BACKGROUND: Recombinant protein production is universally employed as a solution to obtain the milligram to gram quantities of a given protein required for applications as diverse as structural genomics and biopharmaceutical manufacture. Yeast is a well-established recombinant host cell for these purposes. In this study we wanted to investigate whether our respiratory Saccharomyces cerevisiae strain, TM6*, could be used to enhance the productivity of recombinant proteins over that obtained from corresponding wild type, respiro-fermentative strains when cultured under the same laboratory conditions. RESULTS: Here we demonstrate at least a doubling in productivity over wild-type strains for three recombinant membrane proteins and one recombinant soluble protein produced in TM6* cells. In all cases, this was attributed to the improved biomass properties of the strain. The yield profile across the growth curve was also more stable than in a wild-type strain, and was not further improved by lowering culture temperatures. This has the added benefit that improved yields can be attained rapidly at the yeast's optimal growth conditions. Importantly, improved productivity could not be reproduced in wild-type strains by culturing them under glucose fed-batch conditions: despite having achieved very similar biomass yields to those achieved by TM6* cultures, the total volumetric yields were not concomitantly increased. Furthermore, the productivity of TM6* was unaffected by growing cultures in the presence of ethanol. These findings support the unique properties of TM6* as a microbial cell factory. CONCLUSIONS: The accumulation of biomass in yeast cell factories is not necessarily correlated with a proportional increase in the functional yield of the recombinant protein being produced. The respiratory S. cerevisiae strain reported here is therefore a useful addition to the matrix of production hosts currently available as its improved biomass properties do lead to increased volumetric yields without the need to resort to complex control or cultivation schemes. This is anticipated to be of particular value in the production of challenging targets such as membrane proteins.

Spray-dried materials are being used increasingly in industries such as food, detergent and pharmaceutical manufacture. Spray-dried sodium carbonate is an important product that has a great propensity to cake; its moisture-sorption properties are very different to the crystalline and amorphous species, with a great affinity for atmospheric moisture. This work demonstrates how the noncontact surface analysis of individual particles using atomic force microscopy can highlight the possible mechanisms of unwanted agglomeration. The nondestructive nature of this method allows cycling of localised humidity in situ and repeated scanning of the same particle area. The resulting topography and phase scans showed that humidity cycling caused changes in the distribution of material phases that were not solely dependent on topographical changes. © 2013 WILEY-VCH Verlag GmbH & Co. In 2016, the order Mononegavirales was emended through the addition of two new families (Mymonaviridae and Sunviridae), the elevation of the paramyxoviral subfamily Pneumovirinae to family status (Pneumoviridae), the addition of five free-floating genera (Anphevirus, Arlivirus, Chengtivirus, Crustavirus, and Wastrivirus), and several other changes at the genus and species levels. This article presents the updated taxonomy of the order Mononegavirales as now accepted by the International Committee on Taxonomy of Viruses (ICTV).

La anemia por deficiencia de hierro es uno de los diagnósticos más comunes a nivel mundial, afecta tanto a niños como adultos y tiene muy diversas causas. Aparece cuando la ingestión de hierro es inadecuada para cumplir un nivel estándar de demanda, cuando aumentan los requerimientos de hierro o hay una perdida crónica de hemoglobina. El objetivo de la investigación fue determinar anemia por deficiencia de hierro en niños de 8 a 10 años de edad que estudian en el Complejo Educativo Rancho Quemado, Cantón Casa Blanca, municipio de Perquín en el periodo de julio a septiembre de 2014. Para lo cual la metodología que se utilizó fue de tipo prospectiva, transversal, descriptiva y de laboratorio. La población fue constituida por 81 niños y niñas entre 8 a 10 años de edad a los cuales se les realizó las pruebas de laboratorio siguientes: Hemograma completo, conteo de reticulocitos, frotis de sangre periférica, hierro sérico y capacidad de captación del hierro. Resultados obtenidos: en la determinación de hematocrito, hemoglobina e índices hematimétricos no hubo población con valores disminuidos, en la lectura del Frotis de sangre periférica, las células se encontraron con normalidad en el 100% de la población, en el conteo de reticulocitos no hubo disminución , encontrándose normal en el 100% de la población, para la determinación de hierro sérico un niño (1.25%) y una niña (1.25%)de la población resultó con hierro disminuido, capacidad de captación de hierro aumentada y porcentaje de saturación de transferrina disminuida, según la clasificación de las etapas de la anemia el 2.5% (un niño y una niña) de la población se encontró con niveles bajos de hierro sérico, aumento de la capacidad de captación de hierro, y disminución en el porcentaje de saturación de transferrina. Conclusión: En la edad de 8 años no se encontró afectación aparente, resultando con deficiencia de hierro en la edad de 9 años 1.25% y en la edad de 10 años 1.25%.

En el presente trabajo de investigación se refleja de forma sistematizada los elementos que han sido tomados en cuenta en este estudio sobre Los procesos y procedimientos del Centro Escolar Dr. Arturo Romero Nº 2 del distrito 06 -26 del Municipio de Ayutuxtepeque en la formación en valores en los estudiantes del Tercer Ciclo de Educación Básica del Turno Vespertino. Resulta importante destacar la utilidad que tienen los procesos y procedimientos en la enseñanza y aprendizaje de los valores ya que estos contribuyen a la formación moral e integral de los educandos, capacitándolos para que desarrollen conciencia y sean capaces de ponerlos en práctica en su cotidianeidad y ser útil en la sociedad.

Background and Purpose—As a research community, we have failed to demonstrate that drugs which show substantial efficacy in animal models of cerebral ischemia can also improve outcome in human stroke. Summary of Review—Accumulating evidence suggests this may be due, at least in part, to problems in the design, conduct and reporting of animal experiments which create a systematic bias resulting in the overstatement of neuroprotective efficacy. Conclusions—Here, we set out a series of measures to reduce bias in the design, conduct and reporting of animal experiments modeling human stroke.

Este projecto faz uma abordagem à nova museologia e ao seu papel no mercado do turismo, com especial enfoque nos serviços educativos dos museus portugueses e na sua relação com o turismo cultural. O estudo do património artístico e cultural do Museu da Tapeçaria 1e Portalegre Guy Fino e da Manufactura de Tapeçarias de Portalegre, bem como a análise crítica dos serviços educativos de museus importantes a nível nacional e internacional, serviram de suporte à realização do projecto de criação de serviços educativos no referido Museu. O projecto tem como principal objectivo qualificar a oferta turístico-cultural que as Tapeçarias de Portalegre representam e está estruturado de forma a poderem ser desenvolvidas actividades em rede com os outros dois museus da cidade, o Museu Municipal e a Casa Museu José Régio. Foram escolhidas, como áreas temáticas, a História, a História da Arte e a Literatura. As actividades propostas para as áreas temáticas, são de três tipos: visitas dinamizadas, visitas-oficina e vistas-jogo, podendo algumas delas ser desenvolvidas on-line, de forma interactiva. Todas as actividades propostas exploram conteúdos programáticos que integram os planos curriculares do 1° Ciclo do Ensino Básico, podendo algumas delas ser desenvolvidas também por alunos do 2° Ciclo e por famílias de diferentes nacionalidades. O projecto constitui um primeiro passo para uma progressiva transformação das Tapeçarias de Portalegre numa atracção âncora do turismo cultural regional, sem perder de vista as suas inquestionáveis potencialidades de se tornar numa atracção de âmbito nacional e internacional. ABSTRACT: This project makes an approach to the new museology and its role on tourist market, with special focus on the educational services in portuguese museums and its relationship with cultural tourism. The investigation about the artistic and cultural patrimony of Guy Fino Museum and the Manufacture, as well as the critic analysis of educational services in important national and international museums, supported the present project of creation of educational services for Guy Fino Museum. The defined main objective of the project is qualifying the touristic and cultural offer represented by the Tapestries of Portalegre. lt is structured in a way that allows activities to be developed in network with other museums in the city: The Municipal Museum and the Museum House of Jose Regio. ln this way, the following areas were chosen: History, History of Arte and Literature. The activities proposed for the themes are three types: dynamic visits, visits - hands on and visits - games, some being developed on-line, in an interactive way. Twelve activities were conceptualized. Ali the proposed activities explore programmed contents that integrate curricular plans of primary school. Some of the activities can also be developed by students of secondary school and families. The translation of the material used should be in a way that they can be used and developed by foreign visitors. The project constitutes a first step for a progressive transformation of the Tapestries of Portalegre in an anchor attraction of the regional cultural tourism, being sure of their unquestionable potentialities to become a national and international attraction.

This research challenges the origin story of neoliberalism in Latin America. Drawing on archival data from the Mont Pèlerin Society and the personal archives of leading but neglected figures in the post-war push to rebuild economic liberalism, I present a historical geography of elite counter-protest that both predates and broadens the generally accepted “birth” of neoliberalism in 1970s Chile. Beginning in the 1940s, Latin American elites found common cause with key figures from economic liberalism’s most radical wing: the Austrian School. While existing literature links the onset of neoliberalism in Chile to the Austrian School, particularly with respect to the School’s influence on the early Mont Pèlerin Society, this dissertation is the first comprehensive inquiry to place the Austrian tradition in the ideational and organizational landscape of Latin America. Embracing a new mission that promised to save the soul of Western civilization, Latin America’s retro-neoliberal leaders collaborated with transnational actors to build a network of Austrian-inspired think-tanks and institutes of higher learning in the region. These organizations, in turn, served as recruiting mechanisms to found the Hispanic quarter of the Mont Pèlerin Society, which was dominated not (as might be assumed) by Chileans, but rather by retro-neoliberal elites from Mexico, Argentina, Guatemala, and Venezuela. By 1975, when scholars began analyzing how a run-of-the-mill economics department had been transformed into a bastion of free-market thinking in Chile, an entire neoliberal university was up and running in Guatemala, exposing all students, regardless of discipline, to the Austrian tradition – the crowning achievement of Latin America’s retro-neoliberal network. Investigating, and accounting for, the development and impact of this initiative sheds new light on the neoliberal landscape in Latin America, and raises important questions for the study of neoliberalism more broadly.

Off-site manufacture (OSM) offers numerous benefits to all parties in the construction process. The uptake of OSM in Australia has, however, been limited. This limited uptake corresponds to similar trends in the UK and US, although the level of OSM there appears to be increasing. This project undertook three workshops — one each in Victoria, Queensland and Western Australia — and 18 interviews with key stakeholders to assist in identifying the general benefits and barriers to OSM uptake in the Australian construction industry. Seven case studies were also undertaken, involving construction projects that used OSM, ranging from civil projects through to residential. Each of these case studies has been analysed to identify what worked and what didn’t, and suggest the lessons to be learned from each project.

The revival of terracotta and faience in British architecture was widespread, dramatic in its results and, for two decades, the subject of intense debate. However the materials have been frequently denigrated and more generally disregarded by both architects and historians. This study sets out to record and explain the rise and fall of interest in terracotta and faience, the extent and nature of the industry and the range of architectural usage in the Victorian, Edwardian and inter-war periods. The first two chapters record the faltering use of terracotta as an 'artificial stone', until the material gained its own identity, largely through the appreciation of Italian architecture. In the mid-Victorian period, terracotta will be seen to have become symbolic of the philosophy of the Victoria and Albert Museum and its Art School in attempting to reform both architecture and the decorative arts. The adoption of terracotta was furthered as much by industrial as aesthetic factors; three chapters examine how the exploitation of coal-measure clays, developments in the processes of manufacture, the changing motivation of industrialists and differing economics of production served to promote and then to hinder expansion and adaptation. The practical values of economy, durability and fire-resistance and the aesthetic potential, seen in terms of colour and decorative and sculptural modelling, became inter-related in the work of the architects who made extensive use of architectural ceramics. A correlation emerges between the free Gothic style, exemplified by the designs of Alfred Waterhouse and the use of red terracotta supplied from Ruabon, and between the eclectic Renaissance style and a buff material produced by different manufacturers.These patterns were modified as a result of the adoption of faience for facing external walls as well as interiors, and because of the new architectural requirements and tastes of the twentieth century. The general timidity in exploiting the scope for polychromatic decoration and the increasing opposition to architectural ceramics is contrasted with the most successful schemes produced for cinemas, chain-stores and factories. In the last chapter, those undertaken by the Hathern Station Brick and Terracotta Company between 1896 and 1939 are used as a case study; they confirm that manufacturers, architects and clients were all committed to creating a modern and yet decorative architecture, appropriate for new building types and that would appeal to and be comprehensible to the public.

Wynne and Schaffer (2003) have highlighted both the strong growth of gambling activity in recent years, and the revenue streams this has generated for governments and communities. Gambling activities and the revenues derived from them have, unsurprisingly, therefore also been seen as a way in which to increase economic development in deprived areas (Jinkner-Lloyd, 1996). Consequently, according to Brown et al (2003), gambling is now a large taxation revenue earner for many western governments, at both federal and state levels, worldwide (for example UK, USA, Australia). In size and importance, the Australian gambling industry in particular has grown significantly over the last three decades, experiencing a fourfold increase in real gambling turnover. There are, however, also concerns expressed about gambling and Electronic Gaming in particular, as illustrated in economic, social and ethical terms in Oddo (1997). There are also spatial aspects to understanding these issues. Marshall’s (1998) study, for example, highlights that benefits from gambling are more likely to accrue at the macro as opposed to the local level, because of centralised tax gathering and spending of tax revenues, whilst localities may suffer from displacement of activities with higher multipliers than the institutions with EGMs that replace them. This also highlights a regional context of costs, where benefits accrue to the centre, but the costs accrue to the regions and localities, as simultaneously resources leave those communities through both the gambling activities themselves (in the form of revenue for the EGM owners), and the government (through taxes).
