Reabsorption of filtered urea by the kidney is essential for retaining high levels of urea in marine cartilaginous fish. Our previous studies on the shark facilitative urea transporter (UT) suggest that additional UT(s) comprising the urea reabsorption system could exist in the cartilaginous fish kidney. Here, we isolated three cDNAs encoding UTs from the kidney of elephant fish, Callorhinchus milii, and termed them efUT-1, efUT-2 and efUT-3. efUT-1 is orthologous to known elasmobranch UTs, while efUT-2 and efUT-3 are novel UTs in cartilaginous fish. Two variants were found for efUT-1 and efUT-2, in which the NH2-terminal intracellular domain was distinct between the variants. Differences in potential phosphorylation sites were found in the variant-specific NH2-terminal domains. When expressed in Xenopus oocytes, all five UT transcripts including the efUT-1 and efUT-2 variants induced more than a 10-fold increase in [14C] urea uptake. Phloretin inhibited dose-dependently the increase of urea uptake, suggesting that the identified UTs are facilitative UTs. Molecular phylogenetic analysis revealed that efUT-1 and efUT-2 had diverged in the cartilaginous fish lineage, while efUT-3 is distinct from efUT-1 and efUT-2. The present finding of multiple UTs in elephant fish provides a key to understanding the molecular mechanisms of urea reabsorption system in the cartilaginous fish kidney.

Marine cartilaginous fish retain a high concentration of urea to maintain the plasma slightly hyperosmotic to the surrounding seawater. In adult fish, urea is produced by hepatic and extrahepatic ornithine urea cycles (OUCs). However, little is known about the urea retention mechanism in developing cartilaginous fish embryos. In order to address the question as to the mechanism of urea-based osmoregulation in developing embryos, the present study examined the gene expression profiles of OUC enzymes in oviparous holocephalan elephant fish (Callorhinchus milii) embryos. We found that the yolk sac membrane (YSM) makes an important contribution to the ureosmotic strategy of the early embryonic period. The expression of OUC enzyme genes was detectable in the embryonic body from at least stage 28, and increased markedly during development to hatching, which is most probably due to growth of the liver. During the early developmental period, however, the expression of OUC enzyme genes was not prominent in the embryonic body. Meanwhile, we found that the mRNA expression of OUC enzymes was detected in the extra-embryonic YSM; the mRNA expression of cmcpsIII in the YSM was much higher than that in the embryonic body during stages 28-31. Significant levels of enzyme activity and the existence of mitochondrial-type cmgs1 transcripts in the YSM supported the mRNA findings. We also found that the cmcpsIII transcript is localized in the vascularized inner layer of the YSM. Taken together, our findings demonstrate for the first time that the YSM is involved in urea-based osmoregulation during the early to mid phase of development in oviparous cartilaginous fish.

The objective of this study was to evaluate animal performance and carcass characteristics of 64 Nellore young bulls at 22 months of age finished in a feedlot and slaughtered at five body weights (350; 455; 485; 555 and 580 kg) fed diets containing coated or uncoated urea. The experimental design adopted was completely randomized, set in a 4 × 2 factorial arrangement, and for the variables assessed in the control animals, it was 5 × 2. No effect of interaction between slaughter weights and diets were observed, so the variables were analyzed separately, compared by polynomial contrasts and by the F test, respectively. The time animals remained in the feedlot to reach slaughter weights was 66, 88, 145 and 194 days. Average daily gain (ADG) showed quadratic behavior, with a maximum of 1.44 kg/day with animals of 491.7 kg. Dry matter intake (DMI) (kg/day) was similar in all the treatments, but it decreased linearly as body weight increased. The bionutritional efficiency worsened linearly as body weight rose. The elevation in slaughter weight resulted in linear decrease in the percentage of beef round and increase in forequarter. Backfat thickness and rib eye area of the longissimus increased linearly and the percentages of muscle and protein in the carcass reduced and those of fat and ether extract increased linearly as body weight increased. Average daily gain, DMI, feed efficiency and carcass characteristics were not affected by diets containing coated or uncoated urea. However, animals fed coated urea presenter better crude fiber and neutral detergent fiber intake.

Many studies have demonstrated the relationship between dialysis dose and survival. Global mortality is similar among men and women; however, the influence of dialysis dose in the survival could be more intensive between women. Therefore, we conduct an observational study to evaluate the gender-related impact of single pool Kt/V (spKt/V) on the survival of patients submitted to hemodialysis in a university hospital. We found that survival was lower in groups with spKt/V smaller than 1.2 than in those with Kt/V between 1.2 and 1.4. Among female patients, spKt/V smaller than 1.2 had a more adverse effect in survival than among men with a comparable Kt/V. Otherwise, among women, the dialysis dose had an impact in survival even with Kt/V greater than 1.4. Thus, fractional urea clearance more heavily influenced the survival of females than males in hemodialysis patients.

Fibre, crude protein and tannin concentrations were measured in browse species from the semi-arid region of Northeast Brazil during the dry and wet seasons. The effects of oven-, sun- and shade-drying and of urea treatment were also determined. Crude protein (CP) content varied from 103 to 161 g/kg dry matter (DM) and the browses had similar CP content in the two seasons (during 2002) (102-161 and 107-153 g/kg DM in the wet and dry seasons, respectively). Total tannin concentrations ranged from 13 to 201 g/kg DM amongst the browses and were higher in the dry season. A 30-d treatment with urea reduced extractable tannins significantly (P < 0.05). The urea treatment was also most effective at reducing the in vitro effects of tannins compared to the other drying treatments. This was demonstrated by measuring the effect of polyethylene glycol (PEG) on gas production. Addition of PEG increased gas production of oven- (81.4%), sun- (78.5%) and shade-dried (76.7%) samples much more compared to urea treated samples (10.9%). (c) 2005 Elsevier B.V. The objective of the present study was to evaluate the nutrient intake, blood parameters, follicular diameter and performance of pre-puberty crossbred heifers fed isoproteic diets (14.1%CP) containing 0.0; 0.44; 0.88 and 1.32% urea on the total dry matter (DM) of the diet, with a 77:23 roughage:concentrate ratio. Twenty-four 18- month old heifers (Holstein x Zebu), 277.9 kg mean live weight (LW) were used, distributed in four treatments and six replications in a randomized complete design. The following were evaluated: dry matter intake (DM), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), ether extract (EE), hemicellulose (HEM), plasmatic ureic nitrogen (PUN), plasmatic glucose, plasmatic total cholesterol, follicular diameter and daily weight gain (DWG). No influence of the urea levels in the diet was observed on NDF and HEM intake. A maximum intake was obtained of DM (8.75 kg day(-1)), CP (0.88 kg day(-1)), ADF (2.5 kg day(-1)) and EE (0.17 kg day(-1)) respectively for the levels 0.7, 0.8, 0.7 and 0.7% urea in total DM. The 0.6%; 0.77% and 0.6% urea levels in diet were the critical points for obtaining maximum response for the PUN (10.96 mg dL(-1)) and plasmatic glucose (84.97 mg dL(-1)) concentrations and, for follicular diameter (11.08 mm) on the 40(th); 24(th) and 31(st) day, respectively. The plasmatic total cholesterol concentration and DWG were not influenced by the urea added to the diet, with averages of 119.39 mg dL(-1) and 1.66 kg day(-1), respectively. It was concluded that urea can be added up to 1.32% on the total DM of the diet for pre-puberty crossbred heifers.

The preparation of nanometer-sized structures of zinc oxide (ZnO) from zinc acetate and urea as raw materials was performed using conventional water bath heating and a microwave hydrothermal (MH) method in an aqueous solution. The oxide formation is controlled by decomposition of the added urea in the sealed autoclave. The influence of urea and the synthesis method on the final product formation are discussed. Broadband photoluminescence (PL) behavior in visible-range spectra was observed with a maximum peak centered in the green region which was attributed to different defects and the structural changes involved with ZnO crystals which were produced during the nucleation process.
