Two sediment cores of 70 and 252 cm length were recovered from Hjort Sø, a small lake on Store Koldewey, Northeast Greenland, and studied with a multidisciplinary approach in order to reconstruct the local environmental history and to test the relevance of proxies for paleoenvironmental information. The basal sediments from the longer core are dominated by clastic matter, which was likely deposited during deglaciation of the lake basin. These clastic sediments are overlain by gyttja, which is also present throughout the shorter core. AMS radiocarbon dating was conducted on plant macrofossils of 11 samples from the gyttja in both cores. A reliable chronology was established for both cores, which dated the onset of organic accumulation at 9,500 cal. year BP. The Holocene temperature development, with an early to mid Holocene thermal maximum, is best reflected in the grain-size composition. Nutrient availability was apparently low during the early Holocene and led to low productivity in the lake and its vicinity. From ca. 7,000 cal. year BP, productivity in the lake increased significantly, probably induced by external nutrient input from goose excrements. From this time, micro- and macrofossil remains reflect relatively well the climate history of East Greenland, with a cooling during the middle Holocene, the medieval warming, and the Little Ice Age. The amount of organic matter in the sequence seems to be more affected by lake ice cover or by nutrient supply from the catchment than by temperature changes. The record from Hjort Sø thus reveals the difficulties in interpreting sedimentary records from high arctic regions.

Waste biomass is generated during the conservation management of semi-natural habitats, and represents an unused resource and potential bioenergy feedstock that does not compete with food production. Thermogravimetric analysis was used to characterise a representative range of biomass generated during conservation management in Wales. Of the biomass types assessed, those dominated by rush (Juncus effuses) and bracken (Pteridium aquilinum) exhibited the highest and lowest volatile compositions respectively and were selected for bench scale conversion via fast pyrolysis. Each biomass type was ensiled and a sub-sample of silage was washed and pressed. Demineralization of conservation biomass through washing and pressing was associated with higher oil yields following fast pyrolysis. The oil yields were within the published range established for the dedicated energy crops miscanthus and willow. In order to examine the potential a multiple output energy system was developed with gross power production estimates following valorisation of the press fluid, char and oil. If used in multi fuel industrial burners the char and oil alone would displace 3.9 × 105 tonnes per year of No. 2 light oil using Welsh biomass from conservation management. Bioenergy and product development using these feedstocks could simultaneously support biodiversity management and displace fossil fuels, thereby reducing GHG emissions. Gross power generation predictions show good potential.

In the fall of 2005, U.S. Fish and Wildlife Services (USFWS) contracted with Florida International University (FIU) to study the physical and biological drivers underlying the distribution of woody plant species in the marl prairie habitat of the Cape Sable Seaside Sparrow (CSSS). This report presents what we have learned about woody plant encroachment based on studies carried out during the period 2006-2008. The freshwater marl prairie habitat currently occupied by the Cape Sable seaside sparrow (CSSS; Ammodramus maritimus mirabilis) is a dynamic mosaic comprised of species-rich grassland communities and tree islands of various sizes, densities and compositions. Landscape heterogeneity and the scale of vegetative components across the marl prairie is primarily determined by hydrologic conditions, biological factors (e.g. dispersal and growth morphology), and disturbances such as fire. The woody component of the marl prairie landscape is subject to expansion through multiple positive feedback mechanisms, which may be initiated by recent land use change (e.g. drainage). Because sparrows are known to avoid areas where the woody component is too extensive, a better understanding of invasion dynamics is needed to ensure proper management.

Natural CO2 venting systems can mimic conditions that resemble intermediate to high pCO2 levels as predicted for our future oceans. They represent ideal sites to investigate potential long-term effects of ocean acidification on marine life. To test whether microbes are affected by prolonged exposure to pCO2 levels, we examined the composition and diversity of microbial communities in oxic sandy sediments along a natural CO2 gradient. Increasing pCO2 was accompanied by higher bacterial richness and by a strong increase in rare members in both bacterial and archaeal communities. Microbial communities from sites with CO2 concentrations close to today's conditions had different structures than those of sites with elevated CO2 levels. We also observed increasing sequence abundance of several organic matter degrading types of Flavobacteriaceae and Rhodobacteraceae, which paralleled concurrent shifts in benthic cover and enhanced primary productivity. With increasing pCO2, sequences related to bacterial nitrifying organisms such as Nitrosococcus and Nitrospirales decreased, and sequences affiliated to the archaeal ammonia-oxidizing Thaumarchaeota Nitrosopumilus maritimus increased. Our study suggests that microbial community structure and diversity, and likely key ecosystem functions, may be altered in coastal sediments by long-term CO2 exposure to levels predicted for the end of the century.

Hydroxylated glycerol dialkyl glycerol tetraethers (hydroxy-GDGTs) were detected in marine sediments of diverse depositional regimes and ages. Mass spectrometric evidence, complemented by information gleaned from two-dimensional (2D) 1H-13C nuclear magnetic resonance (NMR) spectroscopy on minute quantities of target analyte isolated from marine sediment, allowed us to identify one major compound as a monohydroxy-GDGT with acyclic biphytanyl moieties (OH-GDGT-0). NMR spectroscopic and mass spectrometric data indicate the presence of a tertiary hydroxyl group suggesting the compounds are the tetraether analogues of the widespread hydroxylated archaeol derivatives that have received great attention in geochemical studies of the last two decades. Three other related compounds were assigned as acyclic dihydroxy-GDGT (2OH-GDGT-0) and monohydroxy-GDGT with one (OH-GDGT-1) and two cyclopentane rings (OH-GDGT-2). Based on the identification of hydroxy-GDGT core lipids, a group of previously reported unknown intact polar lipids (IPLs), including the ubiquitously distributed H341-GDGT (Lipp J. S. and Hinrichs K. -U. (2009) Structural diversity and fate of intact polar lipids in marine sediments. Geochim. Cosmochim. Acta 73, 6816-6833), and its analogues were tentatively identified as glycosidic hydroxy-GDGTs. In addition to marine sediments, we also detected hydroxy-GDGTs in a culture of Methanothermococcus thermolithotrophicus. Given the previous finding of the putative polar precursor H341-GDGT in the planktonic marine crenarchaeon Nitrosopumilus maritimus, these compounds are synthesized by representatives of both cren- and euryarchaeota. The ubiquitous distribution and apparent substantial abundance of hydroxy-GDGT core lipids in marine sediments (up to 8% of total isoprenoid core GDGTs) point to their potential as proxies.

O cancro é um problema de saúde crescente no mundo e é a segunda causa de morte depois das doenças cardíacas. De acordo com a Agência Internacional de Investigação em Cancro (IARC) existem atualmente mais de 10 milhões de casos de cancro por ano no mundo. Os produtos naturais oferecem oportunidades de inovação na descoberta de novos fármacos. Neste sentido, os compostos naturais isolados a partir de plantas medicinais, como potenciais fontes de novas drogas anticancerígenas, têm tido um interesse crescente. Os Óleos Essenciais (OEs) são sintetizados pelas plantas e têm sido estudados pelas suas inúmeras atividades biológicas, incluindo anticancerígena, anti-inflamatória, antimicrobiana, antiviral, antioxidante e repelente de insetos. Este estudo tem como objetivos determinar a eficácia de OEs de seis espécies de plantas das dunas de Peniche (Portugal), como potenciais agentes terapêuticos anticancerígenos em linhas celulares de cancro da mama (MCF7) e do colo-rectal (RKO), assim como perceber o mecanismo de ação destes OEs. Neste estudo, partes aéreas de Artemisia campestris subsp. maritima, Crithmum maritimum, Eryngium maritimum, Juniperus turbinata subsp. turbinata, Otanthus maritimus e Seseli tortuosum foram colhidas na praia da Consolação, em Peniche (Portugal), e os seus OEs isolados através de hidrodestilação. A composição química dos OEs foi investigada por cromatografia gasosa (GC) e por cromatografia gasosa com espetrofotometria de massa (GC-MS) e os compostos maioritários foram descritos para cada óleo. Para avaliar a atividade anticancerígena nas linhas celulares MCF7 e RKO, o método MTS (3- (4, 5-dimethyl- 2 -thiazolyl) - 2, 5-dyphenyl-2H-tetrazolium bromide) foi usado e a viabilidade celular avaliada, através de diluições sucessivas, a concentrações iniciais de 5 μL/mL e 1 μL/mL, com diluição de 1:2 e 1:10, respetivamente, comparando com o controlo (DMSO). De todos os OEs testados, a atividade anticancerígena foi descrita, em ambas as linhas celulares, como observado pela diminuição da viabilidade/proliferação celular – exceto o OE Eryngium maritimum a uma concentração inicial de 5 μL/mL.Com o objetivo de avaliar o mecanismo biológico de ação dos OEs, foi realizado um western blot para marcadores relativos ao bloqueio do ciclo celular e apoptose (p53, p21 e caspase 3 clivada), para Seseli tortuosum e Otanthus maritimus. Foi observado um aumento do nível proteína p53 nas células tratadas com estes OEs, sugerindo a indução de stress celular nas células cancerígenas testadas. No entanto, não foi observada caspase 3 clivada, sugerindo que a apoptose não terá sido a causa para a diminuição da viabilidade/proliferação celular observada. Foi ainda observado o aumento da expressão da p21 com os OEs selecionados, sugerindo que o tratamento com OE está associado ao bloqueio do ciclo celular. Para validar estas observações, a análise realizada por FACS, depois do tratamento indica um possível bloqueio do ciclo celular na fase G1. Concluindo, a concentração inicial de 5 μL/mL revelou ser muito tóxica para as linhas celulares testadas. No entanto, a uma concentração final de 1 μL/mL foi demonstrada uma diminuição da viabilidade/proliferação celular para todos os OEs. No estudo preliminar do mecanismo de ação dos OEs, foi demonstrado, face à presença da p21, que os óleos de Seseli tortuosum e Otanthus maritimus atuam bloqueando o ciclo celular. Para comprovar estes resultados, o FACS realizado (apenas no OE de Seseli tortuosum) revelou que este bloqueio pode ocorrer, pelo aumento da percentagem de células observadas, na fase G1. Estes resultados demonstram o interesse destes OEs de Peniche na procura de novos agentes quimo preventivos contra a progressão do cancro da mama e colo-rectal.

Limno-glacial deposits 110 cm in thickness in the upper alpine belt have been investigated. It is established that during the Yuanakhchiri period of glaciation in the region under study a glacial cirque was situated at an altitude of 2750 m a.s.l. After the glacier retreated, a lake was formed in its bed which existed till the second noticeable cooling in the second half of SA2 (1600-1500 years ago).

To better understand Holocene vegetation and hydrological changes in South Africa, we analyzed pollen and microcharcoal records of two marine sites GeoB8331 and GeoB8323 from the Namaqualand mudbelt offshore the west coast of South Africa covering the last 9900 and 2200 years, respectively. Our data corroborate findings from literature that climate developments apparently contrast between the summer rainfall zone (SRZ) and winter rainfall zone (WRZ) over the last 9900 years, especially during the early and middle Holocene. During the early Holocene (9900-7800 cal.yr BP), a minimum of grass pollen suggests low summer rainfall in the SRZ, and the initial presence of Renosterveld vegetation indicates relatively wet conditions in the WRZ. Towards the middle Holocene (7800-2400 cal. yr BP), a rather moist savanna/grassland rich in grasses suggests higher summer rainfall in the SRZ resulting from increased austral summer insolation and a decline of fynbos vegetation accompanied by an increasing Succulent Karoo vegetation in the WRZ possibly suggests a southward shift of the Southern Hemisphere westerlies. During the last 2200 years, a trend towards higher aridity was observed for the SRZ, while the climate in the WRZ remained relatively stable. The Little Ice Age (ca. 700-200 cal. yr BP) was rather cool in both rainfall zones and drier in the SRZ while wetter in the WRZ.

Le climat planétaire évolue à une vitesse alarmante. Selon le Groupe d’experts intergouvernemental sur l’évolution du climat, les températures globales pourraient grimper de 3 à 7°C d’ici 2100. Les zones tempérées, comme le Québec, ne seront pas épargnées. Au sein de la province, le climat typique en forêt boréale pourrait se déplacer de 750 km vers le nord pour atteindre le Haut-Arctique dans quelques décennies. D’autres modifications notables du climat se traduisent dans les précipitations, les glaces, les océans, etc. Tous ces changements climatiques constituent une menace grandissante pour la biodiversité. D’ailleurs, ils pourraient devenir la principale cause d’extinction d’espèces dans le futur, surpassant même la dégradation d’habitats et la pollution. Afin de témoigner de la portée des changements climatiques sur la biodiversité, 6 espèces fauniques à statut précaire au Québec sont étudiées dans le cadre de l’essai. Par leurs fortes réponses aux stress, elles agissent à titre de bioindicateurs des changements climatiques. Ces espèces, de classes taxonomiques différentes, sont : le chevalier cuivré (Moxostoma hubbsi), l’ours blanc (Ursus maritimus), la grive de Bicknell (Catharus Bicknelli), la rainette faux-grillon de l’Ouest (Pseudacris triseriata), la tortue luth (Dermochelys coriacea) et le satyre fauve des Maritimes (Coenonympha nipisiquit). L’étude approfondie de ces espèces a permis de relever un total de 47 impacts potentiels des changements climatiques. En majorité, ces impacts concernent la perte d’habitats, l’augmentation de la compétition, la diminution du succès reproducteur et l’entrave à la survie. L’objectif de cet essai est d’évaluer les mesures de protection actuelles permettant de faire face à ces impacts. Bien qu’elles varient grandement d’une espèce à l’autre, elles peuvent être regroupées en quatre grandes thématiques, soit la recherche et le suivi, la gestion des aires protégées, l’adaptation du cadre législatif et l’aménagement écosystémique. L’évaluation de ces mesures a permis de soulever certains constats. En premier lieu, la recherche et le suivi constituent la principale mesure de protection adoptée. En second lieu, il appert que les mesures de protection actuelles manquent de spécificité. En effet, dans la majorité des cas, elles protègent l’espèce de manière indirecte seulement. Au final, dans le but d’améliorer ou de combler les lacunes des mesures de protection actuelles, des recommandations ont été formulées. D’une part, celles-ci présentent des actions concrètes comme la translocation ou l’aménagement écosystémique. D’autre part, elles suggèrent des modifications quant aux méthodes de suivi, à la gestion des aires protégées et aux cadres législatifs.

Neste trabalho estudou-se a eficiência dos herbicidas Herbipec 500 FL (s.a. Clortolurão) e Dopler Super (s.a. Diclofope-Metilo+Fenoxaprope-P-Etilo+Mefenepir-Dietilo) no controlo, em pós-emergência de infestantes Monocotiledóneas, e na produção de grão e suas componentes, na cultura do trigo mole em sementeira directa, combinando doses inferiores às recomendadas pelos fabricantes. Os ensaios decorreram nos anos agrícolas de 2006/2007 e 2007/2008, na Herdade do Louseiro no concelho de Évora e na Herdade da Revilheira no concelho de Reguengos de Monsaraz, respectivamente. Na experimentação efectuou-se o estudo dos dois herbicidas, com 3 níveis cada, correspondentes a nove tratamentos. O delineamento experimental foi em blocos casualizados com quatro repetições cada. Verificou-se uma maior eficiência no controlo das plantas infestantes de Lolium rigidum Gaud. e de Juncus bufonius L. e, consequentemente, um maior número de grãos e uma produção de grão de trigo elevada com 2 litros ha-l de Herbipec 500 FL e 0,5 litro ha-1 de Dopler Super. ABSTRACT: The purpose of this work was to study the efficiency of the herbicides Herbipec 500 FL (a.i. chlorotoluron) and Dopler Super (a.i. diclofop-methyl + fenoxaprop-P-ethyl + mefenpyr-diethyl) to control grass weeds at post-emergence in no-till bread wheat and consequently to do the evaluation of potential grain yield combining reduced doses to the recommended ones by the manufacturers. The trials were carried out over two growing seasons (2006/2007 and 2007/2008) on the farm "Revilheira" and on a private farm "Louseiro", both in the district of Évora. Trials to study effects of three doses of a two herbicides, with three levels each, corresponding to nine treatments were executed. The experimental design was a randomized block with four replications each. The results showed a great efficiency and grain yield wheat with the mixture with 2 l ha-1 Herbipec 500 FL and 0,5 1 ha-1 Dopler Super to controlling Lolium spp. and Juncus bufonius L..
