An in vitro model, the aggregating brain cell culture of fetal rat telencephalon, has been used to investigate the influence of glial cells on the neurotoxicity of two organophosphorus pesticides (OPs), chlorpyrifos and parathion. Mixed-cell aggregate cultures were treated continuously for 10 days between DIV 5 and 15. Parathion induced astrogliosis at concentration at which MAP-2 immunostaining, found here to be more sensitive than neuron-specific enzyme activities, was not affected. In contrast, chlorpyrifos induced a comparatively weak gliotic reaction, and only at concentrations at which neurons were already affected. After similar treatments, increased neurotoxicity of parathion and chlorpyrifos was found in aggregate cultures deprived of glial cells. These results suggest that glial cells provide neuroprotection against OPs toxicity. To address the question of the difference in toxicity between parathion and chlorpyrifos, the toxic effects of their leaving groups, p-nitrophenol and trichloropyridinol, were studied in mixed-cell aggregates. General cytotoxicity was more pronounced for trichloropyridinol and both compounds had similar toxic effects on neuron-specific enzyme activities. In contrast, trichloropyridinol induced a much stronger decrease in glutamine synthetase activity, the enzymatic marker of astrocytes. Trichloropyridinol may exert a toxic effect on astrocytes, compromising their neuroprotective function, thus exacerbating the neurotoxicity of chlorpyrifos. This is in line with the suggestion that glial cells may contribute to OPs neurotoxicity, and with the view that OPs may exert their neurotoxic effects through different mechanisms.

Brain activity is energetically costly and requires a steady and highly regulated flow of energy equivalents between neural cells. It is believed that a substantial share of cerebral glucose, the major source of energy of the brain, will preferentially be metabolized in astrocytes via aerobic glycolysis. The aim of this study was to evaluate whether uncoupling proteins (UCPs), located in the inner membrane of mitochondria, play a role in setting up the metabolic response pattern of astrocytes. UCPs are believed to mediate the transmembrane transfer of protons, resulting in the uncoupling of oxidative phosphorylation from ATP production. UCPs are therefore potentially important regulators of energy fluxes. The main UCP isoforms expressed in the brain are UCP2, UCP4, and UCP5. We examined in particular the role of UCP4 in neuron-astrocyte metabolic coupling and measured a range of functional metabolic parameters including mitochondrial electrical potential and pH, reactive oxygen species production, NAD/NADH ratio, ATP/ADP ratio, CO2 and lactate production, and oxygen consumption rate. In brief, we found that UCP4 regulates the intramitochondrial pH of astrocytes, which acidifies as a consequence of glutamate uptake, with the main consequence of reducing efficiency of mitochondrial ATP production. The diminished ATP production is effectively compensated by enhancement of glycolysis. This nonoxidative production of energy is not associated with deleterious H2O2 production. We show that astrocytes expressing more UCP4 produced more lactate, which is used as an energy source by neurons, and had the ability to enhance neuronal survival.

Insulin secretion from pancreatic beta cells is stimulated by glucose metabolism. However, the relative importance of metabolizing glucose via mitochondrial oxidative phosphorylation versus glycolysis for insulin secretion remains unclear. von Hippel-Lindau (VHL) tumor suppressor protein, pVHL, negatively regulates hypoxia-inducible factor HIF1alpha, a transcription factor implicated in promoting a glycolytic form of metabolism. Here we report a central role for the pVHL-HIF1alpha pathway in the control of beta-cell glucose utilization, insulin secretion, and glucose homeostasis. Conditional inactivation of Vhlh in beta cells promoted a diversion of glucose away from mitochondria into lactate production, causing cells to produce high levels of glycolytically derived ATP and to secrete elevated levels of insulin at low glucose concentrations. Vhlh-deficient mice exhibited diminished glucose-stimulated changes in cytoplasmic Ca(2+) concentration, electrical activity, and insulin secretion, which culminate in impaired systemic glucose tolerance. Importantly, combined deletion of Vhlh and Hif1alpha rescued these phenotypes, implying that they are the result of HIF1alpha activation. Together, these results identify pVHL and HIF1alpha as key regulators of insulin secretion from pancreatic beta cells. They further suggest that changes in the metabolic strategy of glucose metabolism in beta cells have profound effects on whole-body glucose homeostasis.

The study assessed growth and physiological parameters of 'Sunrise Golden' and 'Tainung 01' papaya seedlings grown in 280mL plastic tubes and watered using a low-cost automatic irrigation system adjusted to operate at substrate water tension for starting irrigation (STI) of 3.0, 6.0 or 9.0 kPa. The water depths applied by the dripping system and drainage were monitored during germination and seedling growth. Germination, emergence velocity index (EVI), leaf area, plant height, shoot and root dry weight, stomatal conductance, relative water content (RWC) and relative chlorophyll content (RCC) were evaluated. Soil nutrient levels were determined by electrical conductivity (EC). Water use efficiency (WUE) corresponded to the ratio of plant dry mass to depth of water applied. STI settings did not affect papaya germination or EVI. System configuration to 3.0 and 6.0 kPa STI exhibited the highest drainage and lowest EC and RCC, indicating soil nutrient loss and plant nutrient deficiency. Drainage was greater in tubes planted with the 'Tainung 01' variety, which developed smaller root systems and lower stomatal conductance than 'Sunrise Golden' seedlings. The highest values for shoot dry weight and WEU were obtained at 6.0 kPa STI for 'Sunrise Golden' (0.62 g and 0.69 g L-1) and at 9.0 kPa in 'Tainung 01' (0.35 g and 0.82 g L-1). RWC at 9.0 kPa STI was lower than at 3.0 kPa in both varieties. The results indicate that the low-cost technology developed for irrigation automation is promising. Even so, new studies are needed to evaluate low-flow irrigation systems as well as the nutrient and water needs of different papaya varieties.

Neutrophil extracellular traps (NETs) formation is a cell death mechanism characterized by the extrusion of DNA fibers associated to antimicrobial peptides such as LL37. Beside their antimicrobial role, NETs are highly immunogenic by their ability to activate plasmacytoid dendritic cells (pDCs). In this context, LL37 binds to NET-DNA, leading to endosomal Toll¬like-receptor (TLR) 9 binding, resulting in Interferon alpha (IFNa) production by pDCs. Uncontrolled pDC activation by NETs is an important player in the pathogenesis of autoimmune disease such as Lupus Erythematosus (LE); however the regulation of NET- driven pDC activation is poorly characterized. Olfactomedin 4 (OLFM4) is a granule protein present in a subset of circulating neutrophils and was shown to bear anti-inflammatory properties in a mouse model, raising the possibility that it may regulate neutrophil-induced inflammation. Therefore, in this project, we aimed at deciphering the mechanism by which OLFM4 may regulate inflammation induced by NET-activated pDC and its relevance in the pathogenesis of Lupus Erythematosus (LE). First, we show that OLFM4 directly interacted with LL37 in neutrophils, impairing LL37/DNA complexes formation and pDC activation to produce IFNa. Then, by using an in vivo model of acute inflammation depending on NET- driven activation of pDCs, we observed that the absence of Olfm4 led to uncontrolled type I IFN production, confirming the regulatory role of neutrophil-derived OLFM4. Beyond controlling NET-induced inflammation, we also show that OLFM4 could inhibit pDC activation mediated by DNA-containing immune complexes (ICs), suggesting that OLFM4 holds anti¬inflammatory properties in the context of LE. Of note, we identified a previously unknown population of OLFM4hi9h neutrophils in healthy individuals that may belong to the immunosuppressive subset of granulocytic myeloid-derived suppressor cells (g-MDSCs). Strikingly, we observed a decreased frequency of OLFM4h'9h cells among inflammatory Low density granulocytes (LDGs) neutrophils in LE patients, suggesting that a disequilibrium between pro- and anti-inflammatory neutrophils may participate to the disease pathogenesis. Altogether, this study demonstrates that OLFM4 is involved in the resolution of inflammation. -- La NETose (formation de Neutrophil Extracellular Traps, NETs) est une réponse à un stimulus inflammatoire caractérisée par l'expulsion de l'ADN lié à des peptides antimicrobiens comme le LL37, induisant la mort de la cellule. Les NETs possèdent des propriétés antibactériennes et sont pro-inflammatoires via leur capacité à activer les cellules dendritiques plasmacytoïdes (pDCs). Dans ce contexte, les complexes ADN/LL37 libérés lient le récepteur Toll-like 9 des pDCs, induisant la production d'Interféron alpha (IFNa). La production incontrôlée d'IFNa par les pDCs est impliquée dans la pathogenèse du Lupus Erythemateux (LE), cependant la régulation de l'activation des pDCs reste mal connue. L'Oflactomédine 4 (OLFM4) est une protéine produite par une sous-population de neutrophiles, avec des propriétés anti-inflammatoires possibles. Le but de ce projet était d'identifier les mécanismes par lesquels l'OLFM4 pourrait réguler l'inflammation induite par les NETs et sa relevance dans la pathogenèse du LE. Tout d'abord, nous avons montré que l'OLFM4 interagissait avec le LL37, empêchant la production des complexes ADN/LL37 qui activent les pDCs. Nous avons vérifié notre hypothèse in vivo en utilisant un modèle murin d'inflammation locale dépendant des pDCs et des NETs. Dans ce contexte, le déficit en Olfm4 était associé à une production accrue d'IFNa, confirmant le rôle de l'OLFM4 dans le contrôle de l'inflammation. De plus, l'OLFM4 pouvait également inhiber l'activation des pDCs induite par des complexes immuns, suggérant que l'OLFM4 serait aussi anti-inflammatoire dans le contexte du LE. Ensuite, nous avons identifié une nouvelle population de neutrophiles OLFM4h'9h chez les sujets sains qui pourraient appartenir au sous-type anti¬inflammatoire des g-MDSCs (granulocytic myeloid-derived suppressor cells). Nous avons observé une diminution de ces cellules parmi les neutrophiles pro-inflammatoires LDGs (Low Density Granulocytes) dans le LE suggérant qu'un déséquilibre entre les sous-types de neutrophiles pourrait participer à l'inflammation excessive de cette maladie. Ces travaux mettent en évidence l'implication de l'OLFM4 dans la résolution de l'inflammation et suggèrent qu'une expression altérée de l'OLFM4 pourrait participer à la pathogenèse du LE. -- Les neutrophils constituent la majorité des globules blancs circulants et sont rapidement mobilisés depuis le sang dans un organe lésé en cas d'infection ou de blessure. Ils représentent la première ligne de défense du système immunitaire. Ils sont indispensables dans la défense contre les infections par leur capacité à tuer les bactéries, par exemple en produisant des peptides antimicrobiens (AMPs) qui fonctionnent comme des antibiotiques naturels. De plus, les neutrophiles recrutent les autres membres du système immunitaire qui sont nécessaires à l'éradication complète des microbes et à la réparation des tissus. Les nombreux outils permettant aux neutrophiles de contrôler les infections ne sont cependant pas sans danger pour les tissus. En effet, diverses molécules comme les AMPs peuvent induire des dommages tissulaires substantiels en participant au développement d'une inflammation chronique. Ceci est particulièrement le cas lorsque les neutrophiles meurent par un processus nommé NETose. Dans ce contexte, la cellule subit une dissolution de sa membrane suivie de l'expulsion de son ADN associé à des AMPs. Ces complexes formés d'ADN et d'AMPs induisent la production de cytokines pro-inflammatoires dont l'Interféron alpha (IFNa). Certaines maladies auto-immunes comme le lupus érythémateux sont associées à un excès de NETose produit par les neutrophiles et à un excès d'IFNa qui participe au développement de la maladie. Dans cette thèse, nous avons montré que l'Olfactomédine 4 (OLFM4), une protéine produite par les neutrophiles eux-mêmes, est un inhibiteur de cette inflammation. Nous avons démontré que TOLFM4 empêchait la formation des complexes ADN/AMPs, réduisant par là la production d'IFNa in vitro et in vivo. Finalement, nos recherches ont suggéré que l'OLFM4 pourrait être insuffisamment produite chez les patients souffrant de lupus, ce qui pourrait participer à l'inflammation chronique associée à la maladie.

Follicle-stimulating hormone (FSH) and insulin regulate glycide metabolism in Sertoli cells, thus stimulating lactate production. These stimulatory effects of FSH and insulin do not require protein synthesis, suggesting a modulation of enzyme activity and/or regulation of glucose transport. The present investigation was performed to characterize the hormonal control of lipid metabolism in Sertoli cells. The data indicate that FSH and insulin have a regulatory effect on lipid metabolism in Sertoli cells. After 8 h of preincubation with insulin (5 µg/ml), the activity of the enzyme ATP-citrate lyase in cultured Sertoli cells was increased from 0.19 to 0.34 nmol NAD+ formed µg protein-1 min-1. FSH (100 ng/ml) had no effect on this enzyme. Glycerol phosphate dehydrogenase activity was not affected by any of the hormones tested. When Sertoli cells from 19-day old rats were incubated with [1,2­14C]acetate for 90 or 360 min, the [14C] label was present predominantly in triglyceride and phospholipid fractions with minor amounts in other lipids. In Sertoli cells pretreated for 16 h with insulin and FSH, an increase in acetate incorporation into lipids was observed. Most of the label was in esterified lipids and this percentage increased with the time of treatment; this increase was remarkable in triglycerides of control cells (18.8% to 30.6%). Since Sertoli cell triglycerides participate in the control of spermatogenesis, the present data suggest that the hormonal control of lipid metabolism in Sertoli cells is important not only for maintaining the energy of the cell itself, but also for the control of the spermatogenesis process.

Dans les élevages laitiers d’aujourd’hui, les déplacements de la caillette sont parmi les conditions chirurgicales les plus souvent rencontrées. Le pronostic pour ces pathologies est cependant très différent que l’on soit en présence d’une dilatation de la caillette à droite (DCD) ou d’un volvulus de la caillette (VC). En pratique, la distinction en période préopératoire entre ces deux conditions est difficile. Afin de limiter les pertes économiques associées aux DCD/VC, l’identification et la réforme précoce des animaux ayant le plus grand risque d’avoir une mauvaise évolution postopératoire deviennent des enjeux de premier plan. Les objectifs de cette étude étaient de déterminer la valeur pronostique de la L-lactatémie (LAC) mesurée à la ferme en préopératoire à l’aide d’un appareil portatif chez des vaches souffrant de DCD ou VC et de suggérer des seuils de LAC cliniquement significatifs. Nos résultats indiquent que la LAC est un bon indicateur pronostique lors de DCD/VC. Une LAC ≤ 2 mmol/L est un bon indicateur de succès chirurgical et s’avère très utile pour appuyer la décision d’opérer l’animal (sensibilité, spécificité, valeurs prédictives positive et négative de respectivement 76.2, 82.7, 53.3 et 93.1%). Par ailleurs, il n’y a aucun avantage économique à effectuer une chirurgie chez des vaches commerciales souffrant de DCD/VC qui ont une LAC ≥ 6 mmol/L (sensibilité, spécificité, valeurs prédictives positive et négative de respectivement 28.6, 97.5, 75 et 84%). Par conséquent, ces animaux devraient, dans la plupart des cas, être réformés en raison de la probabilité élevée qu’ils aient une mauvaise évolution postopératoire.

INTRODUCTION : L’utilisation de la circulation extracorporelle durant la chirurgie cardiaque est associée à des problèmes pulmonaires chez certains patients. L’utilisation d’une pression pulsatile induite par un ballon intra-aortique (BIA) pourrait diminuer la dysfonction endothéliale et la survenue de tels événements. MATÉRIEL ET MÉTHODE : 12 porcs Landrace-Yorkshire ont subi une circulation extracorporelle et ont été divisés en deux groupes et 4 porcs ont servi de contrôles sans CEC. Le premier groupe (n=6) a bénéficié d’un flot pulsatile créé par un BIA en mode interne à 80 battements par minute durant les 90 minutes de l’opération alors que le second groupe (n=6) a subi une CEC standard. Après 60 minutes de reperfusion suivant la CEC, les valeurs hémodynamiques ont été évaluées dont les pressions artérielles, les pressions pulmonaires, l’index cardiaque et la concentration de glucose et de lactate. Les artères pulmonaires sont ensuite montées en chambre d’organe pour évaluer la fonction endothéliale. RÉSULTATS : Les porcs avec pression pulsatile ont tendance à produire moins de lactate sanguin après 60 minutes de reperfusion. Les autres valeurs hémodynamiques sont semblables. Finalement, la relaxation à la bradykinine est significativement meilleure dans le groupe pression pulsatile alors que la relaxation à l’acétylcholine n’est pas significativement différente. CONCLUSION : Ces résultats démontrent que la perfusion pulsatile produite par un BIA protège l’endothélium pulmonaire lors d'une CEC. Cet effet pourrait être dû à une augmentation du flot bronchique qui diminuerait l’ischémie pulmonaire ou à une diminution de la libération de cytokines et de bradykinine qui réduirait les dommages de reperfusion.

L’objectif de cette étude a été d’évaluer l’effet du remplacement total ou partiel du maïs d’une ration alimentaire standard (MS) sur les performances de croissance, le pH ruminal et les paramètres biochimiques sanguins chez les veaux de grain de race Holstein. Quatre groupes de 80 veaux ont été répartisen32 parcs (10 veaux/parc) et ont été assignés au hasard à quatre rations alimentaires. Les rations alimentaires ont été: la ration standard qui est constituée de maïs et un supplément protéique à 43,6% de protéine brute (MS);une ration réduite de maïs, avec tourteau de canola et de drèche de distillerie de maïs avec soluble (MCD); une ration réduite de maïs, avec supplément protéique à 43,6% de protéine brute et de drèche de distillerie de maïs avec soluble (MSD); et finalement une ration d’orge roulé, de tourteau de canola et de drèche de distillerie de maïs avec soluble (OCD). Les rations alimentaires ont été formulées selon une phase de démarrage P1 (j0 à j54), une de croissance P2 (j55 à j85) et une de finition P3 (j86 à j96). Un groupe additionnel de 5 veaux contrôle (CT), a reçu une ration alimentaire non acidogène à base de fourrage et de concentré. Notons qu’avant le début des traitements alimentaires au j0, sauf CT, les veaux ont reçu une ration d’adaptation contenant du maïs et de l’orge (50-50) et un supplément protéique pendant 20j. Les gains moyens quotidiens (GMQ) ont été similaires aux périodes P1 (0j-j27, j28-j54) et P2 (j55-j85), mais à la période P3 (j86-j96), le GMQ de la ration OCD a été plus grand que ceux dans les autres rations (p<0.001). Le rendement carcasse des veaux abattus au poids vifs d’environ 267 kg, de la ration OCD a été plus petit que ceux des rations MS et MSD (p<0.002). La matière sèche ingérée (MSI) a diminué pour le groupe MSD au j96, comparée à celles des groupes MS et BCD (p<0.001). Cependant, les rations alimentaires n’ont pas eu d’effet sur le poids vif des veaux. Les durées moyennes en dessous du pH ruminal de 5.6 (DpH5.6 en h.j.-1) du j68 au j85 (P2) ont été similaires pour les groupes CT et OCD (p=0.09) et plus petites (p<0.001) que celles des groupes du MS, MCD et MSD. Pendant la phase P3, les DpH5.6 des groupes de MS, MCD et MSD, ont été similaires (p>0.83), mais plus grandes que celle du groupe de OCD (p<0.0001). Les DpH5.6 n’ont pas eu d’effet sur les GMQ. Aux j68 et j96, les rations alimentaires n’ont pas eu d’effet sur la L-lactate (p > 0.05), le pH sanguin (p > 0.001; non significatif après l’ajustement de Bonferroni :NSAB) et le trou anionique (p > 0.009; NSAB). La PCO2 des animaux du groupe MS a été plus grande que celle du groupe CT (p = 0.0003). Au j68, HCO3 - du groupe CT a été plus grande que celle du groupe MCD (p = 0.0008). Les traitements alimentaires n’ont pas d’effets sur la lipopolysaccharide binding protein (LBP) aux j0 et j68. Au j96, la LBP du groupe CT a été plus petite que celle du groupe MS et MCD (p=0.001). Les diètes n’ont pas d’effets significatifs sur les épithéliums et les lamina propria du rumen (p0>0.37), ainsi que sur les abcès du foie (p=0.80). Le remplacement total du maïs par l’orge roulé, la drêche de distillerie de maïs avec soluble et le tourteau de canola amélioré le GMQ en phase de finition, a amélioré le pH du rumen, le rapprochant du pH ruminal physiologique, n’a pas modifié les paramètres biochimiques sanguins qui ont été mesurés et a diminué le rendement carcasse moyen de 1,1%.

Ensuite, qui est l’acteur? Parle-t-on d’acteurs ou d’auteurs? Je ne pourrais prétendre donner ici une réponse à ces questions; l’ambition de cet article se limite à en illustrer les enjeux pour approfondir la compréhension de notre monde numérique.

The growth of nine species of Bifidobacterium on media containing glucose, xylose, xylooligosaccharides (XOS), xylan or fructooligosaccharides (FOS) as the sole carbon source were compared in pure culture. The bifidobacteria differed in fermentation profiles when tested on different carbohydrates. All species grew to their highest final optical density (OD) on a glucose containing medium, with the exception of B. catenulatum which demonstrated a preference for xylose over glucose, and XOS over FOS. B. bifidum grew to the highest OD on XOS compared to xylose suggesting a specific transport system for the oligosaccharide over the monomer. This is consistent with a lack of β-xylosidase activity present in the culture medium. Lactate, formate and acetate levels were determined and the ratios of these metabolites altered between and within species growing on different carbohydrates. In general, high lactate production correlated with low formate production and low lactate concentrations were obtained at higher levels of formate. Bifidobacteria may alter their metabolic pathways based upon the carbohydrates that are available for their use.

Here we investigated the effect of lifelong supplementation of the diet with coconut fat (CO, rich in saturated fatty acids) or fish oil (170, rich in n-3 polyunsaturated fatty acids) on tumor growth and lactate production from glucose in Walker 256 tumor cells, peritoneal macrophages, spleen, and gut-associated lymphocytes. Female Wistar rats were supplemented with CO or FO prior to mating and then throughout pregnancy and gestation and then the male offspring were supplemented from weaning until 90 days of age. Then they were inoculated subcutaneously with Walker 256 tumor cells. Tumor weight at 14 days in control rats (those fed standard chow) and CO supplemented was approximately 30 g. Supplementation of the diet with FO significantly reduced tumor growth by 76%. Lactate production (nmol h(-1) mg(-1) protein) from glucose by Walker 256 cells in the group fed regular chow (W) was 381.8 +/- 14.9. Supplementation with coconut fat (WCO) caused a significant reduction in lactate production by 1.6-fold and with fish oil (WFO) by 3.8-fold. Spleen lymphocytes obtained from W and WCO groups had markedly increased lactate production (553 +/- 70 and 635 +/- 150) when compared to non-tumor-bearing rats (similar to 260 +/- 30). FO supplementation reduced significantly the lactate production (297 +/- 50). Gut-associated lymphocytes obtained from W and WCO groups increased lactate production markedly (280 +/- 31 and 276 +/- 25) when compared to non-tumor-bearing rats (similar to 90 +/- 18). FO supplementation reduced significantly the lactate production (168 +/- 14). Lactate production by peritoneal macrophages was increased by tumor burden but there was no difference between the groups fed the various diets. Lifelong consumption of FO protects against tumor growth and modifies glucose metabolism in Walker tumor cells and lymphocytes but not in macrophages. Copyright (C) 2008 John Wiley & Sons, Ltd.

We previously demonstrated an increased liver gluconeogenesis (LG) during insulin-induced hypoglycaemia. Thus, an expected effect of sulphonylureas induced hypoglycaemia (SIH) could be the activation of LG. However, sulphonylureas infused directly in to the liver inhibits LG. Considering these opposite effects we investigated herein LG in rats submitted to SIH. For this purpose, 24 h fasted rats that received glibenclamide (10 mg kg(-1)) were used (SIH group). Control group received oral saline. Glycaemia at 30, 60, 90, 120 and 150 min after oral administration of glibenclamide were evaluated. Since the lowest glycaemia was obtained 120 min after glibenclamide administration, this time was chosen to investigate LG in situ perfused livers. The gluconeogenesis from precursors that enters in this metabolic pathway before the mitochondrial step, i.e. L-alanine (5 mM), L-lactate (2 mM), pyruvate (5 mM) and L-glutamine were decreased (p < 0.05). However, the gluconeogenic activity using glycerol (2 mM), which enters in the gluconeogenesis after the mitochondrial step was maintained. Taken together, the results suggest that the inhibition of LG promoted by SIH overcome the activation of this metabolic pathway promoted by IIH and could be attributed, at least in part, to its effect on mitochondrial function. Contractile activity induces a marked increase in glycolytic activity and gene expression of enzymes and transporters involved in glucose metabolism in skeletal muscle. Muscle contraction also increases the production of reactive oxygen species (ROS). In this study, the effects of treatment with N-acetylcysteine (NAC), a potent antioxidant compound, on contraction-stimulated glycolysis were investigated in electrically stimulated primary rat skeletal muscle cells. The following parameters were measured: 2-[(3)H]deoxyglucose (2-DG) uptake; activities of hexokinase, phosphofructokinase (PFK), and glucose-6-phosphate dehydrogenase (G6PDH); lactate production; and expression of the glucose transporter 4 (GLUT4), hexokinase II (HKII), and PFK genes after one bout of electrical stimulation in primary rat myotubes. NAC treatment decreased ROS signal by 49% in resting muscle cells and abolished the muscle contraction-induced increase in ROS levels. In resting cells, NAC decreased mRNA and protein contents of GLUT4, mRNA content and activity of PFK, and lactate production. NAC treatment suppressed the contraction-mediated increase in 2-DG uptake; lactate production; hexokinase, PFK, and G6PDH activities; and gene expression of GLUT4. HKII, and PFK. Similar to muscle contraction, exogenous H(2)O(2) (500 nM) administration increased 2-DG uptake; lactate production; hexokinase, PFK, and G6PDH activities; and gene expression of GLUT4. HKII, and PFK. These findings support the proposition that ROS endogenously produced play an important role in the changes in glycolytic activity and gene expression of GLUT4, HKII, and PFK induced by contraction in skeletal muscle cells. Increased plasma levels of free fatty acids (FFA) occur in states of insulin resistance such as obesity and type 2 diabetes mellitus. These high levels of plasma FFA are proposed to play an important role for the development of insulin resistance but the mechanisms involved are still unclear. This study investigated the effects of saturated and unsaturated FFA on insulin sensitivity in parallel with mitochondrial function. C2C12 myotubes were treated for 24 h with 0.1 mM of saturated (palmitic and stearic) and unsaturated (oleic, linoleic, eicosapentaenoic, and docosahexaenoic) FFA. After this period, basal and insulin-stimulated glucose metabolism and mitochondrial function were evaluated. Saturated palmitic and stearic acids decreased insulin-induced glycogen synthesis, glucose oxidation, and lactate production. Basal glucose oxidation was also reduced. Palmitic and stearic acids impaired mitochondrial function as demonstrated by decrease of both mitochondrial hyperpolarization and ATP generation. These FFA also decreased Akt activation by insulin. As opposed to saturated FFA, unsaturated FFA did not impair glucose metabolism and mitochondrial function. Primary cultures of rat skeletal muscle cells exhibited similar responses to saturated FFA as compared to C2C12 cells. These results show that in muscle cells saturated FFA-induced mitochondrial dysfunction associated with impaired insulin-induced glucose metabolism. J. Cell. Physiol. 222: 187-194, 2010.
