A presente dissertação descreve a síntese e o estudo de propriedades fotofísicas e biológicas de sistemas baseados em ftalocianinas. Os sistemas sintetizados consistem em ftalocianinas contendo unidades de D-galactose, ciclodextrinas, [60]fulereno ou porfirinas. Na primeira parte da dissertação aborda-se a síntese de ftalocianinas substituídas com unidades de D-galactose, com o intuito de serem utilizadas como fotossensibilizadores em terapia fotodinâmica. A eficiência de geração de oxigénio singuleto induzida pelas glicoftalocianinas, bem como a sua actividade fotodinâmica in vitro em células HeLa são avaliadas. Adicionalmente são descritos os estudos de MALDI-MS/MS de dois pares de glicoftalocianinas isoméricas. Numa segunda parte, descrevem-se a síntese e a caracterização de sistemas supramoleculares de ftalocianinas ligadas a ciclodextrinas ou [60]fulereno. Foram desenvolvidas metodologias sintéticas para obter complexos ftalocianínicos de ruténio(II) substituídos axialmente com derivados de ciclodextrinas, e complexos ftalocianínicos de zinco(II) ligados covalentemente a ciclodextrinas. Descreve-se também a síntese de uma díade ftalocianina- [60]fulereno. A estratégia sintética envolve um peculiar primeiro passo, uma reacção de aminação redutiva, no qual se obtém uma ftalocianina funcionalizada com glicina, seguida da habitual reacção de cicloadição 1,3- dipolar. Na terceira parte, abordam-se sistemas dador-aceitador de díades porfirinaftalocianina, como modelos promissores para mimetizar sistemas fotossintéticos naturais. Diferentes tipos de díades porfirina-ftalocianina, ligadas através do grupo fenilo da posição meso ou da posição β-pirrólica da porfirina, são sintetizadas e caracterizadas. As abordagens sintéticas envolvem reacções de aminação catalisadas por paládio e reacções de condensação cruzada de dois ftalonitrilos adequadamente substituídos. Também se descrevem processos de transferência de energia e electrónica fotoinduzidos que se formam a partir das díades porfirina-ftalocianina.

A number of synthetically useful ring systems can be prepared via the intramolecular insertion of a metal-stabilized carbenoid into a heteroaromatic systems. The chemical outcome of these reactions are dependent not only on the nature of the heteroatom but also on the length of the aliphatic tether linking the carbenoid moiety with the aromatic fragment. Our work with furanyl and thienyl systems containing a single methylene tether have allowed for some rather atypical chemistry. For example, treatment of l-diazo-3-(2-thienyl)-2-propanone (6) with catalytic rhodium (II) acetate yields 5,6- dihydro-4^-cyclopenta[Z>]thiophen-5-one (3) while, the isomeric l-diazo-3-(3-thienyl)-2- propanone(15) gives a spiro-disulphide (20). Novel chemistry was also exhibited in the analogous furanyl systems. While treatment of l-diazo-3-(3-furanyl)-2-propanone (52) with Rh2(OAc)4 resulted in the expected 2-(4-Oxo-2-cyclopentenyliden)acetaldehyde (54), isomeric l-diazo-3-(2- furanyl)-2-propanone (8) undergoes vinylogous Wolff rearrangement to give a mixture of 6a-methyl-2,3,3a,6a-tetrahydrofuro[2,i-^>]furan-2-one (44) and 2-(2-methyl-3-furyl)acetic acid (43). Rhodium acetate catalyzed decomposition of l-diazo-3-(3-benzofuranyl)-2- propanone (84) and l-diazo-3-(2-benzofuranyl)-2-propanone (69)also allows for vinylogous Wolff rearrangement, a chemistry unseen in benzofuranyl systems with longer tethers. A number of interesting products were isolated from the trapping of intermediate ketenes. Decomposition of l-diazo-3-(3-benzothienyl)-2-propanone (100) resulted in the formation of 2,3-dihydro-l//-benzo[^]cyclopenta[^thiophen-2-one (102). However, in addition to (102), a dimer was also generated from the decomposition of l-diazo-3-(2- benzothienyl)-2-propanone (109). The insight into the mechanistic underpinnings of the above reactions are provided by molecular modeling at a PM3 level.

Recent studies have shown that the rhodium (II) acetate decomposition chemistry observed for a-diazoketones tethered to thienyl, furanyl, and benzofuranyl moieties is dependent not only on the nature of the heteroatom but also on the length of the aliphatic tether linking the diazoketone moiety with the aromatic fragment. The present thesis expands on these results and focuses on a-diazoketones tethered to benzothiophenes, pyrroles and indoles by a methylene linker. In the case of benzothiophenes, it was shown that the rhodium catalyst decomposition of I-diazo-4-(3-benzothienyl)-2-butanone (146) and 1-diazo-4-(3benzothienyl)- 2-butanone (152) allow for the isolation of 1,2,3a,3b-tetrahydro-3Hbenzo[ b]cyclopenta[1,3]cyclopropa- [1 ,2-d]thiophen-3-one (147) and 1,2,3a,3btetrahydro- 3H-benzo[b]cyclopenta[1,3]cyclopropa[1,2-d]thiophen-3-one (153). However treatment of 1-diazo-3-(3-Benzothienyl)-2-Propanone (165) with Rh(II) acetate results in the formation of 2,3-Dihydro-1H-benzo[b]cyclopenta[d]thiophen-2-one (159), while 1diazo- 3-(2-Benzothienyl)-2-Propanone with the same condition gives 5,5-bis( 1benzothiophen- 2-ylmethyl)-2(5H)-furanone (166) along with the tricycle 159. The chemistry of the pyrrolyl and the indolyl moieties linked to terminal adiazoketone systems was also investigated. The decomposition of I-diazo-(2-pyrrolyl)-2propanone (173) results in the formation of two products; the N-H insertion product IHpyrrolizin- 2(3H)-one (176) and the alkylation product 4,6-dihydrocyclopenta[b]pyrrol5( 1 H)-one (180). When 1-Diazo-3-(3-indoly)-3-propanone (194) is treated with catalytic amount of Rh (II) 3,4-dihydrocyclopenta[b]indol-2(1H)-one (193) is isolated quantitatively. The later reaction when monitored using IH NMR the intermediate 200 can be seen whose structure was confirmed by the comparison to series of model compounds. The mechanisms underlying these reactions as well as their synthetic utility is discussed.

Ce mémoire présente trois approches différentes vers la synthèse du 3–(trans–2–nitrocyclopropyl)alanine, un intermédiaire synthétique de la hormaomycine. Cette molécule naturelle démontre d’intéressantes activités biologiques et pharmacologiques. Il est intéressant de souligner que ce dérivé donne facilement accès au 3–(trans–2–aminocyclopropyl)alanine, unité centrale de la bélactosine A. Ce composé naturel possédant lui aussi d’intéressantes propriétés biologiques, plusieurs études relationnelles structures-activités menant à des dérivés plus actifs de cette molécule ont été entreprises, démontrant l’intérêt toujours présent de synthétiser de façon efficace et optimale ces dérivés cyclopropaniques. Une méthodologie développée au sein de notre groupe de recherche et basée sur une réaction de cyclopropanation intramoléculaire diastéréosélective sera mise à profit afin d’élaborer une nouvelle voie de synthèse aussi élégante qu’efficace à la construction du 3–(trans–2–nitrocyclopropyl) alanine. En utilisant un carbène de rhodium généré soit par la dégradation d’un dérivé diazoïque, soit par la formation d’un réactif de type ylure d’iodonium, une réaction de cyclopropanation diastéréosélective permettra la formation de deux autres centres contigus et ce, sans même utiliser d’auxiliaire ou de catalyseur énantioenrichis. Ensuite, un réarrangement intramoléculaire précédant deux réactions synchronisées d’ouverture de cycle et de décarboxylation permettront l’obtention du composé d’intérêt avec un rendement global convenable et en relativement peu d’étapes. De cette manière, la synthèse formelle de la bélactosine A et de l’hormaomycine a été effectuée. Cette synthèse se démarque des autres par l’utilisation d’une seule transformation catalytique énantiosélective.

Les dérivés cyclopropaniques di-accepteurs représentent des intermédiaires synthétiques précieux dans l’élaboration de structures moléculaires complexes, ayant des applications dans plusieurs domaines de la chimie. Au cours de cet ouvrage, nous nous sommes intéressés à la synthèse de ces unités sous forme énantioenrichie en utilisant la cyclopropanation d’alcènes par catalyse avec des complexes de Rh(II) utilisant des composés diazoïques di-accepteurs comme substrats. Suite au développement initial d’une méthode de cyclopropanation d’alcènes catalytique asymétrique utilisant des nitro diazocétones, de multiples études expérimentales quant au mécanisme de stéréoinduction dans ce type de réaction ont été effectuées. Nous avons alors pu identifier le groupement p-méthoxyphénylcétone du substrat et le catalyseur Rh2(S-TCPTTL)4 comme étant une combinaison clé pour l’atteinte de diastéréosélectivités et d’excès énantiomères élevés. Ceci a mené au développement de deux autres méthodes de cyclopropanation stéréosélectives distinctes, utilisant soit une cyano diazocétone ou un céto diazoester. Nous avons démontré l’utilité des dérivés cyclopropaniques énantioenrichis obtenus par ces trois méthodes dans une panoplie de manipulations synthétiques, dont l’addition nucléophile d’amines et de cuprates, la cycloaddition formelle avec un aldéhyde, et la synthèse de dérivés cyclopropaniques importants en chimie médicinale. Une étude structurelle approfondie des complexes de Rh(II) chiraux nous a permis de déterminer les facteurs responsables de leur pouvoir d’énantioinduction dans notre système réactionnel, ce qui a d’énormes implications dans d’autres méthodologies utilisant ces mêmes catalyseurs. Le dévoilement d’une conformation inattendue dite ‘All-up’, ainsi que de la présence d’interactions stabilisantes régissant la rigidité de cet arrangement se sont avérés cruciaux dans notre compréhension du mécanisme. Dans le cadre de cette investigation, nous avons développé une méthode générale pour la synthèse de complexes de Rh(II) hétéroleptiques, multipliant ainsi le nombre de catalyseurs accessibles dans l’élaboration éventuelle de nouvelles réactions stéréosélectives, et nous permettant d’effectuer une étude structurelle plus détaillée. De plus, nous avons développé une méthode particulièrement efficace pour la synthèse d’un autre type de dérivé cyclopropanique di-accepteur par catalyse avec des complexes de Rh(II), les cyano-cyclopropylphosphonates. Les produits de cette transformation sont obtenus avec des énantiosélectivités élevées, et sont des substrats intéressants pour des réactions tandem d’ouverture de cycle par addition nucléophile / oléfination de composés carbonylés. De plus, ces composés sont des précurseurs de molécules utiles en chimie médicinale tels que les acides aminocyclopropylphosphoniques.

Absorption spectral measurements of H2Pc, MnPc, NiPc, CoPc, CuPc, MoOPc, ZnPc and FePc doped borate glass matrix have been made in the 200–1100 nm region and the spectra obtained are analyzed in the 2.1–6.2 eV region to obtain the optical band gap (Eg) and the width of the band tail (Et). Other important optical and physical parameters viz. refractive index (n), molar extinction coefficient ("), density (½), glass transition temperature (Tg), molecular concentration (N ), polaron radius (rp), intermolecular separation (R), molar refractivity (Rm) are also reported

Optical absorption and emission spectral studies of free and metal naphthalocyanine doped borate glass matrix are reported for the first time. Absorption spectra recorded in the UV- VIS-NIR region show the characteristic absorption bands, namely, the B-band and Q-band of the naphthalocyanine (Nc) molecule. Some of the important spectral parameters, namely, the optical absorption coefficient (α), molar extinction coefficient (ε) and absorption cross section (σa) of the principal absorption transitions are determined. Optical band gap (Eg) of the materials evaluated from the functional dependence of absorption coefficient on photon energy lies in the range 1.6 eV≤Eg≤2.1 eV. All fluorescence spectra except that of EuNc consist of an intense band in the 765 nm region corresponding to the excitation of Q-band. In EuNc the maximum fluorescence intensity band is observed at 824 nm. The intensity of the principal fluorescence band is maximum in ZnNc, whereas it is minimum in H2Nc. Radiative parameters of the principal fluorescence transitions corresponding to the Q-band excitation are also reported for the naphthalocyanine and phthalocyanine based matrices.

Optical absorption and emission spectral studies of various phthalocyanine (Pc) molecules in PVA matrix have been reported for the first time. The recorded spectra are analyzed to get the important spectral parameters, such as optical absorption cross-section (σa), emission cross-section (σe), oscillator strength (f), fluorescence bandwidth (Δλ), emission wavelength (λ), radiative decay time (τ) and optical gain (G). Analysis shows that the emission cross-section and optical gain are maximum in the NdHPc2-doped PVA matrix. However, a comparison of the calculated emission parameters with that of borate glass matrix show that they are many times smaller in the present matrix.

Optical absorption studies of free base and rare earth incorporated phthalocyanine doped borate glass matrix are reported for the first lime. The absorption spectra recorded in the UV- VIS region show two well defined absorption bands of phthalocyanine (Pc) molecule, namely the Soret band (B) and the Q band. The Q band always shows its characteristic splitting in all the doped glass matrices and the intensities of these components are found to vary from one Pc to another. Some of the important optical parameters, namely optical absorption coefficient (a), molar extinction coefficient (ε), absorption cross section (σa), oscillator strength (f), electric dipole strength (q2), absorption half bandwidth (Δλ) of the principal optical transitions have also been evaluated. Moreover, the spectral dependence of refractive index (n) and thereby the optical dielectric constant (ε) on wavelength yielded values of carrier concentration to effective mass ratio (N/m*) of the phthalocyanine molecule in the present glassy systems. Optical band gap (Eg) and width of the band tail (Et) are computed and their variations among the prepared samples are also discussed.

Optical absorption and emission spectral studies of various phthalocyanine molecules, viz., LaPc, NdPc, SmPc, EuPc, CuPc and ZnPc in a polymer matrix of cyano acrylate are reported for the first time. All the absorption spectra show an intense B band (Soret) in the UV region followed by a weaker Q band in the visible region. The positions of the Q and B bands are found to have dependence on the metallic substitution. Values of the important spectral parameters, viz., molar extinction coefficient (ϵ), oscillator strength (f), radiative transition rate and decay time of the excited singlet state are also presented and compared with other solid matrices. The recorded fluorescence spectrum shows two broad emission bands in the case of NdPc, whereas for ZnPc only a very weak band is observed. The absence of emission bands for the other metallated phthalocyanines is attributed to increased spin orbit interaction and intersystem crossing.

Photoacoustic spectrum of samarium phthalocyanine powder is recorded and compared with previously reported UV–vis absorption spectra of the same dissolved in different liquid and solid host media. The Davidov splitting of Q band is observed in the PA spectrum but the two bands are overlapped considerably and the shorter wavelength band is more intense and dominating one in the powder spectrum.

Wavelength dependence of saturable absorption (SA) and reverse saturable absorption (RSA) of zinc phthalocyanine was studied using 10 Hz, 8 ns pulses from a tunable laser, in the wavelength range of 520–686 nm, which includes the rising edge of the Q band in the electronic absorption spectrum. The nonlinear response is wavelength dependent and switching from RSA to SA has been observed as the excitation wavelength changes from the low absorption window region to higher absorption regime near the Q band. The SA again changes back to RSA when we further move over to the infrared region. Values of the imaginary part of third order susceptibility are calculated for various wavelengths in this range. This study is important in identifying the spectral range over which the nonlinear material acts as RSA based optical limiter.
