La chimie supramoléculaire est un domaine qui suscite depuis quelques années un intérêt grandissant. Le domaine s’appuie sur les interactions intermoléculaires de façon à contrôler l’organisation moléculaire et ainsi moduler les propriétés des matériaux. La sélection et le positionnement adéquat de groupes fonctionnels, utilisés en combinaison avec un squelette moléculaire particulier, permet d’anticiper la façon dont une molécule interagira avec les molécules avoisinantes. Cette stratégie de construction, nommé tectonique moléculaire, fait appel à la conception de molécules appelées tectons (du mot grec signifiant bâtisseur) pouvant s’orienter de façon prévisible par le biais d’interactions faibles et ainsi générer des architectures supramoléculaires inédites. Les tectons utilisent les forces intermoléculaires mises à leur disposition pour s’orienter de façon prédéterminée et ainsi contrecarrer la tendance à s’empiler de la manière la plus compacte possible. Pour ce faire, les tectons sont munies de diverses groupes fonctionnels, aussi appelés groupes de reconnaissance, qui agiront comme guide lors de l’assemblage moléculaire. Le choix du squelette moléculaire du tecton revêt une importance capitale puisqu’il doit permettre une orientation optimale des groupes de reconnaissance. La stratégie de la tectonique moléculaire, utilisée conjointement avec la cristallisation, ouvre la porte à un domaine de la chimie supramoléculaire appelé le génie cristallin. Le génie cristallin permet l’obtention de réseaux cristallins poreux soutenus par des interactions faibles, pouvant accueillir des molécules invitées. Bien que toutes les interactions faibles peuvent être mises à contribution, le pont hydrogène est l’interaction prédominante en ce qui a trait aux réseaux cristallins supramoléculaires. La force, la directionnalité ainsi que la versatilité font du pont hydrogène l’interaction qui, à ce jour, a eu le plus grand impact dans le domaine du génie cristallin. Un des groupements de reconnaissance particulièrement intéressants en génie cristallin, faisant appel aux ponts hydrogène et offrant plusieurs motifs d’interaction, est l’unité 2,4-diamino-1,3,5-triazinyle. L’utilisation de ce groupement de reconnaissance conjointement avec un cœur moléculaire en forme de croix d’Onsager, qui défavorise l’empilement compact, permet l’obtention de valeurs de porosités élevées, comme c’est le cas pour le 2,2’,7,7’-tétrakis(2,4-diamino-1,3,5-triazin-6-yl)-9,9’-spirobi[9H-fluorène]. Nous présentons ici une extension du travail effectué sur les cœurs spirobifluorényles en décrivant la synthèse et l’analyse structurale de molécules avec une unité dispirofluorène-indénofluorényle comme cœur moléculaire. Ce cœur moléculaire exhibe les mêmes caractéristiques structurales que le spirobifluorène, soit une topologie rigide en forme de croix d’Onsager défavorisant l’empilement compact. Nous avons combiné les cœurs dispirofluorène-indénofluorényles avec différents groupements de reconnaissance de façon à étudier l’influence de l’élongation du cœur moléculaire sur le réseau cristallin, en particulier sur le volume accessible aux molécules invitées.

PVC supported liquid membrane and carbon paste potentiometric sensors incorporating an Mn(III)-porphyrin complex as a neutral host molecule were developed for the determination of paracetamol. The measurements were carried out in solution at pH 5.5. Under such conditions paracetamol exists as a neutral molecule. The mechanism of molecular recognition between the Mn(III)-porphyrin and paracetamol, leading to potentiometric signal generation, is discussed.The sensitivity and selectivity toward paracetamol of carbon paste and polymeric liquid membrane electrodes incorporating an Mn(III)-porphyrin host were compared. The applicability of these sensors to the direct determination of paracetamol was checked by performing a recovery test in human plasma.

La present tesi doctoral s'ha basat en la caracterització estructural i en l'estudi de les propietats catalítiques de nous complexos de pal·ladi(0) amb lligands olefínics contenint unitats de ferrocè. En una primera fase s'ha desenvolupat la síntesi d'estructures de tipus (E, E, E)-1,6,11-trisarilsulfonil-1,6,11-triazaciclopentadeca-3,8,13-triè contenint des d'una fins a tres unitats de ferrocè i s'ha estudiat la seva complexació amb pal·ladi(0). Mitjançant estudis electroquímics s'ha avaluat la seva possible aplicació en el reconeixement molecular electroquímic. Encara que aquest tipus d'estructures no han resultat vàlides per aquest tipus de processos de reconeixement, l'estudi electroquímic ha permès determinar que la presència del grup ferrocenil influeix en les propietats redox del metall complexat. S'ha descrit també l'obtenció de macrocicles pentaolefínics anàlegs de 25 membres contenint unitats de ferrocè i s'ha estudiat paral·lelament la seva complexació amb pal·ladi(0).En una segona part del treball s'han avaluat les propietats catalítiques del complex (E, E, E)-6,11-bis[(p-metilfenil)sulfonil]-1-ferrocenilsulfonil-1,6,11-triazaciclopentadeca-3,8,13-trièpal·ladi(0), en reaccions clàssicament catalitzades per pal·ladi tals com l'acoblament creuat de Suzuki o la reacció de Heck, i s'ha pogut determinar que és un catalitzador actiu en aquest tipus de reaccions, essent a més possible, la seva recuperació i reutilització. En el cas concret de la reacció de Heck s'han emprat sals de diazoni com a agents arilants i cal destacar que s'ha descrit el primer sistema catalític recuperable i reutilitzable per aquest tipus de reaccions. Mitjançant espectrometria de masses amb ionització per electrospray ha estat possible determinar el rol mecanístic del nostre complex de pal·ladi en la reacció de Heck amb sals de diazoni.Finalment, en una tercera part estructural del treball s'ha estudiat el comportament espectroscòpic dels complexos de pal·ladi(0) anteriorment mencionats, contenint des de tres unitats aríliques iguals fins a tres unitats aríliques diferents. Mitjançant RMN i difracció de raigs-X s'ha determinat que la complexitat estereoquímica dels complexos deriva dels isòmers que es poden formar mitjançant complexació del metall amb cada una de les cares dels tres dobles enllaços continguts a l'estructura.

Aquesta Tesi Doctoral està dividida en diferents capítols tots ells basats en aplicacions de macrocicles nitrogenats triolefínics de 15 membres i dels seus complexos amb metalls de transició. En primer lloc es descriu l'aplicació d'un complex macrocíclic ferrocenílic de Pd0 com a catalitzador en reaccions de Mizoroki-Heck i Suzuki emprant sals de diazoni com a agents arilants. Aquest catalitzador va proporcionar bons rendiments de productes i va poder ser recuperat i reutilitzat, essent el primer sistema catalític reciclable descrit en la reacció de Mizoroki-Heck amb sals de diazoni. La metodologia es va estendre de forma efectiva a alcohols al·lílics, però en no ser possible recuperar el catalitzador macrocíclic, es va emprar Pd(dba)2. Posteriorment es van preparar estructures macromoleculars mitjançant la introducció de diferents unitats macrocícliques sobre un espaiador. La complexació d'aquestes estructures va permetre preparar complexos tant homo (Pd0) com heterometàl·lics (Pd0, Pt0) amb bons rendiments. Els complexos homometàl·lics van resultar ser bons catalitzadors en reaccions de Suzuki i fàcilment recuperables per simple filtració. En una última part, es van preparar nous macrocicles triolefínics contenint d'un a tres grups amino lliures en l'estructura mitjançant la utilització del grup protector (2-trimetilsilil)etansulfonil (SES). Aquesta modificació estructural va permetre ampliar la capacitat coordinant dels macrocicles a altres metalls de transició, com el PdII, possibilitant noves aplicacions. Concretament es va aplicar el lligand contenint un sol grup amino lliure al reconeixement, transport i preconcentració de PdII i PtIV continguts en dissolucions aquoses.

The tetraprotonated form of the dioxatetraazamacrocycle, 6,19-dioxa-3,9,16,22-tetraaza[22.2.2.2(11,14)]-triaconta-1(26),11,13,24, 27,29-hexaene, (H4L1)(4+), was used as the receptor for binding studies with carboxylate anionic substrates of different shapes, sizes, and charges [succinate (suc(2-)), cyclo- hexanetricarboxylate (cta(3-)), phthalate (ph(2-)), isophthalate (iph(2-)), terephthalate (tph(2-)), and benezenetricarboxylate (btc(3-))]. Association constants were determined by potentiometry in aqueous solution at 298.2 K and 0.10 M KCl and by H-1 NMR titration in D2O. The strongest association was found for the btc3- anion at 5-7 pH region. From both techniques it was possible to establish the binding preference trend of the receptor for the different substrates, and the H-1 NMR spectroscopy gave important suggestions about the type of interactions between partners and the location of the substrates in the supramolecular entities formed. The effective binding constants at pH 6 follow the order: btc(3-)>iph(2-)>cta(3-) =ph(2-)>tph(2-)>suc(2-). All the studies suggest that the anionic substrates bind to the receptor via N-H center dot center dot center dot O = C hydrogen bonds and electrostatic interactions, and the aromatic substrates can also establish pi-pi stacking interactions. The crystal structures of (H4L1)(4+) and its supramolecular assemblies with ph(2-) and tph(2-) were determined by X-ray diffraction. The last two structures showed that the association process in solid state occurs via multiple N-H center dot center dot center dot O = C hydrogen bonds with the anionic substrate located outside the macrocyclic cavity of the receptor. Molecular dynamics simulations carried out for the association of (H4L1)(4+) with tph(2-) and btC(3-) in water solution established at atomic level the existence of all interactions suggested by the experimental studies, which act cooperatively in the binding process. Furthermore, the binding free energies were estimated and the values are in agreement with the experimental ones, indicating that the binding of these two anionic substrates occurs into the receptor cavity. However, the tph(2-) has also propensity to leave the macrocyclic cavity and its molecular recognition can also happen at the top of the receptor. (C) 2008 Elsevier Ltd. All rights reserved.

The 28-membered octaazamacrocycle Me-2[28]py(2)N(6) was used as a receptor for the molecular recognition of aromatic and aliphatic carboxylate substrates. The receptor-substrate binding behaviour of (H6Me2[28]py(2)N(6))(6+) with an aliphatic (-O2C(CH2)(n)CO2-, n=0 to 4) and an aromatic (phthalate, isophthalate, terephthalate, 4,4'-dibenzoate, benzoate, 3- and 4-nitrobenzoate) series of carboxylate anions was evaluated by H-1 NMR spectroscopy (carried out in DMSO-d(6) at 300 K). Two association constants were found for most of the studied cases, except for 3- and 4-nitrobenzoate for which only K-1 was determined. For oxalate, malonate, benzoate and dibenzoate anions only the beta(2) constants could be obtained. The values of the first association constant cover a range from 2.86 to 3.69 (log units), and the second stepwise constant from 2.15 to 2.89 (also in log units). No special selectivity was found but the highest values were determined for adipate and the lowest for the monoprotic 3- and 4-nitrobenzoates. Single crystal X-ray structures of H6Me2[28]py(2)N(6)(6+) with terephthalate, 1, and 4,4'-dibenzoate (2) were determined showing supramolecular entities with general formula (H6Me2[28]py(2)N(6)).(substrate)(2)(PF6)(2).4H(2)O. These anions are the building blocks of an extensive 3-D network of hydrogen bonds.

Ab initio calculations using density functional theory have shown that the reactions that occur between artemisinin, 1, a cyclic trioxane active against malaria, and some metal ions and complexes lead to a series of radicals which are probably responsible for its therapeutic activity. In particular it has been shown that the interaction of Fe(H) with artemisinin causes the O-O bond to be broken as indeed does Fe(III) and Cu(I), while Zn(II) does not. Calculations were carried out with Fe(II) in several different forms including the bare ion, [Fe(H2O)(5)](2+) and [FeP(Im)] (P, porphyrin; Im, imadazole) and similar results were obtained. The resulting oxygen-based radicals are readily converted to more stable carbon-based radicals and/or. stable products. Similar radicals and products are also formed from two simple model trioxanes 2 and 3 that show little or no therapeutic action against malaria although some subtle differences were obtained. This suggests that the scaffold surrounding the pharmacophore may be involved in molecular recognition events allowing efficient uptake of this trioxane warhead into the parasite. This paper investigates dendritic peptides capable of assembling into nanostructured gels, and explores the effect on self-assembly of mixing different molecular building blocks. Thermal measurements, small angle Xray scattering (SAXS) and circular dichroism (CD) spectroscopy are used to probe these materials on macroscopic, nanoscopic and molecular length scales. The results from these investigations demonstrate that in this case, systems with different "size" and "chirality" factors can self-organise, whilst systems with different "shape" factors cannot. The "size" and "chirality" factors are directly connected with the molecular information programmed into the dendritic peptides, whilst the shape factor depends on the group linking these peptides together-this is consistent with molecular recognition hydrogen bond pathways between the peptidic building blocks controlling the ability of these systems to self-recognise. These results demonstrate that mixtures of relatively complex peptides, with only subtle differences on the molecular scale, can self-organise into nanoscale structures, an important step in the spontaneous assembly of ordered systems from complex mixtures.

The terpenoid chiral selectors dehydroabietic acid, 12,14-dinitrodehydroabietic acid and friedelin have been covalently linked to silica gel yielding three chiral stationary phases CSP 1, CSP 2 and CSP 3, respectively. The enantiodiscriminating capability of each one of these phases was evaluated by HPLC with four families of chiral aromatic compounds composed of alcohols, amines, phenylalanine and tryptophan amino acid derivatives and beta-lactams. The CSP 3 phase, containing a selector with a large friedelane backbone is particularly suitable for resolving free alcohols and their derivatives bearing fluorine substituents, while CSP 2 with a dehydroabietic architecture is the only phase that efficiently discriminates 1, 1'-binaphthol atropisomers. CSP 3 also gives efficient resolution of the free amines. All three phases resolve well the racemates of N-trifluoracetyl and N-3,5-dinitrobenzoyl phenylalanine amino acid ester derivatives. Good enantioseparation of beta-lactams and N-benzoyl tryptophan amino acid derivatives was achieved on CSP 1. In order to understand the structural factors that govern the chiral molecular recognition ability of these phases, molecular dynamics simulations were carried out in the gas phase with binary diastereomeric complexes formed by the selectors of CSP 1 and CSP 2 and several amino acid derivatives. Decomposition of molecular mechanics energies shows that van der Waals interactions dominate the formation of the diastereomeric transient complexes while the electrostatic binding interactions are primarily responsible for the enantioselective binding of the (R)- and (S)-analytes. Analysis of the hydrogen bonds shows that electrostatic interactions are mainly associated with the formation of N-(HO)-O-...=C enantio selective hydrogen bonds between the amide binding sites from the selectors and the carbonyl groups of the analytes. The role of mobile phase polarity, a mixture of n-hexane and propan-2-ol in different ratios, was also evaluated through molecular dynamics simulations in explicit solvent. (c) 2006 Elsevier Ltd. The emergence of the mechanical bond during the past 25 years is giving chemistry a fillip in more ways than one. While its arrival on the scene is already impacting materials science and molecular nanotechnology, it is providing a new lease of life to chemical synthesis where mechanical bond formation Occurs as a consequence of the all-important templation Orchestrated by molecular recognition and self-assembly. The way in which covalent bond formation activates noncovalent bonding interactions, switching on molecular recognition that leads to self-assembly, and the template-directed synthesis of mechanically interlocked molecules-of which the so-called catenanes and rotaxanes may be regarded as the prototypes-has introduced a level of integration into chemical synthesis that has not previously been attained jointly at the supramolecular and molecular levels. The challenge now is to carry this I vel of integration during molecular synthesis beyond relatively small molecules into the realms of precisely functionalized extended molecular Structures and superstructures that perform functions in a collective manner as the key sources of instruction, activation, and performance in multi-component integrated Circuits and devices. These forays into organic chemistry by a scientific nomad are traced through thick and thin from the Athens of the North to the Windy City by Lake Michigan with interludes on the edge of the Canadian Shield beside Lake Ontario, in the Socialist Republic of South Yorkshire, on the Plains of Cheshire beside the Wirral, in the Midlands in the Heartland of Albion, and in the City of Angels beside the Peaceful Sea. Impedance spectroscopy has been proven a powerful tool for reaching high sensitivity in sensor arrays made with nanostructured films in the so-called electronic tongue systems, whose distinguishing ability may be enhanced with sensing units capable of molecular recognition. In this study we show that for optimized sensors and bio-sensors the dielectric relaxation processes involved in impedance measurements should also be considered, in addition to an adequate choice of sensing materials. We used sensing units made from layer-by-layer (LbL) films with alternating layers of the polyeletrolytes, poly(allylamine) hydrochloride (PAH) and poly(vinyl sulfonate) (PVS), or LbL films of PAH alternated with layers of the enzyme phytase, all adsorbed on gold interdigitate electrodes. Surprisingly, the detection of phytic acid was as effective in the PVS/PAH sensing system as with the PAH/phytase system, in spite of the specific interactions of the latter. This was attributed to the dependence of the relaxation processes on nonspecific interactions such as electrostatic cross-linking and possibly on the distinct film architecture as the phytase layers were found to grow as columns on the LbL film, in contrast to the molecularly thin PAH/PVS films. Using projection techniques, we were able to detect phytic acid at the micromolar level with either of the sensing units in a data analysis procedure that allows for further optimization.

The immobilization of enzymes in nanostructured films has potential applications, e.g. in biosensing, for which the activity may not only be preserved, but also enhanced if optimized conditions are identified. Optimization is not straightforward because several requirements must be fulfilled, including a suitable matrix and film-forming technique. In this study, we show that horseradish peroxidase (HRP) has its activity enhanced when immobilized in Langmuir-Blodgett (LB) films, in conjunction with dipalmitoylphosphaticlylglycerol (DPPG). Incorporation of HRP into a DPPG monolayer at the air-water interface was demonstrated with compression isotherms, and Polarization-Modulation Infrared Reflection Absorption Spectroscopy (PM-IRRAS). From the PM-IRRAS data, we inferred that HRP was not denatured when adsorbed on a pre-formed, low pressure DPPG monolayer. A change in orientation was induced by the phospholipid matrix, with the amide C=O and NH groups from HRP being oriented perpendicular to the surface, parallel to the DPPG acyl chains, i.e. the alpha-helix was inserted into the monolayer. The mixed DPPG-HRP monolayer could be transferred onto solid supports, to which HRP activity was ca. 23% higher than in solution. The control of molecular architecture and choice of a suitable phospholipid matrix allowed HRP-containing LB films to be used in sensing peroxide. (c) 2008 Elsevier B.V. All rights reserved.

Oligonucleotides have unique molecular recognition properties, being involved in biological mechanisms such as cell-surface receptor recognition or gene silencing. For their use in human therapy for drug or gene delivery, the cell membrane remains a barrier, but this can be obviated by grafting a hydrophobic tail to the oligonucleotide. Here we demonstrate that two oligonucleotides, one consisting of 12 guanosine units (G(12)), and the other one consisting of five adenosine and seven guanosine (A(5)G(7)) units, when functionalized with poly(butadiene), namely PB-G(12) and PB-A(5)G(7), can be inserted into Langmuir monolayers of dipalmitoyl phosphatidyl choline (DPPC), which served as a cell membrane model. PB-G(12) and PB-A(5)G(7) were found to affect the DPPC monolayer even at high surface pressures. The effects from PB-G(12) were consistently stronger, particularly in reducing the elasticity of the DPPC monolayers, which may have important biological implications. Multilayers of DPPC and nucleotide-based copolymers could be adsorbed onto solid supports, in the form of Y-type LB films, in which the molecular-level interaction led to lower energies in the vibrational spectra of the nucleotide-based copolymers. This successful deposition of solid films opens the way for devices to be produced which exploit the molecular recognition properties of the nucleotides. (C) 2010 Elsevier Inc. All rights reserved.

The capability of self-assembly and molecular recognition of biomolecules is essential for many nanotechnological applications, as in the use of alkyl-modified nucleosides and oligonucleotides to increase the cellular uptake of DNA and RNA. In this study, we show that a lipophilic nucleoside, which is an isomer mixture of 2`-palmitoyluridin und 3`-palmitoyluridin, forms Langmuir monolayers and Langmuir-Blodgett films as a typical amphiphile, though with a smaller elasticity. The nucleoside may be incorporated into dipalmitoyl phosphatidyl choline (DPPC) monolayers that serve as a simplified cell membrane model. The molecular-level interactions between the nucleoside and DPPC led to a remarkable condensation of the mixed monolayer, which affected both surface pressure and surface potential isotherms. The morphology of the mixed monolayers was dominated by the small domains of the nucleoside. The mixed monolayers could be deposited onto solid substrates as a one-layer Langmuir Blodgett film that displayed UV-vis absorption spectra typical of aggregated nucleosides owing to the interaction between the nucleoside and DPPC. The formation of solid films with DNA building blocks in the polar heads may open the way for devices and sensors be produced to exploit their molecular recognition properties. (C) 2010 Elsevier B.V. 5-HT(1A) receptor antagonists have been employed to treat depression, but the lack of structural information on this receptor hampers the design of specific and selective ligands. In this study, we have performed CoMFA studies on a training set of arylpiperazines (high affinity 5-HT(1A) receptor ligands) and to produce an effective alignment of the data set, a pharmacophore model was produced using Galahad. A statistically significant model was obtained, indicating a good internal consistency and predictive ability for untested compounds. The information gathered from our receptor-independent pharmacophore hypothesis is in good agreement with results from independent studies using different approaches. Therefore, this work provides important insights on the chemical and structural basis involved in the molecular recognition of these compounds. (C) 2010 Elsevier Masson SAS. All rights reserved.
