Among the goals of the Brazilian soybean improvement programmes, the breeding strategies for cultivars adapted to low latitudes have been included to extend crop areas and to increase production. Seeds of nine Brazilian soybean cultivars adapted to low latitudes were investigated regarding to their composition, and amino acid and antinutritional/toxic protein contents. Protein (394.5 ± 13.1 to 445.3 ± 8.0 g kg-1 dry matter) and oil (200.6 ± 1.2 to 232.3 ± 4.7 g kg-1 dry matter) contents showed low correlation to each other (r = -0.06). The total carbohydrate (141.7 ± 6.1 to 211.1 ± 15.0 g kg-1 dry matter) and ash contents (48.2 ± 4.2 to 52.2 ± 0.5 g kg-1 dry matter) were similar to data available for other soybean cultivars. All soybean cultivars presented low levels of tryptophan and sulphur amino acids. The lectin (1,152 to 147,456 HU kg-1 flour), trypsin inhibitor (34.45 ± 2.28 to 77.62 ± 2.63 g trypsin inhibited kg-1 flour), toxin (6,210 ± 134 to 34,650 ± 110 LD50 kg-1 flour) and urease (0.74 ± 0.02 to 1.22 ± 0.10 g kg¹ flour) presented variations in their contents amongst the cultivars. Compared to other soybean cultivars, urease was higher, the acute toxicity lower and the lectin and trypsin inhibitor contents similar to data available. In general, soybean cultivars showed similar biochemical composition to those developed in different geographic regions. The relevance of these findings to the agronomic features and to choice of soybean cultivars to be used as food or feed is discussed.

This study examined if leucine, arginine or glycine supplementation in adult obese patients (body mass index of 33 ± 4 kg/m²) consuming a Brazilian low energy and protein diet (4.2 MJ/day and 0.6 g protein/kg) affects protein and amino acid metabolism. After four weeks adaptation to this diet, each subject received supplements of these amino acids (equivalent to 0.2 g protein kg-1 day-1) in random order. On the seventh day of each amino acid supplementation, a single-dose 15N-glycine study was carried out. There were no significant differences in protein flux, synthesis or breakdown. The protein flux (grams of nitrogen, gN/9 h) was 55 ± 24 during the nonsupplemented diet intake and 39 ± 10, 44 ± 22 and 58 ± 35 during the leucine-, glycine- and arginine-supplemented diet intake, respectively; protein synthesis (gN/9 h) was 57 ± 24, 36 ± 10, 41 ± 22 and 56 ± 36, respectively; protein breakdown (gN/9 h) was 51 ± 24, 34 ± 10, 32 ± 28 and 53 ± 35, respectively; kinetic balance (gN/9 h) was 3.2 ± 1.8, 4.1 ± 1.7, 3.4 ± 2.9 and 3.9 ± 1.6. There was no difference in amino acid profiles due to leucine, arginine or glycine supplementation. The present results suggest that 0.6 g/kg of dietary protein is enough to maintain protein turnover in obese women consuming a reduced energy diet and that leucine, arginine or glycine supplementation does not change kinetic balance or protein synthesis.

O trabalho teve como objetivos determinar a atividade inibitória de tripsina em produtos comerciais de soja consumidos pela população, tais como bebidas à base de extrato de soja, e fómulas não-lácteas à base de proteína isolada de soja, para os intolerantes ao leite. Os resultados mostraram que, excetuando-se os produtos à base de extrato de soja, os demais apresentam valores baixos de atividade inibitória de tripsina, em torno de 5% do valor encontrado para a farinha desengordurada. Além disso, através de eletroforese em gel de poliacrilamida com revelação para inibidores de tripsina, demonstrou-se que em alguns casos a inibição observada através do método colorimétrico de KAKADE, SIMONS & LIENER [8] se deve provavelmente à inibição não específica provocada por outros componentes da amostra.

Foi estudada a viabilidade de consumo da cultivar de soja IAC PL-1 enlatada como grão verde e sua melhor época de colheita para enlatamento. Para tanto procederam-se cinco colheitas a partir do 48º dia após a floração (DAF) até a extinção da coloração verde. O processamento iniciou-se pelo branqueamento das vagens, debulha e enlatamento. Aos cinco lotes obtidos adicionou-se um sexto lote para estudar o efeito do armazenamento. Para qualificar os seis lotes de grãos enlatados foram efetuadas medidas de peso, cor, textura e características do líquido de enlatamento. Foram realizados estudos das propriedades sensoriais dos grãos enlatados para dimensionar a aceitação pela degustação e aparência. O enlatamento foi otimizado para 121ºC com tempo de esterilização em torno de 4 minutos. A maturidade fisiológica dos grãos ocorreu entre o 61º ao 64º DAF. O processo térmico conservou a cor verde dos grãos enlatados e não induziu perda expressiva da massa dos grãos; a textura firme dos grãos aumentou com a maturação. Os provadores demonstraram boa aceitação dos produtos e não ocorreu preferência em relação aos estádios de maturação. O trabalho conclui que grãos verdes de soja IAC PL-1 proporcionam enlatados com boas características técnicas e gustativas. Foi observado que grãos colhidos no ponto convencional de maturação, armazenados e enlatados obtiveram boa aceitação gustativa, concluindo-se que a cultivar IAC PL-1 é também adequada para o consumo e enlatada após o armazenamento.

Fibras alimentares obtidas de cotilédones de soja original (FAO) e micronizada (FAM) foram fracionadas e avaliadas quanto a polissacarídeos e monossacarídeos constituintes. O componente majoritário foi a hemicelulose, totalizando 59% em FAO e 51% na FAM, a pectina representou em média 14% e a celulose 8,5%. As duas amostras de fibras apresentaram 17 mg.g-1 de ácidos urônicos e a mesma composição de monossacarídeos, sendo galactose, glicose e arabinose/ramnose os principais componentes. As proteínas de concentrado protéico, farinha desengordurada e fibras alimentares (FAO e FAM) foram avaliadas quanto à solubilidade em diferentes solventes (NaCl, água, etanol e NaOH) e quanto ao peso molecular. A farinha desengordurada de soja teve a maior parte das proteínas passível de extração com solução salina, e o concentrado protéico e as fibras de cotilédones com solução alcalina. A fração protéica que não foi extraída com nenhum dos quatro solventes utilizados permaneceu no resíduo, o maior percentual estava no concentrado, seguido pela fibra alimentar micronizada e a farinha, já a menor quantidade estava na fibra alimentar original. A eletroforese das proteínas dos quatro ingredientes alimentares mostrou as subunidades que constituem as frações β-conglicinina e glicinina. Bandas com peso molecular próximo aos 30 kDa foram reveladas nas proteínas extraídas das fibras de cotilédones de soja, sendo provavelmente glicoproteínas de parede celular, ricas em hidroxiprolina.

A soja tem importância para a saúde e seu consumo tem aumentado devido principalmente aos diversos produtos presentes no mercado. Prevenindo a ação das enzimas lipoxigenases há melhora na aceitação dos produtos derivados da soja. Vários processos tecnológicos podem evitar a ação da enzima lipoxigenase, a qual pode causar atributos sensoriais que causam rejeição no consumo de grãos de soja. A pesquisa teve por objetivo verificar a aceitabilidade dos grãos de soja tostados por diferentes tratamentos. Em grãos de soja previamente macerados, com as cascas retiradas, foram realizados cinco tratamentos: T1 e T2 - com secagem prévia + 10 e 15 minutos de forno, respectivamente; T3, T4 e T5 - sem secagem prévia + 45, 50 e 60 minutos de forno. O T6, controle, recebeu o mesmo tratamento, foi seco em estufa (55 °C) durante 31 horas. O comportamento dos provadores em relação ao consumo da soja não condiz com o conhecimento da importância para a saúde e a facilidade de aquisição do hábito de consumo, não havendo diferença para idade ou sexo. O tratamento que promoveu a maior aceitabilidade dos grãos de soja tostados foi o que recebeu o tempo intermediário de 50 minutos, sem secagem prévia.

Physicochemical characteristics and functional properties of vitabosa flour (Mucuna deeringiana) and soybean flour (Glycine max) were determined. Oil absorption capacity was higher in vitabosa. Water absorption capacity was higher in soy and it was affected by the change in the ionic strength of the medium. Emulsifying Activity (EA) decreased with increasing concentration of flour, while Emulsifying Stability (ES) showed an increased. EA and ES of flours have more ionic strength in the range between 0.0 and 0.4 M, but it is reduced afterwards with the higher concentration of NaCl. Foaming stability varied with the concentration of flour solution reaching maximum values of 39 and 33% for vitabosa and soybean, respectively at 10% flour concentration.Vitabosa had the best foaming capacity (56% to 0.6 M) compared with soybeans (47% to 0.4 M). Maximum capacity of gelation was observed in vitabosa at 10% flour concentration. Increases in ionic strength of the flour solution, at low salt concentrations (<0.4 M), improved the gelation of flours.

The reaction of tris(pentafluorophenyl)phosphine [5] with the nucleophiles dimethyl formamide (DMF), hexamethylphosphoric triamide (HMPA), diethyl formamide (DEF), hexaethylphosphoric triamide (HEPA), hydrazine, N,N-dimethyl hydrazine (in presence and/or absence of KF), phenylhydrazine, ammonium hydroxide, formamide, aniline, sodium hydrogen sulfide, and hexaethylphosphorous triamide was investigated. The reaction of [5] with DMF and HMPA gave the same product, namely tris-[4-(N,N-dimethylamino)-2,3,5,6-tetrafluorophenyl]phosphine [12] but in higher yield in the case of HMPA. Compound (5] also reacted with DEF to give tris[4-(N,N-diethylamino)-2,3,5,6-tetrafluorophenyl] phosphine [14]. When [51 was treated with HEPA, it gave a mixture of bis(pentafluorophe~yl)-(N,N-diethylamino-tetrafluorophenyl)phosphine, pentafluorophenyl-bis-(N,N-diethylamino-tetrafluorophenyl)phosphine and tris (N,N-diethylamino-tetrafluorophenyl)phosphine. Treatment of [5] with aqueeus hydrazine solution in excess ethanol gave tris(4-hydrazo-2,3,4,6-tetrafluorophenyl)phosphine [1s1 in high yield while reaction with aqueous hydrazine led to C-P cleavage and production of tetrafluorophenyl hydrazine. With N,N-dimethyl hydrazine, [5] gave tris(4-N,N-dimethylhydrazine-2,3,5,6-tetrafluorophenyl) phosphine {20j. The latter could be obtained in higher yield and shorter reaction time, by the addition of KF. The reaction of compound {51 with phenylhydrazine in THF gave bis(pentafluorophe~yl)-4-S-phenylhydrazino- 2,3,5,6-tetrafluorophenyl phosphine [22] in low yield. Reaction of [5] with ammonium hydroxide in THF at high pressure in the presence of KF gave tris-~4-amino-2,3,5,6-tetrafluorophenyl)phosphine [25]. Similarly, formamide led to a mixture of (C6F4NHZ)3P, (C6F4NHZ)ZPC6FS, (C6F4NHZ)ZPC6F4NHCHO, and C6F4NHZP(C6Fs)(C6F4NHCHO). When [5] was treated with aniline, a mixture of mono-, di-, and tri-substituted products was obtained. Sodium hydrogen sulfide in ethylene glycol/ pyridine led to C-P cleavage and the isolation of pentafluorobenzene and tetrafluorothiophenol. Reaction of [5] and its oxide [35] with different alkoxides in the corresponding alcohols led mainly to C-P bond cleavage products, with the exception of one case where sodium methoxide was used in ether, and which led to tris-(4-methoxy-2,3,9,6-tetrafluorophenyl)phosphine [37]. On the basis of various spectroscopic data, it was concluded that the para position in compound [5] was generally the favoured site of attack.

Soybean (Glycine ~ (L.) Merr. cv. Harosoy 63) plants inoculated with Rhizobium japonicum were grown in vermiculite in the presence or absence of nitrate fertilization for up to 6 weeks after planting. Overall growth of nodulated plants was enhanced in the presence of nitrate fertilization, while the extent of nodule development was reduced. Although the number of nodules was not affected by nitrate fertilization when plants were grown at a light intensity limiting for photosynthesis, at light intensities approaching or exceeding the light saturation point for photosynthesis, nitrate fertilization resulted in at least a 30% reduction in nodule numbers. The mature, first trifoliate leaf of 21 day old plants was allowed to photoassimi1ate 14C02. One hour after·· the initial exposure to 14C02, the , plants were harvested and the 14C radioactivity was determined in the 80% ethanol-soluble fraction: in. o:rider to assess· "the extent of photoassimilate export and the pattern of distribution of exported 14C. The magnitude of 14C export was not affected by the presence of nitrate fertilization. However, there was a significant effect on the distribution pattern, particularly with regard to the partitioning of 14C-photosynthate between the nodules and the root tissue. In the presence of nitrate fertilization, less than 6% of the exported 14C photosynthate was recovered from the nodules, with much larger amounts (approximately 37%) being recovered from the root tissue. In the absence of nitrate fertilization, recovery of exported 14C-photosynthate from the nodules (19 to 27%) was approximately equal to that from the root tissue (24 to 33%). By initiating- or terminating the applications of nitrate at 14 days of age, it was determined that the period from day 14 to day 21 after planting was particularly significant for the development of nodules initiated earlier. Addition of nitrate fertilization at this time inhibited further nodule development while stimulating plant growth, whereas removal of nitrate fertilization stimulated nodule development. The results obtained are consistent with the hypothesis that nodule development is inhibited by nitrate fertilization through a reduction in the availability of photosynthate to the nodules.

Les vertébrés, du poisson à l'homme, possèdent un potentiel membranaire médié en partie par les ions chlorure (Cl-). L’une des premières formes d’activité neuronale lors du développement est la dépolarisation médiée par les ions chlorures extrudés par les canaux glycinergiques (GlyR) et GABAergiques. Cette dépolarisation est rendu possible grâce à l’expression retardée du co-transporteur d’ions chlorure et de potassium KCC2 lors du développement qui génère un gradient hyperpolarisant postnatalement chez les mammifères. Le rôle de cette dépolarisation précoce paradoxale durant le développement est inconnu. En injectant l’ARNm de KCC2 dans des embryons de poissons zébrés nouvellement fertilisé, nous avons devancé l’expression de ce co-transporteur rendant ainsi la glycine hyperpolarisante dans tous les neurones dès les premières phases du développement. Nous avons aussi ciblé le récepteur glycinergique directement en bloquant son activité et son expression à l’aide d’une drogue spécifique, la strychnine et d’un morpholino antisens (Knockdown). Dans les trois cas (KCC2, strychnine et GlyR KD), les perturbations de l’activité neuronale ont provoqués des erreurs dans la neurogenèse, en particulier une diminution du nombre d’interneurones sans avoir d’effets sur les motoneurones et les neurones sensoriels. De plus, en bloquant les canaux calciques activés à bas voltage dans le développement avec la drogue nifedipine, il y a des erreurs dans la neurogénèse semblables à celles remarquées dans les trois conditions précédentes. Nous concluons que la dépolarisation précoce par la glycine permet l’entrée du calcium et l’activation de la neurogénèse chez les interneurones.

Le cancer du pancréas est l’un des plus chimiorésistants, avec un taux de survie sur 5 ans inférieur à 5%. La chimiorésistance pourrait être due à la présence de cellules initiatrices de tumeur (TICs), une petite sous-population des cellules tumorales possédant la capacité de régénérer une nouvelle tumeur. Il a été démontré que la metformine cible les TICs par un mécanisme non élucidé. Il est connu que la metformine affecte le métabolisme du carbone. Il a également été démontré que le métabolisme du carbone, plus précisément la glycine décarboxylase (GLDC), est à la fois nécessaire et suffisant à l’acquisition de propriétés d’initiation tumorale. Nous proposons que la metformine cible les cellules initiatrices de tumeur en affectant le métabolisme du carbone. Nous avons utilisé des lignées cellulaires dérivées d’un modèle murin de cancer du pancréas pour comparer l’expression génique de lésions bénignes versus malignes. Les cellules malignes surexpriment Gldc. La metformine diminue l’expression de Gldc, et la surexpression de Gldc diminue la sensibilité à la metformine dans un essai de sphères tumorales. La metformine induit une augmentation du ratio NADP+/NADPH, et la surexpression de Gldc empêche cette augmentation. Nous proposons que la metformine diminue l’expression de Gldc, ce qui cause une diminution du flux du métabolisme du carbone, et donc une diminution de la production de NADPH par ce dernier. L’augmentation du ratio NADP+/NADPH inhibe la synthèse des acides gras et la régénération de la glutathione, ce qui pourrait expliquer la diminution de la formation de sphères tumorales sous traitement metformine.
