CHAPTER II - This study evaluated the effects of two different types of acute aerobic exercise on the osmotic stability of human erythrocyte membrane and on different hematological and biochemical variables that are associated with this membrane property. The study population consisted of 20 healthy and active men. Participants performed single sessions of two types of exercise. The first session consisted of 60 min of moderate-intensity continuous exercise (MICE). The second session, executed a week later, consisted of high-intensity interval exercise (HIIE) until exhaustion. The osmotic stability of the erythrocyte membrane was represented by the inverse of the salt concentration (1/H50) at the midpoint of the sigmoidal curve of dependence between the absorbance of hemoglobin and the NaCl concentration. The values of 1/H50 changed from 2.29 ± 0.1 to 2.33 ± 0.09 after MICE and from 2.30 ± 0.08 to 2.23 ± 0.12 after HIIE. In MICE has occurred an increase in the mean corpuscular volume, probably due to in vivo lysis of older erythrocytes, with preservation of cells that were larger and more resistant to in vitro lysis. The study showed that a single bout of acute exercise affected the erythrocyte osmotic stability, which increased after MICE and decreased after HIIE.

Studies of authigenic phosphorus (P) minerals in marine sediments typically focus on authigenic carbonate fluorapatite, which is considered to be the major sink for P in marine sediments and can easily be semi-quantitatively extracted with the SEDEX sequential extraction method. The role of other potentially important authigenic P phases, such as the reduced iron (Fe) phosphate mineral vivianite (Fe(II)3(PO4)*8H2O) has so far largely been ignored in marine systems. This is, in part, likely due to the fact that the SEDEX method does not distinguish between vivianite and P associated with Fe-oxides. Here, we show that vivianite can be quantified in marine sediments by combining the SEDEX method with microscopic and spectroscopic techniques such as micro X-ray fluorescence (µXRF) elemental mapping of resin-embedded sediments, as well as scanning electron microscope-energy dispersive spectroscopy (SEM-EDS) and powder X-ray diffraction (XRD). We further demonstrate that resin embedding of vertically intact sediment sub-cores enables the use of synchrotron-based microanalysis (X-ray absorption near-edge structure (XANES) spectroscopy) to differentiate between different P burial phases in aquatic sediments. Our results reveal that vivianite represents a major burial sink for P below a shallow sulfate/methane transition zone in Bothnian Sea sediments, accounting for 40-50% of total P burial. We further show that anaerobic oxidation of methane (AOM) drives a sink-switching from Fe-oxide bound P to vivianite by driving the release of both phosphate (AOM with sulfate and Fe-oxides) and ferrous Fe (AOM with Fe-oxides) to the pore water allowing supersaturation with respect to vivianite to be reached. The vivianite in the sediment contains significant amounts of manganese (~4-8 wt.%), similar to vivianite obtained from freshwater sediments. Our results indicate that methane dynamics play a key role in providing conditions that allow for vivianite authigenesis in coastal surface sediments. We suggest that vivianite may act as an important burial sink for P in brackish coastal environments worldwide.

Nutrient addition experiments were performed during the austral summer in the Amundsen Sea (Southern Ocean) to investigate the availability of organically bound iron (Fe) to the phytoplankton communities, as well as assess their response to Fe amendment. Changes in autotrophic biomass, pigment concentration, maximum photochemical efficiency of photosystem II, and nutrient concentration were recorded in response to the addition of dissolved free Fe (DFe) and Fe bound to different model ligands. Analysis of pigment concentrations indicated that the autotrophic community was dominated by the prymnesiophyte Phaeocystis antarctica throughout most of the Amundsen Sea, although diatoms dominated in two experiments conducted in the marginal ice zone. Few significant differences in bulk community biomass (particulate organic carbon, nitrogen, and chlorophyll a) were observed, relative to the controls, in treatments with Fe added alone or bound to the ligand phytic acid. In contrast, when Fe was bound to the ligand desferrioxamine B (DFB), decreases in the bulk biomass indices were observed. The concentration of the diatom accessory pigment fucoxanthin showed little response to Fe additions, while the concentration of the P. antarctica-specific pigment, 19'-hexanoyloxyfucoxanthin (19'-hex), decreased when Fe was added alone or bound to the model ligands. Lastly, differences in the nitrate:phosphate (NO3- :PO4**3-) utilization ratio were observed between the Fe-amended treatments, with Fe bound to DFB resulting in the lowest NO3- :PO4**3- uptake ratios (~ 10) and the remaining Fe treatments having higher NO3- :PO4**3- uptake ratios (~ 17). The data are discussed with respect to glacial inputs of Fe in the Amundsen Sea and the bioavailability of Fe. We suggest that the previously observed high NO3- :PO4**3- utilization ratio of P. antarctica is a consequence of its production of dissolved organic matter that acts as ligands and increases the bioavailability of Fe, thereby stimulating the uptake of NO3-.

La croissance du phytoplancton est limitée par les faibles concentrations de fer (Fe) dans près de 40% de l’océan mondial. Le Pacifique subarctique Nord-Est représente une de ces zones limitées en fer et désignées High Nutrient - Low Chlorophyll (HNLC). Cet écosystème, dominé par des cellules de petite taille telles les prymnésiophytes, est caractérisé par de très faibles concentrations estivales de chlorophylle a et de fortes concentrations de macronutriments. Il a été maintes fois démontré que les ajouts de fer, sous différentes formes chimiques (habituellement FeSO4), dans les zones HNLC, stimulent la croissance et modifient la structure des communautés planctoniques en favorisant la croissance des cellules de grande taille, notamment les diatomées. Ces effets sur la communauté planctonique ont le potentiel d’influencer les grands mécanismes régulateurs du climat, tels la pompe biologique de carbone et la production de diméthylsulfure (DMS). Les poussières provenant des déserts du nord de la Chine sont reconnues depuis longtemps comme une source sporadique importante de fer pour le Pacifique Nord-Est. Malgré leur importance potentielle, l’influence directe exercée par ces poussières sur l’écosystème planctonique de cette zone HNLC n’a jamais été étudiée. Il s’agit d’une lacune importante puisque le fer associé aux poussières est peu soluble dans l’eau de mer, que la proportion biodisponible n’est pas connue et que les poussières peuvent avoir un effet inhibiteur chez le phytoplancton. Cette thèse propose donc, dans un premier temps, de mesurer pour la première fois l’effet de la fertilisation de la communauté planctonique du Pacifique Nord-Est par un gradient de concentrations de poussières désertiques naturelles. Cette première expérimentation a démontré que le fer contenu dans les poussières asiatiques est biodisponible et qu’une déposition équivalente à celles prenant place au printemps dans le Pacifique Nord-Est peut résulter en une stimulation significative de la prise de nutriments et de la croissance du phytoplancton. Mes travaux ont également montré que l’ajout de 0,5 mg L-1 de poussières peut résulter en la production d’autant de biomasse algale que l’ajout de FeSO4, l’espèce chimique utilisée lors des expériences d’enrichissement en fer à grande échelle. Cependant, les ajouts de FeSO4 favorisent davantage les cellules de petite taille que les ajouts de poussières, observation démontrant que le FeSO4 n’est pas un proxy parfait des poussières asiatiques. Dans un deuxième temps, je me suis intéressée à une source alternative de fer atmosphérique, les cendres volcaniques. Mon intérêt pour cette source de fer a été attisé par les observations d’une floraison spectaculaire dans le Pacifique Nord-Est, ma région d’étude, associée à l’éruption de 2008 du volcan Kasatochi dans les îles Aléoutiennes. Forte de mon expérience sur les poussières, j’ai quantifié l’effet direct de ces cendres volcaniques sur la communauté planctonique du Pacifique Nord-Est. Mes résultats ont montré que le fer contenu dans les cendres volcaniques est également biodisponible pour le phytoplancton. Ils ont également montré que cette source de fer peut être aussi importante que les poussières désertiques dans la régulation de la croissance du phytoplancton dans cette partie de l’océan global à l’échelle millénaire. Dans un troisième temps, j’ai estimé comment l’acidification des océans modulera les réponses des communautés planctoniques aux dépositions naturelles de fer mises en évidence lors de mes expériences précédentes. Pour ce faire, j’ai effectué des enrichissements de poussière dans de l’eau de mer au pH actuel de 8.0 et dans l’eau de mer acidifiée à un pH de 7.8. Mes résultats ont montré une diminution du taux de croissance du phytoplancton dans le milieu acidifié mais pas de changement notable dans la structure de la communauté. Les ajouts de poussières et de cendres, de même que les variations de pH, n’ont pas eu d’effet significatif sur la production de DMS et de son précurseur le diméthylsulfoniopropionate (DMSP), probablement en raison de la courte durée (4 jours) des expériences. L’ensemble des résultats de cette thèse montre que le fer contenu dans diverses sources atmosphériques naturelles est biodisponible pour le phytoplancton du Pacifique Nord-Est et que des taux de déposition réalistes peuvent stimuler la croissance de manière notable dans les premiers jours suivant une tempête désertique ou une éruption volcanique. Finalement, les résultats de mes expériences à stresseurs multiples Fer/acidification suggèrent une certaine résistance des communautés phytoplanctoniques à la diminution du pH prédite d’ici la fin du siècle pour les eaux de surface des océans.

Our aim was the evaluation of serum TE in patients who underwent PEG and its relationship with serum proteins, BMI and nature of underlying disorder.

Recentemente tem-se assistido a um acumular de evidência sugerindo a implicação de uma desregulação do metabolismo do ferro (Fe) na fisiopatologia da doença de Alzheimer (DA). Neste trabalho, pretendemos esclarecer melhor os mecanismos moleculares subjacentes à homeostasia deste metal na DA, particularmente ao nível do efluxo celular. Assim, mediu-se em células mononucleares do sangue periférico de 73 doentes com DA e 74 controlos a expressão de genes diretamente envolvidos na regulação e exportação celular de Fe, utilizando a técnica de PCR quantitativo. Os resultados mostraram uma diminuição significativa na expressão dos genes aconitase (ACO1; P=0,007); ceruloplasmina (CP; P<0,001) e proteína precursora de beta amilóide (APP; P=0,006) em doentes com DA comparativamente com os voluntários saudáveis. Estas observações apontam para uma diminuição significativa da expressão dos genes associados com a exportação de Fe celular mediada pela ferroportina na DA. Assim, o presente estudo reforça resultados anteriores que mostram alterações no metabolismo do Fe e podem estar na origem da retenção intracelular deste metal e aumento de stress oxidativo caraterísticos desta patologia.

The Cluster Variation Method (CVM), introduced over 50 years ago by Prof. Dr. Ryoichi Kikuchi, is applied to the thermodynamic modeling of the BCC Cr-Fe system in the irregular tetrahedron approximation, using experimental thermochemical data as initial input for accessing the model parameters. The results are checked against independent data on the low-temperature miscibility gap, using increasingly accurate thermodynamic models, first by the inclusion of the magnetic degrees of freedom of iron and then also by the inclusion of the magnetic degrees of freedom of chromium. It is shown that a reasonably accurate description of the phase diagram at the iron-rich side (i.e. the miscibility gap borders and the Curie line) is obtained, but only at expense of the agreement with the above mentioned thermochemical data. Reasons for these inconsistencies are discussed, especially with regard to the need of introducing vibrational degrees of freedom in the CVM model. (C) 2008 Elsevier Ltd. All rights reserved.

The reported experimental work on the systems Fe-Zn-O and Fe-Zn-Si-O in equilibrium with metallic iron is part of a wider research program that combines experimental and thermodynamic computer modeling techniques to characterize zinc/lead industrial slags and sinters in the system PbO-ZnO-SiO2-CaO-FeO-Fe2O3. Extensive experimental,investigations using high-temperature equilibration and quenching techniques followed by electron probe X-ray microanalysis (EPMA) were carried out. Special experimental; procedures were developed to enable accurate measurements in these ZnO-containing systems to be performed in equilibrium with metallic iron; The systems Fe-Zn-O and FeZn-Si-O were experimentally investigated in equilibrium with metallic iron in the temperature ranges 900 degreesC to 1200 degreesC (1173 to 1473 K) and from 1000 degreesC to 1350 degreesC (1273 to 1623 K), respectively. The liquidus surface in the system Fe-Zn-Si-O in equilibrium with metallic iron was characterized in the composition ranges 0 to 33 wt pet ZnO and 0 to 40 wt pet SiO2. The wustite (Fe,Zn)O, zincite (Zn,Fe)O, willemite (Zn,Fe)(2)SiO4, arid fayalite: (Fe,Zn)(2)SiO4 solid solutions in equilibrium with metallic iron were measured.

The iron(II) complex [Fe(AMN(3)S(3)sarH)](ClO4)(3).3H(2)O (AMN(3)S(3)sarH = 8-ammonio-1-methyl-3,13,16-trithia-6,10,19-triazabicyclo[6.6.6]icosane) has been synthesized and characterized by single crystal structure and spectroscopic methods. The Fe(II)-S(thiaether) bond lengths are short, indicative of a large degree of metal-ligand orbital mixing (pi-acceptor character) of the thiaether ligand. FeBr2 reacts with the S2C2(CN)22- ion (1:1 ratio) in the presence of an excess of t-BuNC in THF to give the mixed ligand [Fe(S2C2(CN)2)(t-BuNC) 4] compound. This neutral product with a formal oxidation state of two for the iron atom was characterized by conductivity measurements, and, i.r., Mössbauer, 13C and 1H n.m.r. spectroscopy. There is a Fe-C p back-donation strengthened towards isocyanide ligands, according to the data of 13C, 1H n.m.r. and Mössbauer spectroscopy.

Two new iron thioantimonates, [Fe(en)(3)](2)Sb2S5 (.) 0.55H(2)O (1) and [Fe(en)(3)](2)Sb4S8 (2). were synthesised under solvothermal conditions from the reactions of Sb2S3, FeCl2 and S in the presence of ethylenediamine at 413 and 438 K, respectively. The products were characterised by single-crystal X-ray diffraction, elemental analysis and SQUID magnetometry. Compound 1 is unusual in containing isolated Sb2S54- anions formed from two corner-sharing SbS33- trigonal pyramids. These units are arranged in rippled layers, 4 A apart, parallel to the bc-plane. Octahedrally coordinated [Fe(en)(3)](2+) cations lie in depressions within these anionic layers. In compound (2), repeated corner linking of SbS33- trigonal pyramids generates SbS2- chains, which may be considered as a polymerised form of the Sb2S54- anions in 1. The SbS2- chains are separated by [Fe(en)(3)](2+) cations. In both compounds, there is an extensive network of hydrogen bonds between the nitrogen atoms of the ethylenediamine ligands and the sulfur atoms of the anions and, in the case of 1, the uncoordinated water molecule. 1,1′-Diacetylferrocene reacts with neat hydrate over a period of 72 h at 20°C to give the dihydrazone [H2NN(Me)CC5H4FeC5H4C(Me)NNH2] (6) in almost quantitative yield. Either prolonging the reaction time or reacting 6 with fresh hydrazine causes the iron to be stripped from the metallocene and bis(hydrazine)bis(hydrazinecarboxylato-N′,O) iron(II), [Fe(N2H4)2(OOCNHNH2)2] (11), crystallizes. In the presence of Ba2+ or Mo2+ ions two molecules of complex 6 react to give the cyclic diazine [N(Me)CC5H4FeC5H4C (Me)N]2 (7) in high yield. Hydrazine is liberated in this reaction. Complexes 6 and 11 have been characterized crystallographically. The cyclic voltammograms of complexes 6 and 7 contain essentially non-reversible oxidation peaks.
