O tema da adesão ao tratamento tem recebido muita atenção dentro da Psicologia da Saúde nos últimos anos. Estudos nessa área revelam que os pacientes que não aderem ao tratamento podem não se beneficiar dos ganhos trazidos pela intervenção terapêutica. A literatura sobre adesão ao tratamento anti-retroviral revela que é o cuidador que geralmente assume a responsabilidade pelo gerenciamento do tratamento de crianças e adolescentes soropositivos. O objetivo principal do presente estudo foi descrever o padrão de adesão ao tratamento de cuidadores de crianças e adolescentes soropositivos que vivem no Estado do Pará, identificando as variáveis que interferem na adesão. Buscou-se, ainda, revelar os tipos de estratégias de enfrentamento utilizadas para lidar com as condições adversas trazidas pela soropositividade. Para tanto, realizou-se um estudo descritivo, de corte transversal, com 30 cuidadores, matriculados na Unidade de Referência Materno-Infantil e Adolescente do Estado do Pará (UREMIA), utilizando como instrumentos de coleta de dados, junto aos cuidadores, um roteiro de entrevista que investigava aspectos sociodemográficos, clínicos e psicossociais das crianças e adolescentes e a Escala Modos de Enfrentamento de Problemas (EMEP). Em relação à adesão pelos cuidadores, observou-se que algumas condições facilitaram o seguimento do tratamento. Essas condições estiveram relacionadas a variáveis de organização familiar (como o número de pessoas que moravam na residência), à história de interrupção do tratamento e ao auto-relato dos cuidadores sobre o padrão de desempenho a ser emitido mediante o sucesso (ou insucesso) da adesão. A influência dessas variáveis demonstrou a influência do controle do comportamento mediante regras, pois os cuidadores emitiram o desempenho classificado como adesão para evitar entrar em contato com as conseqüências aversivas do não seguimento do tratamento. Em relação às estratégias de enfrentamento, foi demonstrada maior média para o Fator 3, demonstrando a ênfase no uso de estratégias focalizadas em práticas religiosas e/ou pensamentos fantasiosos. Este trabalho propôs que os cuidadores da amostra lidaram com o problema da soropositividade de maneira a esquivar-se de pensar no problema do modo como ele realmente se constituiu diante dos mesmos, utilizando pensamentos religiosos ou mágicos para continuar lidando com a rotina diária de enfrentamento das questões que envolviam a condição de soropositividade da criança e/ou do adolescente. A partir dos dados encontrados, indica-se a realização de estudos que investiguem questões relacionadas ao papel exercido por contingências sociais sobre o comportamento dos cuidadores de crianças e adolescentes que vivem no Pará, bem como a investigação sobre aspectos relacionados à revelação do diagnóstico.

Com o advento da Terapia Anti -Retroviral, a Aids assumiu características de doença crônica, em especial nos países onde o acesso aos medicamentos é efetivamente garantido. O Brasil é tomado como modelo por possuir um programa que tem dado boas respostas à epidemia. Em novembro de 1996, foi promulgada, pelo Sistema Único de Saúde (SUS), a lei que dispõe sobre a obrigatoriedade do acesso gratuito a todos os que necessitarem de medicamentos anti -retrovirais. Os resultados obtidos com o tratamento – a redução progressiva da carga viral e a manutenção e/ou restauração do funcionamento do sistema imunológico – têm sido associados a benefícios marcantes na saúde física das pessoas soropositivas e permitido que elas retomem e concretizem seus projetos de vida. Porém, o acesso universal aos medicamentos que possibilita o tratamento para portadores do HIV gratuitamente ainda enfrenta problemas de adesão. Em uma compreensão mais restrita, adesão pode ser definida como o comportamento de uma pessoa – tomar remédio, seguir uma dieta ou fazer mudanças no estilo de vida – que corresponde às recomendações da equipe de saúde. Nesse contexto, esse estudo se propõe a analisar as representações sociais de sujeitos soropositivos sobre o tratamento anti-retroviral e suas implicações no processo de adesão a este tratamento, caracterizando as imagens e os sentidos que estes sujeitos soropositivos que aderiram ou não aderiram à terapia anti -retroviral possuem sobre este tipo de tratamento e as implicações na sua vida, destacando as objetivações e as ancoragens que compõem suas representações sociais. A metodologia foi pautada nas formulações teóricas sobre pesquisa qualitativa, priorizando -se a entrevista no enfoque do Método de Explicitação do Discurso Subjacente (MEDS), realizadas na Unidade de Referência em Doenças Infecciosas e Parasitárias Especiais (UREDIPE), vinculada à Secretaria de Estado de Saúde do estado do Pará (SESPA) e no Hospital Universitário João de Barros Barreto (HUJBB), mais especificamente na Clinica de Doenças Infecciosas e Parasitárias (DIP).

The gall-forming thrips Gynaikothrips ficorum Marchal (Thysanoptera: Phlaeothripidae) is recorded in all regions where its host plant, Ficus microcarpa (Marchal) (Moraceae), has been cultivated as an urban and interior landscape plant species, including potted plants and bonsai. Similarly, the thrips predator Montandoniola confusa Streito & Matocq (Hemiptera: Anthocoridae) has generally followed the prey distribution. The gall induced by thrips degrades the plant foliage, and the thrips themselves can be annoying for people both outdoors and indoors. The galls, however, create a microcosm with all developmental stages of the thrips and its predator. In this study we present the first records of M. confusa in South America, document the species' widespread concomitant occurrence across Brazil, and report our studies of three aspects of M. confusa predation upon the eggs, larvae/prepupae, and adults of G. ficorum thrips: (i) functional response of the predator adult female as a function of different densities of thrips eggs, larvae/prepupae and adults separately: (ii) predation on eggs by adult M. confusa with adult thrips present in the gall; and (iii) adult M. confusa prey preferences when all thrips stages occurred simultaneously in the gall. For all three thrips life stages tested, the predator exhibited a type II functional response. Despite the availability of different life stages in the gall, M. confusa adults are capable of preying upon all life stages of G. ficorum, predation was preferentially on thrips eggs, with an estimated similar to 10-fold greater predation on eggs compared to larvae/prepupae and adult thrips. Egg predation was unaffected by the presence of defensive adult thrips in the gall under low densities (<30 eggs/gall) but when egg densities were greater than 30 eggs/gall, it was reduced when adult thrips were present. The albendazole and mebendazole drugs are benzimidazole derivatives and belong to the anthelmintic class. These drugs are particularly recommended for the treatment against worms present in the gastrointestinal tract of animals and humans, by acting directly on the worm metabolism. The need for thermally study drugs is related to all the parameters that these analyzes include: presence or absence of polymorphs, possible changes in the crystallinity of the drugs, as well as the quality control during the manufacturing process thereof. In this study the thermal behavior of anthelmintic albendazole and commercial mebendazole and its recrystallisation in organic solvents, such as acetic acid and formic acid in dimethylformamide to mebendazole, and albendazole were studied using TG-DSC techniques, TG-FTIR, FTIR and XRD. TG-DSC techniques were used so it could collect information about the thermal stability of the compounds steps for thermal decomposition process and also prove its melting temperature. For recrystallization of drugs in organic solvents, the TG-DSC curves were analyzed to compare and determine that the occurrence of polymorphs. The coupled TG-FTIR technique allowed the analysis of volatile products which were released during the thermal decomposition of the commercial mebendazole. The absorption spectroscopy in the infrared region was performed to mebendazole, and albendazole in order to show the difference in functional groups of both, comparing the spectra with commercial drugs and see if there was recrystallized changes in the absorption band where the drug was recrystallized or when heated. The diffraction technique by powder X-ray method was used for comparison of the crystal structures of commercial drugs and recrystallization in organic solvents to identify changes in crystallinity both, which might suggest the formation of polymorphs

Curcumin possesses wide-ranging anti-inflammatory and anti-cancer properties and its biological activity can be correlated to its potent antioxidant capacity. Novel maghemite (gamma-Fe3O4) nanoparticles, characterized by a diameter of about 10 nm and possessing peculiar colloidal properties and surface interactions, called Surface Active Maghemite Nanoparticles (SAMN), were superficially modified with curcumin by simple incubation, due to the presence of under-coordinated Fe(III) atoms on nanoparticle surface. The resulting curcumin-modified SAMNs (SAMN@curcumin) were characterized by transmission electron microscopy (TEM), FTIR, Mossbauer, EPR and UV-Vis spectroscopy. The redox properties of bound curcumin were tested by electrochemistry. Finally, SAMN@curcumin was studied in the presence of different electroactive substances, namely hydroquinone, NADH and ferrocyanide, in order to assess its electrochemical behavior. Moreover, SAMN@curcumin was electrochemically tested in the presence of one of the most diffuse reactive oxygen specie, such as hydrogen peroxide, demonstrating its stability. SAMN@curcumin in which curcumin is firmly bound, but still retaining its redox features represents a feasible adduct: a magnetically drivable nano-bio-conjugate mimicking free Curcumin redox behavior. The proposed nanostructured material could be exploited as magnetic drivable curcumin vehicle for biomedical applications.

The hosts for Antricola delacruzi ticks are insectivorous, cave-dwelling bats on which only larvae are found. The mouthparts of nymphal and adult A. delacruzi are compatible with scavenging feeding because the hypostome is small and toothless. How a single blood meal of a larva provides energy for several molts as well as for oviposition by females is not known. Adults of A. delacruzi possibly feed upon an unknown food source in bat guano, a substrate on which nymphal and adult stages are always found. Guano produced by insectivorous bats contains twice the amount of protein and 60 times the amount of iron as beef. In addition, bacteria and chitin-rich fungi proliferate on guano. Comparative data on the transcriptome of the salivary glands of A. delacruzi is nonexistent and would help to understand the physiological adaptations of salivary glands that accompany different sources of food as well as the steps taken by the Acari toward haematophagy, believed to have evolved from scavenging dead animals. Annotation of the transcriptome of salivary glands from female instars of A. delacruzi collected on guano categorized 5.7% of the clusters of expressed genes as putative secreted proteins. They included abundantly expressed TIL-domain-containing proteins (possible anti-microbials), an abundantly expressed protein similar to a serum amyloid found in the sialotranscriptomes of Ornithodoros spp., a savignygrin, a family of mucin/peritrophin/cuticle-like proteins, anti-microbials and an HIV envelope-like glycoprotein also found in soft ticks. When comparing the transcriptome of A. delacruzi with those of blood-feeding female soft and hard ticks some notable differences were observed; they consisted of the following transcripts over- or under-represented or absent in the sialotranscriptome of A. delacruzi that may reflect its source of food: ferritin, mucins with chitin-binding domains and TIL-domain-containing proteins versus lipocalins, basic tail proteins, metalloproteases, glycine-rich proteins and Kunitz protease inhibitors, respectively. Posttraumatic stress disorder (PTSD) is an incapacitating syndrome that follows a traumatic experience. Predator exposure promotes long-lasting anxiogenic effect in rodents, an effect related to symptoms found in PTSD patients. Cannabidiol (CBD) is a non-psychotomimetic component of Cannabis sativa with anxiolytic effects. The present study investigated the anti-anxiety actions of CBD administration in a model of PTSD. Male Wistar rats exposed to a predator (cat) received, 1 h later, singled or repeated i.p. administration of vehicle or CBD. Seven days after the stress animals were submitted to the elevated plus maze. To investigate the involvement of 5HT1A receptors in CBD effects animals were pre-treated with WAY100635, a 5HT1A receptor antagonist. To explore possible neurobiological mechanisms involved in these effects, 5HT1A receptor mRNA and BDNF protein expression were measured in the hippocampus, frontal cortex, amygdaloid complex and dorsal periaqueductal gray. Repeated administration of CBD prevented long-lasting anxiogenic effects promoted by a single predator exposure. Pretreatment with WAY100635 attenuated CBD effects. Seven days after predator exposure 5HT1A mRNA expression was up regulated in the frontal cortex and hippocampus. CBD and paroxetine failed to prevent this effect. No change in BDNF expression was found. In conclusion, predator exposure promotes long-lasting up-regulation of 5HT1A receptor gene expression in the hippocampus and frontal cortex. Repeated CBD administration prevents the long-lasting anxiogenic effects observed after predator exposure probably by facilitating 5HT1A receptors neurotransmission. Our results suggest that CBD has beneficial potential for PTSD treatment and that 5HT1A receptors could be a therapeutic target in this disorder. (C) 2012 Elsevier Ltd. All rights reserved.

Autoimmune and infectious diseases are associated with behavioral changes referred to as sickness behavior syndrome (SBS). In autoimmunity, the generation of anti-self T lymphocytes and autoantibodies critically involves binding of CD40 ligand on T-cells to its receptor CD40 on B-cells, dendritic cells and macrophages. Activation of CD40 leads to production of proinflammatory cytokines and, as shown here, induces SBS. Here we report that these behavioral changes depend on the expression of tumor necrosis factor alpha receptor 1 (TNFR1), but not on interleukin-1 receptor 1 or interleukin-6. Moreover, the intensity of SBS correlates with suppression of E-box controlled clock genes, including Dbp, and upregulation of Bmal1. However, the absence of TNFR1 does not interfere with the development of SBS and dysregulation of clock genes in mice treated with lipopolysaccharide. Thus, our results suggest that TNFR1 mediates SBS and dysregulation of clock genes in autoimmune diseases.

Small pumpkinseed sunfish ( Lepomis gibbosus), were found to be capable of removing the spine of Bythotrephes longimanus, an invasive cladoceran. Because fish consumption may be important in the dispersal or control of Bythotrephes, aquarium feeding experiments were conducted to 1) establish if the spine removal behavior of the pumpkinseeds was locally unique; 2) quantify how frequently pumpkinseeds exhibit the behavior; 3) determine if pumpkinseed handle Bythotrephes more quickly than other species of fish; and 4) verify if Bythotrephes' resting eggs pass through the digestive systems of pumpkinseeds in viable condition. The experiments revealed that pumpkinseeds (45-70 mm TL) from two geographic regions were more successful (100%) at removing Bythotrephes' spine, and handled Bythotrephes more quickly than yellow perch (Perca flavescens) (49-57 mm TL) and smallmouth bass (Micropterus dolomieu) (50-57mm TL) used in the study. Of 244 live Bythotrephes' resting eggs fed to the pumpkinseeds, 104 (42.6%) passed through their digestive systems. From those eggs, only 10 successfully hatched. Preliminary enclosure experiments were carried out and indicated that pumpkinseeds will consume Bythotrephes in natural settings. These findings provide new evidence that certain fish, with specialized morphology for prey manipulation, have the ability to influence the distribution and establishment of Bythotrephes.

BACKGROUND: The nonsteroidal anti-inflammatory drug (NSAID), indomethacin (Indo), has a large number of divergent biological effects, the molecular mechanism(s) for which have yet to be fully elucidated. Interestingly, Indo is highly amphiphilic and associates strongly with lipid membranes, which influence localization, structure and function of membrane-associating proteins and actively regulate cell signaling events. Thus, it is possible that Indo regulates diverse cell functions by altering micro-environments within the membrane. Here we explored the effect of Indo on the nature of the segregated domains in a mixed model membrane composed of dipalmitoyl phosphatidyl-choline (di16:0 PC, or DPPC) and dioleoyl phosphatidyl-choline (di18:1 PC or DOPC) and cholesterol that mimics biomembranes. METHODOLOGY/PRINCIPAL FINDINGS: Using a series of fluorescent probes in a fluorescence resonance energy transfer (FRET) study, we found that Indo induced separation between gel domains and fluid domains in the mixed model membrane, possibly by enhancing the formation of gel-phase domains. This effect originated from the ability of Indo to specifically target the ordered domains in the mixed membrane. These findings were further confirmed by measuring the ability of Indo to affect the fluidity-dependent fluorescence quenching and the level of detergent resistance of membranes. CONCLUSION/SIGNIFICANCE: Because the tested lipids are the main lipid constituents in cell membranes, the observed formation of gel phase domains induced by Indo potentially occurs in biomembranes. This marked Indo-induced change in phase behavior potentially alters membrane protein functions, which contribute to the wide variety of biological activities of Indo and other NSAIDs.

While cancer is one of the greatest challenges to public health care, prostate cancer was chosen as cancer model to develop a more accurate imaging assessment than those currently available. Indeed, an efficient imaging technique which considerably improves the sensitivity and specificity of the diagnostic and predicting the cancer behavior would be extremely valuable. The concept of optoacoustic imaging using home-made functionalized gold nanoparticles coupled to an antibody targeting PSMA (prostate specific membrane antigen) was evaluated on different cancer cell lines to demonstrate the specificity of the designed platform. Two commonly used microscopy techniques (indirect fluorescence and scanning electron microscopy) showed their straightforwardness and versatility for the nanoparticle binding investigations regardless the composition of the investigated nanoobjects. Moreover most of the research laboratories and centers are equipped with fluorescence microscopes, so indirect fluorescence using Quantum dots can be used for any active targeting nanocarriers (polymers, ceramics, metals, etc.). The second technique based on backscattered electron is not only limited to gold nanoparticles but also suits for any study of metallic nanoparticles as the electronic density difference between the nanoparticles and binding surface stays high enough. Optoacoustic imaging was finally performed on a 3D cellular model to assess and prove the concept of the developed platform.
