Resumo:
A invenção se refere a um processo para separação de CTV e CSDaV utilizando citometria de fluxo. Trata-se de um processo rápido de baixo custo e eficiente, relacionado ao principal setor do agronegócio brasileiro, que é a citricultura. Através do produto obtido no trabalho podem-se gerar diagnósticos no sentido aplicado, além de auxiliar no isolamento de outros agentes patógenos dos citros ou outras espécies de plantas.

The tangerine 'Sunki' are highly relevant for the Brazilian citrus industry by presenting important traits for genetic improvement and to be considered a great portaenxento. In this context, the objective of the present work to study the characterization of the fruits of citrus rootstock 'Sunki', depending on the position on the plan and also the germination of their seeds. The physical characteristics evaluated were fruit weight, longitudinal and equatorial diameters of the fruit longitudinal diameter / equatorial diameter, number of buds per fruit, weight of buds and bark and number of viable seeds per fruit. As for the chemical characterization was evaluated titratable, acidity, soluble solid, "Ratio", pH and vitamin C. For the evaluation of these variables, the fruits were obtained from the northeast, southwest, northwest and southeast quadrants. Regarding the evaluation of seed germination, different doses of gibberellic acid and Stimulate were used. The seeds extracted from ripe fruit, washed, dried in the shade and stored in BOD. Immediately after, they were treated for 24 hours with plant growth regulators. After soaking, the seeds were placed in a germinator in germitest paper. After initiation of the germination process, the evaluations were performed daily until the thirty- fourth day. Fruits of 'Sunki'mandarin with uneven as the fruit position within the canopy, the use of gibberellic acid and growth promoter is promising for commercial use on seed germination of this rootstock.

Expansion of agricultural practice, cattle raising and forestry, in a disorderly way and no limits of land use, generates the degradation of natural resources such as soil, water and vegetation. That fact brings consequences, impacts the environment and the rural landscape. This study aimed to identify and quantify the land use in nine watersheds included in the watershed of Faxinal creek, located in western Botucatu, São Paulo State, Brazil, at 22º 51’ 35” and 22º 57’ 02” – Latitude S and 48º 39’ 42” and 48º 38’ 01” – Longitude W. The basin was subdivided into 9 subunits, being eight from second and one from third-order branch. The diagnosis of the subunits was carried out with geospatial technologies, in order to gather data on the use and occupation of the soil. Based on the obtained results, was concluded that the sub watersheds are occupied by the sum of areas of Citrus, horticulture, coffee plantation and small other occupations (25,81%), followed by reforestation (24,80%), as an isolated occupation element, has occupied the largest area.

The great importance of citrus in São Paulo state, particularly in Botucatu city, the and its major expansion in area in recent years and the advancement of techniques for environmental monitoring, such as remote sensing motivated the present research. The aim of this study was the determination the areas and perimeters of citrus in the Botucatu city, São Paulo state, Brazil, obtained through the softwares: Carta Linx, GIS-Idrisi and SPLAN, using aerial colored photographs of 2005, scaled at 1:30000 with longitudinal covering of approximately 60% and 30% laterally. The areas obtained from the aerial photographs and evaluated by Carta Linx presented values closer to the terrestrial reality in determining areas and perimeters, whereas GIS-Idrisi showed discrepancies in the determination of the perimeters, due to working with matrixes. SPLAN and Carta Linx had values for perimeters and areas in close proximity, due to work with vectors.

Huanglongbing (HLB) is associated with Candidatus Liberibacter spp., endogenous, sieve tube-restricted bacteria that are transmitted by citrus psyllid insect vectors. Transgenic expression in the phloem of specific genes that might affect Ca. Liberibacter spp. growth and development may be an adequate strategy to improve citrus resistance to HLB. To study specific phloem gene expression in citrus, we developed three different binary vector constructs with expression cassettes bearing the beta-glucuronidase (GUS) reporter gene (uidA) under the control of one of the three different promoters: Citrus phloem protein 2 (CsPP2), Arabidopsis thaliana phloem protein 2 (AtPP2), and Arabidopsis thaliana sucrose transporter 2 (AtSUC2). Transgenic lines of 'Hamlin', 'Pera', and 'Valencia' sweet oranges [Citrus sinensis (L.) Osbeck] were produced via Agrobacterium tumefaciens transformation. The epicotyl segments collected from in vitro germinated seedlings were used as explants. The gene nptII, which confers resistance to the antibiotic kanamycin, was used for selection. The transformation efficiency was expressed as the number of GUS-positive shoots over the total number of explants and varied from 1.54 to 6.08 % among the three cultivars and three constructs studied. Several lines of the three sweet orange cultivars analyzed using PCR and Southern blot analysis were genetically transformed with the three constructs evaluated. The histological GUS activity in the leaves indicates that the uidA gene was preferentially expressed in the phloem, which suggests that the use of the three promoters might be adequate for producing HLB-resistant transgenic sweet oranges. The results reported here conclusively demonstrate the preferential expression of GUS in the phloem driven by two heterologous and one homologous gene promoters.
