La Nación ha sido una de las más importantes construcciones políticoculturales de la Modernidad. Las premisas que le han dado mayor vitalidad han sido y son, pues, su afirmación como imaginario de identidad para los pueblos y su significación en el desarrollo de las denominadas "relaciones internacionales", en cuanto formas de expresión de las interrelaciones globales de los sectores dominantes. Este estudio se basa en el análisis de esas premisas a la luz de algunas de las nuevas perspectivas emanadas de los paradigmas político y cultural, desarrollados en las ciencias sociales a partir de las últimas décadas del siglo pasado, y aplicadas a los estudios del territorio nacional, su construcción, sus mutaciones y los imaginarios espaciales que de ellos derivan. A partir de algunas de las visiones críticas mencionadas, en la actualidad el concepto de nación parece haber entrado en un proceso de erosión o descomposición. Fenómenos sociales, económicos, políticos y culturales ligados a los procesos de reestructuración global (migraciones intercontinentales, sistemas mediáticos globales, exclusión social, discriminación étnica y religiosa, marginación y falta de participación política, nuevas formas de manifestación social...) y a la construcción de los discursos posmodernos hacen aparecer la nación como un concepto, al menos, puesto en tela de juicio. Los más audaces hablan del virtual ingreso a un mundo "posnacional". ¿Significará esto la extinción de una idea y una realidad con la que se han formado culturalmente millones de personas de numerosas generaciones en los puntos más lejanos del planeta? La respuesta deberá estar planteada en el estudio real y especializado de esos fenómenos y de las nuevas realidades que de ellos emergen.

The Tara Oceans Expedition (2009-2013) sampled the world oceans on board a 36 m long schooner, collecting environmental data and organisms from viruses to planktonic metazoans for later analyses using modern sequencing and state-of-the-art imaging technologies. Tara Oceans Data are particularly suited to study the genetic, morphological and functional diversity of plankton. The present data set provides continuous measurements made with an Aquatic Laser Fluorescence Analyzer (ALFA) (Chekalyuk et al., 2014), connected in-line to the TARA flow through system during 2013. The ALFA instrument provides dual-wavelength excitation (405 and 514 nm) of laser-stimulated emission (LSE) for spectral and temporal analysis. It offers in vivo fluorescence assessments of phytoplankton pigments, biomass, photosynthetic yield (Fv/Fm), phycobiliprotein (PBP)-containing phytoplankton groups, and chromophoric dissolved organic matter (CDOM) (Chekalyuk and Hafez, 2008; 2013A). Spectral deconvolution (SDC) is used to assess the overlapped spectral bands of aquatic fluorescence constituents and water Raman scattering (R). The Fv/Fm measurements are spectrally corrected for non-chlorophyll fluorescence background produced by CDOM and other constituents (Chekalyuk and Hafez, 2008). The sensor was cleaned weakly following the manufacturer recommended protocol.

Stonemasonry of the Gothic vault in its totality is based upon geometry of the line, whereas classic stereotomy relies on the comprehensive knowledge of the surface and the highly sophisticated sides of the voussoirs necessary for its vaults. It is obvious that this leap in the art of construction was paralleled and accompanied by an extension of the horizons of geometry. In Spain, it was made possible thanks to the centuries-old tradition of stone building begun in the most remote medieval times and to the presence of outstanding architects or stonemasons such as Juan de Álava, whose professional work surpassed the established limits and provided the art of building with new instruments.

La evidencia empírica aplicada a países de cierto tamaño y desarrollo económico, muestra que existe una relación directa y positiva entre la intensidad de la industria manufacturera, medida como porcentaje de su PIB, y ciertas variables económicas como, el crecimiento, el desempleo y la balanza exterior de bienes y servicios. En el caso de esta última, se verifica empíricamente, que los países con una proporción de actividad manufacturera inferior al 20%, tienen una marcada tendencia a presentar déficits crónicos de balanza de bienes y servicios, lo que conduce a persistentes déficits por cuenta corriente, al ser el primer déficit el principal componente del segundo. Esto trae consigo un continuado incremento del endeudamiento externo que no cesa, y que terminará en algún momento por desequilibrar el conjunto de la economía de los países con esos déficits crónicos. Las anteriores conclusiones, abren una vía de orientación de la política económica, que tiene como objetivo la promoción de la industria manufacturera de cada país. Y esto es un hecho ya en 2014. Países relevantes, como Alemania o Francia en la UE, incluso los EEUU y últimamente el Reino Unido, y por supuesto países del área asiática como Japón, Corea del Sur y China, llevan años promoviendo su industria manufacturera. Resulta significativo, que el debate ideológico sobre la bondad de la aplicación política industrial por parte de los gobiernos, frente a las teorías liberales de mantener a los poderes públicos lejos de ese tipo de actividades, haya dado paso a un modelo generalizado de corte más bien horizontal, donde los países casi sin excepciones apoyan el desarrollo de sus empresas con numerosos instrumentos, que van bastante más lejos de los habituales de I+D. Se valora por tanto, la industria manufacturera como algo vital para el equilibrio económico. Incluso la UE, defensora durante décadas de la no intervención de los diferentes Estados miembros en actividades de promoción industrial más allá del apoyo a las actividades de I+D, realiza un giro copernicano, que termina en 2012 proclamando que la industria manufacturera es vital para el equilibrio económico de la UE, que hay que promoverla, e incluso marca un objetivo, precisamente del 20%, como contribución manufacturera a su PIB. Es decir, se da por asumido que los servicios no son un sustituto indefinido de la industria y que por tanto tienen un límite, lo que se contrapone frontalmente contra la anterior creencia de que el aumento de la participación de los servicios en la economía, no solo era bueno, sino un síntoma de desarrollo. Esta premisa ya ha dejado de ser cierta para esos y otros países. En cambio, en España nada de esto sucede en las dos últimas décadas, sino que la industria manufacturera no recibe especial atención de los poderes públicos y se desliza en una pendiente de disminución de su contribución al PIB, que incluso se acelera con la crisis económica que comienza en 2007, hasta alcanzar cifras del orden del 12% del PIB en 2013. La política económica que se aplica es la de la deflación de costes, con los efectos consecuentes sobre los salarios y sobre la capacidad de la economía de generar riqueza. Se apuesta por un modelo de mano de obra barata, que recuerda al de los años 60. Como indicador relevante de esta situación, lo que exporta la industria manufacturera española, no ha ganado en contenido tecnológico en los últimos quince años. Esta situación se ve empeorada por un hecho significativo adicional, y es que casi el 40% de las ventas de la industria manufacturera española provienen de empresas de propiedad extranjera, con lo que eso supone por una parte de dependencia tecnológica del exterior como en el caso del automóvil, y de incertidumbre sobre su futuro, al estar basadas en el mantenimiento en el futuro de una mano de obra barata, que frenará que los españoles que trabajan en esas empresas, progresen económicamente. La propuesta de esta Tesis, es en cambio apostar por un modelo de crecimiento para España que tenga como uno de sus pilares el desarrollo de una industria manufacturera sólida y con cada vez mayor contenido tecnológico. Para ello, se propone un Plan de política industrial, donde se incluye la creación de actores impulsores de este plan, que deben ser del máximo nivel político. Si los diferentes gobiernos no entienden y asumen esta necesidad de apoyo a la industria a largo plazo e independiente de los cambios políticos, no será posible llevar a cabo este Plan. Para su puesta en marcha, se propone la creación o refuerzo de numerosos instrumentos de apoyo a la industria manufacturera de carácter fundamentalmente horizontal que van mucho más allá de los habituales del I+D, y que en varios casos, tienen una clara semejanza con otros existentes ya en otros países desarrollados desde hace años. La promoción de la industria manufacturera necesita nuevos instrumentos, como una financiación a largo plazo para las empresas, una promoción ordenada y eficaz de la actividad internacional de las empresas exportadoras, la mayoría de las cuales exportan productos manufacturados, una educación y formación profesional que esté alineada con estos objetivos, unos instrumentos que apoyen en especial el desarrollo la industria manufacturera, o la participación minoritaria pero significativa, del Estado en empresas españolas pertenecientes a sectores estratégicos entre otros. En resumen, esta Tesis propone una alternativa de política económica radicalmente diferente a la de dejar la industria manufacturera española a su suerte, y basar el futuro económico de España en una mano de obra barata. ABSTRACT The empirical evidence, applied to countries of certain size and economic development, shows that there exists a direct and positive relationship between industrial manufacturing activity, measured as a percentage of GDP, and certain economic variables, such as growth, unemployment and the foreign balance of trade. In the case of the latter, it is verified empirically that the countries with a percentage of manufacturing activity below 20% have a marked tendency for chronic deficits of the balance of trade, leading to persistent deficits in the current account, being that the former deficit is the main component of the latter. This brings about a continued increase in foreign debt that does not cease, and that will end at some point by disrupting the economy of the countries with these chronic deficits. The previous conclusions open the way to a new direction for economic policy, which promotes industrial manufacturing in each country. This is already a fact in 2014. Relevant countries, such as Germany or France in the EU, even the US and ultimately the UK, and of course countries of East Asia such as Japan, South Korea and China, have been promoting their industrial manufacturing for years. It becomes significant that the ideological debate about the goodwill of the application of industrial policy by governments, against liberal theories that maintain public powers far from these kinds of activities, has taken a step towards a horizontal-cut generalized model, where countries, with almost no exception, rely on various instruments to develop their companies that go much further than the usual R&D. Industrial manufacture is therefore valued as vital for economic stability. Even the EU, proponent for decades of non-intervention policy that goes beyond R&D, has gone full circle, ending in 2012 by proclaiming that industrial manufacture is vital for the economic stability of the EU, that it must be promoted. They even mark precisely 20% as an objective for manufacturing as a percentage of GDP. In other words, it is a given that services are not an indefinite substitute for industry, and that therefore it has a limit as such. This rejects the notion that the increase in services at the cost of manufacture is not only healthy, but is also a symptom of development. This premise is no longer true for these and other countries. On the other hand, none of this happens in Spain, where industrial manufacture receives no special attention from the public authorities, and it slides on a downward slope of percentage contribution to GDP, which accelerates the economic crisis that begins in 2007, until manufacture reaches values of around 12% of GDP in 2013. The economic policy applied is that of cost deflation, with consequential effects on wages and the capacity of the economy to generate wealth. A model is proposed for cheaper labor, akin to that of the 1960s. As a relevant indicator of this situation, manufacturing exports from Spain have not grown technologically in the last 15 years. The situation is made worse by another significant fact: almost 40% of sales of the manufacturing industry originate from companies of foreign origin, which supposes on one hand a technological dependence on foreign countries, such as in the case of the automotive industry, and on the other hand uncertainty in its future, being that they are based on maintaining cheap labor in the future, which will slow economic progress of Spaniards working in these companies. The proposition of this Thesis is to bet on a growth model for Spain that has as one of its pillars the development of a solid manufacturing industry, with increasing technological content. For this, an industrial policy plan is proposed, which includes the creation of driving agents for this plan, which must be of maximum political level. If the various governments don’t understand and assume this necessity for support of industry in the long term, independent of political change, this plan will not be accomplished. To start it, the creation or reinforcement of numerous instruments to promote the manufacturing activities are proposed, with a fundamentally horizontal nature that goes far beyond the usual R&D, and that, in several cases, have a clear similarity with others existing in other countries, having been developed for years. The promotion of the manufacturing industry needs new instruments, such as the long-term financing of companies, an orderly and efficient promotion of international activity of exporting companies, the most of which export manufactured goods, education and professional training which is in tune with these objectives, some instruments which support in particular the development of the manufacturing industry, or the minor yet significant participation of the State in Spanish companies belonging to strategic sectors, among others. In summary, this Thesis proposes an different alternative to the economic policy of leaving the manufacturing industry of Spain to its chances, and to base the economic future of Spain on a cheaper labor force.

La degradación del suelo ha adquirido una magnitud preocupante. Los métodos tradicionales de descontaminación, son costosos e insuficientes. La fitorremediación representa una alternativa eficaz, de bajo coste, respetuosa con el medio ambiente, que además mejora las propiedades del suelo, si bien ha habido desarrollos relevantes en la última década. Desde el punto de vida científico, el reto principal es descifrar las rutas metabólicas implicadas en respuesta a contaminantes y comprender su regulación. Esta información es imprescindible si aspiramos a mejorar las capacidades naturales de algunas especies vegetales para remediar los suelos contaminados. Los estudios de esta Tesis se han centrado en Populus, el mejor modelo forestal disponible a raíz de la secuenciación de su genoma completo. Por otra parte, Populus tiene una gran capacidad natural para la degradación de contaminantes orgánicos, lo que explica su predominio en los programas forestales de fitorremediación que se desarrollan actualmente. Hemos elegido en concreto al híbrido Populus tremula x P. alba, por la facilidad con que se cultiva y su particular interés biotecnológico. La presente Tesis plantea un estudio comprehensivo de la respuesta molecular a bifenilos policlorados (PCBs), una familia de contaminantes orgánicos persistentes de particular relevancia a escala mundial. Se ha utilizado para ello una aproximación transcriptómica, basada en tecnología RNA-seq, para identificar los genes implicados en el metabolismo de los compuestos in planta y cuantificar sus niveles de activación en distintas situaciones controladas. La tesis pretende asimismo definir el control transcripcional subyacente a la respuesta bioquímica frente a este tipo de contaminantes. Resulta sorprendente que dicha respuesta sea prácticamente desconocida a nivel molecular, a pesar de su gran potencial aplicado en el contexto de la tecnología fitorremediadora. Para desarrollar este proyecto aplicamos a nuestros cultivos de chopo híbridos concentraciones diferentes de Aroclor 1221, una mezcla de PCBs muy utilizada a nivel comercial durante décadas, su uso está prohibido hoy internacionalmente. Y tomamos muestras de RNA a dos concentraciones y dos momentos distintos de exposición al contaminante, generando así una matriz de cuatro elementos con sus controles correspondientes. Con el fin de incrementar la especificidad de nuestro análisis, consideramos sobre todo los genes diferencialmente expresados más significativos según cuatro algoritmos estadísticos distintos. Por otra parte, realizamos análisis funcionales con herramientas bioinformáticas basadas en comparaciones de secuencias y en redes de co-expresión génica. La respuesta de los genes de particular interés fue validada mediante tecnología qRT-PCR (reacción de la polimerasa en cadena cuantitativa en tiempo real). Se trata del primer estudio comprehensivo de la respuesta de un organismo vegetal ante la presencia de PCBs. Este estudio nos ha permitido identificar una cantidad considerable de genes estructurales y reguladores, definiendo nuevos factores de transcripción cuya expresión es proporcional a la concentración de contaminante en el medio o al tiempo de exposición al mismo. Los análisis de correlación nos permiten afirmar en que la respuesta metabólica a PCBs, incluyendo posibles rutas degradadoras, participan en al menos quince factores de transcripción y unas cuarenta proteínas o enzimas que resultan particularmente inducidas. Entre las familias implicadas destacan los citocromos P450, la glutatión transferasas, las deshidrogenasas reductasas (short-chain dehydrogenase reductase) y las proteínas MDR (multi-drug resistance). Mientras que los factores de transcripción encontrados pertenecen a la familia de ZF-TF, MYBs, WRKYs entre otros. También identificamos proteínas de función desconocida que no se habían vinculado previamente a este tipo de respuestas en plantas, como la CSP (cold-shock domain proteins). Para estudiar su posible relación con la presencia de PCBs, se caracterizó un gen de esta familia detectado mediante espectrometría de masas en tándem (MS/MS) a partir de mapas IEF x SDS-PAGE (isoelectro focusing x sodium dodecyl sulphate- polyacrylamide gel electrophoresis) de alta resolución. Mediante qRT-PCR pudimos confirmar la inducción del gen correspondiente, ortólogo a PtCSP4 de P. trichocarpa (Potri.004g172600), en respuesta a Aroclor 1221. El análisis fenotípico de las líneas transgénicas de Arabidopsis thaliana que sobre-expresaba la proteína CSP de chopo híbrido confirmó un papel para la misma tolerancia a PCBs, posiblemente a través de mecanismos reguladores que activan proteínas MDR. Este trabajo, además de aportar datos novedosos sobre los mecanismos moleculares desencadenados por la presencia de un PCB en Populus, utilizado aquí como sistema modelo. Con ello se demuestra el potencial de las especies arbóreas no solo como agentes descontaminantes, ya explotado comercialmente, sino también como fuente potencial de genes interesantes. Entre los genes identificados en esta Tesis hay candidatos evidentes a participar en mecanismos de tolerancia al estrés inducido por la contaminación y también rutas metabólicas degradadores de PCBs. Precisamente la posibilidad de degradar al contaminante confiere particular interés a este tipo de estudios frente a la fitorremediación de metales pesados y otros contaminantes elementales. La comparación de los datos generados en este estudio con estudios análogos que se realicen en el futuro con otras especies y xenobióticos, contribuirán a definir mejor la respuesta de las plantas ante la contaminación orgánica y mejorar su potencial descontaminante. ABSTRACT Soil degradation has acquired a disturbing magnitude. Traditional methods of decontamination are expensive and insufficient. Phytoremediation represent an effective alternative, low cost, respectful of the environment, that also improves soil properties, although there have been relevant developments in the last decade. From a life scientist, the challenge is to decipher the major metabolic pathways involved in response to pollutants and understand their regulation. This information is essential if we desire to enhance the natural abilities of some plant species to remediate contaminated soils. This thesis studies have focused on Populus, the best available forestry model following the sequencing of the entire genome. Moreover, Populus has a natural ability to degrade organic pollutants, which explains its predominance in phytoremediation forestry programs currently being developed. We have chosen specifically to hybrid Populus tremula x P. alba, the ease with which it is grown and its particular biotechnological interest. This thesis presents a comprehensive study of the molecular response to polychlorinated biphenyls (PCBs), a family of persistent organic pollutants of particular relevance worldwide. It has been used for a transcriptomic approach using RNA-seq technology, to identify genes involved in the metabolism of compounds in plant and quantify their levels of activation in different controlled situations. The thesis also aims to define the underlying transcriptional control the biochemical response to these pollutants. It is surprising that the response is virtually unknown at the molecular level, despite its great potential applied in the context of phytoremediation technology. To develop this project we applied our hybrid poplar crops different concentrations of Aroclor 1221, a mixture of PCBs widely used commercially for decades, its use is now banned internationally. And we RNA samples at two different concentrations and times of exposure to the pollutant, generating an array of four elements with their corresponding controls. In order to increase the specificity of our analysis, we consider mainly the most significant differentially expressed genes in four different statistical algorithms. Moreover, functional analyzes conducted with bioinformatics tools based on sequence comparisons and networks gene co-expression. The response of genes of particular interest was validated by qRT-PCR (polymerase reaction chain in real-time quantitative. This is the first comprehensive study of the response of a plant organism in the presence of PCBs. This study allowed us to identify a considerable amount of structural and regulatory genes, defining new transcription factors whose expression is proportional to the concentration of contaminant in the middle or at the time of exposure. Correlation analyzes allow us to affirm that the metabolic response to PCBs, including possible degradative pathways, at least fifteen involved in transcription factors and forty proteins or enzymes which are particularly induced. Among the families involved include cytochromes P450, the glutathione transferases, dehydrogenases reductases (short -chain dehydrogenase reductase) and MDR proteins (multi - drug resistance). While transcription factors belong to the family found ZF-TF, MYBs, WRKYs among others. We also identify proteins of unknown function that had not been previously linked to such responses in plants such as CSP (cold- shock domain proteins). To study their possible relationship with the presence of PCBs, a gene in this family was characterized and was detected by tandem mass spectrometry (MS/MS) from maps IEF x SDS -PAGE (sodium dodecyl isoelectro x sulphate- polyacrylamide gel electrophoresis) of high resolution. By qRT -PCR could confirm the induction of the corresponding gene, ortholog to PtCSP4 of P. trichocarpa (Potri.004g172600), in response to Aroclor 1221. Phenotypic analysis of transgenic Arabidopsis thaliana lines over- expressing the protein CSP poplar hybrid confirmed a role for PCBs same tolerance, possibly through regulatory mechanisms activated MDR proteins. This work, in addition to providing new data on the molecular mechanisms triggered by the presence of PCBs in Populus, used here as a model system. Thus the potential of tree species not only as decontamination agents, and commercially exploited, but also as a potential source of interesting genes is shown. Among the genes identified in this thesis there are evident candidates to participate in tolerance mechanisms to stress induced by pollution and degrading metabolic pathways of PCBs. Precisely the possibility of degrading the pollutant attaches particular interest to this type of study off the phytoremediation of heavy metals and other elemental pollutants. The comparison of the data generated in this study with similar studies carried out in the future with other species and xenobiotics contribute to better define the response of plants to organic pollution and improve their decontamination potential.

Spodoptera species, representing widespread polyphagous insect pests, are resistant to Bacillus thuringiensis δ-endotoxins used thus far as insecticides in transgenic plants. Here we describe the chemical synthesis of a cryIC gene by a novel template directed ligation–PCR method. This simple and economical method to construct large synthetic genes can be used when routine resynthesis of genes is required. Chemically phosphorylated adjacent oligonucleotides of the gene to be synthesized are assembled and ligated on a single-stranded, partially homologous template derived from a wild-type gene (cryIC in our case) by a thermostable Pfu DNA ligase using repeated cycles of melting, annealing, and ligation. The resulting synthetic DNA strands are selectively amplified by PCR with short specific flanking primers that are complementary only to the new synthetic DNA. Optimized expression of the synthetic cryIC gene in alfalfa and tobacco results in the production of 0.01–0.2% of total soluble proteins as CryIC toxin and provides protection against the Egyptian cotton leafworm (Spodoptera littoralis) and the beet armyworm (Spodoptera exigua). To facilitate selection and breeding of Spodoptera-resistant plants, the cryIC gene was linked to a pat gene, conferring resistance to the herbicide BASTA.

Death-associated protein kinase (DAP-kinase) is a Ca+2/calmodulin-regulated serine/threonine kinase with a multidomain structure that participates in apoptosis induced by a variety of signals. To identify regions in this protein that are critical for its proapoptotic activity, we performed a genetic screen on the basis of functional selection of short DAP-kinase-derived fragments that could protect cells from apoptosis by acting in a dominant-negative manner. We expressed a library of randomly fragmented DAP-kinase cDNA in HeLa cells and treated these cells with IFN-γ to induce apoptosis. Functional cDNA fragments were recovered from cells that survived the selection, and those in the sense orientation were examined further in a secondary screen for their ability to protect cells from DAP-kinase-dependent tumor necrosis factor-α-induced apoptosis. We isolated four biologically active peptides that mapped to the ankyrin repeats, the “linker” region, the death domain, and the C-terminal tail of DAP-kinase. Molecular modeling of the complete death domain provided a structural basis for the function of the death-domain-derived fragment by suggesting that the protective fragment constitutes a distinct substructure. The last fragment, spanning the C-terminal serine-rich tail, defined a new regulatory region. Ectopic expression of the tail peptide (17 amino acids) inhibited the function of DAP-kinase, whereas removal of this region from the complete protein caused enhancement of the killing activity, indicating that the C-terminal tail normally plays a negative regulatory role. Altogether, this unbiased screen highlighted functionally important regions in the protein and revealed an additional level of regulation of DAP-kinase apoptotic function that does not affect the catalytic activity.

The aim of this paper is to contribute to the estimation of the potential effects of the CAP reform on propensity to transaction, particularly comparing the effect of different new instruments/policy settings with the current policy (CAP health check) used as a baseline. The work is focused on three of new policy instruments within the post 2013 CAP reform proposal: regionalization, greening and capping. The first and second are analysed in more detail. The analysis will be based on a survey of farmers in the Province of Bologna, Emilia Romagna, Italy. The questionnaire focuses on mechanism of access to land and related incentives towards different land use/economic behaviour. The survey includes information about respondent characteristics (farm, farmer, household and payments received) and stated intention about potential changes in land operated under alternative agricultural policy scenarios (particularly the post-2013 reform proposals).

In the aftermath of the crisis, new instruments of economic governance have been adopted at the EU level. Until recently, these have been strongly dominated by what I assume to be the ECFIN coalition. However, at least since 2011, this coalition’s supremacy has been challenged by the competing coalition’s (EPSCO) willingness to rebalance the economic governance so that social concerns are better taken into account. Hence, drawing on the agenda-setting literature in the EU context, this working paper aims at retracing the process that has led to put this issue of the social dimension of the EMU on to the EU political agenda. Three hypotheses are made concerning the rise of this issue, the strategies employed by agenda-setters, and the policy subsystem of the economic governance. First, this study shows that the interest in this issue has been gradually fostered ‘from below’, at the level of the European Parliament and the European Commission. Second, due to its ‘high politics’ nature, this issue could only be initiated ‘from above’ (European Council) and then expanded to lower levels of decision-making (Commission). Specifically, DG EMPL has managed to attract attention to this issue and to build its credibility in dealing with it by strategically framing the issue and directing it towards the EPSCO venue. Finally, I analyze the outcome of this agenda-setting process by assessing to what extent the two new social scoreboards which form part of this social dimension have been taken into account during the 2014 European semester. The result of this analysis is that the new economic governance has not been genuinely rebalanced insofar as its dominant policy core remains that of the ECFIN coalition.

The intracellular stages of apicomplexan parasites are known to extensively modify their host cells to ensure their own survival. Recently, considerable progress has been made in understanding the molecular details of these parasite-dependent effects for Plasmodium-, Toxoplasma- and Theileria-infected cells. We have begun to understand how Plasmodium liver stage parasites protect their host hepatocytes from apoptosis during parasite development and how they induce an ordered cell death at the end of the liver stage. Toxoplasma parasites are also known to regulate host cell survival pathways and it has been convincingly demonstrated that they block host cell major histocompatibility complex (MHC)-dependent antigen presentation of parasite epitopes to avoid cell-mediated immune responses. Theileria parasites are the masters of host cell modulation because their presence immortalises the infected cell. It is now accepted that multiple pathways are activated to induce Theileria-dependent host cell transformation. Although it is now known that similar host cell pathways are affected by the different parasites, the outcome for the infected cell varies considerably. Improved imaging techniques and new methods to control expression of parasite and host cell proteins will help us to analyse the molecular details of parasite-dependent host cell modifications.

We describe the creation process of the Minimum Information Specification for In Situ Hybridization and Immunohistochemistry Experiments (MISFISHIE). Modeled after the existing minimum information specification for microarray data, we created a new specification for gene expression localization experiments, initially to facilitate data sharing within a consortium. After successful use within the consortium, the specification was circulated to members of the wider biomedical research community for comment and refinement. After a period of acquiring many new suggested requirements, it was necessary to enter a final phase of excluding those requirements that were deemed inappropriate as a minimum requirement for all experiments. The full specification will soon be published as a version 1.0 proposal to the community, upon which a more full discussion must take place so that the final specification may be achieved with the involvement of the whole community. The need to measure the response of the oculomotor system, such as ocular accommodation, accurately and in real-world environments is essential. New instruments have been developed over the past 50 years to measure eye focus including the extensively utilised and well validated Canon R-1, but in general these have had limitations such as a closed field-of-view, a poor temporal resolution and the need for extensive instrumentation bulk preventing naturalistic performance of environmental tasks. The use of photoretinoscopy and more specifically the PowerRefractor was examined in this regard due to its remote nature, binocular measurement of accommodation, eye movement and pupil size and its open field-of-view. The accuracy of the PowerRefractor to measure refractive error was on averaging similar, but more variable than subjective refraction and previously validated instrumentation. The PowerRefractor was found to be tolerant to eye movements away from the visual axis, but could not function with small pupil sizes in brighter illumination. The PowerRefractor underestimated the lead of accommodation and overestimated the slope of the accommodation stimulus response curve. The PowerRefractor and the SRW-5000 were used to measure the oculomotor responses in a variety of real-world environment: spectacles compared to single vision contract lenses; the use of multifocal contact lenses by pre-presbyopes (relevant to studies on myopia retardation); and ‘accommodating’ intraocular lenses. Due to the accuracy concerns with the PowerRefractor, a purpose-built photoretinoscope was designed to measure the oculomotor response to a monocular head-mounted display. In conclusion, this thesis has shown the ability of photoretinoscopy to quantify changes in the oculomotor system. However there are some major limitations to the PowerRefractor, such as the need for individual calibration for accurate measures of accommodation and vergence, and the relatively large pupil size necessary for measurement.

Food allergy affects 6% of children but there is no cure, and strict avoidance of index allergens along with immediate access to rescue medication is the current best management. With specialist care, morbidity from food allergy in children is generally low, and mortality is very rare. However, there is strong evidence that food allergy and food hypersensitivity has an impact on psychological distress and on the quality of life (QoL) of children and adolescents, as well as their families. Until recently, the measurement of QoL in allergic children has proved difficult because of the lack of investigative tools available. New instruments for assessing QoL in food allergic children have recently been developed and validated, which should provide further insights into the problems these children encounter and will enable us to measure the effects of interventions in patients. This review examines the published impact of food allergy on affected children, adolescents and their families. It considers influences such as gender, age, disease severity, co-existing allergies and external influences, and examines how these may impact on allergy-related QoL and psychological distress including anxiety and depression. Implications of the impact are considered alongside avenues for future research.

This study uses a purpose-built corpus to explore the linguistic legacy of Britain’s maritime history found in the form of hundreds of specialised ‘Maritime Expressions’ (MEs), such as TAKEN ABACK, ANCHOR and ALOOF, that permeate modern English. Selecting just those expressions commencing with ’A’, it analyses 61 MEs in detail and describes the processes by which these technical expressions, from a highly specialised occupational discourse community, have made their way into modern English. The Maritime Text Corpus (MTC) comprises 8.8 million words, encompassing a range of text types and registers, selected to provide a cross-section of ‘maritime’ writing. It is analysed using WordSmith analytical software (Scott, 2010), with the 100 million-word British National Corpus (BNC) as a reference corpus. Using the MTC, a list of keywords of specific salience within the maritime discourse has been compiled and, using frequency data, concordances and collocations, these MEs are described in detail and their use and form in the MTC and the BNC is compared. The study examines the transformation from ME to figurative use in the general discourse, in terms of form and metaphoricity. MEs are classified according to their metaphorical strength and their transference from maritime usage into new registers and domains such as those of business, politics, sports and reportage etc. A revised model of metaphoricity is developed and a new category of figurative expression, the ‘resonator’, is proposed. Additionally, developing the work of Lakov and Johnson, Kovesces and others on Conceptual Metaphor Theory (CMT), a number of Maritime Conceptual Metaphors are identified and their cultural significance is discussed.
