O objetivo do experimento foi testar a eficácia de diferentes diluidores, a base de Ringer-Lactato, Leite Desnatado, TRIS (hidroxi-methil-amino-methan) e Lactose/TRIS, na criopreservação de sêmen bubalino. Foram utilizados três machos bubalinos da raça Murrah em plena atividade sexual. O sêmen foi colhido por vagina artificial totalizando 71 ejaculados. Após a colheita, cada amostra foi submetida às análises qualitativas e quantitativas do sêmen. Os ejaculados foram fracionados e diluídos nos quatro diluidores. O sêmen diluído foi envasado em palhetas de 0,25ml e submetidos a um tempo de equilíbrio de até quatro horas a 5ºC, com posterior congelação em nitrogênio líquido. As amostras identificadas foram descongeladas em banho maria à temperatura de 40°C por 30 segundos e seqüencialmente avaliadas quanto a motilidade, vigor, lesão de acrossoma, e percentual de patologias espermáticas. As amostras também foram submetidas ao teste de termo-resistência, permanecendo incubadas à temperatura de 40°C durante 30 segundos, 3-5 minutos, 30 minutos, 1 hora, 2 horas e 3 horas, onde foram avaliadas quanto a motilidade e o vigor espermático. As características físicoquímicas, após análise do sêmen in natura, encontraram-se dentro dos valores preconizados para a espécie bubalina e satisfatórios para o processo de congelação. Após descongelação do sêmen, observou redução numérica estatística (p<0,05) na motilidade espermática, sendo que no sêmen in natura se observou 86,67±6,17% reduzindo para 70±6,92% em TRIS, 67,4±8,01% em Ringer/lactato, 67,09±9,03% em Lactose/TRIS e 59,7±9,05% em leite desnatado e ao comparar entre os quatro tratamentos apenas o Leite desnatado mostrou diferença estatística significativa (p<0,05). Após descongelação, o vigor espermático também diminuiu estatisticamente (p<0,05) nos quatro tratamentos (3,50±0,53 TRIS; 3,38±0,49 Ringer-lactato; 3,3±0,46 Lactose/TRIS e 3,25±0,44 Leite desnatado) versus (4±0,39 sêmen in natura) e ao comparar entre os tratamento, apenas entre Leite desnatado e TRIS se observou diferença estatística (p<0,05). Quanto aos defeitos maiores (4,15±1,9% sêmen in natura; 10,51±4,4% TRIS; 11,94±4,2% Ringer-Lactato; 11,88±4,8% Lactose/TRIS; 12,01±5% Leite desnatado), menores (3,81±1,2% sêmen in natura; 4,67±1,1% TRIS; 4,98±1,7% Ringer-Lactato; 4,93±2,0% Lactose/TRIS; 4,93±2,0% Leite desnatado) e totais (7,91±2,1% sêmen in natura; 15,18±4,7% TRIS; 16,92±4,8% Ringer-Lactato; 16,82±5,6% Lactose/TRIS; 17,11±5,6% Leite desnatado), após descongelação houve aumento significativo dos defeitos nos quatro tratamentos (p<0,05), e entre eles não foram observadas diferenças estatísticas entre si (p>0,05). Na fase pós-TTR, após 3 horas de incubação, a motilidade progressiva (TRIS 21,13±7,5%; Ringer-Lactato 20,78±7,4%; Lactose/TRIS 20,25±5,3%; Leite desnatado 20,12±6,6%) e vigor espermático (TRIS 2,04±0,5; Ringer-Lactato 2,07±0,5; Lactose/TRIS 2,02±0,4; Leite desnatado 2,00±0,5) não apresentaram diferença estatística entre os tratamentos (p<0,05). Quanto a intergridade do acrossoma, após a descongelação (Sêmen in natura 97,85±0,6%; TRIS 91,65±4,3%; Ringer-Lactato 90,46±4,8%; Lactose/TRIS 89,76±5,4%; Leite desnatado 90,56±5,6%), houve diminuição estatística (p<0,05) e quando se comparou tal parâmetro entre os tratamentos não foi observado diferenças estatísticas significativas entre os tratamento (p>0,05). Diante dos resultados observados é possível concluir que a congelação de sêmen de búfalo com os diluidores TRIS (Trishydroxy- methyl-amino-methan), Ringer-Lactato, Lactose/TRIS, e Leite desnatado mostraram satisfatória função de crioproteção na viabilidade espermática do sêmen nas diferentes etapas da criopreservação.

O objetivo deste trabalho foi avaliar a característica do queijo de coalho, produzido a partir do leite bovino pasteurizado, mediante a utilização de bactérias láticas mesofílicas do gênero Lactococcus lactis ssp. cremoris e Lactococcus lactis ssp. lactis específicas. As culturas láticas oriundas do Banco de Bactérias Láticas da Universidade Estadual do Ceará foram ativadas junto às instalações do Laboratório de Bactérias Láticas da Universidade Federal Rural da Amazônia-UFRA, Campus de Belém. As culturas láticas foram ativadas durante três dias consecutivos em Leite Desnatado Reconstituído (LDR) 12% esterilizado e incubadas a 30 °C ± 2 °C, até a coagulação do leite. Após reativação, a cultura industrial foi obtida pela transferência do inoculo de 1% (v/v) para frascos de vidros contendo 500 ml de LDR 12% esterilizado, seguida de incubação a 30 °C ± 2 °C até a coagulação do leite, em seguida a cultura (fermento lático) foi adicionada diretamente no tanque de fabricação contendo o leite pasteurizado, mantendo-se a proporção de 1:1. Para avaliação tecnológica foram utilizados as seguintes culturas láticas isoladas de leite cru: Lactococcus lactis ssp. lactis (LL); Lactococcus lactis (atípico) (LLA); Lactococcus lactis ssp. cremoris (atípico) (LLCA); Lactococcus lactis ssp. cremoris (LLC),. As porções de Amostras foram retiradas, colocadas em processador de alimentos e processadas até formar uma amostra. Em seguida, foram acondicionadas em frascos estéreis, identificadas e mantidas em freezer para posterior análises de determinação do extrato seco, umidade (%), extrato seco total (EST), gordura (G), gordura no extrato seco (GES), acidez, pH, cloretos, nitrogênio total (NT), nitrogênio solúvel em pH 4,6, nitrogênio solúvel em TCA 12%. O índice de proteólise ou extensão da maturação foi avaliado pela divisão do NT. Para o teste de aceitação utilizou-se a escala hedônica estruturada de nove pontos, para avaliar o produto quanto ao aroma, aspecto geral, gosto e textura. O teste de fritura de acordo com metodologia descrita por Cavalcante et al., (2007). As análises microbiológicas das amostras de queijos experimentais nos 1º e 30º dia de maturação, encaminhadas ao Laboratório Central – LACEN, Divisão de Análises de Produtos – DEP. E consistiram em Contagem de bactérias Aeróbias Mesófilas, Determinação de Coliformes. Para o teste de fritura não houve análise estatística. O delineamento utilizado foi o Inteiramente Casualisado e foi utilizada a metodologia de modelos mistos para dados longitudinais, com objetivo de modelar a estrutura de (co)variância entre medidas coletadas na mesma unidade experimental em tempos diferentes, por meio do modelo yijk=μ+αi+ δ(i)k+ α βikijk. Utilizando-se o programa estatístico Statistical Analysis Systems - SAS (SAS INSTITUTE INC., 1992). Os tratamentos LL e LLA foram reprovados no teste de fritura. Houve ligação entre a característica derretimento com a umidade, acidez e proteólise. Os queijos que apresentaram maiores valores de proteólise apresentaram maior capacidade de derretimento. As amostras de queijo coalho tiveram boa aceitabilidade no teste de aceitação.

This work consists of the implementation of the steps Define, Measure , Analyze , Improve and Control ( DMAIC ) to develop a Six Sigma project in an industry the food industry. The objective was to demonstrate a potential for reducing the occurrence of crushed cans in potting milk powder industry in a White Belt project. The food industry accounts for about 9 % of Brazil's gross domestic product ( GDP ), generating thousands of jobs . Among the major sectors of the food industry is the manufacturing sector of milk , occupies approximately 10 % of the total turnover of the food industry . Brazil is considered today one of the eight largest producers of milk powder in the world. The milk powder is packed , mostly for aluminum cans that are lined internally with varnishes and other materials to protect the milk of metals from aluminum. When the cans are dented food protection is compromised and may lead ingestion causing dis-eases such as botulism. Aiming to solve the problem of dented cans methodology was used as a case study with a quantitative approach through the DMAIC method. Some quality tools used in each step of the project as brainstorming , cause and effect diagram , flowchart , ef-fort and impact matrix, 5W1H , among other Pareto diagram is presented . A survey about the disposal of cans in the company verifying a mean loss and, from this histor-ical , a goal loss was calculated was performed . With the target set we calculated the annual saving design . During application of DMAIC was found that the highest rate of loss occurred in transportation between the factory and the factory that fills cans milk . Several actions were taken to resolve problems that resulted in dented cans and the first two months of phase control it was found that the smaller losses calculated target resulting in a saving for the company. The penis and prepuce of the stallion have a high bacterial load on its surface, forming a natural microbial flora that contaminates the semen during ejaculation. Bacterial growth in semen may cause a decline on sperm quality, viability, and fertility and predisposes the occurrence of endometritis in inseminated mares. Thus, the aim of this study was to evaluate the effect of penile wash before semen collection, the addition of different commercial skim milk-based extenders containing antibiotics (BotuSemen and INRA96), and the removal of seminal plasma by filtration on the quality, viability, and bacterial proliferation on fresh and cooled stallion semen. Animals that were never submitted to penile wash before semen collection tended to have lower bacterial contamination in the ejaculate. Semen samples extended in BotuSemen showed superiority in total motility, progressive motility, average path velocity, and rapid sperm and lower bacterial contamination in relation to semen samples extended in INRA96 after 24 hours of cooling. No difference was found in these parameters between the storage temperatures (5 degrees C and 15 degrees C). Furthermore, the removal of seminal plasma by filtration reduced the bacterial load in semen after cooling. In conclusion, the penile wash before semen collection tended to reduce the bacterial growth in fresh semen. The use of a semen extender with appropriate antibiotics and removal of seminal plasma by filtration were effective in reducing the bacterial contamination and preserved the quality of cooled stallion semen. The development of a reliable technique to freeze epididymal semen would provide a unique opportunity to preserve valuable genetic material from unexpectedly lost stallions. The aim of this study was to compare the apoptotic indices of sperm obtained from ejaculate, sperm recently recovered from the epididymides (EP), and sperm recovered from epididymides stored at 5 C for 24 hours (EP-stored). For the first category, two ejaculates from seven stallions were collected and then submitted to cryopreservation using an egg yolk-based extender. One week after the last semen collection, the stallions were submitted to bilateral orchiectomy, and sperm from one of the cauda epididymis was harvested immediately after castration (EP). The remaining testicle was stored in a passive refrigeration container at 5 C for 24 hours before the cauda epididymal sperm was harvested (EP-stored). Sperm harvesting from the epididymis for EP and EP-stored was performed by retrograde flushing of the caudal portion of the epididymis using a skim milk-based extender. The recovered sperm was then cryopreserved using the egg yolk-based extender. Sperm motility parameters were studied by computerassisted semen analysis, and apoptosis was estimated by measuring caspase activity and membrane phospholipid translocation using epifluorescence microscopy. The samples were evaluated immediately (0 hour) and 8 hours after thawing. At 0 hour, no differences in sperm parameters were observed among the groups, but after 8 hours, significant statistical differences were observed in sperm motility parameters and plasma membrane integrity among the treatment groups. In addition, viable cells with no apoptotic signs were more prevalent in EP and EP-stored, suggesting that epididymal sperm is less sensitive to the cold shock caused by sperm cryopreservation.

The organic acids profile, sugar metabolism and biomass growth of Streptococcus thermophilus (St) and Bifidobacterium lactis (BI) have been studied in pure cultures or binary co-culture (St-BI) in skim milk either containing 40 mg/g of inulin or not. With inulin, the time required by St. BI and St-BI to complete fermentation (i.e., when the pH reached 4.5) was about 14, 8 and 49% shorter than without inulin, respectively. This prebiotic also enhanced the levels of lactic and acetic acids and volatile compounds, showing a positive synbiotic effect between pre- and probiotics. In particular, the St-BI co-culture showed final concentrations of both microorganisms about 15 and 38% higher than in their respective pure cultures, thus highlighting a clear synergistic effect between these microorganisms due to mutual interactions. This study evaluated the effect of the supplementation of total dietary fiber from apple, banana or passion fruit processing by-products on the post-acidification, total titratable acidity, bacteria counts and fatty acid profiles in skim milk yoghurts co-fermented by four different probiotics strains: Lactobacillus acidophilus L10 and Bifidobacterium animalis subsp. lactis BL04, HN019 and B94. Apple and banana fibers increased the probiotic viability during shelf-life. All the fibers were able to increase the short chain and polyunsaturated fatty acid contents of yoghurts compared to their respective controls. A synergistic effect between the type of fiber and the probiotic strain on the conjugated linoleic acid content was observed, and the amount of alpha-linolenic acid was increased by banana fiber. The results of this study demonstrate, for the first time, that fruit fibers can improve the fatty acid profile of probiotic yoghurts and point out the suitability of using fibers from fruit processing the by-products to develop new high value-added fermented dairy products. Metabolic studies are very important to improve quality of functional dairy products. For this purpose, the behaviors of pure cultures of Streptococcus thermophilus (St) and Lactobacillus rhamnosus (Lr) as well a co-culture of them (St-Lr) were investigated during skim milk fermentation, and the inulin effect as prebiotic was assessed. Lr was able to metabolize 6 g/100 g more galactose than St and St-Lr. Final lactic acid production by Lr was higher (9.8 g/L) compared to St (9.1 g/L) and St-Lr (9.1 g/L). Acetic acid concentration varied from 0.8 g/L (St-Lr) to 1.5 g/L (Lr) and that of ethanol from only 0.2 g/L (St-Lr) to 0.4 g/L (Lr), which suggests the occurrence in Lr of a NADH oxidase activity and citrate co-metabolization via pyruvate, both dissipating a part of the reducing power. Diacetyl and acetoin accumulated at the highest levels (18.4 and 0.8 mg/L, respectively) with St-Lr, which suggests possible synergistic interactions between these microorganisms as well as the Lr capability of co-metabolizing citrate in the presence of lactose. Inulin stimulated both biomass growth and levels of all end-products, as the likely result of fructose release from its partial hydrolysis and subsequent metabolization as an additional carbon and energy source.
