Pt(cur)(NH3)(2)](NO3) (1), a curcumin-bound cis-diammineplatinum(II) complex, nicknamed Platicur, as a novel photoactivated chemotherapeutic agent releases photoactive curcumin and an active platinum(II) species upon irradiation with visible light. The hydrolytic instability of free curcumin reduces upon binding to platinum(II). Interactions of 1 with 5'-GMP and ct-DNA indicated formation of platinum-bound DNA adducts upon exposure to visible light (lambda = 400-700 nm). It showed apoptotic photocytotoxicity in cancer cells (IC50 approximate to 15 mu M), thus forming (OH)-O-center dot, while remaining passive in the darkness (IC50 > 200 mu M). A comet assay and platinum estimation suggest Pt-DNA crosslink formation. The fluorescence microscopic images showed cytosolic localization of curcumin, thus implying possibility of dual action as a chemo-and phototherapeutic agent.

Ternary copper(Il) complexes of salicylaldehyde-histamine Schiff base (HL) and pyridyl ligands, viz. Cu(bpy)(L)](ClO4) (1) and Cu(dppz)(L)](C104) (2), where bpy is 2,2'-bipyridine (in 1) and dppz is dipyrido3,2-a:2',3'-c]phenazine (in 2), were synthesized, characterized and their DNA binding, photo-activated DNA cleavage activity and photocytotoxicity studied. The 1:1 electrolytic one-electron paramagnetic complexes showed a d-d band near 670 nm in aqueous DMF (1:1 v/v). The crystal structure of complex 1 showed the metal in CuN4O distorted square-pyramidal geometry. Complex 2 intercalatively binds to calf-thymus (ct) DNA with a binding constant (K-b) of similar to 10(5) M-1. It exhibited moderate chemical nuclease activity but excellent DNA photocleavage activity in red light of 647 nm forming (OH)-O-center dot radicals. It showed remarkable photocytotoxicity in human cervical cancer cells (HeLa) giving IC50 of 1.6 mu M in visible light (400-700 nm) with low dark toxicity. The photo-induced cell death is via generation of oxidative stress by reactive oxygen species.

Vitamin-B6 (VB6) Schiff base (H2L) copper(II) complexes of pyridyl bases, viz. Cu(bpy)(L)] (1), Cu(phen)(L)] (2) and Cu(dppz)(L)] (3), where bpy is 2,2'-bipyridine, phen is 1,10-phenanthroline and dppz is dipyrido3,2-a:2',3'c]phenazine are synthesized, characterized and their phto-induced anticancer activity studied. The non-electrolytic one electron paramagnetic complexes exhibit a d-d band near 700 nm in DMF. The dppz complex intercalatively binds to calf-thymus DNA with binding constant (K-b) values of similar to 10(6) M-1. This complex exhibits low chemical nuclease activity but excellent DNA photocleavage activity when irradiated with red light of 705 nm forming (OH)-O-center dot radical. It displays remarkable photocytotoxicity in human cervical cancer cells (HeLa) giving IC50 value of 0.9 mu M in visible light (400-700 nm) while being less toxic in darkness (IC50 : 23 mu M). The cellular uptake of the complexes seems to be via VB6 transporting membrane carrier mediated diffusion pathway. Photo-induced cell death follows apoptotic pathway involving photo-generated intracellular reactive oxygen species.

In the laser induced thermal fatigue simulation test on pistons, the high power laser was transformed from the incident Gaussian beam into a concentric multi-circular pattern with specific intensity ratio. The spatial intensity distribution of the shaped beam, which determines the temperature field in the piston, must be designed before a diffractive optical element (DOE) can be manufactured. In this paper, a reverse method based on finite element model (FEM) was proposed to design the intensity distribution in order to simulate the thermal loadings on pistons. Temperature fields were obtained by solving a transient three-dimensional heat conduction equation with convective boundary conditions at the surfaces of the piston workpiece. The numerical model then was validated by approaching the computational results to the experimental data. During the process, some important parameters including laser absorptivity, convective heat transfer coefficient, thermal conductivity and Biot number were also validated. Then, optimization procedure was processed to find favorable spatial intensity distribution for the shaped beam, with the aid of the validated FEM. The analysis shows that the reverse method incorporated with numerical simulation can reduce design cycle and design expense efficiently. This method can serve as a kind of virtual experimental vehicle as well, which makes the thermal fatigue simulation test more controllable and predictable. (C) 2007 Elsevier Ltd. Recent emphasis on ecosystem approaches to fisheries management renews interest in, and the need for, trophic information about fish communities. A program was started in 1980 at the National Marine Fisheries Service Galveston Laboratory to develop a trophic database for continental shelf fishes. Collections were made during 1982-1983 that were processed but never published, yet the data remain valid today for historical purposes and for delimiting food web components within ecosystem assessments. I examined spring, summer, and fall foods in offshore populations of nine common species of trawl-susceptible fishes, with particular reference to predation on commercial penaeid shrimps (Farfantepenaeus and Litopenaeus). Diets were evaluated with the Index of Relative Importance (IRI) which combines the occurrence, number, and weight of each food item. Bank sea bass (Centropristis ocyurus) and bighead searobin (Prionotus tribulus) primarily consumed crabs, more so by larger than smaller fish. Inshore lizardfish (Synodus foetens) was almost entirely piscivorous. Ocellated flounder (Ancylopsetta ommata) consumed fishes, crabs, and stomatopods. Dwarf sand perch (Diplectrum bivittatum), blackwing searobin (Prionotus rubio), rock sea bass (Centropristis philadelphica), southern kingfish (Menticirrhus americanus), and red snapper (Lutjanus campechanus) fed mainly on shrimps. Most fish diets varied with respect to size (age), time of day, area sampled, depth, or season. Rimapenaeus and Sicyonia were the most frequently identified shrimp genera - only five Farfantepenaeus and no Litopenaeus were identified in almost 4,300 fish stomachs. I also examined gonadal development and documented fish length-weight relationships. Ripe gonads were most frequently found during summer in dwarf sand perch, during fall in ocellated flounder and bighead searobin, and during spring for other species, except no ripe red snapper or bank sea bass were collected. Rock sea bass was found to be a protogynous hermaphrodite, while dwarf sand perch is a synchronous hermaphrodite. Only ocellated flounder and southern kingfish exhibited sex-related differences in length-weight relationships. (PDF contains 40 pages.)

A study was conducted to assess the status of ecological condition and potential human-health risks in subtidal estuarine waters throughout the North Carolina National Estuarine Research Reserve System (NERRS) (Currituck Sound, Rachel Carson, Masonboro Island, and Zeke’s Island). Field work was conducted in September 2006 and incorporated multiple indicators of ecosystem condition including measures of water quality (dissolved oxygen, salinity, temperature, pH, nutrients and chlorophyll, suspended solids), sediment quality (granulometry, organic matter content, chemical contaminant concentrations), biological condition (diversity and abundances of benthic fauna, fish contaminant levels and pathologies), and human dimensions (fish-tissue contaminant levels relative to human-health consumption limits, various aesthetic properties). A probabilistic sampling design permitted statistical estimation of the spatial extent of degraded versus non-degraded condition across these estuaries relative to specified threshold levels of the various indicators (where possible). With some exceptions, the status of these reserves appeared to be in relatively good to fair ecological condition overall, with the majority of the area (about 54%) having various water quality, sediment quality, and biological (benthic) condition indicators rated in the healthy to intermediate range of corresponding guideline thresholds. Only three stations, representing 10.5% of the area, had one or more of these indicators rated as poor/degraded in all three categories. While such a conclusion is encouraging from a coastal management perspective, it should be viewed with some caution. For example, although co-occurrences of adverse biological and abiotic environmental conditions were limited, at least one indicator of ecological condition rated in the poor/degraded range was observed over a broader area (35.5%) represented by 11 of the 30 stations sampled. In addition, the fish-tissue contaminant data were not included in these overall spatial estimates; however, the majority of samples (77% of fish that were analyzed, from 79%, of stations where fish were caught) contained inorganic arsenic above the consumption limits for human cancer risks, though most likely derived from natural sources. Similarly, aesthetic indicators are not reflected in these spatial estimates of ecological condition, though there was evidence of noxious odors in sediments at many of the stations. Such symptoms reflect a growing realization that North Carolina estuaries are under multiple pressures from a variety of natural and human influences. These data also suggest that, while the current status of overall ecological condition appears to be good to fair, long-term monitoring is warranted to track potential changes in the future. This study establishes an important baseline of overall ecological condition within NC NERRS that can be used to evaluate any such future changes and to trigger appropriate management actions in this rapidly evolving coastal environment. The coastal shrimp trawl fisheries have long been the focus of conservation actions to reduce turtle bycatch and mortality in the Gulf of Mexico and the U.S. Atlantic (NRC, 1990). Calculation of catch rates of sea turtles in shrimp trawls is necessary to evaluate the impact on sea turtle populations. In this paper we analyze sea turtle bycatch to provide an estimate of the current number of interactions with otter trawl gear as well as an estimate of the number of fatal inions in Southeast U.S. waters and the Gulf of Mexico. We also provide an estimate of the number of individuals likely to die in the future with the new regulations that will require an increase in the size of the escape openings in trutle excluder devices (TEDs). The new regulations will allow many more turtles to escape. Other gears also are discussed. (PDF contains 24 pages)

This assessment applies to cobia (Rachycentron canadum) located in the territorial waters of the U.S. Gulf of Mexico. Separation of the Gulf of Mexico and Atlantic Ocean is defined by the seaward extension of the Dade/Monroe county line in south Florida. Mixing of fish between the Atlantic and Gulf of Mexico occurs in the Florida Keys during winter months. Cobia annually migrate north in early spring in the Gulf to spawning grounds in the northern Gulf of Mexico, returning to the Florida Keys by winter. Catches of cobia in the Gulf of Mexico are dominated by recreational landings, accounting for nearly 90% of the total. Since 1980, the landings of cobia in the recreational fishery have remained fairly stable at around 400-600 mt with a slight peak of 1,014 mt in 1997. The recreational fishery was estimated to have landed 471 mt in 2000. The landings from the commercial fishery have shown a steady increase from 45 mt in 1980 to a peak of 120 mt in 1994, followed by a decline to 62 mt in 2000. The previous assessment of cobia occurred in 1996 using a virtual population analysis (VPA) model. For this analysis a surplus-production model (ASPIC) and a forward-projecting, age-structured population model programmed in the AD Model Builder (ADMB) software were applied to cobia data from the Gulf of Mexico. The primary data consisted of four catch-per-unit-effort (CPUE) indices derived from the Marine Recreational Fisheries Statistics Survey (MRFSS) (1981-1999), Southeast region headboat survey (1986-1999), Texas creel survey (1983-1999), and shrimp bycatch estimates (1980-1999). Length samples were available from the commercial (1983-2000) and recreational (1981-2000) fisheries. The ASPIC model applied to the cobia data provided unsatisfactory results. The ADMB model fit described the observed length composition data and fishery landings fairly well based on graphical examination of model residuals. The CPUE indices indicated some disagreement for various years, but the model fit an overall increasing trend from 1992-1997 for the MRFSS, headboat, and Texas creel indices. The shrimp bycatch CPUE was treated as a recruitment index in the model. The fit to these data followed an upward trend in recruitment from 1988-1997, but did not fit the 1994-1997 data points very well. This was likely the result of conflicting information from other data sources. Natural mortality (M) for cobia is unknown. As a result, a range of values for M from 0.2-0.4, based on longevity and growth parameters, were selected for use in the age-structured model. The choice of natural mortality appears to greatly influence the perceived status of the population. Population status as measured by spawning stock biomass in the last year relative to the value at maximum sustainable yield (SSB2000/SSBMSY), spawning stock biomass in the last year relative to virgin spawning stock biomass (SSB2000/S0), and static spawning stock biomass per recruit (SSBR) all indicate the population is either depleted, near MSY, or well above MSY depending on the choice of M. The variance estimates for these benchmarks are very large and in most cases ranges from depleted to very healthy status. The only statement that can be made with any degree of certainty about cobia in the Gulf of Mexico is that the population has increased since the 1980s. (PDF contains 61 pages)

The Scientific Forum on the Gulf of Mexico: The Islands in the Stream Concept took place in January 2008 in Sarasota, Florida. The purpose of the meeting was to bring together scientists and managers from around the Gulf of Mexico to discuss a range of topics on our knowledge of the Gulf of Mexico, from its geology to larger-scale connectivity to the Caribbean region, and their applications to the concept of a more integrated approach to area-based management. The forum included six panels of invited experts who spoke on the oceanographic and biological features in the Gulf of Mexico, including connections with Mexico and the Mesoamerican barrier reef system, and the legal and regulatory structure currently in place. The charge to the group was to share information, identify gaps in our knowledge, identify additional potential areas for protection, and discuss available science about connectivity and the potential value of establishing a marine protected area network in the Gulf of Mexico. In Central California, and elsewhere around the world, a great deal of discussion is occurring about the use of marine protected areas (MPAs) as a tool to help manage marine resources. This discussion is taking place because there is growing evidence that humans have depleted marine resources in many parts of the world, often despite strong regulatory efforts. Moreover, there is also mounting evidence that the degradation of marine resources began long ago, and we do not fully realize how much humans have altered “natural” environments. This uncertainty has led people to discuss the use of MPAs as a precautionary tool to prevent depletion or extinction of marine resources, and as a means of redressing past damages. The discussion about the use of marine reserves is increasing in intensity in California because several resource management agencies are considering reserves as they create or revise management plans. Often, the discussions surrounding this important public policy debate lead to questions about the biological or ecological value of existing marine protected areas. More than 100 MPAs exist along the coast of California. Many of these were established arbitrarily and lack specific purposes. Some California marine protected areas also have co-occurring or overlapping boundaries, have conflicting designations for use, and have conflicting rules and regulations. Because few of the existing marine protected areas have clearly articulated goals or objectives, however, it is difficult or impossible to evaluate their ecological effectiveness. We present data on ichthyoplankton distribution, abundance, and seasonality and supporting environmental information for four species of coastal pelagics from the family Clupeidae: round herring Etrumeus teres, scaled sardine Harengula jaguana, Atlantic thread herring Opisthonema oglinum, and Spanish sardine Sardinella aurita. Data are from 1982 and 1983 cruises across the northern Gulf of Mexico sponsored by the Southeastern Area Monitoring and Assessment Program (SEAMAP). This is the first such examination for these species on a multiyear and gulfwide scale. Bioproflles on reproductive biology, early life history, meristics, adult distribution, and fisheries characteristics are also presented for these species. During the summer, larval Atlantic thread herring and scaled and Spanish sardines were abundant on the inner shelf <40 m depth), but were rare or absent in deeper waters. Scaled sardine and thread herring were found virtually everywhere inner-shelf waters were sampled, but Spanish sardines were rare in the north-central Gulf. During 1982, larval Atlantic thread herring were the most abundant of the four target c1upeid species, whereas Spanish sardine were the most abundant during 1983. On the west Florida shelf, Spanish sardine dominated larval c1upeid populations both years. Scaled sardine larvae were the least abundant of the four species both years, but were still captured in 25% of inner-shelf bongo net collections. Round herring larvae, collected February-early June (primarily March-April), were abundant on the outer shelf (40-182 m depth) and especially off Louisiana. Over the 2-year period, outer-shelf mean abundance for round herring was 40.2 larvae/10 m2; inner-shelf mean abundances for scaled sardine, Atlantic thread herring, and Spanish sardine were 14.9, 39.2, and 41.9 larvae/l0 m2, respectively. ENGLISH: The map method, the Jones method, the variance-covariance method, and the Skellam method were used to study the migrations of tagged yellowfin tuna released off the southern coast of Mexico in 1960 and 1969. The first three methods are all useful, and each presents information which is complementary to that presented by the others. The Skellam method, as used in this report, is less useful. The movements of the tagged fish released in 1960 appeared to have been strongly directed, but this was probably caused principally by the distribution of the fishing effort. The effort was much more widely distributed in 1970, and the movements of the fish released in 1969 appeared to have been much less directed. The correlation coefficients derived from the variance-covariance method showed that it was not random, however. The small fish released in the Acapulco and 10°N-100°W areas in 1969 migrated to the Manzanillo area near the beginning of February 1970. The medium and large fish released in the same areas in the same year tended to migrate to the southeast throughout the first half of 1970, however.
