Using a short-term bulk culture protocol designed for an intracellular-staining method based on a flow cytometry approach to the frequencies of cytokine-producing cells from tuberculosis and leprosy patients, we found distinct patterns of T cell subset expression. The method also reveals the profile of peak cytokine production and can provide simultaneous information about the phenotype of cytokine-producing cells, providing a reliable assay for monitoring the immunity of these patients. The immune response of Mycobacterium leprae and purified protein derivative (PPD) in vitro to a panel of mycobacteria-infected patients from an endemic area was assessed in primary mononuclear cell cultures. The kinetics and source of the cytokine pattern were measured at the single-cell level. IFN-gamma-, TNF-alpha-, IL-4- and IL-10-secreting T cells were intracytoplasmic evaluated in an attempt to identify M. leprae- and PPD-specific cells directly from the peripheral blood. The analysis by this approach indicated that TNF-alpha was the first (8 h) to be produced, followed by IFN-gamma (16 h), IL-10 (20 h) and IL-4 (24 h), and double-staining experiments confirmed that CD4+ were a greater source of TNF-alpha than of CD8+ T cells (P < 0.05). Both T cell subsets secreted similar amounts of IFN-gamma. We conclude that the protocol permits rapid evaluation of cytokine production by different T cell populations. The method can also be used to define immune status in non-infected and contact individuals.

Evidence based on immunological cross-reactivity and anti-diabetic properties has suggested the presence of insulin-like peptides in plants. The objective of the present study was to investigate the presence of insulin-like proteins in the leaves of Bauhinia variegata ("pata-de-vaca", "mororó"), a plant widely utilized in popular medicine as an anti-diabetic agent. We show that an insulin-like protein was present in the leaves of this plant. A chloroplast protein with a molecular mass similar to that of bovine insulin was extracted from 2-mm thick 15% SDS-PAGE gels and fractionated with a 2 x 24 cm Sephadex G-50 column. The activity of this insulin-like protein (0.48 mg/mL) on serum glucose levels of four-week-old Swiss albino (CF1) diabetic mice was similar to that of commercial swine insulin used as control. Further characterization of this molecule by reverse-phase hydrophobic HPLC chromatographic analysis as well as its antidiabetic activity on alloxan-induced mice showed that it has insulin-like properties. Immunolocalization of the insulin-like protein in the leaves of B. variegata was performed by transmission electron microscopy using a polyclonal anti-insulin human antibody. Localization in the leaf blades revealed that the insulin-like protein is present mainly in chloroplasts where it is also found associated with crystals which may be calcium oxalate. The presence of an insulin-like protein in chloroplasts may indicate its involvement in carbohydrate metabolism. This finding has strengthened our previous results and suggests that insulin-signaling pathways have been conserved through evolution.

Nitric oxide (NO) is a molecular messenger involved in several events of synaptic plasticity in the central nervous system. Ca2+ influx through the N-methyl-D-aspartate receptor (NMDAR) triggers the synthesis of NO by activating the enzyme neuronal nitric oxide synthase (nNOS) in postsynaptic densities. Therefore, NMDAR and nNOS are part of the intricate scenario of postsynaptic densities. In the present study, we hypothesized that the intracellular distribution of nNOS in the neurons of superior colliculus (SC) superficial layers is an NMDAR activity-dependent process. We used osmotic minipumps to promote chronic blockade of the receptors with the pharmacological agent MK-801 in the SC of 7 adult rats. The effective blockade of NMDAR was assessed by changes in the protein level of the immediate early gene NGFI-A, which is a well-known NMDAR activity-dependent expressing transcription factor. Upon chronic infusion of MK-801, a decrease of 47% in the number of cells expressing NGFI-A was observed in the SC of treated animals. Additionally, the filled dendritic extent by the histochemical product of nicotinamide adenine di-nucleotide phosphate diaphorase was reduced by 45% when compared to the contralateral SC of the same animals and by 64% when compared to the SC of control animals. We conclude that the proper intracellular localization of nNOS in the retinorecipient layers of SC depends on NMDAR activation. These results are consistent with the view that the participation of NO in the physiological and plastic events of the central nervous system might be closely related to an NMDAR activity-dependent function.

Toxoplasma, which infects all eukaryotic cells, is considered to be a good system for the study of drug action and of the behavior of infected host cells. In the present study, we asked if thiosemicarbazone derivatives can be effective against tachyzoites and which morphological and ultrastructural features of host cells and parasites are associated with the destruction of Toxoplasma. The compounds were tested in infected Vero cell culture using concentration screens (0.1 to 20 mM). The final concentration of 1 mM was chosen for biological assay. The following results were obtained: 1) These new derivatives decreased T. gondii infection with an in vitro parasite IC50% of 0.2-0.7 mM, without a significant effect on host cells and the more efficient compounds were 2, 3 (thiosemicarbazone derivatives) and 4 (thiazolidinone derivative); 2) The main feature observed during parasite elimination was continuous morphological disorganization of the tachyzoite secretory system, progressive organelle vesiculation, and then complete disruption; 3) Ultrastructural assays also revealed that progressive vesiculation in the cytoplasm of treated parasites did not occur in the host cell; 4) Vesiculation inside the parasite resulted in death, but this feature occurred asynchronously in different intracellular tachyzoites; 5) The death and elimination of T. gondii was associated with features such as apoptosis-like stage, acidification and digestion of parasites into parasitophorous vacuoles. Our results suggest that these new chemical compounds are promising for the elimination of intracellular parasites by mainly affecting tachyzoite development at 1 mM concentration for 24 h of treatment.

The transient receptor potential channels family (TRP channels) is a relatively new group of cation channels that modulate a large range of physiological mechanisms. In the nervous system, the functions of TRP channels have been associated with thermosensation, pain transduction, neurotransmitter release, and redox signaling, among others. However, they have also been extensively correlated with the pathogenesis of several innate and acquired diseases. On the other hand, the omega-3 polyunsaturated fatty acids (n-3 fatty acids) have also been associated with several processes that seem to counterbalance or to contribute to the function of several TRPs. In this short review, we discuss some of the remarkable new findings in this field. We also review the possible roles played by n-3 fatty acids in cell signaling that can both control or be controlled by TRP channels in neurodegenerative processes, as well as both the direct and indirect actions of n-3 fatty acids on TRP channels.

Resveratrol (Resv) is natural polyphenol found in grapes. This study evaluated the protective effect of Resv against the effects of uric acid (UA) in immortalized human mesangial cells (ihMCs). ihMCs were preincubated with Resv (12.5 µM) for 1 h and treated with UA (10 mg/dL) for 6 or 12 h. The intracellular calcium concentration [Ca2+]i was quantified by fluorescence using flow cytometry. Angiotensinogen (AGT) and pre-pro endothelin-1 (ppET-1) mRNA were assayed by quantitative real-time RT-PCR. Angiotensin II (AII) and endothelin-1 (ET-1) were assayed by ELISA. UA significantly increased [Ca2+]i. Pre-incubation with Resv significantly reduced the change in [Ca2+]i induced by UA. Incubation with UA for 6 or 12 h also increased AGT mRNA expression and AII protein synthesis. Resv blunted these increases in AGT mRNA expression and AII protein. Incubation with UA in the ihMCs increased ppET-1 expression and ET-1 protein synthesis at 6 and 12 h. When ihMCs were pre-incubated with Resv, UA had a significantly diminished effect on ppET-1 mRNA expression and ET-1 protein synthesis at 6 and 12 h, respectively. Our results suggested that UA triggers reactions including AII and ET-1 production in mesangial cells. The renin-angiotensin system may contribute to the pathogenesis of renal function and chronic kidney disease. Resv can minimize the impact of UA on AII, ET-1 and the increase of [Ca2+]i in mesangial cells, suggesting that, at least in part, Resv can prevent the effects of soluble UA in mesangial cells.

GABA (4-aminobutyrate) is synthesized through the decarboxylation of LGlu- (L-Glu-+ H+ ---> GABA + C02), and compared to many free amino acids is present in high concentrations in plant cells. GABA levels rise rapidly and dramatically in response to varied stress conditions including anaerobiosis. Recent papers suggest that GABA production and associated H+ consumption are parts of a metabolic pH-stat mechanism which ameliorates the intracellular pH decline associated with anaerobiosis or other treatments. To test this hypothesis GABA production and efflux have been measured in isolated Asparagus sprengeri cells in response to three treatments which potentially cause intracellular acidification. Acid loads were imposed using 60 min of (i) anaerobiosis, (ii) H+/LGlu- cotransport, and (iii) treatment with permeant weak acids (butyric, acetic and propionic). Both intra- and extracellular GABA concentrations increased more than 100% after anaerobiosis, almost 1000% after H+/L-Glu- cotransport (light or dark) and almost 5000/0 after addition of 5 mM butyric acid at pH 5.0. HPLC analysis of amino acids indicates that as GABA concentrations increased in response to butyric acid addition, glutamate concentrations decreased. Time-course studies demonstrated that added butyric acid stimulates GABA production by 2800/0 within 15 seconds. A fluorescent determination of cytosolic pH indicates that addition of butyric or other weak acids resulted in a rapid reduction in cytosolic pH of 0.6 pH units. The half time for the response to butyric acid addition is 2.1 seconds, indicating that the decline in cytosolic pH is rapid enough to account for the rapid stimulation of GABA production. The acid load in response to butyric acid addition was assayed by measurements of 14C-butyric acid uptake. Calculations indicate that GABA production accounted for 45% of the imposed acid load. The biological significance of GABA efflux is not yet understood. The results support the original hypothesis suggesting a role for GABA production in cellular pH regulation.

Since its discovery nearly a century ago, a-tocopherol (vitamin E) research has been mainly focused on its ability to terminate the cycle of lipid peroxidation in membranes. Nitrobenzoxadiazole fluorescent analogues were made previously to study the intracellular transfer of vitamin E in cells. However, these molecules were reportedly susceptible to photobleaching while under illumination for transfer assays and microscopy. Here is reported the synthesis of a series of fluorescent analogues of vitamin E incorporating the more robust dipyrrometheneboron difluoride fluorophore (BDP-a-Tocs; Aex = 507 nm, Aem = 511 nm). C8-BDP-a-Toc 42c, having an eight-carbon chain between the chromanol and fluorophore, wa<; shown to bind specifically to a-tocopherol transfer protein with a dissociation constant of approximately 100 nM. Another fluorescent analogue of vitamin E with a thienyl derivative of BODIPY that is excited and fluoresces at longer wavelengths (Aex = 561 nm, Aem = 570 nm) is in development.

In animals, both stress resistance and longevity appear to be influenced by the insulin/insulin-like growth factor-l signaling (lIS) pathway, the basic organization of which is highly conserved from invertebrates to vertebrates. Reduced lIS or genetic disruption of the lIS pathway leads to the activation of forkhead box transcription factors, which is thought to upregulate the expression of genes involved in enhancing stress resistance, including perhaps key antioxidant enzymes as well as DNA repair enzymes. Enhanced antioxidant and DNA repair capacities may underlie the enhanced cellular stress resistance observed in long-lived animals, however little data is available that directly supports this idea. I used three. experimental approaches to test the association of intracellular antioxidant and DNA base excision repair (BER) capacities with stress resistance and longevity: (1) a comparison of multiple vertebrate endotherm species of varying body masses and longevities; (2) a comparison of long-lived Snell dwarf mice and their normallittermates; and (3) a comparison of hypometabolic animals undergoing hibernation or estivation with their active counterparts. The activities of the five major intracellular antioxidant enzymes as well as the two rate-limiting enzymes in the BER pathway, apurininc/apyrimidinic (AP) endonuclease and polymerase ~, were measured. These measurements were performed in one or more of the following: (1) cultured dermal fibroblasts; (2) brain tissue; (3) heart tissue; (4) liver tissue. My results indicate that antioxidant enzymes are not universally upregulated in association with enhanced stress resistance and longevity. I also did not find that BER enzyme activity was positively correlated with longevity, in an inter-species context, though there was evidence for enhanced BER in long-lived Snell dwarf mice. Thus, while there were instances in which enhanced antioxidant and BER enzyme activities were associated with increased stress resistance and/or longevity, this was not universally the case, indicating that other mechanisms must be involved. These results suggest the need to re-examine existing 'oxidative stress' hypotheses of longevity and probe further into the molecular physiology of longevity to discover its mechanistic basis.

Le dogme voulant que les récepteurs couplés aux protéines G (GPCRs) activent des voies de signalisation seulement lorsqu’ils sont localisés à la membrane plasmatique, a récemment été remis en question. Des données récentes indiquent que certains GPCRs peuvent également induire une réponse intracellulaire à partir des compartiments intracellulaires dont le noyau. Les récepteurs activés par la protéase (PAR) sont des membres de la famille GPCR. Les PARs sont activés par le clivage de la partie N–terminale du récepteur ce qui permet au ligand attaché sur le récepteur de se lier à sa poche réceptrice. Quatre PARs ont été décrits : PAR1, PAR2, PAR3 et PAR4. PAR2 peut susciter des effets mitogéniques et participer aux processus comme l’angiogenèse et l'inflammation. Alors que beaucoup d'effets intracellulaires de PAR2 peuvent être expliqués lorsqu’il est localisé à la membrane plasmatique, une fonction intracrine de PAR2 a aussi été proposée. Pourtant les mécanismes par lesquels PAR2 peut provoquer l’expression de gènes ciblés sont toujours inconnus. Le but de notre étude était de vérifier l’existence d’une population nucléaire de PAR2. Nous avons également émis l’hypothèse que les voies activées par l’activation de PAR2 dépendent de sa localization cellulaire. En utilisant des techniques de microscopie confocale et de "Western Blot" nous avons démontré la présence d’une population nucléaire de PAR2. À la suite de la stimulation de PAR2, nous avons observé une augmentation de la translocation du récepteur de la membrane plasmatique au noyau. En utilisant la technique de "RT – PCR", nous avons observé des rôles différents de PAR2 à la surface de la cellule et du noyau dans l’initiation de l’expression des gènes. Afin d’identifier les mécanismes responsables de la translocation nucléaire de PAR2, nous avons évalué l’implication des membres de la famille de "Sorting Nexins (SNX)" dans la translocation nucléaire de PAR2. "Sorting Nexins" est un groupe de protéines avec des fonctions de transport bien établies. SNX1 et SNX2 ont été identifiés comme responsables du transfert de PAR1 vers les lysosomes. SNX11 n'a pas encore été étudié et nous avons émis l’hypothèse qu'il pourrait être un autre membre de la famille des SNX impliqué dans la signalisation de PAR2. Pour ce faire, nous avons développé des "knockdowns" stables pour SNX1, SNX2 et SNX11 dans les cellules HEK293. En utilisant les essais d’immunofluorescence, "Western Blot" et de cytométrie en flux, nous avons déterminé que tous les trois membres du groupe SNX sont des partenaires d'interaction de PAR2. Toutefois, seul SNX11 se co-localise avec son partenaire au noyau et est responsable de sa translocation nucléaire. Les expériences de "RT - PCR" sur les lignées de cellule de SNXs "knockdowns" ont démontré que la fonction de PAR2 nucléaire dépend surtout de SNX11; néanmoins SNX1 et SNX2 peuvent aussi l’influencer, suggérant qu'ils font aussi partie du réseau signalétique de PAR2. En conclusion, PAR2 est déplacé de la membrane plasmatique à la membrane nucléaire après sa stimulation avec un agoniste. La translocation nucléaire de PAR2 par un mécanisme impliquant SNX11, initie des effets intracellulaires différents de sa signalisation membranaire. The present investigation is dedicated to understanding various mechanisms of salinity tolerance in the estuarine clam V. cyprinoides var. cochinensis. Even though V. cyprinoids var. cochinensis and V. cyprinoides are found to coexist in the same area, V. cyprinoids is reported to tolerate higher salinities than variety cochinenesis. Variations in the salinity of sea water may affect the aquatic organisms through specific gravity control and variations in osmotic pressure. The specific gravity of most soft tissues is close to that of normal seawater. Many bottom living forms, both attached and motile, have very high specific gravities eg.villorita cyprinoids. Villorita spp. Occurs abundantly in the reaches of the estuary and backwaters of Kerala. In both marine and estuarine forms, it is observed that mantle employs a lesser quantity of amino acids compared to adductor and foot. The regulation of cell volume is not carried out equally in all types of tissues. The capability of salinity tolerance is an aggregate of both the capabilities of extra cellular anisosmotic and intracellular isosmotic regulations in osmoconforming animals. The ultimate aim of water regulation is to regulate the cell volume.T here are slight changes occur in cell volume even in osmoregulators. These studies can also help in revealing the changes brought about in the cellular organelles like lysosomes, which were found to have a role in the osmoregulatory process. The osmoregulatory machinery of estuarine animals is more streamlined for a successful life in the estuarine regime.

Aquesta tesi doctoral s'engloba dins d'un projecte general d'estudi de gens implicats en l'embriogènesi del blat de moro. L'embriogènesi del blat de moro, i en general la de totes les plantes superiors, es dóna en tres etapes: una primera etapa on es diferencien tots els diversos teixits que formaran l'embrió, una segona etapa on l'embrió acumula productes de reserva i un tercer període, la dormància, que finalitza quan les condicions ambientals són les idònies per a la germinació. En el laboratori estàvem interessats, concretament, en l'estudi de gens implicats en la primera etapa morfogenètica, on els diferents teixits i estructures embrionàries queden definides. Per tal d'estudiar gens que s'expressaven en aquest període, una de les estratègies que es va realitzar fou un crivellat diferencial entre teixit embrionari i teixit de planta adulta. D'entre els diferents clons obtinguts, un corresponia a un clon parcial que presentava similitud amb receptors quinasa i que fou objecte d'estudi. A partir d'aquest clon es va obtenir el clon complet i es va anomenar MARK (per Maize Atypical Receptor Kinase). MARK presenta una estructura típica d'un receptor quinasa amb un domini extracel.lular, que conté 6 còpies imperfectes de LRR (Leucine- Rich Repeats), un únic domini transmembrana i un domini quinasa intracel.lular. El domini quinasa de MARK presenta, però, algunes variacions en els residus aminoacídics que es consideren claus per a la funció catalítica dels dominis quinasa. En concret cinc dels aminoàcids considerats essencials per a la fosforilació es troben substituits en el domini quinasa de MARK (DK-MARK). Els experiments de fosforilació in vitro que es van realitzar al laboratori, van mostrar com MARK era incapaç de fosforilar in vitro. Aquesta característica no és, però, exclusiva de MARK. Una búsqueda en les bases de dades ens van permetre identificar altres seqüències que també presentaven els mateixos o altres canvis en aquestes posicions aminoacídiques. En les bases de dades de plantes es van identificar un conjunt de seqüències genòmiques o ESTs amb aquestes característiques i només una d'elles, la proteïna TMKL1 d'Arabidopsis, ha sigut descrita com un receptor quinasa incapaç de fosforilar in vitro. Respecte a la búsqueda de receptors similars a MARK en les bases de dades d'animals, es van identificar també un conjunt de proteïnes que, en alguns casos, s'ha descrit que no tenen activitat quinasa in vivo. Per exemple, un dels casos més ben estudiats és el del receptor erbB3 que forma part de la família de receptors del EGF (Epidermal Growth Factor). Aquesta família de receptors està formada per 4 receptors: erbB1, erbB2, erbB3 i erbB4, dels quals només l'erbB3 no presenta activitat catalítica. S'ha descrit que erbB3 és capaç, tot i no fosforilar in vivo, de participar activament en la transducció del senyal formant heterodímers amb els altres membres de la família. Així, erbB3 és fosforilat pel seu partner i pot iniciar la cascada de transducció del senyal. La participació d'erbB3 en la transducció del senyal és essencial ja que embrions de ratolí knock-out pel gen erbB3 són inviables. Així doncs, el fet que receptors quinasa catalíticament inactius participin en les cascades de transducció del senyal, suggereix l'existència de nous mecanismes d'acció per a la transducció del senyal. Per tant, l'objectiu d'aquest treball fou l'estudi del mecanisme d'acció de MARK mitjançant la caracterització les proteïnes capaces d'interaccionar amb el seu domini quinasa. Per tal d'assolir aquest objectiu, es va realitzar un crivellat de doble-híbrid amb una llibreria de cDNA d'embrions de blat de moro de 7 DAP. D'aquest crivellat es va obtenir un conjunt de possibles clons positius que foren seqüenciats i entre els quals es van escollir per un estudi més detallat aquells que s'havien obtingut més vegades com a clons independents. Aquests clons codificaven per: una SAMDC (S-Adenosil Descarboxilasa), una eIF5 (Eukaryotic translation initiation), una hypothetical protein, una unknown protein, una gamma-adaptina i una MAP4K. Amb aquests 6 clons es van fer estudis in vitro i in vivo per tal de confirmar al seva interacció amb DK-MARK. Els estudis in vivo es van realitzar amb la soca de llevat AH109, una soca més astringent que la utilitzada en el crivellat, ja que presenta tres gens marcadors: Histidina, Adenina i Lacz. Els resultats obtinguts van mostrar que els clons codificants per SAMDC i eIF5 no van créixer en un medi selectiu per His i Ade i, per tant o es tracta de falsos positius del sistema o la seva interacció amb DK-MARK és dèbil. D'altra banda, la resta dels clons analitzats (proteïna hipotètica, una proteïna de funció desconeguda, la gamma-adaptina i una MAP4K) van créixer en medis en absència de Histidina i Adenina. Els assatjos de b-galactosidasa van ser tots positius a excepció de la proteïna hipotètica suggerint que potser aquesta interacció sigui més feble. D'altra banda també es van realitzar estudis in vitro amb la tècnica del pull-down. Els resultats obtinguts amb aquesta tècnica van recolzar els obtinguts en cèl.lules de llevat, ja que tots els clons analitzats a excepció dels codificants per SAMDC i eIF5 van donar un resultat d'interacció amb KD-MARK in vitro positiu. Davant aquests resultats ens vam centrar en l'estudi de la proteïna similar a MAP4K, doncs algunes proteïnes de la seva família s'han relacionat amb receptors de membrana. Els clons que es va obtenir del crivellat codificaven per una proteïna similar amb el domini C-terminal a les proteïnes BnMAP4Ka1 i a2 de Brassica napus. Aquestes proteïnes presenten una forta similitud de seqüència amb proteïnes de la família GCK/SPS1 que formen part d'un grup particular de MAPK relacionades amb la proteïna Ste20 (sterile 20 protein) de llevat. Ste20p activa la MAP3K de llevat Ste11 directament per fosforilació, transduint d'aquesta manera el senyal del receptor de feromones de creuament de les cèl.lules de llevat i es pot, doncs, considerar com una proteïna del tipus MAP4K (mitogen-activated protein kinase kinase kinase kinase). En els darrers anys, s'han identificat un gran nombre de proteïnes similars a Ste20: fins a una trentena en mamífers, en Drosophila, en Caenorhabditis elegans i en altres organismes. Segons la seva estructura aminoacídica, la família Ste20 s'ha classificat en dues subfamílies: les proteïnes STE20/PAK (p21-activated kinases) i la subfamília GCK/SPS1 (germinal center kinases). Les dues subfamílies estan formades per proteïnes que contenen un domini quinasa i un domini regulador, però, mentre que les proteïnes PAK presenten el domini quinasa en la part C-terminal, les GCKs el presenten en la regió N terminal. Les proteïnes GCK presenten una elevada diversitat estructural en el domini regulador permetent la seva classificació en 6 subfamílies. Mitjançant la tècnica del RACE es va obtenir el clon de cDNA complet que es va anomenar MIK (MARK Interacting Kinase). Amb la tècnica del Southern blot es va poder determinar que el gen MIK és un gen de còpia única en el genoma de blat de moro. Per tal d'analitzar la possible interacció entre DK-MARK i MIK, es va estudiar tant el patró d'expressió d'ambdós gens com el seu patró d'acumulació d'ambdues proteïnes durant l'embriogènesi del blat de moro. El patró d'expressió, analitzat per Northen blot va mostrar uns patrons coincidents al llarg de l'embriogènesi des del seu inici fins als 20 DAP amb una acumulació màxima de mRNA en embrions de 15 DAP. D'altra banda per tal d'estudiar el patró d'acumulació de la proteïna MIK així com per comparar-lo amb el de MARK, es van realitzar estudis de Westerns blot. Els resultats també van mostrar una coincidència en el temps de l'acumulació de les proteïnes MARK i MIK durant l'embriogènesi de blat de moro amb una major acumulació en embrions de 15 i 20 DAP. Es van dur a terme també estudis d'immunolocalitzacions sobre embrions de blat de moro de 15 DAP per tal d'estudiar en quins teixits s'acumulaven ambdues proteïnes. Les immunolocalitzacions van mostrar una major acumulació tant de MARK com de MIK en les zones meristemàtiques i en el teixit vascular sobretot del coleòptil on s'aprecia una forta co-localització de MARK i MIK. Totes aquestes dades són compatibles, doncs, amb una possible interacció de les proteïnes MARK i MIK, tot i que no la demostren. Per tal de demostrar la interacció es van realitzar experiments d'immunoprecipitació in vivo a partir d'extractes d'embrions. Malauradament, els resultats no són clars i en aquests moments en el laboratori s'estan posant a punt aquests experiments. També es van realitzar estudis comparatius de seqüència amb diferents proteïnes de la família GCK, mostrant una major similitud amb les proteïnes de la subfamília GCK-III. La subfamília GCK-III ha estat molt poc estudiada i en formen part un conjunt de proteïnes amb funcions molt diverses des de l'apoptosi, la citoquinesi o l'anòxia cel.lular. Per tant, la similitud de seqüència possiblement fa referència a una conservació en el mecanisme d'acció més que no pas a una conservació funcional. La possible interacció de MARK amb el domini C-terminal de MIK (el domini regulador) podria activar aquesta última iniciant una cascada de transducció del senyal en un model en el que una proteïna del tipus GCK-III faria de lligam directa entre un receptor de membrana i una cascada de senyalització intracel.lular. Aquest tipus de lligam entre un recepctor de membrana i mòduls intracel.lulars de senyalització s'ha descrit per a altres proteïnes GCK, si bé no directament sinó a través de proteïnes adaptadores. D'altra banda, la interacció directa de MARK, un receptor quinasa atípic que no té activitat catalítica, amb MIK suggereix un mecanisme on receptors atípics podrien interaccionar en la transducció del senyal activant la via de les MAPK.

The focus of the present review is to assimilate current knowledge concerning the differing signalling transduction cascades that control muscle mass development and affect skeletal muscle phenotype following exercise or nutritional uptake. Effects of mechanical loading on protein synthesis are discussed. Muscle growth control is regulated by the interplay of growth promoting and growth suppressing factors, which act in concert. Much emphasis has been placed on understanding how increases in the rate of protein synthesis are induced in skeletal muscle during the adaptive process. One key point to emerge is that protein synthesis following resistance exercise or increased nutrient availability is mediated through changes in signal transduction involving the phosphorylation of mTOR and sequential activation of downstream targets. On the other hand, AMPK activation plays an important role in the inhibition of protein synthesis by suppressing the function of multiple translation regulators of the mTOR signalling pathway in response to cellular energy depletion and low metabolic conditions. The effects of exercise and/or nutritional uptake on the activation of signalling molecules that regulate protein synthesis are highlighted, providing a better understanding of the molecular changes in the cell.
