Current treatment strategies for the treatment of brain tumor have been hindered primarily by the presence of highly lipophilic insurmountable blood-brain barrier (BBB). The purpose of current research was to investigate the efficiency of engineered biocompatible polymeric nanoparticles (NPs) as drug delivery vehicle to bypass the BBB and enhance biopharmaceutical attributes of anti-metabolite methotrexate (MTX) encapsulated NPs. The NPs were prepared by solvent diffusion method using cationic bovine serum albumin (CBA), and characterized for physicochemical parameters such as particle size, polydispersity index, and zeta-potential; while the surface modification was confirmed by FTIR, and NMR spectroscopy. Developed NPs exhibited zestful relocation of FITC tagged NPs across BBB in albino rats. Further, hemolytic studies confirmed them to be non-toxic and biocompatible as compared to free MTX. In vitro cytotoxicity assay of our engineered NPs on HNGC1 tumor cells proved superior uptake in tumor cells; and elicited potent cytotoxic effect as compared to plain NPs and free MTX solution. The outcomes of the study evidently indicate the prospective of CBA conjugated poly (D,L-lactide-co-glycolide) (PLGA) NPs loaded with MTX in brain cancer bomber with amplified capability to circumvent BBB.

Cette étude concerne un écosystème paralique, la lagune d’Aveiro (Portugal). Elle vise à déterminer l’organisation des peuplements de poissons en fonction des caractéristiques et du fonctionnement de cet écosystème. L’ichtyofaune a été échantillonnée mensuellement en 10 stations d’août 1987 à juillet 1988 et de janvier 1999 à décembre 2000, avec une seine de plage traditionnelle. La répartition des peuplements de poissons est étudiée au moyen de descripteurs populationnels (richesses spécifique et familiale, densité, biomasse et indice de diversité) et d’analyses statistiques (groupements et ordination). La lagune d’Aveiro présente de fortes variations, dans l’espace et le temps, de ses paramètres physico- chimiques reflétant ainsi les variations climatiques annuelles. Si l’on considère la mobilité des poissons et la géomorphologie et l’hydrologie du système étudié, nous pouvions nous attendre à une forte homogénéité de la distribution des poissons. À l’inverse, une diminution de l’influence marine a pour conséquence une diminution des richesses spécifiques et familiales, de la densité et de la biomasse. Nous avons également observé une modification de composition de l’assemblage de poissons et la présence d’espèces dominantes caractéristiques des différents niveaux de confinement (taux de renouvellement des eaux marines en un point donné du système). Le peuplement de poissons présente une organisation semblable à la zonation biologique, indépendamment des paramètres physico-chimiques tels que la salinité, décrite par la macrofaune benthique et induite par le confinement. La comparaison des résultats avec des données obtenues douze ans plus tôt, montre que l’organisation générale de la lagune est demeurée inchangée, illustrant ainsi la stabilité des écosystèmes paraliques. De plus, des modifications du niveau de confinement dans les marges nord et sud, induites principalement par des changements locaux de l’hydrodynamisme, ont été constatées. Le déconfinement de la zone nord est la conséquence de l’entretien des canaux de navigation par dragage. À l’inverse, le confinement de la zone sud est l’évolution naturelle des bassins paraliques soumis souvent à une sédimentation élevée et rapide. Cette étude montre que l’organisation du peuplement de poissons valide le concept du confinement pour l’organisation biologique des milieux paraliques, et peut être employé pour expliquer les changements de ces écosystèmes.

A atividade vitícola praticada na região do Douro é uma das mais importantes atividades agrícolas do país, não só devido a fatores socioculturais, como também pelo seu valor económico. A Região Demarcada do Douro (RDD) é uma das regiões vitícolas mais antiga do mundo, onde é produzido um dos vinhos generosos mais famosos do mundo, o Vinho do Porto. Contudo, no cultivo intensivo da vinha, o uso recorrente a pesticidas para o controlo de pragas das vinhas pode ter repercussões a nível da qualidade dos solos e dos cursos de água na envolvente às vinhas. O presente estudo foi desenvolvido numa área-piloto da sub-região Baixo Corgo da RDD, com o intuito de averiguar os possíveis efeitos e estimar os prováveis riscos ambientais promovidos pela viticultura. Foram selecionadas três vinhas, com idades de plantação distintas, onde foram recolhidas amostras de solos à superfície, tendo-se ainda feito uma recolha de amostras de água, sedimentos e águas intersticiais dos rios e da barragem na envolvente da área de estudo. Diferentes parâmetros químicos e físico-químicos foram analisados para uma avaliação global da área e prováveis fontes dos elementos. Especial destaque foi dado ao Cu, C, P, N, NO3 e S quer pelos teores anómalos e elevada variação espacial, quer pela sua importância como traçadores das atividades vitícolas e alguns como potenciais contaminantes. O Cu é o elemento potencialmente contaminante com mais destaque, visto que os seus teores são consideravelmente elevados nos solos e, a par, o P e N tendem a apresentar teores mais elevados na vinha mais velha, o que pode indicar efeitos de acumulação. Já os nitratos são mais elevados na vinha mais nova, visto que os processos de acumulação são mais difíceis neste caso devido à sua alta solubilidade e mobilidade. Apesar da elevada aplicação de S nas vinhas, os teores nestes solos são mais baixos do que nos solos sem atividades vitícolas, o que pode dever-se à sua baixa temperatura de sublimação e migração para níveis mais profundos dos solos. As baixas concentrações determinadas nos sedimentos e águas permitem concluir que as práticas vitícolas não parecem estar a interferir na qualidade dos mesmos. Contudo, novas campanhas de amostragem, abrangendo períodos sazonais distintos e próximos de épocas de aplicação de fertilizantes e pesticidas, deverão ser realizadas para validação temporal destes resultados.

Au Québec, les drains installés dans les sols sableux-limoneux sont sensibles au colmatage par ensablement et/ou par ocre de fer. Dans le passé, les drains avec des pertuis inférieurs à 2 mm étaient utilisés au Québec avec un filtre tissé de 110 microns ou un filtre tricoté de 450 microns. Récemment, des drains avec des pertuis supérieurs à 2 mm ainsi qu’un filtre de 250 microns ont été introduits sur le marché mais n’ont jamais été testés. Le projet avait pour objectif de déterminer les vitesses auxquelles se feront l’ensablement et le colmatage ferrique pour différentes combinaisons de drains (pertuis de 1,8 mm et de 3 mm) et de filtres (110 μm, 250 μm et 450 μm) dans un sol sableux à Bécancour. Un dispositif expérimental en blocs complets (3) aléatoires a été utilisé. Les débits ont été mesurés à la sortie des drains de chaque parcelle et les hauteurs des nappes ont été mesurées avec un bulleur dans des puits d’observation. Le suivi du pH, du potentiel d’oxydoréduction et du contenu en Fe2+ a été réalisé dans l’eau de la nappe et celle des drains. Les drains excavés 13 mois après leur installation ne montrent que des traces de sédiments et de colmatage ferrique. Les niveaux de Fe2+ sont significativement plus faibles dans l’eau à la sortie des drains que dans l’eau de la nappe. Le processus de colmatage ferrique ne semble que commencer et son impact n’est pas mesurable au niveau des débits unitaires à la sortie des drains.

Aquatic ecosystems are final collectors of all kinds of pollution as an outcome of anthropogenic inputs, such us untreated industrial and municipal sewage and agricultural pollutants. There are several aquatic ecosystems that are threatened by mineral and organic pollution. In Northeastern Portugal, near Bragança, different watercourses are suffering negative impacts of human activities. It has been developed several studies in the monitoring of environmental impacts in these river basins, namely in Rio Fervença, affected by organic pollution, and in Portelo stream, affected, since 2009, by the collapse and continuous input of mining deposits. In this sense, the present study aimed to continue the monitoring study of ecological status of freshwater ecosystems of Northeastern Portugal, namely the following objectives: a) mineral pollution effects of mining deposits sudden incorporated into Portelo stream; b) organic pollution due to domestic and industrial inputs in River Fervença. Also, since fish are useful experimental models to evaluate toxicological mechanisms of contaminants, c) acute toxicity tests with Cu were conducted in laboratory conditions. During 2015/2016, it was made abiotic and biotic characterization of 16 sampling sites distributed by both Portelo and Fervença rivers, tributaries of main River Sabor (Douro Basin). Several physicochemical parameters were determined and Riparian Quality (QBR Index) and Channel Quality (GQC) Indexes were determined for habitat evaluation. Fish and invertebrate communities were sampled, according to protocols of Water Framework Directive (WFD). Several metrics were determined, with particular emphasis on the Biotic Index IBMWP and the Northern Portuguese Invertebrate Index (IPtIN). Acute toxicity tests were conducted with an Iberian fish species, common barbel (Luciobarbus bocagei) and some plasmatic electrolytes levels were evaluated, to assess their contribution to mitigate osmoregulatory adverse effects of Cu. Also, same electrolytes were measured after changing to clean water, in attempt to assess fish capacity to reverse this situation. Results obtained for both rivers showed a significant level of disturbance that affected decisively water, habitat and biological quality of aquatic ecosystems. Mineral and Organic Pollution in River Sabor (NE Portugal): Ecotoxicological Effects on Freshwater Fauna Due to this change of environmental conditions in Portelo stream (extreme pH values, high conductivity and presence of heavy metals), several biological metrics (e.g. taxonomic richness, abundance, diversity, evenness) confirmed, comparatively with reference sites, a substantial decrease on ecological integrity status. The same pattern was found for Fervença River; however other water parameters, namely the content of most limiting nutrients (e.g. N and P) seemed to have more influence in the composition and structure of macroinvertebrate and fish communities. In fact, despite the operation of the Sewage Treatment Plant of Bragança, Fervença River presented significant levels of disturbance that affected decisively the quality and ecological integrity of the aquatic ecosystem. The synergic effect of domestic and industrial pollution, intensive agriculture, regulation and degradation of aquatic and riparian habitats contributed to the decrease of ecological condition, namely in the downstream zones (after Bragança). The results for acute toxicity, showed that fish can change Na+ and K+ levels face to Cu exposition and, depending of Cu concentration tested, can also return to normal levels, providing some insights to that are believed to occurred in fish population, near the Portelo mines. The low ecological integrity status detected in the lotic ecosystems in NE Portugal as a result of mineral and organic pollution deserves the development of several measures for rehabilitation and improving of water quality. On the other hand, environmental education actions are needed to contribute to improvement of ecological integrity of the river and its conservation.

La actividad industrial y el desarrollo material y económico, han traído como consecuencia la contaminación del aire, agua y el suelo; lo que ocasiona modificaciones físicas químicas y biológicas que han producido un deterioro en la calidad del agua, dando como resultado problemas de contaminación que afectan tanto la productividad de los sistemas como la salud humana. A las aguas de composición variada provenientes de uso municipal, industrial, comercial, agrícola, pecuario, o de cualquier otra índole, ya sea privada o pública que han sufrido una degradación o alteración en su calidad original se le conoce como agua residual. Este trabajo tiene como objetivo “Caracterizar las bacterias filamentosas en el funcionamiento de un reactor aerobio de la planta de tratamiento de aguas residuales de origen cervecero” para ello se estudió durante varios meses los parámetros físico-químicos y biológicos para poder así controlar de las bacterias filamentosas y así evitar en un futuro problemas de operación o poder controlar su crecimiento y por ende, ecológicos dentro del sistema de lodos activados. La identificación se realizó tomando muestras en el reactor aerobio y realizándole la tinción de Gram para posteriormente ser vistos en un microscopio de una resolución de 100x; luego se caracterizaron las bacterias juntas con los parámetros de operación del tanque de lodos por muestra y los resultados encontrados fueron la presencia de filamentos Microtrix Parvicella, Tipo 021N, y Sp1 (llamada así por no ser identificada, ni encontrada en la literatura), esta es una nueva especie encontrada, ya que es propia de este reactor aerobio y crecen principalmente en aguas cerveceras y por deficiencia de nutrientes en el sistema. Este trabajo nos permitió conocer la dinámica que existe entre los parámetros fisicoquímicos y los microorganismos que se encuentran en un biorreactor Aerobio. Esto nos llevó a comprender mejor el funcionamiento de estos sistemas dentro de plantas de aguas residuales de tipo industrial. A varied composition waters from municipal, industrial, commercial, agricultural, livestock, or any other use, whether private or public who have suffered a degradation or alteration in their original quality is known as residual water. tratamiento debe ser evaluado y controlado periódicamente, para garantizar su calidad para el consumo humano. El objetivo principal del presente estudio fue evaluar mediante un estudio físico-químico y microbiológico el Sistema de Calidad y Tratamiento del Agua que se efectúa en la Planta Potabilizadora del cantón Chordeleg. Se realizó un estudio no experimental, de campo, descriptivo y de corte no longitudinal. Para el análisis físico-químico se evaluaron 176 muestras en las 8 semanas, realizando 1 muestreo al día. En cada muestreo se realizó el análisis de 11 muestras que corresponden: 2 muestra de agua cruda, 2 muestras de agua pre-filtrada, 6 muestras de agua filtrada y 1 muestra de agua tratada realizándose un total de 22 muestras a la semana, en los cuales se determinó los parámetros físico-químicos: temperatura, sólidos totales disueltos, turbiedad y color, pH, dureza, alcalinidad, hierro, sulfato, nitritos, nitratos y cloro libre residual; parámetros microbiológicos: Coliformes totales y fecales. Los resultados obtenidos en las pruebas realizadas, mediante el análisis estadístico demostraron que los parámetros fisicoquímicos cumplen con lo establecido en la norma INEN 1108-2014, mientras que en los parámetros microbiológicos se estableció que los Coliformes fecales se encuentran dentro del rango establecido por la norma INEN 1108-2014 y los Coliformes totales fuera de lo establecido por la OMS(ausencia).

Physicochemical properties, consumer acceptance, antioxidant and angiotensin-converting enzyme (ACE) inhibitory activities of infusions and fermented beverages of Eucalyptus camaldulensis and Litsea glaucescens were compared. Among physicochemical parameters, only the pH of fermented beverages decreased compared with the unfermented infusions. No relevant changes were reported in consumer preference between infusions and fermented beverages. Phenolic profi le measured by UPLC MS/MS analysis demonstrated signifi cant concentration changes of these compounds in plant infusions and fermented beverages. Fermentation induced a decrease in the concentration required to stabilize 50 % of DPPH radical (i.e. lower IC50). Additionally, it enhanced the antioxidant activity measured by the nitric oxide scavenging assay (14 % of E. camaldulensis and 49 % of L. glaucescens); whereas relevant improvements in the fermented beverage were not observed in the lipid oxidation assay compared with unfermented infusions. The same behaviour was observed in the inhibitory activity of ACE; however, both infusions and fermented beverages had lower IC50 than positive control (captopril). The present study demonstrated that fermentation has an infl uence on the concentration of phenolics and their potential bioactivity. E. camaldulensis and L. glaucescens can be considered as natural sources of biocompounds with antihypertensive potential used either as infusions or fermented beverages.

The objective of this study was to evaluate the physicochemical and microbiological parameters of pork meat submitted to dry salting. Sodium chloride (NaCl) was added at levels of 0%, 2.5%, 5%, 7.5% or 10% by the meat weight. Dry salting technique was used, which consists of rubbing the sodium chloride manually, followed by a rest period. The data were submitted to analysis of variance using a completely randomized experimental design. The means were compared by Duncan test at 5%. The salting process reduced (P < 0.05) humidity and water activity, and it increased (P < 0.05) ash, chloride, palmitic acid, and water holding capacity levels compared to those of the control. Luminosity (L*) was lower (P < 0.05) in the control, and a* color was more intense in samples with 2.5% NaCl. Cooking loss was lower (P < 0.05) in the samples salted with 5% and 10% NaCl, and similarity was observed between the levels 0 and 7.5% salt. The treatments with levels 0% and 2.5% NaCl had higher mesophilic counts. The other microbiological parameters were within limits established by law. Therefore, salting with 5% NaCl can be used in pork meat in order to maintain the physicochemical and microbiological characteristics of the final product.

Focusing on the therapeutic and cosmetic potentials of the thermal water, several processes were developed to achieve a raw material known as fango which presents in its constitution water, clay and organic soil. This research work aimed at characterizing turf, sulphur mud and fango from Araxa, MG, Brazil, through physical, physicochemical, inorganic and organic assessments for cosmetic and topical product proposes. The characterization permitted the determination of relevant parameters to suggest the efficacy (presence, of ions) and safety (absence of toxic metals) of those raw materials for cosmetic and pharmaceutical utilization.

This study aims to optimize the water quality monitoring of a polluted watercourse (Leça River, Portugal) through the principal component analysis (PCA) and cluster analysis (CA). These statistical methodologies were applied to physicochemical, bacteriological and ecotoxicological data (with the marine bacterium Vibrio fischeri and the green alga Chlorella vulgaris) obtained with the analysis of water samples monthly collected at seven monitoring sites and during five campaigns (February, May, June, August, and September 2006). The results of some variables were assigned to water quality classes according to national guidelines. Chemical and bacteriological quality data led to classify Leça River water quality as “bad” or “very bad”. PCA and CA identified monitoring sites with similar pollution pattern, giving to site 1 (located in the upstream stretch of the river) a distinct feature from all other sampling sites downstream. Ecotoxicity results corroborated this classification thus revealing differences in space and time. The present study includes not only physical, chemical and bacteriological but also ecotoxicological parameters, which broadens new perspectives in river water characterization. Moreover, the application of PCA and CA is very useful to optimize water quality monitoring networks, defining the minimum number of sites and their location. Thus, these tools can support appropriate management decisions.

ABSTRACTThe composition of propolis depends on time, vegetation and the location of the collection area. The objective of this study was to determine the physicochemical characteristics, the concentration of phenol compounds and the antioxidant capacity of propolis of native stingless bees (Meliponinae)and Apis from the State of Tocantins. Extraction with 80% ethanol (v/v) was performed in order to obtain the extracts. Parameters examined were: propolis mass loss by desiccation at 105 ºC, ashes, wax concentration and pH. Furthermore, the propolis antioxidant activity was measured, as well as the total concentration of phenol compounds. The extracts were also analyzed by high performance liquid chromatography. The total concentration of phenol compounds varied between 121.78 and 631.29 (mg GAE g-1). The antioxidant activity expressed by the value of CE50 varied between 29.81 and 845.38 µg mL-1. High performance liquid chromatography analysis allowed us to infer the existence of phenol compounds. The results indicated that the studied propolis samples constitute good sources of natural antioxidants. The variety of phenol compounds identified in this study, and the diverse biological functions reported in literature for these compounds indicated that this stingless bee propolis (Meliponinae) and Apis has a high pharmacological potential.

OBJECTIVES: This study aimed at measuring the lipophilicity and ionization constants of diastereoisomeric dipeptides, interpreting them in terms of conformational behavior, and developing statistical models to predict them. METHODS: A series of 20 dipeptides of general structure NH(2) -L-X-(L or D)-His-OMe was designed and synthetized. Their experimental ionization constants (pK(1) , pK(2) and pK(3) ) and lipophilicity parameters (log P(N) and log D(7.4) ) were measured by potentiometry. Molecular modeling in three media (vacuum, water, and chloroform) was used to explore and sample their conformational space, and for each stored conformer to calculate their radius of gyration, virtual log P (preferably written as log P(MLP) , meaning obtained by the molecular lipophilicity potential (MLP) method) and polar surface area (PSA). Means and ranges were calculated for these properties, as was their sensitivity (i.e., the ratio between property range and number of rotatable bonds). RESULTS: Marked differences between diastereoisomers were seen in their experimental ionization constants and lipophilicity parameters. These differences are explained by molecular flexibility, configuration-dependent differences in intramolecular interactions, and accessibility of functional groups. Multiple linear equations correlated experimental lipophilicity parameters and ionization constants with PSA range and other calculated parameters. CONCLUSION: This study documents the differences in lipophilicity and ionization constants between diastereoisomeric dipeptides. Such configuration-dependent differences are shown to depend markedly on differences in conformational behavior and to be amenable to multiple linear regression.
