In this paper we argue that intentional curriculum design in the first year of law should encourage law students to develop an emergent sense of a positive professional identity. When first year law students engage with a nascent notion of a positive professional identity, their well-being is supported because their studies are informed and contextualised by a sense of purpose for their future professional life. In a first year law subject run for the first time at the QUT Law School in 2011, reflective practice was successfully used to achieve these goals. The paper discusses the subject, the opportunity of using reflective practice to teach a positive sense of professional identity, and some student perspectives on the subject’s design.

2005年3月-7月对南充市金翅雀的繁殖生态进行了研究.结果表明:金翅雀3月开始繁殖;雌雄共同筑巢,营巢期4-5d;窝卵数:3.67±0.58(n=3),孵化率63.64%;雌孵卵,孵卵期12-13d,孵卵期间食物由雄鸟提供;双亲共同育雏,育雏期14-15d.雏鸟生长曲线呈“S”型.

利用从滇池蓝藻水华生物量中提取的微囊藻毒素试验溶液,接种滇池沉积物的微生物,研究其在有氧条件下的生物降解过程.结果表明,水体中的微囊藻毒素易被生物降解,其降解反应服从方程C=A/(1+Be-Ct).当温度在12~25℃,加入的沉积物量为1~10g时,藻毒素粗提液的平均降解反应速率为3.181.13d-1,平均半衰期t1/2为2.661.27d,且藻毒素的生物降解速度随反应温度和沉积物量的增加而提高.研究结果还表明,生物降解是去除滇池水环境中微囊藻毒素的一个重要机制.

白介素-4受体(IL-4R)在实体瘤和血癌等许多肿瘤细胞表面表达量很高构建导向IL-4R的免疫毒素,是研究肿瘤治疗的一个重要方向。天然IL-4分子N末端和C末端含有很多与受体结合的活性位点,为了降低连接蛋白毒素时对这些位点的影响,将天然IL-4分子的N端和c端用寡肚GGNGG相连,并在非活性部位形成新的开口,构建了cpIL-4;为了进一步提高cpIL-4与IL-4R的亲和力,通过重叠PCR引入13位点突变,得到cpIL-4(13D);为了增强IL-4免疫毒素对淋巴细胞的选择性,在121位引入点突变,得到cpIL-4(13D121E)。将上述三种重组IL-4分子分别于大肠杆菌表达系统进行表达。ELISA分析表明,三种重组蛋白均可与人IL-4抗体特异性结合。由于PE的DNA序列的GC含量很高,很难用常规手段进行改造,本文采用特殊条件的PCR反应和酶切反应进行绿脓杆菌外毒素PE的改造,得到PE38KDEL,并于大肠杆菌表达系统进行表达。其特点是分子量较小,不含结合区,C末端氨基酸KDEL有利于提高其跨膜能力和细胞毒作用。将靶向分子分别与毒素分子相连,得到三种免疫毒素:cpIL4-PE38KDEL,cpIL4(13D)-PE38KDEL,cpIL4(13D121B)-PE38KDEL。将之分别于表达载体pET32a(+)进行表达,目的蛋白表达量均约为菌体总蛋白的30%。western blotting分析表明,诱导后表达的三种IL-4免疫毒素均可与hIL-4抗体特异性结合。采用Ni-NTA亲和层析和阴离子交换层析纯化上述三种IL-4免疫毒素,纯度均在95%以上。用MTT法检测其细胞毒作用,结果显示,免疫毒素cpIL4(13D)-PE38KDEL可特异性地靶向产生IL-4R的细胞株,_且其活性与未突变的免疫毒素cpIIL4-PE38KDEL相比有2-3倍的提高;免疫毒素cpIL4(13D121E)-PE38KDEL对表达I型IL-4R的淋巴瘤细胞结合力较强,对表达II型IL-4R的内皮细胞结合力较弱,因此对淋巴瘤具有一定的选择性,这对于提高药物疗效,降低血管渗漏症等毒副作用具有重要意义。

以盆栽法研究了不同浓度1,2,4-三氯苯(TCB)胁迫对大豆下胚轴膜脂过氧化作用的影响。结果表明,100—300μg·g-1TCB胁迫初期(1—3d)促使萌发大豆下胚轴内过氧化氢(H2O2)的积累显著增加,同时伴随质膜电解质渗漏率和组织自动氧化速率升高,显示发生膜脂质过氧化作用。TCB胁迫1—6d使活性氧清除酶功能紊乱,其中过氧化物酶(POD)活性升高,过氧化氢酶(CAT)活性开始上升后转为下降。推测大豆下胚轴受TCB胁迫伤害过程中,活性氧代谢失衡造成的膜脂质过氧化起着重要作用。

1960—2005年期间,沈阳气候受全球气候变暖的影响,年平均气温总体呈上升趋势,气温升幅为0.96℃,但以1982年为界,1960—1982年为偏冷阶段,气温降幅最大值为1.95℃;1983—2005年为偏暖阶段,气温升幅最大值为1.60℃。一年四季气温处于上升阶段,以冬、春季气温升幅最大,分别为2.3℃和1.35℃。城市森林主要树种的春季物候期,在气候偏冷阶段,春季物候期出现较晚,而在偏暖阶段,春季物候期提前发生。同时,物候春季开始日期与结束日期有密切的同步相关性,并与物候季节节奏的长短呈负相关。树木萌动期早晚与冬季和早春气温高低呈显著的负相关,冬春季气温越高,芽萌动越提前。展叶早晚主要受展叶前的春季气温高低的影响,与冬季气温相关性不显著。始花前2—8旬,特别是2—4旬气温对始花期影响最显著。寒冷指数(CI)与树木芽萌动期、展叶始期呈显著正相关,而与开花始期相关性不显著。预测了CO2倍增和气温升高条件下,沈阳城市森林主要树种的萌动期、展叶始期和开花始期分别提前40.41、43.08和24.13d。

利用开顶式气室研究了1年生小青杨绿叶、细根、粗根和树皮内营养成分对CO2浓度升高的响应,并检验了上树取食的舞毒蛾幼虫的生长反应。结果表明:CO2浓度升高环境下小青杨绿叶、细根、树皮和粗根中的氮含量显著降低,绿叶、树皮和粗根中的碳氮比显著增加,但细根中碳氮比增加未达到显著水平;CO2浓度升高对所有杨树组织的总碳含量均无影响;CO2浓度升高显著增加了绿叶中总可溶性糖和总非结构碳水化合物的浓度,而绿叶淀粉含量仅在部分采样日期发生显著增加,除细根可溶性糖含量在CO2浓度升高处理下降低外,根系和树皮中各碳水化合物指标的浓度均未发生显著变化;CO2浓度升高环境下2龄舞毒蛾幼虫上树取食13d后体重显著低于自然CO2浓度处理下幼虫的体重,但幼虫继续在各自的实验处理下取食11d后,体重未表现出显著性差异。大气CO2浓度升高对杨树各组织内的营养成分含量均有影响,绿叶营养品质变差可能是导致低龄舞毒蛾幼虫生长受抑制的主要原因。

本文主要以花鲈[Lateolabrax.japonicus (C.& V.)]和褐牙鲆[Paralichthys olivaceus(T. & S.)]作为海洋肉食性鱼类的代表种类,根据鱼类生态生理学理论,通过设定不同饥饿时问下因子水平,研究两种鱼类摄食率、排粪率、转化效率和SGR等生态生理效率的状态变化。其目的在于研究高营养级鱼类在海洋生态系统中的下行控制作用(Top-down Effect),以及肉食性鱼类的生态对策与鱼类资源补充机制的相互关系,为深入解析鱼类资源生产力及其持续利用海洋生物资源,提供科学依据。其主要研究结果概述如下:1.花鲈: 饥饿0(对照组)、4、8、12、16d后恢复投喂,过量投喂淡水桡足类,温度为19.5±2.0℃,初始体重为0.61-0.93g,平均体重为0.79g实验周期为28d。1.1饥饿时间对花鲈的体重损失率产生显著影响,受过饥饿的个体的湿重失率

通过对青海省8个农气观测站的土壤表面始冻期和解冻期连续观测资料的分析,结果表明:青海各地土壤表面始冻期和解冻期存在着明显的地域差异,多数站点的封冻期呈现出缩短的趋势,互助、德令哈略有延长.始冻期变化受气候变化的明显影响,当3-9月平均气温升高1℃,除贵德、诺木洪、互助站响应不明显外,其余站点的始冻期推迟1.5~7.5d,平均推迟2.3d;当3-9月日照时数增多10h,诺木洪地区始冻期提早0.5d,其余站点的响应不明显;当上年10月至当年2月平均气温升高1℃,除门源、德令哈站的解冻期响应不明显外,其余站点提前2~13d,平均提早2.5d;降水量增多10mm,除诺木洪推迟12d,其余站点推迟2~6d,平均推迟2.3d;当上年10月至当年2月平均气温升高1℃,封冻期日数除河南、门源、贵德响应不明显外,其余缩短2.4~23.1d,平均缩短1.8d.

Daily ultrasound examinations were conducted from Days 10 to 60 (ovulation = Day 0) of pregnancy to monitor the conceptus in jennies (n = 12). The embryonic vesicle was first detected on Day 11.5 +/- 0.9 (mean +/- SD; range 10 to 13d) and was mobile until movement ceased (fixation) on Day 15.5 +/- 1.4 (range, 13 to 18d). The vesicle was spherical from Days 10 to 18 (mean growth rate, 3.2 mm/d), non spherical (irregular) with a reduced growth rate (0.5 mm/d) from Days 19 to 29, and then grew at a moderate rate (1.6 mm/d) up to Day 46. on average, detection of the embryo proper (consistently located on the ventral aspect of the yolk sac) and embryonic heartbeat were Days 20.7 +/- 1.2 and 23.5 +/- 1.3, respectively. Formation of the allantoic sac was first detected on Day 24.4 +/- 1.7 and was complete on Day 36.8 +/- 1.6. Descent of the fetus (and formation of the umbilical cord) began on Day 37.9 +/- 1.7 and was complete on Day 44.1 +/- 2.1. Crown-ramp length averaged 3.7, 15.4, 22.7, 37.5 and 59.6 mm on Days 20, 30, 40, 50 and 60, respectively. Structure and composition of sub-surface bottom sediments from the southwest Barents Sea have been under study. The study has revealed heterogeneity of sediment structure resulted from temporal irregularity and variability of sedimentation processes. The study of the heavy minerals from 0.1-0.01 mm grain size fraction has shown prevalence of green hornblende, epidote, garnet, and ilmenite in all types of sediments; these minerals are the basis of terrigenous-mineralogical province. At the same time in different areas local terrigenous-mineralogical associations have been identified. Clay mineral composition of in the sediments was quite uniform: biotite, chlorite, hydromica, smectite. Despite this, a number of features indicating initial stages of clay mineral transformation has been identified. Differences in material composition and structure of the studied sediments are associated with rapid change in paleogeographic situation on the land - ice cover melting on the Kola Peninsula and subsequent Holocene climatic situation.
