Chitin-binding vicilins from legume seeds (Erythrina velutina. Canavalia ensiformes and Phaseolus vulgares) were isolated by ammonium sulfate followed by affinity chromatography on a chitin column. Effect of these vicilins on female adults of Ceratitis capitata was examined by bioassay and in a semi-field assay model. Mechanism of action of the vicilins was determined by in vivo digestibility and chitin affinity. Among the tested vicilins, E. velutina when added to diet caused strong effect on mortality at 10% dose. This insecticidal property was tested in a semi-field assay which showed the same effect observed in laboratory conditions, where doses of 10% and 15% were lethal to female adults of C. capitata. These deleterious effects were not only associated to the binding to chitin structures present in peritrophic membrane, but principally to its low digestibility in the C. capitata digestive tract. This fact was confirmed because chiting binding proteins as WGA and the other tested vicilins were not toxic to female adults of C. capitata due susceptibility of these proteins to digestive enzymes of the insects. By other side EvV was more resistant to digestive enzymes, causing deleterious effects on female adults of C. capitata. These results showed that EvV may be part of the pest management programs or an alternative in plant improvement program in the population control of this fruticulture pest

Avaliou-se o efeito dos inseticidas endossulfam e deltametrina sobre a formiga predadora Azteca chartifex spiriti For., em cultivo do cacaueiro no Sudeste da Bahia, conforme padronização da International Organization for Biological and Integrated Control (IOBC), West Palaearctic Regional Section (WPRS), com algumas modificações. Os testes foram: a) contato; b) pulverização direta; c) persistência. Nos testes de contato, operárias foram expostas a um filme fresco e seco dos inseticidas, aplicado sobre cristalizadores. Nos testes de aspersão, as formigas foram diretamente pulverizadas com os agroquímicos às concentrações recomendadas para o controle de tripes e percevejos. Os insetos foram mantidos a 25±2°C, 65±10% UR e fotofase de 12h. A sobrevivência foi avaliada após 24h e a intervalos variáveis de tempo. Nos testes de persistência, as operárias foram confinadas em cristalizadores e expostas ao contato com folhas de cacaueiro tratadas previamente no campo, sob as mesmas condições controladas. Desenvolveram-se experimentos a um, três, seis, 10, 18 e 32 dias após a aplicação (DAA). Deltametrina foi considerado seletivo nos testes de contato e pulverização direta, não sendo necessária a condução dos testes de persistência em folhas para este inseticida. Endossulfam foi altamente tóxico nos testes de contato e pulverização direta, entretanto, apresentou-se levemente persistente no terceiro teste. A análise conjunta dos três tipos de testes sugere que ambos os inseticidas são seletivos para a espécie benéfica, podendo ser recomendadosem Programas de MIP em agroecossistema cacaueiro. A metodologia proposta pela IOBC/WPRS possibilita estabilidade dos resultados, permitindo sua adaptação e utilização para o fim proposto.

Use of antibiotics as an additive in poultry diets to improve growth has been discussed in relation to bacterial resistance and the development of new products and management practices. This study was carried out to test the efficacy of a new substance (Saccharomyces cereviside cell walls, var. Calsberg- SCCW) obtained from the brewery industry, added (at 0.1 and 0.2%) to broiler chicken diets (based on corn and soybean meal), on performance and intestinal mucosa development. In Experiment 1 (carried out in litter-floor pens) the results revealed higher body weight gain,for the total experimental period and higher villus height at 7 d of age for the birds fed 0.2%,SCCW. In a field test using 44,000 broilers that,received feed containing 0.2% SCCW,. The results also showed higher body weight gain and better feed conversion for SCCW-supplemented birds. The present findings show that SCCW improved body weight gain in broiler chickens and that this effect can be attributed to the trophic effect of this product on the intestinal mucosa, because it increases villus height, particularly during the first 7. d of a chicken's life.

The aim was to study the efficiency of electrical conductivity test in evaluating the quality of castor seeds. Emergence of seedlings was evaluated in the field, with five seed lots of cv. AL Guarany 2002. The seeds were submitted to the following tests: germination; first count of germination; accelerated aging (45 degrees C by 24 to 100% UR), emergence of seedlings in the field, rate of speed of development of seedlings and electrical conductivity, testing the periods (2, 4, 6, 8 and 24 hours) and the number of seeds (25,50 and 75) in 75ml of distilled water conditioning to 25 degrees C. A completely randomized design was used. The averages were compared by the Tukey test at 5%. The mean laboratory and field test data were correlated. Electrical conductivity test for 4 hours with 25 seeds and 6 hours with 50 seeds, and the rate of speed emergency proved efficient in the selection of lots vigor by providing information equivalent to the emergence of seedlings in the field.

Um equipamento denominado Unidade Móvel de Ensaio da Barra de Tração - UMEB, foi desenvolvido na FCA/UNESP de Botucatu para realizar ensaios de tratores em solo agrícola. Construída a partir de um reboque (trailer), a UMEB foi adaptada para servir como carro dinamométrico instrumentado, utilizado na avaliação do desempenho de tratores submetidos a ensaios na barra de tração. Sua massa total é de 10.500 kg sustentados por um conjunto de seis rodados pneumáticos. Ensaios de campo mostraram que a UMEB proporcionou força de tração acima de 35 kN, mantendo-a constante, em diferentes condições de superfície do solo, mesmo quando a velocidade de deslocamento foi modificada.

Low voltage solar panels increase the reliability of solar panels due to reduction of in series associations the configurations of photovoltaic cells. The low voltage generation requires DCDC converters devices with high efficiency, enabling raise and regulate the output voltage. This study analyzes the performance of a photovoltaic panel of Solarex, MSX model 77, configured to generate an open circuit voltage of 10.5 V, with load voltage of 8.5 V, with short circuit current of 9 A and a power of 77 W. The solar panel was assembled in the isolated photovoltaic system configuration, with and without energy storage as an interface with a DCDC converter, Booster topology. The converter was designed and fabricated using SMD (Surface Mounted Devices) technology IC (integrated circuit) that regulates its output voltage at 14.2 V, with an efficiency of 87% and providing the load a maximum power of 20.88 W. The system was installed and instrumented for measurement and acquisition of the following data: luminosities, average global radiation (data of INPE Instituto Nacional de Pesquisas Espaciais), solar panel and environment temperatures, solar panel and DC-DC converter output voltages, panel, inverter, and battery charge output currents. The photovoltaic system was initially tested in the laboratory (simulating its functioning in ideal conditions of operation) and then subjected to testing in real field conditions. The panel inclination angle was set at 5.5°, consistent with the latitude of Natal city. Factors such as climatic conditions (simultaneous variations of temperature, solar luminosities and ra diation on the panel), values of load resistance, lower limit of the maximum power required by the load (20.88 W) were predominant factors that panel does not operate with energy efficiency levels greater than 5 to 6%. Anxiety is an emotional phenomenon, and normally it is interpreted as an adaptative behavior front to adversities. In its pathological form, anxiety can severely affect aspects related to the personal and professional life. Studies have shown a close relationship between anxiety disorders and aversive memory processing. Considering that the pharmacotherapy of anxiety disorders is still limited, innovative anxiolytic agents are needed. In this regard, neuropeptides systems are interesting therapeutic targets to the treatment of psychopathologies. Neuropeptide S (NPS), a 20-aminoacid peptide, is the endogenous ligand of a G-protein coupled receptor (NPSR), which has been reported to evoke hyperlocomotion, awakefull states, besides anxiolysis and memory improvements in rodents. This study aimed to investigate the effects of biperiden (BPR; an amnesic drug), diazepam (DZP; an anxiolytic drug) and NPS at three distinct times: pre-training, post-training, and pre-test, in order to assess anxiety and memory process in the same animal model. The elevated Tmaze (ETM) is an apparatus derived from the elevated plus-maze test, which consists of one enclosed and two open arms. The procedure is based on the avoidance of open spaces learned during training session, in which mice were exposed to the enclosed arm as many times as needed to stay 300 s. In the test session, memory is assessed by re-exposing the mouse to the enclosed arm and the latency to enter an open arm was recorded. When injected pre-training, BPR (1 mg/kg) impaired learning and memory processing; DZP (1 and 2 mg/kg) evoked anxiolysis, but only at the dose of 2 mg/kg impaired memory; and NPS 0.1 nmol induced anxiolysis without affecting memory. Post-training injection of DZP (2 mg/kg) or BPR (1 and 3 mg/kg) did not affect memory consolidation, while the post-trainning administration of NPS 1 nmol, but not 0.1 nmol, improved memory in mice. Indeed, pre-trainning administration of NPS 1 nmol did not prevent memory impairment elicited by BPR (2 mg/kg, injected before training). In the open field test, BPR 1 mg/kg and NPS 1 nmol induced hyperlocomotion in mice. In conclusion, the proposed ETM task is practical for the detection of the anxiolytic and amnesic effects of drugs. The anxiolytic and memory enhancement effects of NPS were detected in the ETM task, and reinforce the role of NPS system as an interesting therapeutic target to the treatment of anxiety disorders

Neuropeptide S (NPS) is an endogenous 20-aminoacid peptide which binds a G protein-coupled receptor named NPSR. This peptidergic system is involved in the modulation of several biological functions, such as locomotion, anxiety, nociception, food intake and motivational behaviors. Studies have shown the participation of NPSR receptors in mediating the hyperlocomotor effects of NPS. A growing body of evidence suggests the participation of adenosinergic, dopaminergic and CRF systems on the hyperlocomotor effects of NPS. Considering that little is known about the role of dopaminergic system in mediating NPS-induced hyperlocomotion, the present study aims to investigate the locomotor actions of intracerebroventricular (icv) NPS in mice pretreated with α-metil-p-tirosine (AMPT, inhibitor of dopamine synthesis), reserpine (inhibitor of dopamine vesicle storage) or sulpiride (D2 receptor antagonist) in the open field test. A distinct group of animals received the same pretreatments described above (AMPT, reserpine or sulpiride) and the hyperlocomotor effects of methylphenidate (dopamine reuptake inhibitor) were investigated in the open field. NPS and methylphenidate increased the mouse locomotor activity. AMPT per se did not change the locomotion of the animals, but it partially reduced the hyperlocomotion of methylphenidate. The pretreatment with AMPT did not affect the psychostimulant effects of NPS. Both reserpine and sulpiride inhibited the stimulatory actions of NPS and methylphenidate. These findings show that the hyperlocomotor effects of methylphenidate, but not NPS, were affected by the pretreatment with AMPT. Furthermore, methylphenidate- and NPS-induced hyperlocomotion was impaired by reserpine and sulpiride pretreatments. Together, data suggests that NPS can increase locomotion even when the synthesis of catecholamines was impaired. Additionally, the hyperlocomotor effects of NPS and methylphenidate depend on monoamines vesicular storaged, mainly dopamine, and on the activation of D2 receptors. The psychostimulant effects of NPS via activation of dopaminergic system display clinical significance on the treatment of diseases which involves dopaminergic pathways, such as Parkinson s disease and drug addiction

Artemisia annua tem sido utilizada tradicionalmente para o tratamento de malária e febre na China devido à presença do princípio ativo, artemisinina. O presente trabalho avaliou a atividade central de do óleo essencial obtido por hidrodestilação e do extrato etanólico bruto de folhas frescas de A. annua em modelo in vivo como parte de um screening farmacológico dessa espécie. Sono induzido por pentobarbital, nado forçado e o ensaio de campo aberto são modelos de estudo conhecidos para o estudo de fármacos sobre depressão induzida. A administração do óleo essencial ou extrato bruto etanólico de A. annua aumentaram o tempo de imobilidade no teste do nado forçado. Por outro lado, diminuíram outros parâmetros no campo aberto, como ambulação, exploração, o ato de lamber as patas ou se lamber. Ambos produtos aumentaram o tempo de sono induzido por pentobarbital, com o óleo essencial apresentando um efeito superior ao do extrato. Pela análise dos resultados, é possível sugerir que tanto o extrato bem como o óleo essencial podem atuar como depressores do Sistema Nervoso Central (SNC).

Os testes de vigor são empregados pelas empresas produtoras de sementes, no controle interno de qualidade. Dentre estes, o teste de comprimento de plântulas tem potencial para ser usado para este fim. Assim, o trabalho objetivou verificar a possibilidade de utilização do teste de comprimento de plântulas como um método de vigor para classificar lotes de sementes de soja de acordo com a sua qualidade fisiológica. Para tanto, variou-se a forma de apresentação dos resultados médios de comprimento de plântulas e de suas partes (raiz e hipocótilo). A qualidade fisiológica de três lotes de soja (Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA) 48), de mesma classe de tamanho e com teor de água semelhante, foi avaliada por testes de viabilidade e vigor, realizados em laboratório e campo. Empregou-se delineamento inteiramente casualizado. As médias foram comparadas pelo teste de Tukey a 5% de probabilidade. Correlacionaram-se os dados médios dos testes realizados em laboratório e em campo. O comprimento da raiz de soja foi mais sensível para diferenciar lotes, apresentando maior correlação com a emergência das plântulas de soja em campo. O comprimento de plântulas ou de suas partes distinguiram os três lotes de soja somente quando seus resultados médios foram divididos pelo número de sementes em teste, mostrando ser a forma mais eficaz para classificar lotes de alta qualidade e com diferenças sutis de vigor.
