The freshwater phytoplankton, responsible for a significant part of primary production in rivers and lakes, is of great importance for matter and energy available to other trophic levels of structuring aquatic food chains. In recent decades, the construction of dams, especially for the generation of electricity, produced changes not only in physical and chemical characteristics of rivers, but also in biocoenosis of these ecosystems. Thus, research is needed to expand the basic knowledge about these environments and subsidize actions appropriate conservation and management. Considering that lotic environments are relatively little studied, and the river-reservoir interaction, the work aims to analyze the limnological conditions and assemblies phytoplankton along the gradient established by the Itaipu reservoir and downstream stretch of the Paraná River. During two periods of the year, winter and summer, we analyzed nine sampling stations: one in lentic environment (the dam), six on the Parana River (each 5 km), at the mouth of a river Monday and another at the mouth of the Iguazu river. The phytoplankton community was composed of 315 taxa, with the class Bacillariophyceae most specious. At every point the richness was higher in summer and with a longitudinal trend in the increasing points after falling after the dam and the contribution of the tributaries. The number of taxa common to the three rivers was low in both study periods. The higher taxa can be explained by the variety of environments sampled, different residence times of water and nutrient concentrations, whereas the rivers Iguazu and Monday suffer high human influence. The greater abundance of organisms in the dam, in both periods, it should be the class Cryptophyceae contributing over 75% of total phytoplankton in point. In cluster analysis (composition and abundance)...(Complete abstract click electronic access below)

Studies regarding the quality and quantity of water have become more and more important because of the increasing negative environmental impacts occurred and their scarcity increase as well. At the Center of Tropical Fish Research (Centro de Pesquisas de Peixes Tropicais - CEPTAlIBAMA) located in the city of Pirassununga, SP, there is a dam named Represa Velha which supplies 128 tanks that are fish hatcheries. This dam takes in part of the Barrinha stream micro-basin where it has been observed strong impacts due to the deforestation of its support forest, as well an the intensive agricultural activities in the neighboring areas. There has been established and distributed 13 points of water-collectors disposed inside the stream and the dam. The water samples were taken from the stream only from its surface because of the low depth and, inside. In the dam, the water samples were taken at the water column, at a 50cm distance over the sediment. In every point of collection inside the dam, it was placed a chamber of sedimentation, vertically in the water column at a 70% height from the total depth. The collections took place during the dry/cold and raining/warm periods of the years 2000 and 2001. Then, it were analyzed the physical, physical-chemical, chemical and micro-biological parameters of the water samples. The obtained results were statistically analyzed throughout the SAS system (Statistical Analisys System). Parameters such as total phosphate, nitrate, nitrite and turbidity were within the established limits for the Class 2 water, according to the n° 20/1986 CONAMA Resolução. However, the total and Escherichia coli coliforms, mainly inside the Barrinha stream, had showed values above of the established limits for the same... The aim of this paper was to evaluate the effect of retention and detention reservoirs along with the regulation in channel flow upgrade on flood for an urban watershed located at Rio Claro, SP. For this purpose, modeling and simulation techniques were applied for runoff determination and its propagation in channel. The Soil Conservation Service – SCS hydrologic model as well as Pulz and non-linear Muskingum-Cunge model were used. The software IPHS1 was applied on simulations. The results pointed out that the combination of retention increasing and detention reservoir implementation (120,000 m3, corresponding to 1.5% of the watershed area) with the streamflow upgrade (n decreasing from 0,04 to 0,02) can minimize the flood on the investigated Servidão watershed. Further, after the proposed intervention, the flood was eliminated for the investigated times of recurrence: 5, 20, 50 and 100 years. The prognostic indicated that the available area occupation had a minor effect on flow increasing due to the observed high urbanization.

Interventions in nature made by man are very common since the beginnings of human history, especially in the case regarding the storage of water. The construction of dams have been and still are fundamental in maintaining human life due to the vital importance that water plays. The size of these structures vary according to need, such as water catchment, fish-farming or electricity generation. Embankment dams are the most common type of these structures. Can be defined as dams natural material obtained from loan chambers located near the dam site. This type of barrier may be divided into earth dams and rockfill dams. The study area covers an earth dam located in Cordeirópolis (SP) and is essentially composed of diabases altered soil of the Serra Geral Formation of the Paraná Basin. With the aid of geophysics, more specifically of the Electrical Resistivity method, the aim is to check any water infiltration zones in the dam's body. Given the risks associated with water seepage in earth dams, that may generate breakdown structures, erosion, and consequently accidents and / or harmful factors in the nearby areas. One of the main structural problems married by water infiltration is the possible generation of pipes that could jeopardize the dam structure. This work aims to contribute towards the evaluation of the effectiveness of using an indirect technique of research and monitoring in aid to direct research techniques such as piezometers and drive stakes. The results are presented in the form of 2D and 3D geophysical models, the analysis shows a low resistivity zone with typical values of the presence of humidity originating upstream of the dam and downstream bottleneck trend, that are the basis for interpretation by percolation or not water in the dam

Wireless networks rapidly became a fundamental pillar of everyday activities. Whether at work or elsewhere, people often benefits from always-on connections. This trend is likely to increase, and hence actual technologies struggle to cope with the increase in traffic demand. To this end, Cognitive Wireless Networks have been studied. These networks aim at a better utilization of the spectrum, by understanding the environment in which they operate, and adapt accordingly. In particular recently national regulators opened up consultations on the opportunistic use of the TV bands, which became partially free due to the digital TV switch over. In this work, we focus on the indoor use of of TVWS. Interesting use cases like smart metering and WiFI like connectivity arise, and are studied and compared against state of the art technology. New measurements for TVWS networks will be presented and evaluated, and fundamental characteristics of the signal derived. Then, building on that, a new model of spectrum sharing, which takes into account also the height from the terrain, is presented and evaluated in a real scenario. The principal limits and performance of TVWS operated networks will be studied for two main use cases, namely Machine to Machine communication and for wireless sensor networks, particularly for the smart grid scenario. The outcome is that TVWS are certainly interesting to be studied and deployed, in particular when used as an additional offload for other wireless technologies. Seeing TVWS as the only wireless technology on a device is harder to be seen: the uncertainity in channel availability is the major drawback of opportunistic networks, since depending on the primary network channel allocation might lead in having no channels available for communication. TVWS can be effectively exploited as offloading solutions, and most of the contributions presented in this work proceed in this direction.

Benzodiazepines act at the major isoforms of GABA type A receptors where they potentiate the current evoked by the agonist GABA. The underlying mechanism of this potentiation is poorly understood, but hypothesized to be related to the mechanism that links agonist binding to channel opening in these ligand activated ion channels. The loop F of the ?(1) and the ?(2) subunit have been implicated in channel gating, and loop F of the ?(2) subunit in the modulation by benzodiazepines. We have identified the conservative point mutation Y168F located N-terminally of loop F in the ?(1) subunit that fails to affect agonist properties. Interestingly, it disrupts modulation by benzodiazepines, but leaves high affinity binding to the benzodiazepine binding site intact. Modulation by barbiturates and neurosteroids is also unaffected. Residue ?(1) Y168 is not located either near the binding pockets for GABA, or for benzodiazepines, or close to the loop F of the ?(2) subunit. Our results support the fact, that broader regions of ligand gated receptors are conformationally affected by the binding of benzodiazepines. We infer that also broader regions could contribute to signaling from GABA agonist binding to channel opening.

The transient receptor potential channel, TRPM4, and its closest homolog, TRPM5, are non-selective cation channels that are activated by an increase in intracellular calcium. They are expressed in many cell types, including neurons and myocytes. Although the electrophysiological and pharmacological properties of these two channels have been previously studied, less is known about their regulation, in particular their post-translational modifications. We, and others, have reported that wild-type (WT) TRPM4 channels expressed in HEK293 cells, migrated on SDS-PAGE gel as doublets, similar to other ion channels and membrane proteins. In the present study, we provide evidence that TRPM4 and TRPM5 are each N-linked glycosylated at a unique residue, Asn(992) and Asn(932), respectively. N-linked glycosylated TRPM4 is also found in native cardiac cells. Biochemical experiments using HEK293 cells over-expressing WT TRPM4/5 or N992Q/N932Q mutants demonstrated that the abolishment of N-linked glycosylation did not alter the number of channels at the plasma membrane. In parallel, electrophysiological experiments demonstrated a decrease in the current density of both mutant channels, as compared to their respective controls, either due to the Asn to Gln mutations themselves or abolition of glycosylation. To discriminate between these possibilities, HEK293 cells expressing TRPM4 WT were treated with tunicamycin, an inhibitor of glycosylation. In contrast to N-glycosylation signal abolishment by mutagenesis, tunicamycin treatment led to an increase in the TRPM4-mediated current. Altogether, these results demonstrate that TRPM4 and TRPM5 are both N-linked glycosylated at a unique site and also suggest that TRPM4/5 glycosylation seems not to be involved in channel trafficking, but mainly in their functional regulation.

This dataset present result from the DFG- funded Arctic-Turbulence-Experiment (ARCTEX-2006) performed by the University of Bayreuth on the island of Svalbard, Norway, during the winter/spring transition 2006. From May 5 to May 19, 2006 turbulent flux and meteorological measurements were performed on the monitoring field near Ny-Ålesund, at 78°55'24'' N, 11°55'15'' E Kongsfjord, Svalbard (Spitsbergen), Norway. The ARCTEX-2006 campaign site was located about 200 m southeast of the settlement on flat snow covered tundra, 11 m to 14 m above sea level. The permanent sites used for this study consisted of the 10 m meteorological tower of the Alfred Wegener Institute for Polar- and Marine Research (AWI), the international standardized radiation measurement site of the Baseline Surface Radiation Network (BSRN), the radiosonde launch site and the AWI tethered balloon launch sites. The temporary sites - set up by the University of Bayreuth - were a 6 m meteorological gradient tower, an eddy-flux measurement complex (EF), and a laser-scintillometer section (SLS). A quality assessment and data correction was applied to detect and eliminate specific measurement errors common at a high arctic landscape. In addition, the quality checked sensible heat flux measurements are compared with bulk aerodynamic formulas that are widely used in atmosphere-ocean/land-ice models for polar regions as described in Ebert and Curry (1993, doi:10.1029/93JC00656) and Launiainen and Cheng (1995). These parameterization approaches easily allow estimation of the turbulent surface fluxes from routine meteorological measurements. The data show: - the role of the intermittency of the turbulent atmospheric fluctuation of momentum and scalars, - the existence of a disturbed vertical temperature profile (sharp inversion layer) close to the surface, - the relevance of possible free convection events for the snow or ice melt in the Arctic spring at Svalbard, and - the relevance of meso-scale atmospheric circulation pattern and air-mass advection for the near-surface turbulent heat exchange in the Arctic spring at Svalbard. Recommendations and improvements regarding the interpretation of eddy-flux and laser-scintillometer data as well as the arrangement of the instrumentation under polar distinct exchange conditions and (extreme) weather situations could be derived.

A rock salt-lamprophyre dyke contact zone (sub-vertical, NE-SW strike) was investigated for its petrographic, mechanic and physical properties by means of anisotropy of magnetic susceptibility (AMS) and rock magnetic properties, coupled with quantitative microstructural analysis and thermal mathematical modelling. The quantitative microstructural analysis of halite texture and solid inclusions revealed good spatial correlation with AMS and halite fabrics. The fabrics of both lamprophyre and rock salt record the magmatic intrusion, "plastic" flow and regional deformation (characterized by a NW-SE trending steep foliation). AMS and microstructural analysis revealed two deformation fabrics in the rock salt: (1) the deformation fabrics in rock salt on the NW side of the dyke are associated with high temperature and high fluid activity attributed to the dyke emplacement; (2) On the opposite side of the dyke, the emplacement-related fabric is reworked by localized tectonic deformation. The paleomagnetic results suggest significant rotation of the whole dyke, probably during the diapir ascent and/or the regional Tertiary to Quaternary deformation.
