The development of scaffolds based on biomaterials is a promising strategy for Tissue Engineering and cellular regeneration. This work focuses on Bone Tissue Engineering, the aim is to develop electrically tailored biomaterials with different crystalline and electric features, and study their impacts onto cell biological behavior, so as to predict the materials output in the enhancement of bone tissue regeneration. It is accepted that bone exhibits piezoelectricity, a property that has been proved to be involved in bone growth/repair mechanism regulation. In addition electrical stimulations have been proved to influence bone growth and repair. Piezoelectric materials are therefore widely investigated for a potential use in bone tissue engineering. The main goal is the development of novel strategies to produce and employ piezoelectric biomaterials, with detailed knowledge of mechanisms involved in cell-material interaction. In the current work, poly (L-lactic) acid (PLLA), a synthetic semi-crystalline polymer, exhibiting biodegradibility, biocompatibility and piezoelectricity is studied and proposed as a promoter of enhanced tissue regeneration. PLLA has already been approved for implantation in human body by the Food and Drug Administration (FDA), and at the moment it is being used in several clinical strategies. The present study consists of first preparing films with different degrees of crystallinity and characterizing these PLLA films, in terms of surface and structural properties, and subsequently assessing the behavior of cells in terms of viability, proliferation, morphology and mineralization for each PLLA configuration. PLLA films were prepared using the solvent cast technique and submitted to different thermal treatments in order to obtain different degrees of crystallinity. Those platforms were then electrically poled, positively and negatively, by corona discharge in order to tailor their electrical properties. The cellular assays were conducted by using two different osteoblast cell lines grown directly onto the PLLA films:Human osteoblast Hob, a primary cell culture and Human osteosarcoma MG-63 cell line. This thesis gives also a comprehensive introduction to the area of Bone Tissue Engineering and provides a review of the work done in this field in the past until today, in that same field, including the one related with bone’s piezoelectricity. Then the experimental part deals with the effects of the crystallinity degrees and of the polarization in terms of surface properties and cellular bio assays. Three different degrees of crystallinity, and three different polarization conditions were prepared; which results in 9 different configurations under investigation.

O poli (ácido L-láctico) (PLLA) é um material de grande interesse na área científica, por ser um polímero biodegradável e bioabsorvível, e pela sua grande utilização na área biomédica. Este estudo teve como objetivo a síntese de PLA através de policondensação direta e em meio supercrítico (scCO2) e sua caracterização. As duas rotas buscam um processo limpo de síntese dos polímeros, livre de solvente orgânico. Além disso, procurou-se reduzir os custos de obtenção utilizando matéria-prima nacional (ácido láctico P.A. comercial). As reações de polimerização de PLLA realizada por policondensação e em scCO2 foram feitas em dois estágios. No primeiro estágio da síntese, para ambos os processos, o pré-polímero do ácido láctico foi obtido através de uma reação simples de condensação, com retirada de água, sob atmosfera inerte de N2 e 160ºC. Na segunda etapa, para a reação de policondensação, o polímero PLLA, foi obtido a partir do pré-polímero em uma temperatura de 140ºC e sob pressão reduzida por o tempo desejado (100h, 200h e 350h). As reações em meio scCO2 foram realizadas em um reator de aço inoxidável de 100 mL equipado com uma barra de agitação magnética, sob pressão de CO2 (80atm) e a 90ºC por 4 horas. Em ambas as reações de polimerização foram usadas complexos de estanho como catalisador. No início deste trabalho foram realizadas algumas reações utilizando o D, L-ácido láctico em scCO2. Também foi realizada a síntese do lactide e a polimerização deste por abertura de anel. Para efeito comparativo, também foi realizada a polimerização do lactide comercial. Os polímeros obtidos foram caracterizados por espectroscopia de Ressonância Magnética Nuclear de Próton (1HRMN), por espectroscopia de infravermelho (IV), por cromatografia de permeação em gel (GPC), por calorimetria exploratória de varredura (DSC) e pela técnica de análise termogravimétrica (TGA) A reação de policondensação revelou ser uma rota sintética excelente na obtenção de polímeros de PLA com peso molecular variados. A formação dos polímeros a partir do monômero de ácido láctico foi confirmada pelo desaparecimento da banda de OH em 3500 cm-1 no espectro de infravermelho. O espectro de 1H-RMN de ambos os polímeros, mostrou um sinal atribuído ao hidrogênio do grupo CH3 em 1,52 ppm e um sinal atribuído aos hidrogênios do grupo do CH em 5,16 ppm, que estão de acordo com a literatura. Os polímeros obtidos possuem potencial para uso clínico como materiais de implante.

O objectivo deste estudo consistiu na análise de diferentes parâmetros de formulação e processo e a sua influência no tamanho e potencial zeta de nanopartículas de poli(D,L-láctico-co-glicólico) (PLGA) preparadas por dois métodos diferentes, especificamente, emulsificação espontânea com difusão de solvente (m-SESD) e deslocamento de solventes (SD). As nanopartículas foram produzidas com um tamanho entre um intervalo de 200-300 nm para os dois métodos. O processo de liofilização causou o aumento do tamanho das nanopartículas para 300-500 nm. A presença de agente emulsivo aumentou a estabilidade das nanopartículas. Por outro lado, o tamanho de partículas dependeu do tipo de fármaco utilizado (ibuprofeno ou ácido azelaico). O rendimento de produção das nanopartículas foi de 96,0% para as nanopartículas produzidas com poloxamer 188 e de 94,0% para as partículas produzidas com laurilsulfato de sódio. Este estudo demonstrou esta formulação poderá ser considerada muito promissora sendo que o método m-SESD reveou-se mais eficaz para os parâmetros analisados.

The copolymer poly (L-co-D,L lactic acid), PLDLA, has gained prominence in the field of temporary prostheses due to the fact that their time of degradation is quite compatible with the requirement in the case of osseous fracture. In this work the in vivo degradation of devices from copolymer, as a system of plates and screws, used in fixation of the tibia of rabbits was studied. The devices were implanted in 15 adult rabbits, albinos, New Zealand race, and they were used as control devices of alloys of titanium (Ti-6Al-4V/ V grade). The use of copolymers, synthesized in the laboratory, was tested in the repair of fracture in rabbits'tibias, being assessed in the following times: 2 weeks, 2 months and 3 months. Morphological analysis of tissue surrounding the plate and screw system, for 2 weeks of implantation, showed the presence of osteoblasts, indicating a pre bone formation. After 2 months there was new bone formation in the region in contact with the polymer. This bone growth occurred simultaneously with the process of PLDLA degradation, invading the region where there was polymer and after 3 months there was an intense degradation of the copolymer and hence greater tissue invasion compared to 2 months which characterized bone formation in a region where the polymer degraded. The in vivo degradation study of the devices for PLDLA by means of histological evaluations during the period of consolidation of the fracture showed the efficiency of plate and screw system, and it was possible to check formation of bone tissue at the implantation site, without the presence of inflammatory reaction

Carbon nanotubes (CNT) have been studied for biomedical applications due to their unique properties. However, pristine CNT have structural features and impurities that can cause toxicity to biological systems. In this work, we describe a method to purify multiwalled carbon nanotubes (MWCNT) by chemical modification and subsequent attachment of hydroxyl and carboxyl groups to improve dispersion and to decrease toxic effects. Nanocomposites from poly (L-lactic acid) (PLLA) and nanotubes were produced by the solvent casting method and characterized and evaluated for cytocompatibility with Vero cells. The nanocomposite interactions with Vero cells demonstrated that the cells were able to adhere and sustain proliferation and showed favorable cytocompatibility. In vitro studies also revealed an increase in fibroblast cell viability in the nanocomposites, compared with neat PLLA.

In this paper we describe the preparation poly (L-lactide) (PLA) nanocapsules as a drug delivery system for the local anesthetic benzocaine. The characterization and in vitro release properties of the system were investigated. The characterization results showed a polydispersity index of 0.14, an average diameter of 190.1± 3 nm, zeta potential of -38.5 mV and an entrapment efficiency of 73%. The release profile of Benzocaine loaded in PLA nanocapsules showed a significant different behavior than that of the pure anesthetic in solution. This study is important to characterize a drug release system using benzocaine for application in pain treatment.

We report the single-step derivatization reaction of a biopolymer based onL -lysine with D -biotin analogs:Co -poly(L -lysine)-graft-(ε-N -[X-D-biotinyl]-L -lysine) (PLL-X-Biotin). The valeric acid carboxylate of D -biotin is activated to an NHS ester for direct modification of amine groups in proteins and other macromolecules. NHS esters react by nucleophilic attack of an amine in the carbonyl group, releasing the NHS group, and forming a stable amide linkage. NHS-X-Biotin is the simplest biotinylation reagent commercially available. In contrast withD -biotin, it has a longer spacer arm off the valeric acid side chain allowing better binding potential for avidin or streptavidin probes. Derivatization of poly(L -lysine) (PLL) with NHS-X-Biotin led to a copolymer PLL-X-Biotin. UV-Visible, IR-FT and 1H NMR characteristics derived from synthesis are briefly discussed.
