A esteatose hepática, que se caracteriza pelo acúmulo excessivo de gordura nas células do fígado, é um problema que vem preocupando a comunidade médico-científica, pois sua incidência vem aumentando a nível global, com expectativa de se tornar a doença crônica hepática de maior predominância em várias partes do mundo. Apesar de ser considerada uma doença benigna, a esteatose pode evoluir para doenças mais graves como cirrose, fibrose avançada, esteato hepatite (com ou sem fibrose) ou carcinoma. Entretanto, é potencialmente reversível, mesmo em quadros mais graves, o que reforça a urgência de se desenvolver métodos confiáveis para detecção e avaliação, inclusive ao longo de tratamento. Os métodos atuais para diagnóstico e quantificação da gordura hepática ainda são falhos: com a ultrassonografia não se é capaz de realizar quantificação; a tomografia computadorizada faz uso de radiação ionizante; a punção (biópsia), considerada o padrão ouro, é precisa, mas invasiva e pontual. A Ressonância Magnética (RM), tanto com espectroscopia (MRS) como com imagem (MRI), são alternativas completamente não invasivas, capazes de fornecer o diagnóstico e quantificação da gordura infiltrada no fígado. Entretanto, os trabalhos encontrados na literatura utilizam sequências de pulsos desenvolvidas especialmente para esse fim, com métodos de pós-processamento extremamente rebuscados, o que não é compatível com o estado atual dos equipamentos encontrados em ambientes clínicos nem mesmo ao nível de experiência e conhecimento das equipes técnicas que atuam em clínicas de radiodiagnóstico. Assim, o objetivo central do presente trabalho foi avaliar o potencial da RM como candidato a método de diagnóstico e de quantificação de gordura em ambientes clínicos, utilizando, para isso, sequências de pulsos convencionais, disponíveis em qualquer sistema comercial de RM, com protocolos de aquisição e processamento compatíveis com àqueles realizados em exames clínicos, tanto no que se refere à simplicidade como ao tempo total de aquisição. Foram avaliadas diferentes abordagens de MRS e MRI utilizando a biópsia hepática como padrão de referência. Foram avaliados pacientes portadores de diabetes tipo II, que apresentam alta prevalência de esteatose hepática não alcoólica, além de grande variabilidade nos percentuais de gordura. Foram realizadas medidas de correlação, acurácia, sensibilidade e especificidade de cada uma das abordagens utilizadas. Todos os métodos avaliados apresentaram alto grau de correlação positiva (> 87%) com os dados obtidos de maneira invasiva, o que revela que os valores obtidos utilizando RM estão de acordo com aquilo observado pela biópsia hepática. Muito embora os métodos de processamento utilizados não sejam tão complexos quanto seriam necessários caso uma quantificação absoluta fosse desejada, nossas análises mostraram alta acurácia, especificidade e sensibilidade da RM na avaliação da esteatose. Em conclusão, a RM se apresenta, de fato, como uma excelente candidata para avaliar, de forma não invasiva, a fração de gordura hepática, mesmo quando se considera as limitações impostas por um ambiente clínico convencional. Isso sugere que essas novas metodologias podem começar a migrar para ambientes clínicos sem depender das sequências complexas e dos processamentos exóticos que estão descritos na literatura mais atual.

Foram conduzidos dois experimentos com o intuito de se avaliar o desempenho, características de carcaça e parâmetros ruminais de bovinos Nelore recebendo dietas contendo grãos de milho flint processados de diferentes formas em associação a diferentes aditivos e níveis de FDN de silagem de milho (FDNf). No Experimento 1, 239 tourinhos (PCI=350±26,79kg) foram alojados em 40 baias de acordo com o peso corporal inicial e receberam dietas contendo dois tipos de processamento dos grãos de milho (moagem fina e floculação) e diferentes aditivos (monensina sódica; 25 ppm e formulação A062 a base de virginiamicina; produto teste, não comercial, fornecido pela Ourofino Saúde Animal; nas dosagens de 17 e 25 ppm), totalizando 8 tratamentos. Os dados foram analisados pelo PROC MIXED do SAS. A floculação aumentou o peso final, o ganho de peso diário, a eficiência alimentar, o peso da carcaça quente e tendeu a aumentar o rendimento de carcaça em comparação a moagem fina. Houve tendência de interação entre processamento e fornecimento de aditivos para a ingestão de matéria seca e ganho de peso diário. A floculação reduziu o teor de amido fecal, aumentou a digestibilidade do amido no trato total, aumentou os valores de energia liquida para manutenção e ganho da dieta e o numero médio de papilas ruminais. Os aditivos aumentaram a altura, largura e área das papilas e reduziram a espessura de faixa de queratina em relação ao tratamento controle. Houve tendência das formulações a base de virginiamicina aumentarem o peso da carcaça quente dos animais em comparação com a monensina sódica, mas não em relação ao tratamento controle sem aditivo. A floculação foi mais efetiva para aumentar a digestiblidade do amido no trato total, o valor energético do milho e o desempenho dos bovinos enquanto que os aditivos não foram efetivos para aumentar o valor energético das dietas e a eficiência alimentar dos animais. No Experimento 2, 237 tourinhos (PCI=350±28,49kg) foram alojados em 32 baias de acordo com o peso corporal inicial e receberam dietas contendo dois tipos de processamento (moagem grosseira e floculação) e diferentes níveis de FDNf (4; 7; 10 e 13% MS), totalizando 8 tratamentos. Os dados foram analisados pelo PROC MIXED do SAS. A floculação tendeu a aumentar a eficiência alimentar, o peso da carcaça quente e a espessura de gordura subcutânea, reduziu o teor de amido fecal, aumentou a digestibilidade do amido no trato total como também aumentou os valores de energia liquida para manutenção e para ganho das dietas. Os níveis de FDNf afetaram ingestão de matéria seca e o peso da carcaça quente independente do método de processamento. Houve tendência de aumento linear no GPD e tendência de redução quadrática na eficiência alimentar com inclusão crescente de FDNf. A inclusão de silagem de milho em dietas com alto teor de milho flint reduziu a eficiência alimentar dos animais, mas aumentou o peso da carcaça quente. A floculação aumentou o valor energético do cereal para tourinhos Nelore em dietas com alto teor de energia.

Listeria monocytogenes is a food-borne Gram-positive bacterium that is responsible for a variety of infections (worldwide) annually. The organism is able to survive a variety of environmental conditions and stresses, however, the mechanisms by which L. monocytogenes adapts to environmental change are yet to be fully elucidated. An understanding of the mechanism(s) by which L. monocytogenes survives unfavourable environmental conditions will aid in developing new food processing methods to control the organism in foodstuffs. We have utilized a proteomic approach to investigate the response of L. monocytogenes batch cultures to the transition from exponential to stationary growth phase. Proteomic analysis showed that batch cultures of L. monocytogenes perceived stress and began preparations for stationary phase much earlier (approximately A(600) = 0.75, mid-exponential) than predicted by growth characteristics alone. Global analysis of the proteome revealed that the expression levels of more than 50% of all proteins observed changed significantly over a 7-9 h period during this transition phase. We have highlighted ten proteins in particular whose expression levels appear to be important in the early onset of the stationary phase. The significance of these findings in terms of functionality and the mechanistic picture are discussed.

Background: The mismatch negativity (MMN) is a fronto-centrally distributed event-related potential (ERP) that is elicited by any discriminable auditory change. It is an ideal neurophysiological tool for measuring the auditory processing skills of individuals with aphasia because it can be elicited even in the absence of attention. Previous MMN studies have shown that acoustic processing of tone or pitch deviance is relatively preserved in aphasia, whereas the basic acoustic processing of speech stimuli can be impaired (e.g., auditory discrimination). However, no MMN study has yet investigated the higher levels of auditory processing, such as language-specific phonological and/or lexical processing, in individuals with aphasia. Aims: The aim of the current study was to investigate the MMN response of normal and language-disordered subjects to tone stimuli and speech stimuli that incorporate the basic auditory processing (acoustic, acoustic-phonetic) levels of non-speech and speech sound processing, and also the language-specific phonological and lexical levels of spoken word processing. Furthermore, this study aimed to correlate the aphasic MMN data with language performance on a variety of tasks specifically targeted at the different levels of spoken word processing. Methods M Procedures: Six adults with aphasia (71.7 years +/- 3.0) and six healthy age-, gender-, and education-matched controls (72.2 years +/- 5.4) participated in the study. All subjects were right-handed and native speakers of English. Each subject was presented with complex harmonic tone stimuli, differing in pitch or duration, and consonant-vowel (CV) speech stimuli (non-word /de:/versus real world/deI/). The probability of the deviant for each tone or speech contrast was 10%. The subjects were also presented with the same stimuli in behavioural discrimination tasks, and were administered a language assessment battery to measure their auditory comprehension skills. Outcomes O Results: The aphasic subjects demonstrated attenuated MMN responses to complex tone duration deviance and to speech stimuli (words and non-words), and their responses to the frequency, duration, and real word deviant stimuli were found to strongly correlate with performance on the auditory comprehension section of the Western Aphasia Battery (WAB). Furthermore, deficits in attentional lexical decision skills demonstrated by the aphasic subjects correlated with a word-related enhancement demonstrated during the automatic MMN paradigm, providing evidence to support the word advantage effect, thought to reflect the activation of language-specific memory traces in the brain for words. Conclusions: These results indicate that the MMN may be used as a technique for investigating general and more specific auditory comprehension skills of individuals with aphasia, using speech and/or non-speech stimuli, independent of the individual's attention. The combined use of the objective MMN technique and current clinical language assessments may result in improved rehabilitative management of aphasic individuals.

Starchy plant foods are significant in the diet of almost all peoples. Archaeologically, however, preservation of such plants is limited, and direct evidence of plant use by past people is also rare. Although starch grains can be preserved on artefacts used to process starchy plants, it is very difficult to identify grains damaged by processing methods such as milling or cooking. We present a method for identifying such damaged starch grains using Congo Red staining to identify cooking or milling activities in the past subsistence behaviour of Aboriginal people of southeast Queensland, Australia. Listeria monocytogenes has previously been shown to adapt to a wide variety of environmental niches, principally those associated with low pH, and this compromises its control in food environments. An understanding of the mechanism(s) by which L. monocytogenes survives unfavourable environmental conditions will aid in developing new food processing methods to control the organism in foodstuffs. The present Study aimed to gain a further understanding of the physiological basis for the differential effects of one control strategy, namely the use of the lantibiotic nisin. Using propidium iodide (PI) to probe membrane integrity it was shown that L. monocytogenes Scott A was sensitive to nisin (8 ng mL(-1)) but this was growth phase dependent with stationary phase cells (OD600=1.2) being much more resistant than exponential phase cells (OD600=0.38). We demonstrate that, using a combination of techniques including fluorescence activated cell sorting (FACS), the membrane adaptations underpinning nisin resistance are triggered much earlier (OD600 < 0.5) than the onset of stationary phase. We present an application of Mathematical Morphology (MM) for the classification of astronomical objects, both for star/galaxy differentiation and galaxy morphology classification. We demonstrate that, for CCD images, 99.3 +/- 3.8% of galaxies can be separated from stars using MM, with 19.4 +/- 7.9% of the stars being misclassified. We demonstrate that, for photographic plate images, the number of galaxies correctly separated from the stars can be increased using our MM diffraction spike tool, which allows 51.0 +/- 6.0% of the high-brightness galaxies that are inseparable in current techniques to be correctly classified, with only 1.4 +/- 0.5% of the high-brightness stars contaminating the population. We demonstrate that elliptical (E) and late-type spiral (Sc-Sd) galaxies can be classified using MM with an accuracy of 91.4 +/- 7.8%. It is a method involving fewer 'free parameters' than current techniques, especially automated machine learning algorithms. The limitation of MM galaxy morphology classification based on seeing and distance is also presented. We examine various star/galaxy differentiation and galaxy morphology classification techniques commonly used today, and show that our MM techniques compare very favourably.

Frequent Itemsets mining is well explored for various data types, and its computational complexity is well understood. There are methods to deal effectively with computational problems. This paper shows another approach to further performance enhancements of frequent items sets computation. We have made a series of observations that led us to inventing data pre-processing methods such that the final step of the Partition algorithm, where a combination of all local candidate sets must be processed, is executed on substantially smaller input data. The paper shows results from several experiments that confirmed our general and formally presented observations.

The use of ontologies as representations of knowledge is widespread but their construction, until recently, has been entirely manual. We argue in this paper for the use of text corpora and automated natural language processing methods for the construction of ontologies. We delineate the challenges and present criteria for the selection of appropriate methods. We distinguish three ma jor steps in ontology building: associating terms, constructing hierarchies and labelling relations. A number of methods are presented for these purposes but we conclude that the issue of data-sparsity still is a ma jor challenge. We argue for the use of resources external tot he domain specific corpus.

Through the application of novel signal processing techniques we are able to measure physical measurands with both high accuracy and low noise susceptibility. The first interrogation scheme is based upon a CCD spectrometer. We compare different algorithms for resolving the Bragg wavelength from a low resolution discrete representation of the reflected spectrum, and present optimal processing methods for providing a high integrity measurement from the reflection image. Our second sensing scheme uses a novel network of sensors to measure the distributive strain response of a mechanical system. Using neural network processing methods we demonstrate the measurement capabilities of a scalable low-cost fibre Bragg grating sensor network. This network has been shown to be comparable with the performance of existing fibre Bragg grating sensing techniques, at a greatly reduced implementation cost.

This thesis documents the design, manufacture and testing of a passive and non-invasive micro-scale planar particle-from-fluid filter for segregating cell types from a homogeneous suspension. The microfluidics system can be used to separate spermatogenic cells from testis biopsy samples, providing a mechanism for filtrate retrieval for assisted reproduction therapy. The system can also be used for point-of-service diagnostics applications for hospitals, lab-on-a-chip pre-processing and field applications such as clinical testing in the third world. Various design concepts are developed and manufactured, and are assessed based on etched structure morphology, robustness to variations in the manufacturing process, and design impacts on fluid flow and particle separation characteristics. Segregation was measured using image processing algorithms that demonstrate efficiency is more than 55% for 1 µl volumes at populations exceeding 1 x 107. the technique supports a significant reduction in time over conventional processing, in the separation and identification of particle groups, offering a potential reduction in the associated cost of the targeted procedure. The thesis has developed a model of quasi-steady wetting flow within the micro channel and identifies the forces across the system during post-wetting equalisation. The model and its underlying assumptions are validated empirically in microfabricated test structures through a novel Micro-Particle Image Velocimetry technique. The prototype devices do not require ancillary equipment nor additional filtration media, and therefore offer fewer opportunities for sample contamination over conventional processing methods. The devices are disposable with minimal reagent volumes and process waste. Optimal processing parameters and production methods are identified with any improvements that could be made to enhance their performance in a number of identified potential applications.

The thesis examines the possibilities for the beneficiation of steel making slags by using mineral processing methods. Chemical and Mineralogical investigations have been carried out by SEM and EP}ffi to determine the most suitable separation methods in terms of crystal size, chemical composition and surface properties. Magnetic separation was applied in connection with size reductions for the extraction of the metallic iron prills and other iron containing phases and the results were related to the feed size and operating conditions. The behaviour of the slags in flotation tests was studied with respect to the recovery and grade. It was found that the free lime presence in the slags caused a high acid consumption of both weak and strong acids. It also reacted with acids and consequently produced a white precipitate, (CaS04 for H2 S04 ). The poor response of the phases to the flotation by different types of collector was found to be due to surface alteration caused by the free lime. The flocculation tests were carried out at the natural pH of the slags to prevent surface alterations. Settling tests were done to determine the suitable flocculants for the separation tests. The effect of the settling period, flocculant concentration, conditioning period and number of cleaning cycles were determined to optimize the separation tests. The discussion brings together this study with previous theoretically based work cited in the literature to elucidate the factor5governing the utilisation of steel making slags.

Scaffolds derived from processed tissues offer viable alternatives to synthetic polymers as biological scaffolds for regenerative medicine. Tissue-derived scaffolds provide an extracellular matrix (ECM) as the starting material for wound healing and the functional reconstruction of tissues, offering a potentially valuable approach for the replacement of damaged or missing tissues. Additionally, acellular tissue may provide a natural microenvironment for host-cell migration and the induction of stem cell differentiation to contribute to tissue regeneration. There are a number of processing methods that aim to stabilize and provide an immunologically inert tissue scaffold. Furthermore, these tissue-processing methods can often be applied to xenogenic transplants because the essential components of the ECM are often maintained between species. In this study, we applied several tissue-processing protocols to the cornea in order to obtain a decellularized cornea matrix that maintained the clarity and mechanical properties of the native tissue. Histology, mechanical testing and electron microscopy techniques were used to assess the cell extraction process and the organization of the remaining ECM. In vitro cell seeding experiments confirmed the processed corneas’ biocompatibility.

Fine-fraction (<63 µm) grain-size analyses of 530 samples from Holes 1095A, 1095B, and 1095D allow assessment of the downhole grain-size distribution at Drift 7. A variety of data processing methods, statistical treatment, and display techniques were used to describe this data set. The downhole fine-fraction grain-size distribution documents significant variations in the average grain-size composition and its cyclic pattern, revealed in five prominent intervals: (1) between 0 and 40 meters composite depth (mcd) (0 and 1.3 Ma), (2) between 40 and 80 mcd (1.3 and 2.4 Ma), (3) between 80 and 220 mcd (2.4 and 6 Ma), (4) between 220 and 360 mcd, and (5) below 360 mcd (prior to 8.1 Ma). In an approach designed to characterize depositional processes at Drift 7, we used statistical parameters determined by the method of moments for the sortable silt fraction to distinguish groups in the grainsize data set. We found three distinct grain-size populations and used these for a tentative environmental interpretation. Population 1 is related to a process in which glacially eroded shelf material was redeposited by turbidites with an ice-rafted debris influence. Population 2 is composed of interglacial turbidites. Population 3 is connected to depositional sequence tops linked to bioturbated sections that, in turn, are influenced by contourite currents and pelagic background sedimentation.

Botanically, green composites belong to an economically important seed plant family that includes maize, wheat, rice, and sorghum known as Saccharum offi cinarum. There are so many natural fibers available in the environment such as rice husk, hemp fibers, flax fibers, bamboo fibers, coconut fiber, coconut coir, grawia optiva and many others also. Life Cycle Assessment (LCA) is a process to estimate the environmental feature and potential impacts related to a product, by organizing a directory of pertinent inputs and outputs of a product system, assessing the potential environmental impacts related with the said inputs and outputs, explaining the results of the inventory analysis and impact evaluation phases in connection to the objectives of the study. Particularly Bagasse, an agricultural residue not only becomes a problem from the environmental point of view, but also affects the profitability of the sugarcane industries. This chapter discusses the properties, processing methods and various other aspects including economic and environmental aspects related to green composites.
