Biogeochemical measurements in sediment cores collected with the submersible JAGO (pusch cores) and a TV-MUC in the Black Sea during MSM15/1, Northwest Crimea (HYPOX Project), at water depths between 152-156 m. A series of microbial mats were sampled on the hypoxic region of the Crimean Shelf. Concentrations of organic carbon (Corg) and nitrogen (N) were measured on finely powdered, freeze-dried subsamples of sediment using a using a Fisons NA-1500 elemental analyzer. For organic carbon determination samples were pre-treated with 12.5% HCl to remove carbonates. Chlorophyll a (chl a), phaeopigments (PHAEO) and chloroplastic pigment equivalents (CPE) was measured according to Schubert et al., (2005) and total hydrolyzable amino acids (THAA) and single amino acid: ASP, GLU, SER, HIS, GLY, THR, ARG, ALA, TYR, MET, VAL, PHE, ILE, LEU, LYS following Dauwe et al., 1998.

En el artículo introduciremos en primer lugar el nuevo entorno en que se enmarca el cambio sustancial en la naturaleza de la Web tal y como la conocemos. Adoptaremos para ello un enfoque híbrido, diseccionando con un poco más de detalle los elementos tecnológicos, socioeconómicos, administrativos y legislativos que, a nuestro parecer, pueden conformar el contexto que enmarcaría un escenario factible para la introducción de una Gobernanza 2.0 ?de nueva generación? capaz de integrar de una manera sostenible ?ecosistémicamente viable? tecnologías y procesos que soporten de forma natural el paso de una era post-industrial de la información en la que vivimos hace unos años, hacia una sociedad de la información, poblada por verdaderos infociudadanos (nativos digitales en una creciente mayoría) y que debería ser sólo el paso previo para la última utopía: la sociedad del conocimiento, un estadio de la evolución sociotécnica al que se pretende llegar por el ancestral método de la profecía autocumplida, ignorando una realidad vigente considerablemente compleja.

En una Web dominada por los medios sociales para la información, la relación y la comunicación, la dinámica que se establece entre contenidos, personas y tecnología cambia radicalmente. Ante la relevancia que cobra el contenido generado por usuarios en este escenario –esencialmente relacional-, la localización de las mejores fuentes de información requiere sistemas recomendadores que incorporen la naturaleza social de una Web que va más allá de la primigenia internet. Se revisan las aproximaciones actuales a los procesos de recomendación, poniéndolas en el contexto de las tendencias asociadas al fenómeno del social computing. Asimismo, se destacan algunas líneas de actuación en la redefinición del problema de la recomendación en un panorama dominado por las redes sociales y la generación de contenidos por los usuarios

En este trabajo se han identificado los deslizamientos y desprendimientos que se pueden desarrollar en el acantilado de El Rincón (Gran Canaria) y sus repercusiones sobre la Autovía GC-2, en la base del acantilado. El análisis de estabilidad realizado para las condiciones actuales indica que la ladera es estable. El análisis a largo plazo considera el sustrato saturado en agua y se limita a los dos bloques rocosos más susceptibles de experimentar deslizamiento: uno a media ladera y otro en coronación. El deslizamiento del bloque de coronación parece más probable ya que sólo requiere la saturación de los piroclastos y está favorecido por la progresiva apertura de la grieta subvertical. Por último, la evaluación de los desprendimientos rocosos que afectan al acantilado indica que los bloques no alcanzarían la calzada de la autovía GC-2; sino que se acumularían en los conos de deyección presentes al pie de la ladera.

Este artículo trata dos temas estrechamente vinculados: por un lado, se presenta parte del trabajo realizado en la Tesis denominada “Modelo Híbrido de Ayuda al Capitán”, cuyos planteamientos han servido de base para la realización del proyecto de I+D+i denominado “e- Flow. Sistema integral de soporte a la evacuación”, segundo tema que se trata en este artículo. Se describirá una herramienta desarrollada para la creación de un modelo de las redes de evacuación (Macromodelo) y otra para realizar simulaciones de comportamiento de individuos (Micromodelo), se hablará de alternativas a los simulacros y se especificarán los requerimientos de una infraestructura física y de servicios para un sistema de ayuda al capitán. Sobre el proyecto e-Flow, se dará una visión general, describiendo la infraestructura física y de servicios. Se mostrará el potencial que puede suponer la implantación de un sistema cuyas posibilidades van más allá del soporte a la evacuación.

Previously metal-ion sites have been used as structural and functional probes in seven transmembrane receptors (7TM), but as yet all the engineered sites have been inactivating. Based on presumed agonist interaction points in transmembrane III (TM-III) and -VII of the β2-adrenergic receptor, in this paper we construct an activating metal-ion site between the amine-binding Asp-113 in TM-III—or a His residue introduced at this position—and a Cys residue substituted for Asn-312 in TM-VII. No increase in constitutive activity was observed in the mutant receptors. Signal transduction was activated in the mutant receptors not by normal catecholamine ligands but instead either by free zinc ions or by zinc or copper ions in complex with small hydrophobic metal-ion chelators. Chelation of the metal ions by small hydrophobic chelators such as phenanthroline or bipyridine protected the cells from the toxic effect of, for example Cu2+, and in several cases increased the affinity of the ions for the agonistic site. Wash-out experiments and structure–activity analysis indicated, that the high-affinity chelators and the metal ions bind and activate the mutant receptor as metal ion guided ligand complexes. Because of the well-understood binding geometry of the small metal ions, an important distance constraint has here been imposed between TM-III and -VII in the active, signaling conformation of 7TM receptors. It is suggested that atoxic metal-ion chelator complexes could possibly in the future be used as generic, pharmacologic tools to switch 7TM receptors with engineered metal-ion sites on or off at will.

Hereditary hemochromatosis (HH) is the most common autosomal recessive disorder known in humans. A candidate gene for HH called HFE has recently been cloned that encodes a novel member of the major histocompatibility complex class I family. Most HH patients are homozygous for a Cys-282→Tyr (C282Y) mutation in HFE gene, which has been shown to disrupt interaction with β2-microglobulin; a second mutation, His-63→Asp (H63D), is enriched in HH patients who are heterozygous for C282Y mutation. The aims of this study were to determine the effects of the C282Y and H63D mutations on the cellular trafficking and degradation of the HFE protein in transfected COS-7 cells. The results indicate that, while the wild-type and H63D HFE proteins associate with β2-microglobulin and are expressed on the cell surface of COS-7 cells, these capabilities are lost by the C282Y HFE protein. We present biochemical and immunofluorescence data that indicate that the C282Y mutant protein: (i) is retained in the endoplasmic reticulum and middle Golgi compartment, (ii) fails to undergo late Golgi processing, and (iii) is subject to accelerated degradation. The block in intracellular transport, accelerated turnover, and failure of the C282Y protein to be presented normally on the cell surface provide a possible basis for impaired function of this mutant protein in HH.

The cell adhesion molecule L1 is a potent inducer of neurite outgrowth and it has been implicated in X-linked hydrocephalus and related neurological disorders. To investigate the mechanisms of neurite outgrowth stimulated by L1, attempts were made to identify the neuritogenic sites in L1. Fusion proteins containing different segments of the extracellular region of L1 were prepared and different neuronal cells were assayed on substrate-coated fusion proteins. Interestingly, both immunoglobulin (Ig)-like domains 2 and 6 (Ig2, Ig6) promoted neurite outgrowth from dorsal root ganglion cells, whereas neural retinal cells responded only to Ig2. L1 Ig2 contains a previously identified homophilic binding site, whereas L1 Ig6 contains an Arg-Gly-Asp (RGD) sequence. The neuritogenic activity of Ig6 was abrogated by mutations in the RGD site. The addition of RGD-containing peptides also inhibited the promotion of neurite outgrowth from dorsal root ganglion cells by glutathione S-transferase-Ig6, implicating the involvement of an integrin. The monoclonal antibody LM609 against αvβ3 integrin, but not an anti-β1 antibody, inhibited the neuritogenic effects of Ig6. These data thus provide the first evidence that the RGD motif in L1 Ig6 is capable of promoting neurite outgrowth via interaction with the αvβ3 integrin on neuronal cells.

The cDNAs of two new human membrane-associated aspartic proteases, memapsin 1 and memapsin 2, have been cloned and sequenced. The deduced amino acid sequences show that each contains the typical pre, pro, and aspartic protease regions, but each also has a C-terminal extension of over 80 residues, which includes a single transmembrane domain and a C-terminal cytosolic domain. Memapsin 2 mRNA is abundant in human brain. The protease domain of memapsin 2 cDNA was expressed in Escherichia coli and was purified. Recombinant memapsin 2 specifically hydrolyzed peptides derived from the β-secretase site of both the wild-type and Swedish mutant β-amyloid precursor protein (APP) with over 60-fold increase of catalytic efficiency for the latter. Expression of APP and memapsin 2 in HeLa cells showed that memapsin 2 cleaved the β-secretase site of APP intracellularly. These and other results suggest that memapsin 2 fits all of the criteria of β-secretase, which catalyzes the rate-limiting step of the in vivo production of the β-amyloid (Aβ) peptide leading to the progression of Alzheimer's disease. Recombinant memapsin 2 also cleaved a peptide derived from the processing site of presenilin 1, albeit with poor kinetic efficiency. Alignment of cleavage site sequences of peptides indicates that the specificity of memapsin 2 resides mainly at the S1′ subsite, which prefers small side chains such as Ala, Ser, and Asp.

The reaction center (RC) from Rhodobacter sphaeroides converts light into chemical energy through the reduction and protonation of a bound quinone molecule QB (the secondary quinone electron acceptor). We investigated the proton transfer pathway by measuring the proton-coupled electron transfer, kAB(2) [QA⨪QB⨪ + H+ → QA(QBH)−] in native and mutant RCs in the absence and presence of Cd2+. Previous work has shown that the binding of Cd2+ decreases kAB(2) in native RCs ≈100-fold. The preceding paper shows that bound Cd2+ binds to Asp-H124, His-H126, and His-H128. This region represents the entry point for protons. In this work we investigated the proton transfer pathway connecting the entry point with QB⨪ by searching for mutations that greatly affect kAB(2) (≳10-fold) in the presence of Cd2+, where kAB(2) is limited by the proton transfer rate (kH). Upon mutation of Asp-L210 or Asp-M17 to Asn, kH decreased from ≈60 s−1 to ≈7 s−1, which shows the important role that Asp-L210 and Asp-M17 play in the proton transfer chain. By comparing the rate of proton transfer in the mutants (kH ≈ 7 s−1) with that in native RCs in the absence of Cd2+ (kH ≥ 104 s−1), we conclude that alternate proton transfer pathways, which have been postulated, are at least 103-fold less effective.

Neutral residue replacements were made of 21 acidic and basic residues within the N-terminal half of the Halobacterium salinarium signal transducer HtrI [the halobacterial transducer for sensory rhodopsin I (SRI)] by site-specific mutagenesis. The replacements are all within the region of HtrI that we previously concluded from deletion analysis to contain sites of interaction with the phototaxis receptor SRI. Immunoblotting shows plasmid expression of the htrI-sopI operon containing the mutations produces SRI and mutant HtrI in cells at near wild-type levels. Six of the HtrI mutations perturb photochemical kinetics of SRI and one reverses the phototaxis response. Substitution with neutral amino acids of Asp-86, Glu-87, and Glu-108 accelerate, and of Arg-70, Arg-84, and Arg-99 retard, the SRI photocycle. Opposite effects on photocycle rate cancel in double mutants containing one replaced acidic and one replaced basic residue. Laser flash spectroscopy shows the kinetic perturbations are due to alteration of the rate of reprotonation of the retinylidene Schiff base. All of these mutations permit normal attractant and repellent signaling. On the other hand, the substitution of Glu-56 with the isosteric glutamine converts the normally attractant effect of orange light to a repellent signal in vivo at neutral pH (inverted signaling). Low pH corrects the inversion due to Glu-56 -> Gln and the apparent pK of the inversion is increased when arginine is substituted at position 56. The results indicate that the cytoplasmic end of transmembrane helix-2 and the initial part of the cytoplasmic domain contain interaction sites with SRI. To explain these and previous results, we propose a model in which (i) the HtrI region identified here forms part of an electrostatic bonding network that extends through the SRI protein and includes its photoactive site; (ii) alteration of this network by photoisomerization-induced Schiff base deprotonation and reprotonation shifts HtrI between attractant and repellent conformations; and (iii) HtrI mutations and extracellular pH alter the equilibrium ratios of these conformations.

The VPAC(1) receptor belongs to family B of G protein-coupled receptors (GPCR-B) and is activated upon binding of the vasoactive intestinal peptide (VIP). Despite the recent determination of the structure of the N terminus of several members of this receptor family, little is known about the structure of the transmembrane (TM) region and about the molecular mechanisms leading to activation. In the present study, we designed a new structural model of the TM domain and combined it with experimental mutagenesis experiments to investigate the interaction network that governs ligand binding and receptor activation. Our results suggest that this network involves the cluster of residues Arg(188) in TM2, Gln(380) in TM7, and Asn(229) in TM3. This cluster is expected to be altered upon VIP binding, because Arg(188) has been shown previously to interact with Asp(3) of VIP. Several point mutations at positions 188, 229, and 380 were experimentally characterized and were shown to severely affect VIP binding and/or VIP-mediated cAMP production. Double mutants built from reciprocal residue exchanges exhibit strong cooperative or anticooperative effects, thereby indicating the spatial proximity of residues Arg(188), Gln(380), and Asn(229). Because these residues are highly conserved in the GPCR-B family, they can moreover be expected to have a general role in mediating function.

Lower jaws (containing the teeth), eyes, and skin samples were collected from harp seals (Pagophilus groenlandicus) in the southeastern Barents Sea for the purpose of comparing age estimates obtained by 3 different methods, the traditional technique of counting growth layer groups (GLGs) in teeth and 2 novel approaches, aspartic acid racemization (AAR) in eye lens nuclei and telomere sequence analyses as a proxy for telomere length. A significant correlation between age estimates obtained using GLGs and AAR was found, whereas no correlation was found between GLGs and telomere length. An AAR rate (k Asp) of 0.00130/year ± 0.00005 SE and a D-enantiomer to L-enantiomer ratio at birth (D/L 0 value) of 0.01933 ± 0.00048 SE were estimated by regression of D/L ratios against GLG ages from 25 animals (12 selected teeth that had high readability and 13 known-aged animals). AAR could prove to be useful, particularly for ageing older animals in species such as harp seals where difficulties in counting GLGs tend to increase with age. Age estimation by telomere length did not show any correlation with GLG ages and is not recommended for harp seals.
