Soil management and crop rotations can affect P and K budget in soil, decreasing losses, and increasing fertilizer use efficiency. The P and K budget in the soil-plant system at depths up to 60. cm was studied for different soil managements and crop rotations under no-till for three years in Botucatu, São Paulo, Brazil. The investigated crop rotations were: triticale (X Triticosecale) and sunflower (Helianthus annuus) cropped in autumn-winter; pearl millet (Pennisetum glaucum), forage sorghum (Sorghum bicolor), and Sunn hemp (Crotalaria juncea) were grown in the spring, as well as an additional treatment with chiseling followed by a fallow period; and soybean (Glycini max, L., Merril) was cropped in the summer. Each year triticale and sunflower were grown in plots and pearl millet, forage sorghum, Sunn hemp and of chisel/fallow in sub-plots. The triticale/millet rotation led to the largest decrease in available P within the 0-0.60. m layer of the soil profile and the largest K increase within the 0-0.05. m layer. Potassium mobility in the soil profile and the increases in the available K content in the 0.40-0.60. m layer were independent of the management system. Crop rotations with or without chiseling are not effective in preventing soil P losses. There is considerable K leaching below 0.60. m, but chiseling and the use of high K accumulating plants as triticale results in lower K losses. © 2012 Elsevier B.V.

Il vigore è un aspetto rilevante della qualità delle sementi, strettamente connesso al loro status fisiologico, al genotipo e alle condizioni di stoccaggio. Stress ossidativi e danni alle macromolecole sono alla base del deterioramento del seme, che è equipaggiato con sistemi protettivi e di riparazione. Uno di questi coinvolge lL-isoaspartil metiltransferasi (PIMT) che ripara i misfolding proteici catalizzando la riconversione in aspartato dell’isoaspartile anomalo accumulato. Scopo di questo studio era valutare il possibile ruolo del meccanismo di riparazione di PIMT nel vigore del seme in girasole. Per questo il relativo gene è stato isolato e caratterizzato, la variabilità allelica determinata su un campione di linee inbred e lespressione genica misurata in risposta all’invecchiamento accelerato (aging) e al priming. La sequenza codificante ottenuta è costituita da 4 esoni e contiene i 5 domini caratteristici delle metiltransferasi. Il gene mostra elevata similarità con gli ortologhi vegetali e scarsa diversità nucleotidica nei genotipi coltivati rappresentativi della variabilità della specie. Nella sequenza aminoacidica, comunque, sono state rinvenute tre sostituzioni che potrebbero influenzare la funzionalità enzimatica. Dal punto di vista fisiologico i genotipi considerati hanno esibito notevole variabilità di risposte ai trattamenti, sia in termini di vigore che di espressione genica. Aging e priming hanno prodotto generalmente gli effetti attesi, rispettivamente negativi e positivi, sulla germinabilità e sulla sua velocità. In generale lespressione di PIMT è risultata massima nel seme secco, come riportato altrove, e ridotta dall’aging. Anche il priming ha diminuito lespressione rispetto al seme quiescente, mentre il suo effetto dopo laging è risultato genotipo-dipendente. Tuttavia, nelle condizioni descritte, non si sono evidenziate correlazioni significative tra vigore ed espressione di PIMT, tali da suggerire un chiaro ruolo di questo meccanismo nella qualità fisiologica del seme.

Os elementos potencialmente tóxicos (EPTs) estão presentes nos solos em concentrações dependentes do material de origem e das ações antrópicas. A adição de EPTs ao solo pelas atividades antrópicas pode ocasionar risco à saúde humana, já que estes elementos podem ser acumulados no organismo por meio do contato dérmico com o solo, da inalação de partículas em suspensão, de ingestão de solo e de alimentos contaminados. A contaminação dos alimentos ocorre pelo cultivo em áreas com alta biodisponibilidade de EPTs, e nessa condição ocorre absorção e translocação para a parte aérea, com possível acúmulo dos metais nas porções comestíveis, como raízes, frutos e grãos. A biodisponibilidade dos EPTs é regulada pelas características químicas dos elementos e por atributos do solo, como a CTC, o pH e a matéria orgânica (MO). Sintomas de toxicidade e alterações morfolgicas e fisiolgicas podem aparecer dependendo da absorção e da movimentação dos EPTs nas plantas. Objetivou-se neste trabalho avaliar o efeito da adição de bário (Ba), de cádmio (Cd), de cobre (Cu), de níquel (Ni) e de zinco (Zn) em amostras de um Neossolo Quartzarênico e um Latossolo Vermelho distrófico, sob duas condições de saturação por bases (30% e 50 ou 70%, dependendo da cultura), no cultivo de arroz (Oryza sativa), alface (Lactuca sativa), girassol (Helianthus annuus) e tomate (Solanum lycopersicum). Os EPTs nos solos foram extraídos com EPA 3051a, Água Régia, DTPA, Mehlich 1, Mehlich 3, HNO3 (0,43 mol L-1) e CaCl2 (0,01 mol L-1), e seus teores correlacionados com os presentes nas raízes, na parte aérea, nos frutos e com a quantidade acumulada pelas plantas. Os fatores de bioconcentração (FBC) e de transferência (FT) foram calculados para as culturas. O índice SPAD (Soil Plant Analysis Development - Chlorophyll Meter) foi determinado na fase vegetativa da alface, do arroz e do girassol, enquanto a atividade fotossintética foi determinada pelo IRGA (Infrared gas analyzer). Os maiores teores de EPTs foram observados nas plantas cultivadas no Neossolo. As quantidades de Cu, Ni e Zn acumuladas nas plantas apresentaram correlação positiva com os teores extraídos pelo EPA 3051a e pela Água Régia. Os teores extraídos com HNO3 (0,43 mol L-1) apresentaram elevada correlação positiva com os teores reativos extraídos com DTPA e com Mehlich 3, e também com as quantidades de EPTs acumuladas pelas plantas. Os FBCs foram mais altos nos solos com baixa CTC, baixos teores de MO e baixos valores de pH. O arroz apresentou a menor translocação de Cd do sistema radicular para os grãos. O Cu, o Ni e o Zn causaram alterações no desenvolvimento da alface e do girassol, e diminuíram a transpiração e a condutância estomática da alface. O arroz apresentou a menor absorção de EPTs e a maior tolerância ao Ba, ao Cd, ao Ni e ao Zn, no entanto, as plantas apresentaram maiores condutividade estomática e transpiração.

Flavonoids in Australian honeys from five botanical species (Melaleuca, Guioa, Lophostemon, Banksia and Helianthus) have been analyzed in relation to their floral origins. Tea tree (Melaleuca quinquenervia) and heath (Banksia ericifolia) honeys show a common flavonoid profile comprising myricetin (3,5,7,3',4',5'-hexahydroxyflavone), tricetin (5,7,3',4,5'-pentahydroxyflavone), querectin (3,5,7,3',4'-pentahydroxyflavone) and luteolin (5,7,3',4'-tetrahydroxyflavone), which was previously suggested as a floral marker for an Australian Eucalyptus honey (bloodwood or Eucalyptus intermedia honey). These honeys of various floral species can be differentiated by their levels of total flavonoids, being 2.12 mg/100 g for heath honey and 6.35 m/100 g for tea tree honey. In brush box (Lophostemon conferta) honey, the flavonoid profile comprising mainly tricetin, luteolin and quercetin is similar to that of another Eucalyptus honey (yellow box or Eucalyptus melliodora honey). These results indicate that the flavonoid profiles in some of the Australian non-Eucalyptus honeys may contain more or less certain flavonoids from Eucalyptus floral sources because of the diversity and extensive availability of Eucalyptus nectars for honeybee foraging yearly around or a possible cross contamination of the monofloral honeys during collection, transportation and/or storage. Further analyses are required to differentiate and/or verify the botanical sources of the flavonoids that contribute to the flavonoid profiles of these honeys, by restricting honey sampling areas and procedures, employing other complementary analytical methods (e.g. pollen analysis, sugar profile) and using materials (e.g. nectar) directly sourced from the flowering plant for comparative studies. In Australian crow ash (Guioa semiglauca) honey, myricetin, tricetin, quercetin, luteolin and an unknown flavonoid have been found to be the main flavonoids, which is characteristic only to this type of honey, and could thus be used as the floral marker, while in Australian sunflower (Helianthus annuus) honey, the content of total flavonoids is the smallest amount comparing to those in the other honeys analysed in this study. However, the flavonoid quercetin and the flavonoid profile mainly consisting of quercetin, quercetin 3,3'-dimethyl ether (5,7,4'-trihydroxy3,3'-dimethoxyflavone), myricetin and luteolin are characteristic only to this sunflower honey and could thus be used for the authentication.

SFTI-1 is a bicyclic 14 amino acid peptide that was originally isolated from the seeds of the sunflower Helianthus annuus. It is a potent inhibitor of trypsin, with a sub-nanomolar K, value and is homologous to the active site region of the well-known family of serine protease inhibitors known as the Bowman-Birk trypsin inhibitors. It has a cyclic backbone that is cross-braced by a single disulfide bridge and a network of hydrogen bonds that result in a well-defined structure. SFTI-1 is amenable to chemical synthesis, allowing for the creation of synthetic variants. Alterations to the structure such as linearising the backbone or removing the disulfide bridge do not reduce the potency of SFTI-1 significantly, and minimising the peptide to as few as nine residues results in only a small decrease in reactivity. The creation of linear variants of SFTI-1 also provides a tool for investigating putative linear precursor peptides. The mechanism of biosynthesis of SFTI-1 is not yet known but it seems likely that it is a gene-coded product that has arisen from a precursor protein that may be evolutionarily related to classic Bowman-Birk inhibitors.

Eight phenolic acids and two abscisic acid isomers in Australian honeys from five botanical species (Melaleuca, Guioa, Lophostemon, Banksia and Helianthus) have been analyzed in relation to their botanical origins. Total phenolic acids present in these honeys range from 2.13 mg/100 g sunflower (Helianthus annuus) honey to 12.11 mg/100 g tea tree (Melaleuca quinquenervia) honey, with amounts of individual acids being various. Tea tree honey shows a phenolic profile of gallic, ellagic, chlorogenic and coumaric acids, which is similar to the phenolic profile of an Australian Eucalyptus honey (bloodwood or Eucalyptus intermedia honey). The main difference between tea tree and bloodwood honeys is the contribution of chlorogenic acid to their total phenolic profiles. In Australian crow ash (Guioa semiglauca) honey, a characteristic phenolic profile mainly consisting of gallic acid and abscisic acid could be used as the floral marker. In brush box (Lophostemon conferta) honey, the phenolic profile, comprising mainly gallic acid and ellagic acid, could be used to differentiate this honey not only from the other Australian non-Eucalyptus honeys but also from a Eucalyptus honey (yellow box or Eucalyptus melliodora honey). However, this Eucalyptus honey could not be differentiated from brush box honey based only on their flavonoid profiles. Similarly, the phenolic profile of heath (Banksia ericifolia) honey, comprising mainly gallic acid, an unknown phenolic acid (Phl) and coumaric acid, could also be used to differentiate this honey from tea tree and bloodwood honeys, which have similar flavonoid profiles. Coumaric acid is a principal phenolic acid in Australian sunflower honey and it could thus be used together with gallic acid for the authentication. These results show that the HPLC analysis of phenolic acids and abscisic acids in Australian floral honeys Could assist the differentiation and authentication of the honeys. © 2005 Elsevier Ltd. RESUMO: O teste de condutividade eltrica é um teste de vigor de sementes utilizado para avaliar a qualidade das sementes e complementar o teste de germinação. O tamanho da amostra necessária para a estimação da média de condutividade eltrica de sementes é diretamente proporcional à variabilidade dos dados e confiabilidade desejada na estimativa. Uma técnica adequada para dimensionamento amostral é a utilização de intervalos de confiança, obtidos por reamostragem. Essa técnica tem a vantagem de não depender da distribuição de probabilidade dos dados. Assim, o objetivo deste trabalho foi estimar o tamanho de amostra necessário para o teste de condutividade eltrica individual em sementes de girassol. Foram utilizadas sete lotes de sementes de girassol, sendo três lotes da cultivar Hélio 250 (A, B e C), dois lotes da cultivar BRS 323 (D e E) e dois lotes da cultivar BRS 324 (F e G). O teste de condutividade eltrica individual foi realizado com 400 sementes de cada lote e o tamanho de amostra foi determinado pelo número de sementes a partir do qual a amplitude do intervalo de confiança de 95% (AIC95%) foi menor ou igual a 10, 15 e 20% da estimativa da média. Tempos de embebição de vinte e quatro hora possibilitam utilizar os menores tamanhos de amostras. O tamanho de amostra de 257 sementes é recomendado para a amplitude do intervalo de confiança de 95% igual a 20% da estimativa da média. ABSTRACT: The aim of this study was to estimate the sample size required for the individual electrical conductivity test in sunflower seeds. The sample size of 257 seeds is recommended for the amplitude of the 95% confidence interval of 20% of the mean estimate.

The objective of this work was to evaluate the biological variables of Spodoptera frugiperda on species of cover crops. The experiments were conducted in laboratory and greenhouse using the following species: sunflower (Helianthus annuus), sun hemp (Crotalaria juncea), brachiaria (Urochloa decumbens e Urochloa ruziziensis), millet (Pennisetum americanum), black oat (Avena stringosa), white lupin (Lupinus albus), forage turnip (Rafanus sativus) and maize (Zea mays). In laboratory the S. frugiperda larval survival varied from 57%, on L. albus, to 93% on H. annuus and the survival of the pre-imaginal phase varied from 45% on U. decumbens to 81.6% on Z. mays. On C. juncea the larval biomass was lower and the development period of the young and larval stage was higher. The adaptation index was less on C. juncea in greenhouse and laboratory. In greenhouse the larval survival at 14 days was similar for all plants and at 21 days was the lowest on C. juncea. There was less accumulation of biomass at 14 days on C. juncea and at 21 days on C. juncea and A. stringosa. Regarding damage, C. juncea presented less susceptibility to Spodoptera frugiperda attack, which together with the other evaluated parameters, indicated this plant as the most appropriate for soil cover before cultivation of maize.

Sunflower is one of the most important oilseed crops and produces a high-quality edible oil. Balance of fatty acids in standard sunflower oil shows preponderance of linoleic rather than oleic acid, and conditions during seed development, such as temperature, changes the oleic/linoleic ratio of the oil. This work aimed to evaluate the environmental effect on fatty acid profile in a group of standard and high oleic varieties and hybrids. Seeds were produced during regular season crop and during off-season crop featuring different temperatures from anthesis to maturity. Fatty acid composition was determined by gas chromatography. Levels of oleic acid, in standard oil genotypes, raised as the crop developed in warmer environment while levels of linoleic acid decreased, and the opposite was observed when the crop was grown under lower temperature. High oleic genotypes were less sensitive to environment switching and showed lower variation on fatty acid composition.

The continuous soybean-maize crop succession in the tropical region of Brazil has led to significant increases in the population size of root-knot (Meloidogyne incognita and M. javanica ) and root-lesion nematodes (Pratylenchus brachyurus), which make soils unsuitable for soybean cropping. A greenhouse study was conducted to identify sunflower genotypes adapted to the tropical region of Brazil and that are resistant to M. incognita, M. javanica and/or P. brachyurus . Two experiments for each nematode were conducted in a completely randomized design with six replicates. Gall index was calculated from visual scores (0?5) of gall intensity on roots for the root-knot nematode. Initial and final population density and reproduction factor were also measured for each nematode. Sunflower genotypes varied in resistance to the nematodes. Sunflower hybrids BRS 321 and BRS 323 were resistant to M. javanica and P. brachyurus and exhibited low gall index for M. incognita . The cultivars are good alternatives to using in the succession of soybean in nematode-infested areas of the tropical regions of Brazil. No sunflower genotype was identified as resistant to M. incognita and thus sunflower cropping is not indicated in areas infested with this nematode.

A specimen of downy mildew on leaves of Sphagneticola trilobata found in northern Queensland was identified by a systematic approach as a novel species of Plasmopara. A new species, Plasmopara sphagneticolae, is proposed for this specimen, which differs from other species of Plasmopara by morphology, host range, and sequence data from nuclear-ribosomal DNA and mitochondrial DNA. Plasmopara sphagneticolae, together with P. halstedii, are downy mildews found on host species in the tribe Heliantheae (Asteraceae). Plasmopara halstedii causes downy mildew on Helianthus annuus, and is not present on sunflower in Australia. Phylogenetic analysis of the large subunit region of ribosomal DNA showed that P. sphagneticolae was sister to P. halstedii on sunflower.

Seeds of 14 plant species extracted from food products proposed for field trips in Galapagos were tested for viability. Strawberry Fragaria ananasa and Blackberry Rubus glaucus jams (Snob and Gustadina brands) contained no viable seeds. Schullo brand granola contained inviable Sesame Sesamum indicum seed, but Sesame in granolas prepared in Galapagos was viable. Sesame seed in bread was viable but Flax Linum usitatissimum seed in bread was not. Brown Rice Oryza sativa and Sunflower seeds Helianthus annuus were both viable. Fresh Apple Malus domestica, Naranjilla Solanum quitoense, Cucumber Cucumis sativus, Pineapple Ananas comosus, Pear Pyrus communis, Bell Pepper Capsicum annuum, Tomato Solanum lycopersicum, Grape Vinis vinifera all contained viable seeds. We recommend prohibiting any product with viable seeds from field trips to uninhabited areas. CDF Contribution Number 1009.

中国资源植物丰富,蕴藏着优异的基因资源,开发和利用这些优异资源是植物学研究的重要课题。本文面向国家重大需求选择两种资源植物一羊草(Leymuschinensis (Trin.)Tzvel)和向日葵(llelia thus annuus L),采用分子标记技术和分子生物学方法对其进行评价和研究,以期为资源利用提供依据。由于两种植物本身的差别和采用的研究方法各异,故分别论述。 羊草,隶属禾本科赖草属,是欧亚大陆草原区东部重要建群种之一。羊草是牧草之王,是我国比较有优势的战略性生物资源,对我国北方畜牧业的发展以及生态环境的保育均具有重要意义。近年来,由于缺乏科学管理、过度放牧等不利影响,加之羊草本身固有的“三低”问题(即抽穗率低、结实率低、发芽率低)已对羊草生物多样性维持构成了严重的威胁,限制了我国人工草地建设和天然草地的改良及沙化治理的步伐。因此,如何通过形态调查结合生物技术手段评价羊草遗传多样性为建立核心种质及改良羊草、快速评价和创造新的种质、如何加快育种进程便成为当前亟待解决的问题。本文围绕这些问题开展了系统的研究并取得如下结果: 1. 对羊草的形态调查和AFLP分析,表明羊草是一种形态变异较大但是遗传变 异较小的物种。两种生态型的表现显著差异,其中灰绿生态型羊草比黄绿生 态型差异大。羊草遗传多样性与包括长期的栽培驯化、地理分布有很大的相 关性,地理来源相同的几乎全部聚到了一组。 2. 通过主成分分析和通径分析,简化了羊草31个性状分析的复杂性,了解到 羊草无性繁殖受好的营养生长促进。 3. AFLP分子标记技术在分析羊草遗传多样性方面有显著优势,尤其是对于羊 草这样多态性不高的物种是一种非常有效的分析工具。在分析AFLP数据时 采用聚类分析和主坐标分析相结合的方法,既兼顾了亲缘关系较近的种质之 间的关系调查也兼顾了亲缘关系较远的种质之间的关系调查。 4.羊草AFLP反应,不同引物所获得的总带数和多态性带数差别明显。羊草基因 组对3’端有选择性碱基TN的所有EcoRJ选择性引物扩增效果很差,前人 的有关赖草属的遗传研究也支持这一结果。 向日葵(n=17),属于菊科( Compositae)向日葵属(Helia thus),向日葵的研究重要领域是向日葵杂种生产,而细胞质雄性不育系的使用是杂种优势育种的核心。全世界90%以上的向日葵杂交种生产仍然在使用同一个细胞质类型PETI,玉米遗传单一给生产带来的毁灭性打击仍然令研究者和生产者记忆犹新,因此寻找更多的细胞质类型仍然是研究者的重要任务。本研究围绕一个新的不育源(G20023)的发现及鉴定,通过使用不育的G20023的保持系、恢复系、恢复的Fi代、回交一代之间比较以及与属于PETI细胞质类型的不育系的相应材料进行比较,找出与这一新的细胞质类型不育表型有关的可能差异序列,来探讨其不育机制,得到如下结果: 1、 通过田间杂交试验,证明G20023的保持系有很多(已证实有24份), 目前找到的恢复系只有一个,H.maximiliani。G20023不育源作为一 种新的细胞质类型可以成为将来杂交育种的候选资源。同时,我们找 到一些表型证据,除了无花粉之外,G20023与PETI表型的典型不同 之处还在于前者的花药上下均为分离状态,而后者花药的基部联合, 顶部分离。显然,不同的细胞质类型在解剖结构上可能表现不尽相同。 2、 与线粒体基因组特异基因的核酸序列比较,结果表明,G20023线粒体 基因组上没有orfH522序列,与PETI表现出差异;此外,在基因atp6 位点也与PETI不同,而且在该位点也与同属向日葵ANTI不相同。同 时由于orf873并没有出现在ANTI中而出现在G20023中,因此我们可 以认为G20023这一个新的不育系是与ANTI和PETI不同的细胞质类 型。 3、 在参考常规线粒体DNA提取方法的基础上,我们做了很多改进,建立 了自己的向日葵线粒体DNA提取方法。该方法更快更简单,提取的线 粒体DNA完全可以用于酶切和杂交。 4、 G20023不育源由于其稳定的不育性状,可以作为培育无花粉彩色向日 葵杂交种的亲本材料,我们通过此不育源选育适当花色的无花粉观赏 向日葵生产杂交种。
