Background: Eicosanoids are biologically active, oxygenated metabolites of three C20 polyunsaturated fatty acids. They act as signalling molecules within the autocrine or paracrine system in both vertebrates and invertebrates mainly functioning as important mediators in reproduction, the immune system and ion transport. The biosynthesis of eicosanoids has been intensively studied in mammals and it is known that they are synthesised from the fatty acid, arachidonic acid, through either the cyclooxygenase (COX) pathway; the lipoxygenase (LOX) pathway; or the cytochrome P450 epoxygenase pathway. However, little is still known about the synthesis and structure of the pathway in invertebrates. Results: Here, we show transcriptomic evidence from Daphnia magna (Crustacea: Branchiopoda) together with a bioinformatic analysis of the D. pulex genome providing insight on the role of eicosanoids in these crustaceans as well as outlining a putative pathway of eicosanoid biosynthesis. Daphnia appear only to have one copy of the gene encoding the key enzyme COX, and phylogenetic analysis reveals that the predicted protein sequence of Daphnia COX clusters with other invertebrates. There is no current evidence of an epoxygenase pathway in Daphnia; however, LOX products are most certainly synthesised in daphnids. Conclusion: We have outlined the structure of eicosanoid biosynthesis in Daphnia, a key genus in freshwater ecosystems. Improved knowledge of the function and synthesis of eicosanoids in Daphnia and other invertebrates could have important implications for several areas within ecology. This provisional overview of daphnid eicosanoid biosynthesis provides a guide on where to focus future research activities in this area.

In Drosophila, telomere retrotransposons counterbalance the loss of telomeric DNA. The exceptional mechanism of telomere recovery characterized in Drosophila has not been found in lower dipterans (Nematocera). However, a retroelement resembling a telomere transposon and termed ""RaTART"" has been described in the nematoceran Rhynchosciara americana. In this work, DNA and protein sequence analyses, DNA cloning, and chromosomal localization of probes obtained either by PCR or by screening a genomic library were carried out in order to examine additional features of this retroelement. The analyses performed raise the possibility that RaTART represents a genomic clone composed of distinct repetitive elements, one of which is likely to be responsible for its apparent enrichment at chromosome ends. RaTART sequence in addition allowed to assess a novel subtelomeric region of R. americana chromosomes that was analyzed in this work after subcloning a DNA fragment from a phage insert. It contains a complex repeat that is located in the vicinity of simple and complex tandem repeats characterized previously. Quantification data suggest that the copy number of the repeat is significantly lower than that observed for the ribosomal DNA in the salivary gland of R. americana. A short insertion of the RaTART was identified in the cloned segment, which hybridized preferentially to subtelomeres. Like RaTART, it displays truncated sequences related to distinct retrotransposons, one of which has a conceptual translation product with significant identity with an endonuclease from a lepidopteran retrotransposon. The composite structure of this DNA stretch probably reflects mobile element activity in the subtelomeric region analyzed in this work.

Uma significativa quantidade de proteínas vegetais apresenta-se compartimentalizada nas diversas estruturas celulares. A sua localização pode conduzir à elucidação do funcionamento dos processos biossintéticos e catabólicos e auxiliar na identificação de genes importantes. A fim de localizar produtos gênicos relacionados à resistência, foi utilizada a fusão de cDNAs de arroz (Oryza sativa L.) ao gene da proteína verde fluorescente (GFP). Os cDNAs foram obtidos a partir de uma biblioteca supressiva subtrativa de genes de arroz durante uma interação incompatível com o fungo Magnaporthe grisea. Estes cDNAs foram fusionados a uma versão intensificada de gfp e usados para transformar 500 plantas de Arabidopsis thaliana. Outras 50 plantas foram transformadas com o mesmo vetor, porém sem a fusão (vetor vazio). Foram obtidas aproximadamente 25.500 sementes oriundas das plantas transformadas com as fusões EGFP::cDNAs e 35.000 sementes das transformadas com o vetor vazio, produzindo, respectivamente, 750 e 800 plantas tolerantes ao herbicida glufosinato de amônio. Após a seleção, segmentos foliares das plantas foram analisados por microscopia de fluorescência, visando o estabelecimento do padrão de localização de EGFP. Foram observadas 18 plantas transformadas com a fusão EGFP::cDNAs e 16 plantas transformadas com o vetor vazio apresentando expressão detectável de GFP. Uma planta transformada com uma fusão EGFP::cDNA apresentou localização diferenciada da fluorescência, notadamente nas células guarda dos estômatos e nos tricomas. Após seqüenciamento do cDNA fusionado, foi verificado que esta planta apresentava uma inserção similar a uma seqüência codificante de uma quinase, uma classe de enzimas envolvidas na transdução de sinais em resposta à infecção por patógenos.

The first reference map of the proteome of pooled normal dog tears was created using 2-dimensional polyacrylamide gel electrophoresis and the identity of a number of the major species determined using matrix-assisted laser desorption time of flight mass spectrometry (MALDI-TOF) and peptide mass fingerprint matching on protein sequence databases. In order to understand the changes in protein expression in the tear film of dogs with cancer, tears from such animals were similarly examined. A number of differences were found between the tears of healthy dogs and the dogs with cancer. Differences were found in levels of actin and albumin and in an unidentified protein which may be analogous to human lacryglobulin. These findings suggest that it may be possible to develop tear film analysis to provide a simple non-invasive test for the diagnosis and/or management of canine cancers. (C) 2007 Elsevier Ltd. Envenomation by arachnids of the genus Loxosceles leads to local dermonecrosis and serious systemic toxicity mainly induced by sphingomyelinases D (SMase D). These enzymes catalyze the hydrolysis of sphingomyelin resulting in the formation of ceramide-phosphate and choline as well as the cleavage of lysophosphatidyl choline generating the lipid mediator lysophosphatidic acid. We have, previously, cloned and expressed two functional SMase D isoforms, named P1 and P2, from Loxosceles intertnedia venom and comparative protein sequence analysis revealed that they are highly homologous to SMase I from Loxosceles laeta which folds to form an (alpha/beta)(8) barrel. In order to further characterize these proteins, pH dependence kinetic experiments and chemical modification of the two active SMases D isoforms were performed. We show here that the amino acids involved in catalysis and in the metal ion binding sites are strictly conserved in the SMase D isoforms from L. intermedia. However, the kinetic studies indicate that SMase P1 hydrolyzes sphingomyelin less efficiently than P2, which can be attributed to a substitution at position 203 (Pro-Leu) and local amino acid substitutions in the hydrophobic channel that could probably play a role in the substrate recognition and binding. The unique carbohydrate-binding property of lectins makes them invaluable tools in biomedical research. Here, we report the purification, partial primary structure, carbohydrate affinity characterization, crystallization, and preliminary X-ray diffraction analysis of a lactose-specific lectin from Cymbosema roseum seeds (CRLII). Isolation and purification of CRLII was performed by a single step using a Sepharose-4B-lactose affinity chromatography column. The carbohydrate affinity characterization was carried using assays for hemagglutination activity and inhibition. CRLII showed hemagglutinating activity toward rabbit erythrocytes. O-glycoproteins from mucine mucopolysaccharides showed the most potent inhibition capacity at a minimum concentration of 1.2 A mu g mL(-1). Protein sequencing by mass spectrometry was obtained by the digestion of CRLII with trypsin, Glu-C, and AspN. CRLII partial protein sequence exhibits 46% similarity with the ConA-like alpha chain precursor. Suitable protein crystals were obtained with the hanging-drop vapor-diffusion method with 8% ethylene glycol, 0.1 M Tris-HCl pH 8.5, and 11% PEG 8,000. In order to identify genes expressed in the pistil that may have a role in the reproduction process, we have established an expressed sequence tags project to randomly sequence clones from a Nicotiana tabacum stigma/style cDNA library. A cDNA clone (MTL-8) showing high sequence similarity to genes encoding glycine-rich RNA-binding proteins was chosen for further characterization. Based on the extensive identity of MTL-8 to the RGP-1a sequence of N. sylvestris, a primer was defined to extend the 5′ sequence of MTL-8 by RT-PCR from stigma/style RNAs. The amplification product was sequenced and it was confirmed that MTL-8 corresponds to an mRNA encoding a glycine-rich RNA-binding protein. Two transcripts of different sizes and expression patterns were identified when the MTL-8 cDNA insert was used as a probe in RNA blots. The largest is 1,100 nucleotides (nt) long and markedly predominant in ovaries. The smaller transcript, with 600 nt, is ubiquitous to the vegetative and reproductive organs analyzed (roots, stems, leaves, sepals, petals, stamens, stigmas/styles and ovaries). Plants submitted to stress (wounding, virus infection and ethylene treatment) presented an increased level of the 600-nt transcript in leaves, especially after tobacco necrosis virus infection. In contrast, the level of the 1,100-nt transcript seems to be unaffected by the stress conditions tested. Results of Southern blot experiments have suggested that MTL-8 is present in one or two copies in the tobacco genome. Our results suggest that the shorter transcript is related to stress while the larger one is a flower predominant and nonstress-inducible messenger.

Aborda a classificação automática de faltas do tipo curto-circuito em linhas de transmissão. A maioria dos sistemas de transmissão possuem três fases (A, B e C). Por exemplo, um curto-circuito entre as fases A e B pode ser identicado como uma falta\AB". Considerando a possibilidade de um curto-circuito com a fase terra (T), a tarefa ao longo desse trabalho de classificar uma série temporal em uma das 11 faltas possíveis: AT, BT, CT, AB, AC, BC, ABC, ABT, ACT, BCT, ABCT. Estas faltas são responsáveis pela maioria dos distúrbios no sistema elétrico. Cada curto-circuito é representado por uma seqüência (série temporal) e ambos os tipos de classificação, on-line (para cada curto segmento extraído do sinal) e off-line (leva em consideração toda a seqüência), são investigados. Para evitar a atual falta de dados rotulados, o simulador Alternative Transient Program (ATP) é usado para criar uma base de dados rotulada e disponibilizada em domínio público. Alguns trabalhos na literatura não fazem distinção entre as faltas ABC e ABCT. Assim, resultados distinguindo esse dois tipos de faltas adotando técnicas de pré-processamento, diferentes front ends (por exemplo wavelets) e algoritmos de aprendizado (árvores de decisão e redes neurais) são apresentados. O custo computacional estimado durante o estágio de teste de alguns classificadores é investigado e a escolha dos parâmetros dos classificadores é feita a partir de uma seleção automática de modelo. Os resultados obtidos indicam que as árvores de decisão e as redes neurais apresentam melhores resultados quando comparados aos outros classificadores.
