mário Covas declara que o Brasil perde um excepcional ministro, que o governo perde um extraordinário colaborador e o partido perde um político de grande estatura. O Deputado Fernando Lira (PMDB-PE) lembra da sua principal luta, a Reforma Agrária. O Deputado Egídio Ferreira Lima (PMDB-PE) ressalta que Marcos Freire era o porta-voz dos oprimidos. Women's Club of Rock Hill Records - Accession 219

The Women's Club of Rock Hill's mission is to further the cultural, educational, and social interest of its members and to promote interest in Rock Hill’s civic development and welfare. The Women's Club of Rock Hill Records consist of constitutions and bylaws, histories, minutes, reports, correspondence, memoranda, program notes, brochures, newsletters, membership lists, yearbooks, questions, certificates, awards, scrapbooks, newspaper clippings, and photographs. The records also relate to the thirteen affiliated clubs comprising the Woman’s Club of Rock Hill: Amateur Arts, Arts Appreciation, Book Discussion, Book Lovers, Crafts, Hearthstone, Hermitage, Home Study, Lantern, Literary, Outlook, Palmetto, and Politeia, and to other records for the South Carolina Federation of Women’s Club and the General Federation of Women’s Clubs. Records of various organizations not directly related to the Woman’s Club of Rock Hill are also included, such as the Tri-County Parents Without Partners, the Rock Hill Community Council, the Rock Hill Model Cities Commission, the Rock Hill Senior Center, and the South Carolina Conference on the Status of Women.

Acute stress reactions in the first 3 weeks postpartum: A study of 219 parturients

OBJECTIVES: There is increasing research on posttraumatic stress (PS) 4-6 weeks and 3 months postpartum, but, there are no data on acute stress reactions (ASR) in the first 3 weeks postpartum, i.e. the potential precursors of PS. However, ASR may have long-term effects, e.g., on a subsequent pregnancy without having manifested as PS in the meantime. We propose: (i) to describe the patterns of ASR after childbirth, (ii) to explore differences between women with normal and traumatogenic ASR, and (iii) to provide data on the early detection of traumatogenic ASR 2 and 3 weeks postpartum. STUDY DESIGN: Intra-event variables (relationship with caregivers, overall birth experience, and dissociative experiences, as well as obstetric variables) were assessed 48-96h. postpartum, as were ASR (by means of the Impact-of-Event Scale IES) in weeks 1, 2, and 3 postpartum. According to research on PS the upper 33%-range of ASR in weeks 2 and 3 was defined as 'traumatogenic'. RESULTS: Normal ASR in week 1 are at a level which in non-obstetric trauma-situations is considered as the upper range of low stress or lower range of medium distress. ASR decline constantly from week 1 to week 3. However, high ASR in week 1 do not drop faster than do low ones, thus indicating a prolonged stress reaction in women with high ASR in week 1. Low ASR (IES-scores <10) and high ASR (IES-scores >20) in week 1 are highly predictive for normal ASR, and traumatogenic ASR in weeks 2 and 3, respectively. Medium ASR (IES-scores 10-20) in week 1 are of uncertain predictive value for stress reactions in weeks 2 and 3 and have to be re-assessed at that time. CONCLUSIONS: Clinical screening for ASR appears to be helpful in detecting women with a compromised ability to process childbirth-related stress. The association between ASR and long-term development should be further explored.
