Seed reserve mobilization during germination of the Amazonian species Myciaria dubia (camu-camu), Eugenia stipitata(araca-boi), Dipteryx odorata (cumaru) and Hymenaea courbaril (jatoba) was evaluated. Seeds were placed in germination chambers at 30 degrees C with it 12 h photoperiod. Analysis of primary metabolites (carbohydrates, lipids and proteins) and fatty acid composition were carried out in quiescent seeds and at four germination stages after radicle protrusion. Germination was high in all species but there were statistically significant differences between species. Differences were also observed with regard to the duration of the germination period. The seeds showed variation in the content and composition of the analyzed compounds. indicating that the mobilization rates of these compounds may affect germination velocity.

Seeds sprouts have been used as a good source of basic nutrients and nutraceutical compounds. The high nutritional value of seeds derives from the deposition of compounds during development. However some of these molecules are used in metabolic processes like germination, which leads to a considerable variation in their concentrations once these events are completed. In this work, we investigate the levels of inositols (myo-inositol, D-pinitol and ononitol), soluble carbohydrates and proteins in cotyledons of Phaseolus vulgaris and Vigna unguiculata sprouts. Sprouting increased myo-inositol and glucose content and reduction of raffinose and ononitol was observed. The protein levels increased in P. vulgaris and decreased in V. unguiculata sprouting. The level of sucrose was maintained in both sprouts. D-Pinitol was detected only in quiescent seeds. Our results suggested that bean sprout is an important source of proteins, sucrose, glucose and myo-inositol. Additionally, bean sprouts have low levels of raffinose, an antinutritional compound.

Primary cultures of vascular smooth muscle cells (VSMCs) from rats offer a good model system to examine the molecular basis of mechanism of vascular contraction-relaxation. However, during pathological conditions such as atherosclerosis and hypertension, VSMCs characteristically exhibit phenotypic modulation, change from a quiescent contractile to a proliferative synthetic phenotype, which impairs this mechanism of vascular contraction-relaxation. Taking in account that Myosin light chain (MLC) and ERK1/2 directly participate in the process of vascular contraction, the aim of the current study was to analyze the involvement of MLC and ERK1/2 signaling during the process of VSMCs phenotypic modulation. Primary cultures of VSMCs from rat thoracic aortas were isolated and submitted to different number of passages or to freezing condition. Semi-quantitative RT-PCR was used to evaluate the mRNA levels of VSMCs differentiation markers, and western blot assays were used to determine the MLC and ERK1/2 phosphorylation levels during VSMCs phenotypic modulation. Also, immunocytochemical experiments were performed to evaluate morphological alterations occurred during the phenotypic modulation. Elevated number of passages (up to 4) as well as the freezing/thawing process induced a significant phenotypic modulation in VSMCs, which was accompanied by diminished MLC and ERK1/2 phosphorylation levels. Phosphorylation of MLC was suppressed completely by the treatment with a synthetic inhibitor of MEK-1, a direct upstream of ERK1/2, PD98059. These findings provide that ERK1/2-promoted MLC phosphorylation is impaired during VSMCs phenotypic modulation, suggesting that ERK1/2 signaling pathway may represent a potential target for understanding the pathogenesis of several vascular disease processes frequently associated to this condition.

p53 activation is one of the main signals after DNA damage, controlling cell cycle arrest, DNA repair and apoptosis. We have previously shown that confluent nucleotide excision repair (NER)-deficient cells are more resistant to apoptosis induced by ultraviolet irradiation (UV). Here, we further investigated the effect of cell confluence on UV-induced apoptosis in normal and NER-deficient (XP-A and XP-C) cells, as well as the effects of treatments with the ATWATR inhibitor caffeine, and the patterns of p53 activation. Strong p53 activation was observed in either proliferating or confluent cells. Caffeine increased apoptosis levels and inhibited p53 activation in proliferating cells, suggesting a protective role for p53. However, in confluent NER-deficient cells no effect of caffeine was observed. Transcription recovery measurements showed decreased recovery in proliferating XPA-deficient cells, but no recovery was observed in confluent cells. The levels of the cyclin/Cdk inhibitor, p21(Waf1/Cip1), correlated well with p53 activation in proliferating cells. Surprisingly, confluent cells also showed similar activation of p21(Waf1/Cip1). These results indicate that reduced apoptosis in confluent cells is associated with the deficiency in DNA damage removal, since this effect is not clearly observed in NER-proficient cells. Moreover, the strong activation of p53 in confluent cells, which barely respond to apoptosis, suggests that this protein, under these conditions, is not linked to UV-induced cell death signaling. The impact of ultraviolet (UV-C) photoproducts on apoptosis induction was investigated in growth arrested (confluent) and proliferating human primary fibroblasts. Confluent fibroblasts were more resistant to UV-C-induced apoptosis than proliferating cells, and this was observed for normal human cells and for cells from patients with Cockayne and trichothiodystrophy syndromes, deficient in transcription coupled repair. This resistance was sustained for at least seven days and was not due to DNA repair efficiency, as the removal of CPDs in the genome was similar under both growth conditions. There was no correlation between reduced apoptosis and RNA synthesis recovery. Following UV-C treatment, proliferating and confluent fibroblasts showed a similar level of RNA synthesis inhibition and recovery from transcription blockage. These results support the hypothesis that the decrease of DNA replication, in growth arrested cells, protects cell from UV-C-induced apoptosis, even in the presence of DNA lesions. The study proposed to describe sexual development in pelagic stage loggerhead sea turtles Caretta caretta and compare this to hatchlings and adults. It is meant as an ontogenic approach, in order to understand reproductive development and population composition and their dynamics in the pelagic environment. The study focused on the pelagic loggerheads that are found in the waters offshore Madeira Island (Portugal) in the North-eastern Atlantic and use it as a developmental habitat. The innovating character of this work relied on the lack of any description regarding the gonad ontogenesis and reproductive development for the pelagic stage in any of the 7 existing sea turtle species, all of them in danger of extinction. Three methods were used to diagnose the sex of each juvenile individual and asses the level of reproductive development: (1) laparoscopy, (2) gonad biopsy and (3) the assessment of two sex steroids circulating levels, namely testosterone and estradiol. In order to cover all life stages and compare data obtained for the juvenile stage, hatchlings and nesting female adults were sampled at the nearest nesting rookery at Boa Vista Island in the Cape Verde Archipelago. Gonads from dead hatchlings were collected for gonad histology and blood was collected from nesting females for sex steroids assessment. Laparoscopies revealed to be a valid sexing method for the juvenile stage, since gonads are morphologically differentiated at these size classes. Moreover, laparoscopy was validated using gonad histology. Gonad histology of juveniles showed that gonads are already completely differentiated into ovaries or testes at the size classes examined, but development seems to be quiescent. Males present already developed seminiferous tubules with spermatogonia lining the interior of the seminiferous tubule. Female gonads present oocytes at different development stages, but only oocytes up to stage III were observed. The maximum oocyte diameter in each individual correlated with body size, suggesting that reproductive development is an on-going process in juvenile females. The circulating levels of both testosterone and estradiol in juveniles of both sexes were very low and consistently lower than the ones observed in the nesting females from Boa Vista Island. No bimodal distribution was found for any of the sex steroids analysed and thus circulating hormone levels were not a reliable tool for sexing juvenile individuals with a non-invasive technique. The ratio testosterone:estradiol did not show a bimodal distribution either. The levels of testosterone correlated with sea surface temperature. The fact that temperatures observed during this study were below 24ºC might have hindered a differential testosterone pattern between juvenile males and females. Sex ratios for this population were generated according to laparoscopy results and compared among years and size classes. An overall sex ratio of 2 females for each male was found, but they varied among size classes but not among years. Possible causes for the sex ratios observed are discussed. This study is a contribution to our knowledge on the pelagic stage of loggerhead turtles, namely on the population structure regarding sex ratio, which is a vital tool for implementing conservation strategies.

Este trabalho teve por objetivo determinar a ocorrência e a freqüência de fungos em banana 'Prata anã' e elucidar o agente causal das podridões em pós-colheita de frutos provenientes do norte de Minas Gerais. Dois métodos de isolamento foram adotados: diluição em placas, a partir da lavagem de frutos verdes, e direto de frutos maduros. Os fungos Colletotrichum musae, Trichoderma harzianum, Fusarium equisetii, Penicillium sp. Aspergillus parasiticus, Trichothecium roseum, Colletotrichum acutatum, Alternaria sp., Cladosporium musae e Curvularia lunata foram os mais freqüentemente associados aos frutos. A patogenicidade desses fungos foi testada pela substituição de discos da casca de frutos verdes por discos de micélio. Colletotrichum musae apresentou área média lesionada em torno do ponto de inoculação igual a 5,8 cm², enquanto para os demais fungos testados não passou de 1,50 cm². Os resultados mostraram que C. musae é o agente primário das podridões dos frutos examinados com 100 % de incidência e os demais fungos limitaram-se a necrosar os ferimentos em torno do ponto de inoculação. O modo de infecção latente, causada por C. musae, parece favorecer, primeiramente, a colonização interna dos tecidos e, posteriormente, a ação dos fungos oportunistas, que aceleram as podridões nos frutos e na coroa.
Indução da expressão precoce de sintomas de Guignardia citricarpa em frutos de laranjeira 'pêra-rio'

O presente trabalho teve por objetivo avaliar a influência de diferentes concentrações de ethephon na expressão precoce de sintomas de Guignardia citricarpa em frutos de laranjeira 'Pêra-Rio'. Para tal, frutos assintomáticos e isentos de aplicações com fungicidas, com 20 e 28 semanas após a queda de pétalas, foram coletados em área de comprovada existência da doença, no município de Conchal-SP e levados ao Laboratório de Fitopatologia da FCAV/UNESP, em Jaboticabal-SP, onde foram tratados com soluções nas seguintes doses de ethephon: i) 1,57 g L-1; ii) 2,10 g L-1; iii) 2,42 g L-1; iv) Testemunha (água). Todas acrescidas de imazalil a 0,25 g L-1, para prevenir podridões de pós-colheita. Após os tratamentos, os frutos foram mantidos em câmara incubadora para B.O.D., calibrada à temperatura de 25ºC ±1ºC, por 15 dias. Posteriormente, os frutos foram submetidos a quatro avaliações, em intervalos semanais, sendo atribuídas notas que variaram de zero (ausência de sintomas) a 6 (sintomas severos). Os dados da severidade da doença observados nos frutos colhidos prematuramente e submetidos aos diferentes tratamentos com ethephon foram comparados aos observados em frutos ensacados e não ensacados, mantidos no campo até a maturação natural. Constatou-se maior equivalência de sintomas nos frutos com idade entre 20 e 28 semanas, quando estes foram tratados com 2,10 g L-1 de ethephon e avaliados entre 28 e 35 dias. Concluiu-se que o emprego de ethephon, nestas condições, viabilizou a expressão precoce dos sintomas da mancha preta em frutos contendo infecções quiescentes de G. citricarpa, com antecedência de, pelo menos, 105 dias antes da colheita. Tal resultado constitui-se, portanto, em alternativa de grande aplicabilidade na detecção precoce de sintomas de mancha preta.

O leiteiro é uma planta daninha que ocorre em pastagens, de importância crescente no Brasil e que reproduz-se por sementes. Com o objetivo de conhecer o comportamento germinativo dessa espécie, as sementes foram colocadas para germinar sobre papel, em temperaturas constantes de 15, 20, 25, 30, 35 e 40ºC, sem ou com luz (78 µmol s-1m-2/8h). Para a avaliação dos tratamentos, foram contabilizadas, semanalmente, o número de sementes germinadas (G), considerando-se a protrusão de 1cm de raiz primária, até que cessasse a germinação em todos os tratamentos (63 dias após a semeadura) e, determinou-se o índice de velocidade de germinação (IVG). Após esse período, as sementesforam transferidas para a temperatura de 30ºC com luz (condição favorável detectada na etapa anterior) e realizou-se contagem semanal até que a germinação cessasse novamente, em todos os tratamentos (91 dias após a semeadura), quando foram contabilizadas as sementes mortas (M) e as sementes quiescentes (Q). Os resultados indicaram que as sementes de leiteiro foram indiferentes à luz. A temperatura ótima de germinação para a espécie situou-se entre 25 e 30ºC (G=85%, M=15% e IVG=34). Nas temperaturas extremas de 15ºC (G=0%; Q=78%; M=22%) e 40 ºC (G=0%; Q=0% e M=100%) não houve a germinação das sementes.

The study of Brazilian sedimentary basins concentrates on their rift phase, whereas the Post-rift phase has been considered a tectonic quiescent period. The post-rift sequence of the Potiguar Basin, in the far northeastern Brazil, was once considered little deformed, however several studies have shown how that it was affected by major fault systems. The purpose of this thesis is to characterize the post-rift tectonic. The specific objectives are: to characterize the Neogene and Quaternary sedimentary units that outcrop of the Potiguar Basin; to show how the NW-SEtrending Afonso Bezerra Faults System deformed outcrop rocks in the Basin; to describe soft-sediment deformation in gravels of the Quaternary Alluvial Deposits from Açu River. Facies analyses, grain-size studies, luminescence dating, remote sensing, structural mapping, shallow geophysics (georadar), paleostress and petrography were carried out. The structural mapping and the georadar sections indicated that the Carnaubais and Afonso Bezerra fault systems formed fractures, silicified and non-silicified faults or deformation bands, affecting mainly the Açu, Jandaíra and Barreiras formations. The petrographic data indicate that the strong silicification resulted in a sealant character of the faults. Paleostress analysis indicates that two stress fields affected the Basin: the first presented N-S-trending compression, occurred from the Neocretaceous to the Miocene; the second stress field presents E-W-trending compression, acts from the Miocene to the present. It was verified once the Afonso Bezerra System Faults was reactivated in periods post-Campanian and affects all post-rift lithostratigraphic units of Potiguar Basin, including Quaternary sedimentary covers. The study about soft-sediment deformation structures indicates that they are similar in morphology and size to modern examples of seismically-induced deformation strutures in coarse sediments. TL and OSL ages indicate that sediment deposition and associated soft-sediment deformation occurred at least six times from ~352 Ka to ~9 Ka. Finally these studies demonstrate how recent is tectonics in the Basin Potiguar

Through an integrated approach, using litho, chrono and biostratigraphic data, the relative importance of climate variations and tectonics were recognized in rift sediments of the onshore Potiguar Basin, Northeast Brazil. Concepts of sequence stratigraphy were applied as a template to integrate sedimentological and geochemical data (oxygen isotopes), as well as quantitative palynologic methods to address and recognize the main depositional patterns produced in a rift basin. The main objective was to address the relative importance of climate changes and tectonics to the resultant stratigraphic architecture. The results of computer simulations of sedimentary basin fills of rift basins were quite useful to test working hypothesis and mimic the process of filling a half graben during a rift event. The studied section includes a neovalanginian-eobarremian (Lower Cretaceous) rift interval from the Pendência Formation, located in the southwestern portion of Umbuzeiro Graben, in the offshore Potiguar Basin. The depositional setting is interpreted as progradational deltaic system entering a lake from its flexural margin. Sismoestratigraphyc and well logs analyses allowed to interpret two regressive intervals (Green and Yellow Sequences), separated by a broad transgressive interval (Orange Sequence), known as the Livramento Shale. The depositional history encompass three stages: two tectonically active phases, during the deposition of the Green and Yellow Sequences, and a tectonically quiescent phase, during the deposition of the Orange Sequence. Paleoclimatic interpretation, based on quantitative palynology and geochemical data (18O), suggests a tendency to arid conditions during the tectonically active phases and wet conditions during the tectonically quiescent phase. Stratigraphic modeling and backstripping techniques, supported by paleoclimatic/paleoecologic interpretations provide a powerful methodology to evaluate the tectonic and climatic controls on tectonic lakes

In amphibia, steroidogenesis remains quiescent in distinct seasonal periods, but the mechanism by which spermatogenesis is maintained under low steroidogenic conditions is not clear. In the present study, testosterone location in the testes of Rana catesbeiana was investigated immunohistochemically during breeding (summer) and nonbreeding (winter) periods. In winter, the scarce interstitial tissue exhibited occasional testosterone immunopositivity in the interstitial cells but the cytoplasm of primordial germ cells (PG cells) was clearly immunopositive. By contrast, in summer, PG cells contained little or no immunoreactivity whereas strong immunolabelling was present in the well-developed interstitial tissue. These results suggest that PG cells could retain testosterone during winter. This androgen reservoir could be involved in the control of early spermatogenesis in winter and/or to guarantee spermiogenesis and spermiation in the next spring/summer. The weak or negative immunoreaction in the summer PG cells might reflect consumption of androgen reservoir by the intense spermatogenic activity from spring to summer. Thus, besides acting as stem cells, PG cells of R. catesbeiana could exert an androgen regulatory role during seasonal spermatogenesis.
