A fast neutron-mutagenized population of Arabidopsis ( Arabidopsis thaliana) Columbia-0 wild-type plants was screened for floral phenotypes and a novel mutant, termed hawaiian skirt ( hws), was identified that failed to shed its reproductive organs. The mutation is the consequence of a 28 bp deletion that introduces a premature amber termination codon into the open reading frame of a putative F-box protein ( At3g61590). The most striking anatomical characteristic of hws plants is seen in flowers where individual sepals are fused along the lower part of their margins. Crossing of the abscission marker, Pro(PGAZAT):beta-glucuronidase, into the mutant reveals that while floral organs are retained it is not the consequence of a failure of abscission zone cells to differentiate. Anatomical analysis indicates that the fusion of sepal margins precludes shedding even though abscission, albeit delayed, does occur. Spatial and temporal characterization, using Pro(HWS):beta-glucuronidase or Pro(HWS):green fluorescent protein fusions, has identified HWS expression to be restricted to the stele and lateral root cap, cotyledonary margins, tip of the stigma, pollen, abscission zones, and developing seeds. Comparative phenotypic analyses performed on the hws mutant, Columbia-0 wild type, and Pro(35S):HWS ectopically expressing lines has revealed that loss of HWS results in greater growth of both aerial and below-ground organs while overexpressing the gene brings about a converse effect. These observations are consistent with HWS playing an important role in regulating plant growth and development.

The development of novel intervention strategies for the control of zoonoses caused by bacteria such as Salmonella spp. in livestock requires appropriate experimental models to assess their suitability. Here, a novel porcine intestinal in vitro organ culture (IVOC) model utilizing cell crown (CC) technology (CCIVOC) (Scaffdex) was developed. The CCIVOC model was employed to investigate the characteristics of association of S. enterica serovar Typhimurium strain SL1344 with porcine intestinal tissue following exposure to a Lactobacillus plantarum strain. The association of bacteria to host cells was examined by light microscopy and electron microscopy (EM) after appropriate treatments and staining, while changes in the proteome of porcine jejunal tissues were investigated using quantitative label-free proteomics. Exposure of porcine intestinal mucosal tissues to L. plantarum JC1 did not reduce the numbers of S. Typhimurium bacteria associating to the tissues but was associated with significant (P < 0.005) reductions in the percentages of areas of intestinal IVOC tissues giving positive staining results for acidic mucins. Conversely, the quantity of neutrally charged mucins present within the goblet cells of the IVOC tissues increased significantly (P < 0.05). In addition, tubulin- was expressed at high levels following inoculation of jejunal IVOC tissues with L. plantarum. Although L. plantarum JC1 did not reduce the association of S. Typhimurium strain SL1344 to the jejunal IVOC tissues, detection of increased acidic mucin secretion, host cytoskeletal rearrangements, and proteins involved in the porcine immune response demonstrated that this strain of L. plantarum may contribute to protecting the pig from infections by S. Typhimurium or other pathogens.

The role of ethylene in regulating organ senescence in Arabidopsis has been investigated by studying the development of mutants that have an attenu- ated capacity to perceive the gas. The onset of leaf senescence and floral organ abscission was delayed in the ethylene-insensitive mutant etr1. The photosynthetic life span of rosette leaves was similarly extended in the gain- of-function mutant ers2, and this mutant also exhibited a delay in the timing of pod dehiscence primarily as a con- sequence of an extension in the final stages of senescence. A detailed analysis of yield revealed that whilst thousand grain weight was increased, by as much as 20 %, in etr1, ein4, and the loss-of-function mutant etr2, only the latter showed a significant increase in total weight of seeds produced per plant. The other studied mutants exhibited a reduction in total seed yield of almost 40 %. These observations are discussed in the context of the possible role of ethylene in regulating organ senescence and their significance in the breeding of crop plants with enhanced phenotypic characteristics.

Two new species of Pamphobeteus Pocock 1901 are described from Brazil: Pamphobeteus crassifemur sp. nov. and Pamphobeteus grandis sp. nov.; Pamphobeteus nigricolor, formerly described from Colombia, Ecuador and Bolivia, is recorded from Brazil and its distribution in Ecuador and Bolivia is questioned. A new type of stridulatory organ is described from legs III and IV of P. crassifemur sp. nov. The structure consists of spiniform setae. Stridulation occurs when the spider moves the legs III and IV, sometimes while shedding urticating hairs.

This paper described the production of a novel biosynthetic material using the manufacturing technique of electro spinning for the construction of scaffold for organ replacement. This electrostatic technique uses an electric field to control the deposition of polymer fibres onto a specific substrate to fabricate fibrous polymer constructs composed of fibre diameters ranging from several microns down to 100 nm or less. Two areas of research, in particular, heart valve leaflets and blood vessel will be discussed. Here, a sandwich structure nanofibre mesh was used to construct materials for leaflets of heart valve and blood vessel. In the case of heart valve leaflet, the randomly oriented polyurethane nanofibres were prepared as the first layer, followed by gelatin-chitosan complex layer. Complex nanofibres were initially used to spin on the PU layer with cross orientation to mimic the fibrosa layer. A gelatin and chitosan complex was then spun onto the other side of PU nanofibre mesh to mimic the ventricularis layer. This particular sandwich structure using the PU layer was designed to simulate the mechanical properties of natural tissue. In addition, this design was aimed to provide good biocompatibility and improved cellular environment to assist in adhesion and proliferation. Smooth muscle cells adhered and flattened out onto the surface of the gelatin-chitosan complex as early as 1 day post seeding. There is great potential for this biosynthetic biocompatible nanofibrous material to be developed for various clinical applications.

UK statistics show that whilst waiting lists for transplantation surgery continue to increase, donor numbers are static. This paper describes the hermeneutic phase of a mixed method study and puts forward the concept of protection behaviour as one explanation for nurses’ reticence to discuss post-mortem donation wishes with patients’ relatives. The desire to protect appears to influence attitudes, confidence levels and perceived ability to become involved in donor identification and donation discussion, consequently affecting the availability of transplantable organs and tissue. By understanding more fully why protective behaviours are employed, it increases the likelihood of a solution being found.

This theory-building research suggests that the key to unlocking Australia's potential organ donor pool is to reduce the ethical ambiguity and legal uncertainty plaguing physicians' interepretation of the patients's best interests standard. This may generate a greater acceptance of organ donation as part of end-of-life patient care amongst our physicians.

Next of kin who are aware of the deceased's organ donation wishes usually will honor those wishes, while next of kin who are unaware of these wishes typically withhold consent for posthumous donation. Encouraging individuals to communicate or register their organ donation wishes is therefore important. Using a sample of 409 participants, the current study sought to develop a profile of Australian adults who had communicated their organ donation wishes to family members. Christian participants and those who had a higher income were more likely to have communicated their donation wishes. Conversely, participants were less likely to have communicated their donation wishes if they were unregistered and undecided/opposed to organ donation, unregistered but willing to donate, or fearful of death. Finally, whether participants had communicated, registered, or communicated and registered their donation wishes was associated with their age, religion, attitude toward organ donation, and recall of media content about organ donation. Messages encouraging the communication of organ donation wishes to family members should therefore be targeted toward those individuals who are most likely to be receptive toward enacting this behavior.

This paper considers the practice of learning-by-heart and argues for its relevance to learning, to thought (as defined by Gilles Deleuze) and as a way of turning towards the ‘new’ or ‘the future’, via the operation of repetition. It considers two modes in which rote learning can be productive and provocative—firstly, when the content itself is something worth retaining, and secondly, when the actual process of the learning itself and then the repeating align themselves with the criteria of ‘practice’, as framed by the author here. In the face of rote learning’s reputation as an out-moded pedagogical tool, the paper argues that it inhabits a paradoxical and productive site, whereby what begins as a repetition of the same, can open towards pure repetition (as Deleuze frames this notion), and facilitate inventiveness and a courting of the new. In this way, poetry, and the learning of it by rote, constitute a unique constellation, disputing the platitude that learning is ‘only discovering what one already knew’ and instead proposing that learning is closer to an awesome ordeal, one that leads to concepts and collisions that did not exist before and cannot be predicted in advance.
