Coleus blumei Benth (Lamiaceae), popularly known in Brazil as heart-hurt or coleus-of-Java is an ornamental plant widely used due to the color of its leaves. Several species of the genus Coleus present compounds with antioxidant, antimicrobial and allopathic activities due to the presence of chemicals such coleonol, forskolin and rosmarinic acid. Therefore, this study aimed to carry out phytochemical study and biological tests to evaluate the antibacterial, antioxidant and allopathic activities of the 70% ethanol extract and its fractions ethyl acetate, dichloromethane and end aqueous fraction leaf of Coleus blumei Benth. The plant extract was prepared by turbolisis from the powder of dried leaves, as a solvent for extraction using 70% ethanol. The ethanol extract was fractionated with solvents dichloromethane and ethyl acetate. The phytochemical study identified the presence of saponins, free anthraquinones, flavonoids and anthocyanidins. In evaluating the antibacterial dichloromethane fraction showed activity against all microorganisms tested both in agar diffusion test and microdilution test. The 70% ethanol extract showed activity against the micro-organisms S. aureus and S. epidermidis in agar diffusion test and against all microorganisms tested in microdilution test. The ethyl acetate fraction showed activity against the micro-organism S. aureus in the agar diffusion test and against all microorganisms tested in microdilution test. The end aqueous fraction showed no activity against any micro-organism tested. In the test of evaluation activity allopathic dichloromethane fraction showed greater inhibition of seed germination and growth of tomato (Lycopersicon esculentum Mill), followed by 70% ethanolic extract, the ethyl acetate fraction and the end aqueous fraction. In this work evaluated the content of vitamin C and total carotenoid in tomatoes fresh and dehydrated for conserve, with posterior immersion in sauce, verifying its sensorial acceptability. Dehydration was performed in dryer with airflow at 70 °C to obtain a product with final moisture of 45%. Determinations were carried out ascorbic acid, soluble solids, acidity, sugars and carotenoid before and after the dehydration process. The dehydrated tomatoes had been immersed in three types of sauces (pepper sauce, oregano with parrot leaves sauce and garlic sauce) per 25 days. After this period, the dry tomatoes in conserve had been submitted to the sensorial analysis in the pate form, applying themselves preference test. The results had been showed that retention of vitamin C and total carotenoid in the dehydrated product showed retention of 14.0% and 44.3%, respectively, in tomatoes after the dehydration process. Con el objetivo de evaluar la influencia de las bandejas de celdas sobre la producción de tomate tipo italiano en el campo, se realizó este trabajo en Piracicaba, SP, Brasil, de mayo a agosto del 2005. Los tratamientos consistieron en cuatro volúmenes de recipiente, tres bandejas de poliestireno expandido de 121,2; 34,6 y 12,0 cm³ y de una bandeja de plástico rígido de 14,0 cm³ (72, 128, 288 y 450 celdas, respectivamente) combinadas con cuatro edades para el trasplante (19, 24, 29 y 34 días después de la siembra). El delineamiento para la producción de mudas fue completamente al azar, con cinco plantas por parcela y tres repeticiones. Se analizaron área foliar, altura, masa verde y seca de la parte aérea y raíz y la calidad de las mudas. En la producción a campo, el delineamiento fue en bloques al azar con diez plantas por parcela y tres repeticiones. Fueron evaluadas la precocidad para inicio de cosecha, producción comercial y total por planta. Volúmenes mayores de recipiente (121,2 y 34,6 cm³) presentaron mejor calidad de mudas. En la producción de frutos, el número comercial y total de frutos por planta fue superior en la muda de 24 días de edad, sin embargo, en la producción total de frutos, no hubo diferencia entre los tratamientos. Por otro lado, también se obtuvo precocidad para la cosecha en los tratamientos realizados en los volúmenes de 121,2 y 34,6 cm³. El volumen de recipiente de 14,0 cm³ (450 celdas) resultó en mudas de calidad muy inferior, alongadas y raquíticas.

Tomato (Lycopersicon esculentum Mill., Solanum lycopersicon L.) is one of the most popular vegetable throughout the world, and the importance of its cultivation is threatened by a wide array of pathogens. In the last twenty years this plant has been successfully used as a model plant to investigate the induction of defense pathways after exposure to fungal, bacterial and abiotic molecules, showing triggering of different mechanisms of resistance. Understanding these mechanisms in order to improve crop protection is a main goal for Plant Pathology. The aim of this study was to search for general or race-specific molecules able to determine in Solanum lycopersicon immune responses attributable to the main systems of plant defense: non-host, host-specific and induced resistance. Exopolysaccharides extracted by three fungal species (Aureobasidium pullulans, Cryphonectria parasitica and Epicoccum purpurascens), were able to induce transcription of pathogenesis-related (PR) proteins and accumulation of enzymes related to defense in tomato plants cv Money Maker,using the chemical inducer Bion® as a positive control. During the thesis, several Pseudomonas spp. strains were also isolated and tested for their antimicrobial activity and ability to produce antibiotics. Using as a positive control jasmonic acid, one of the selected strain was shown to induce a form of systemic resistance in tomato. Transcription of PRs and reduction of disease severity against the leaf pathogen Pseduomonas syringae pv. tomato was determined in tomato plants cv Money Maker and cv Perfect Peel, ensuring no direct contact between the selected rhizobacteria and the aerial part of the plant. To conclude this work, race-specific resistance of tomato against the leaf mold Cladosporium fulvum is also deepened, describing the project followed at the Phytopathology Laboratory of Wageningen (NL) in 2007, dealing with localization of a specific R-Avr interaction in transfected tomato protoplast cultures through fluorescence microscopy.

Our previous work has shown that localised activity of the cell-wall-loosening protein expansin is sufficient to induce primordia on the apical meristem of tomato, consistent with the hypothesis that tissue expansion plays a key role in leaf initiation. In this paper we describe the earliest morphogenic events visible on the surface of the apical meristem of tomato (Lycopersicon esculentum Mill.) following treatment with expansin and report on the spectrum of final structures formed. Our observations are consistent with a proposed primary function of expansin effecting morphogenesis via altered biophysical stress patterns in the meristem. The primordia induced by expansin do not complete the full program of leaf development. We present data indicating that one reason for this might be the inability of exogenous expansin to mimic the endogenous pattern of expansin activity in the meristem. These data provide the first detailed analysis at the cellular level of expansin action on living tissue, the first description of the spectrum of structures induced by expansin on the apical meristem, and give an insight into a potentially fundamental mechanism in plant development.

In order to determine the presence of Fusarium spp. in atmospheric dust and rainfall dust, samples were collected during September 2007, and July, August, and October 2008. The results reveal the prevalence of airborne Fusarium species coming from the atmosphere of the South East coast of Spain. Five different Fusarium species were isolated from the settling dust: Fusarium oxysporum, F. solani, F. equiseti, F. dimerum, and F. proliferatum. Moreover, rainwater samples were obtained during significant rainfall events in January and February 2009. Using the dilution-plate method, 12 fungal genera were identified from these rainwater samples. Specific analyses of the rainwater revealed the presence of three species of Fusarium: F. oxysporum, F. proliferatum and F. equiseti. A total of 57 isolates of Fusarium spp. obtained from both rainwater and atmospheric rainfall dust sampling were inoculated onto melon (Cucumis melo L.) cv. Piñonet and tomato (Lycopersicon esculentum Mill.) cv. San Pedro. These species were chosen because they are the main herbaceous crops in Almeria province. The results presented in this work indicate strongly that spores or propagules of Fusarium are able to cross the continental barrier carried by winds from the Sahara (Africa) to crop or coastal lands in Europe. Results show differences in the pathogenicity of the isolates tested. Both hosts showed root rot when inoculated with different species of Fusarium, although fresh weight measurements did not bring any information about the pathogenicity. The findings presented above are strong indications that long-distance transmission of Fusarium propagules may occur. Diseases caused by species of Fusarium are common in these areas. They were in the past, and are still today, a problem for greenhouses crops in Almería, and many species have been listed as pathogens on agricultural crops in this region. Saharan air masses dominate the Mediterranean regions. The evidence of long distance dispersal of Fusarium spp. by atmospheric dust and rainwater together with their proved pathogenicity must be taken into account in epidemiological studies.

Crop irrigation is a major consumer of energy. Only a few countries are self-sufficient in conventional non-renewable energy sources. Fortunately, there are renewable ones, such as wind, which has experienced recent developments in the area of power generation. Wind pumps can play a vital role in irrigation projects in remote farms. A methodology based on daily estimation balance between water needs and water availability was used to evaluate the feasibility of the most economic windmill irrigation system. For this purpose, several factors were included: three-hourly wind velocity (W3 h, m/s), flow supplied by the wind pump as a function of the elevation height (H, m) and daily greenhouse evapotranspiration as a function of crop planting date. Monthly volumes of water required for irrigation (Dr, m3/ha) and in the water tank (Vd, m3), as well as the monthly irrigable area (Ar, ha), were estimated by cumulative deficit water budgeting taking in account these factors. An example is given illustrating the use of this methodology on tomato crop (Lycopersicon esculentum Mill.) under greenhouse at Ciego de Ávila, Cuba. In this case two different W3 h series (average and low wind year), three different H values and five tomato crop planting dates were considered. The results show that the optimum period of wind-pump driven irrigation is with crop plating in November, recommending a 5 m3 volume tank for cultivated areas around 0.2 ha when using wind pumps operating at 15 m of height elevation.

Wind power can play an interesting role in irrigation projects in different areas. A methodology can determine the feasibility of the technology and the levels of daily irrigation demand satisfied by windmills at different levels of risk, using tomato (Lycopersicon esculentum Mill) as greenhouse crop. The present work compared the feasibility of the technology and the critical factors involved in three different countries: Cuba, Spain and Pakistan. The study considered as factors the wind speed level, the energy cost, the tomato prices, the reliability and distance to the electrical grid, and the crop development dates, determining the economic feasibility for each combination of factors in each country.

Experiments were performed on three abscisic acid (ABA)-deficient tomato (Lycopersicon esculentum Mill.) mutants, notabilis, flacca, and sitiens, to investigate the role of ABA and jasmonic acid (JA) in the generation of electrical signals and Pin2 (proteinase inhibitor II) gene expression. We selected these mutants because they contain different levels of endogenous ABA. ABA levels in the mutant sitiens were reduced to 8% of the wild type, in notabilis they were reduced to 47%, and in flacca they were reduced to 21%. In wild-type and notabilis tomato plants the induction of Pin2 gene expression could be elicited by heat treatment, current application, or mechanical wounding. In flacca and sitiens only heat stimulation induced Pin2 gene expression. JA levels in flacca and sitiens plants also accumulated strongly upon heat stimulation but not upon mechanical wounding or current application. Characteristic electrical signals evolved in the wild type and in the notabilis and flacca mutants consisting of a fast action potential and a slow variation potential. However, in sitiens only heat evoked electrical signals; mechanical wounding and current application did not change the membrane potential. In addition, exogenous application of ABA to wild-type tomato plants induced transient changes in membrane potentials, indicating the involvement of ABA in the generation of electrical signals. Our data strongly suggest the presence of a minimum threshold value of ABA within the plant that is essential for the early events in electrical signaling and mediation of Pin2 gene expression upon wounding. In contrast, heat-induced Pin2 gene expression and membrane potential changes were not dependent on the ABA level but, rather, on the accumulation of JA.

Fruit tissues of tomato (Lycopersicon esculentum Mill.) contain both photosynthetic and heterotrophic ferredoxin (FdA and FdE, respectively) isoproteins, irrespective of their photosynthetic competence, but we did not previously determine whether these proteins were colocalized in the same plastids. In isolated fruit chloroplasts and chromoplasts, both FdA and FdE were detected by immunoblotting. Colocalization of FdA and FdE in the same plastids was demonstrated using double-staining immunofluorescence microscopy. We also found that FdA and FdE were colocalized in fruit chloroplasts and chloroamyloplasts irrespective of sink status of the plastid. Immunoelectron microscopy demonstrated that FdA and FdE were randomly distributed within the plastid stroma. To investigate the significance of the heterotrophic Fd in fruit plastids, Glucose 6-phosphate dehydrogenase (G6PDH) activity was measured in isolated fruit and leaf plastids. Fruit chloroplasts and chromoplasts showed much higher G6PDH activity than did leaf chloroplasts, suggesting that high G6PDH activity is linked with FdE to maintain nonphotosynthetic production of reducing power. This result suggested that, despite their morphological resemblance, fruit chloroplasts are functionally different from their leaf counterparts.

A tomato gene that is induced early after infection of tomato (Lycopersicon esculentum Mill.) with root-knot nematodes (Meloidogyne javanica) encodes a protein with 54% amino acid identity to miraculin, a flavorless protein that causes sour substances to be perceived as sweet. This gene was therefore named LeMir (L. esculentum miraculin). Sequence similarity places the encoded protein in the soybean trypsin-inhibitor family (Kunitz). LeMir mRNA is found in root, hypocotyl, and flower tissues, with the highest expression in the root. Rapid induction of expression upon nematode infection is localized to root tips. In situ hybridization shows that LeMir is expressed constitutively in the root-cap and root-tip epidermis. The LeMir protein product (LeMir) was produced in the yeast Pichia pastoris for generation of antibodies. Western-blot analysis showed that LeMir expression is up-regulated by nematode infection and by wounding. LeMir is also expressed in tomato callus tissue. Immunoprint analysis revealed that LeMir is expressed throughout the seedling root, but that levels are highest at the root/shoot junction. Analysis of seedling root exudates revealed that LeMir is secreted from the root into the surrounding environment, suggesting that it may interact with soil-borne microorganisms.

A cDNA encoding for a functional ornithine decarboxylase has been isolated from a cDNA library of carpels of tomato (Lycopersicon esculentum Mill.). Ornithine decarboxylase in tomato is represented by a single-copy gene that we show to be up-regulated during early fruit growth induced by 2,4-dichlorophenoxyacetic acid and gibberellic acid.
