Protoplast fusion between sweet orange and mandarin/mandarin hybrids scion cultivars was performed following the model "diploid embryogenic callus protoplast + diploid mesophyll-derived protoplast". Protoplasts were isolated from embryogenic calli of 'Pera' and 'Westin' sweet orange cultivars (Citrus sinensis) and from young leaves of 'Fremont', Nules', and 'Thomas' mandarins (C. reticulata), and 'Nova' tangelo [C. reticulata x (C. paradisi x C. reticulata)]. The regenerated plants were characterized based on their leaf morphology (thickness), ploidy level, and simple sequence repeat (SSR) molecular markers. Plants were successfully generated only when 'Pera' sweet orange was used as the embryogenic parent. Fifteen plants were regenerated being 7 tetraploid and 8 diploid. Based on SSR molecular markers analyses all 7 tetraploid regenerated plants revealed to be allotetraploids (somatic hybrids), including 2 from the combination of 'Pera' sweet orange + 'Fremont' mandarin, 3 'Pera' sweet orange + 'Nules' mandarin, and 2 'Pera' sweet orange + 'Nova' tangelo, and all the diploid regenerated plants showed the 'Pera' sweet orange marker profile. Somatic hybrids were inoculated with Alternaria alternata and no disease symptoms were detected 96 h post-inoculation. This hybrid material has the potential to be used as a tetraploid parent in interploid crosses for citrus scion breeding.

Despite considerable research conducted on 'Tahiti' lime [Citrus latifolia (Yu Tanaka) Tanaka] in several countries, few long-term studies have focused on rootstock effects on fruit production and quality under non-irrigated conditions. As for many other fruit crops, rootstock studies for 'Tahiti' lime are often based on the evaluation of several horticultural responses simultaneously, instead of considering multivariate statistical approaches which may provide with more comprehensive information. Consequently, a trial was installed to evaluate the horticultural performance of non-irrigated 'Tahiti' lime trees budded onto the following 12 rootstocks: 'HRS 801' and 'HRS 827' hybrids; 'Rubidoux', 'FCAV' and 'Flying Dragon' trifoliates; 'Sun Chu Sha Kat' and 'Sunki' mandarins; 'Cravo Limeira' and 'Cravo FCAV' 'Rangpur' limes; 'Carrizo' citrange, 'Swingle' citrumelo, and 'Orlando' tangelo. The trial was installed in 2001, in an 8 m x 5 m spacing with no supplementary irrigation. Measurements of yield, fruit quality oriented to different consuming markets, canopy volume and tree tolerance to drought, were performed. A multivariate cluster analysis identified both 'Rangpur' lime rootstocks as those inducing larger cumulative yield and higher percentage of fruits for the domestic market, with highest drought tolerance to the trees. Despite of their high susceptibility to drought stress under non-irrigated conditions, the 'Flying Dragon' and 'FCAV' trifoliate rootstocks performed outstandingly for 'Tahiti' lime, inducing higher yield efficiency, early bearing and larger percentage of high-quality fruits for foreign markets, with smaller trees more suitable for high-density plantings. El objetivo de este trabajo fue evaluar la tolerancia al estrés salino en el portainjerto Citrumelo cultivar 75 AB (Citrus paradisi x Poncirus trifoliata), en comparación con Mandarino Cleopatra (Citrus retuculata), conocido por su elevada tolerancia. Se incubaron plántulas entre toallas de papel, humedecidas con agua destilada o soluciones de NaCl 30 mM. El diseño experimental fue completamente aleatorizado con 3 repeticiones, y los datos se analizaron mediante ANOVA y Test de Tukey. Luego de 41 días de ensayo se determinó el peso fresco, contenido relativo de agua (CRA), concentración de prolina y composición mineral. El crecimiento de Cleopatra fue más sensible a la salinidad que el de Citrumelo cultivar 75 AB. El CRA se mantuvo constante en ambos portainjertos. El ajuste osmótico se realizó mediante la acumulación de prolina en hojas; su concentración fue mayor en Citrumelo 75 AB. Este último excluyó los iones Na+ y Cl- de la parte aérea, restringiéndolos al sistema radicular, mientras que en Cleopatra se observó lo opuesto. Se concluye que Citrumelo cultivar 75 AB es más tolerante a la salinidad que Cleopatra, y excluye los iones Na+ y Cl- de la parte aérea.

High quality snap bean ( Phaseolus vulgaris L. ) can be produced under rain-fed conditions, provided that adequate moisture is available. However, drought may occur at any stage of growth of snap bean. The objective of this study was to evaluate the effect of drought stress at different growth stages on pod physical quality and nutrient concentrations. An experiment was conducted at the Horticulture Greenhouse, Hawassa University in Ethiopia. Drought stress (50% of field capacity [FC]) was applied at the unfolding of the fourth trifoliate leaf, flowering and pod formation, against a control with no drought stress. The drought stress treatments and eight cultivars were arranged as a factorial experiment in a completely randomised design, with three replications. Drought stress (50% FC) during reproductive stages significantly (P<0.05) reduced pod texture, appearance, and pod curvature. Drought stress increased protein and zinc concentrations by 41 and 15%, respectively; but reduced iron concentration by 15% in snap bean pods. All the tested cultivars had relatively similar responses to drought stress.

A produção de citrinos encontra-se amplamente disseminada por todos os continentes, representando uma atividade económica geradora de milhões de euros. As crescentes exigências dos consumidores conduzem ao aparecimento de novas variedades, em detrimento das cultivares tradicionais. A produção de tangerineira ‘Setubalense’ tem vindo a sofrer um acentuado declínio, mas os consumidores mais exigentes em aromas cítricos e fragrâncias intensas continuam a apreciar esta cultivar. Por isso, esta cultivar tem algum espaço comercial para mercados diferenciados em que se valorizam as cultivares tradicionais. Estas características e a sua época de maturação, que implica ser pouco afetada pela Ceratitis capitata, fazem da ‘Setubalense’ uma cultivar recomendável para o modo de produção biológico. Com o objetivo de caracterizar o padrão produtivo alternante e tendo por objeto de estudo um pomar de 5 anos de tangerineira ‘Setubalense’ enxertado sobre citranjeira ‘Troyer’ [Citrus sinensis (L.) Osbeck x Poncirus trifoliata L.], foi avaliada a composição da rebentação e da floração nos anos de safra e de contrassafra. A intensidade e as características da rebentação dependem do quadrante da árvore e do ano (safra ou contrassafra). Nos anos de safra a percentagem de nós rebentados oscila entre 67,6 a 79,5 %, que contrasta com os 25,4 a 28,2 % dos anos de contrassafra, função do quadrante analisado. Nos anos produtivos os rebentos formados são essencialmente florais e nos improdutivos quase exclusivamente vegetativos. A formação de elevado número de flores nos anos de safra conduz a uma intensa abcisão de órgãos generativos. O número de folhas formada por cada 100 nós é similar nos anos de safra e de contrassafra. Os resultados obtidos refletem o padrão alternante desta cultivar, evidenciando o cuidado particular que esta cultura tem de ter na sua condução.

RESUMO: O objetivo deste trabalho foi avaliar o desempenho de inseticidas autorizados emergencialmente para o controle de Helicoverpa armigera (Lepidoptera: Noctuidae) em soja. Sete inseticidas foram pulverizados em campo e, após 24 horas, folhas do ponteiro foram coletadas e oferecidas para lagartas de 2o instar em laboratório. Lagartas do 4o instar receberam a última folha trifoliolada que se encontrava completamente expandida no momento da pulverização. Outro grupo foi exposto a folhas coletadas a partir de 72 horas da pulverização. Em campo, seis inseticidas foram pulverizados e, em seguida, as plantas foram infestadas com lagartas de 2o e 3o instar. No primeiro estudo, flubendiamida, clorantraniliprole, clorfenapir, indoxacarbe e metoxifenozida causaram 100% de mortalidade do 4o instar aos oito dias após o início da exposição, enquanto baculovírus e Bacillus thuringiensis (Bt) propiciaram mortalidade de 60-75%, que evoluiu para 88?90% ao final da fase de pupa. Para o 2o instar, apenas flubendiamida e clorantraniliprole proporcionaram mortalidade de 100%. Flubendiamida, clorantraniliprole e clorfenapir apresentaram o menor tempo letal para o 4o instar, e flubendiamida e clorantraniliprole, para o 2o instar. Após 72 horas da pulverização, o desempenho dos inseticidas foi insatisfatório. Em campo, houve eficiência satisfatória de flubendiamida, espinosade, baculovírus e Bt sobre lagartas de 2o e 3o instar. ABSTRACT:The objective of this work was to evaluate the performance of insecticides authorized on an emergency basis to control of Helicoverpa armigera (Lepidoptera: Noctuidae) in soybean. Seven insecticides were sprayed on the field and, 24 hours after that, soybean pointer leaves were collected and offered to 2nd instar larvae in the laboratory. Fourth instar larvae received the last trifoliate leaf that was fully expanded at the time of spraying. Another larvae group was exposed to leaves collected from 72 hours onwards after spraying. In the field, six insecticides were sprayed, and then the plants were infested with 2nd and 3rd instar larvae. In the first study, flubendiamide, chlorantraniliprole, chlorfenapyr, indoxacarb, and methoxyfenozide caused 100% mortality of the 4th instar, eight days after the beginning of exposure, while baculovirus and Bacillus thuringiensis (Bt) caused 60?75% mortality, which reached 88?90% at the end of the pupal stage. For 2nd instar larvae, only flubendiamide and chlorantraniliprole caused 100% mortality. Flubendiamide, chlorantraniliprole, and chlorfenapyr showed the lowest lethal time for the 4th instar, and flubendiamide and chlorantraniliprole for the 2nd instar. Seventy-two hours after spraying, the performance of insecticides was not satisfactory. In the field, there was satisfactory efficiency of flubendiamide, spinosad, baculovirus, and Bt on 2nd and 3rd instar larvae.
