Purpose: Anti-oxidation and exocytosis are important for maintaining exocrine tissue homeostasis. During aging, functional and structural alterations occur in the lacrimal gland (LG), including oxidative damage to proteins, lipids, and DNA. The aims of the present study were to determine in the aging LG: a) the effects of aging on LG structure and secretory activity and b) changes in the expression of oxidative stress markers. Methods: To address these goals, tear secretion composition and corneal impression cytology were compared between male Wistar rats of 2 (control) and 24 (aged) months. LG morphology and the expression levels of vitamin E and malonaldehyde (MDA) were evaluated to determine the anti-oxidant activity and lipid peroxidation, respectively. RT-PCR and western blot analysis were used for the analysis of Ras related in brain GTPase protein (Rab) and soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) proteins of the secretory machinery (i.e.; Rab 3d, Rab 27, vesicle-associated membrane protein-2 (Vamp-2), and syntaxin). Results: Histological analysis of aged rats revealed a higher frequency of corneal epithelia metaplasia. In the acinar cells, organelles underwent degeneration, and lipofucsin-like material accumulated in the cytoplasm along with declines in the anti-oxidant marker vitamin E. Rab3d and Rab27b mRNA levels fell along with Rab3d protein expression, whereas syntaxin levels increased. Conclusions: These findings indicate that exocytotic and anti-oxidant mechanisms become impaired with age in the rat LG. In parallel with these structural alterations, functional declines may contribute to the pathophysiology caused by tear film modification in dry eye disease.

Many pathways can be used to synthesize polythiophenes derivatives. The polycondensation reactions performed with organometallics are preferred since they lead to regioregular polymers (with high content of heat-to-tail coupling) which have enhanced conductivity and luminescence. However, these pathways have several steps; the reactants are highly moisture sensitive and expensive. On the other hand, the oxidative polymerization using FeCl3 is a one-pot reaction that requires less moisture sensitive reactants with lower cost, although the most common reaction conditions lead to polymers with low regioregularity. Here, we report that by changing the reaction conditions, such as FeCl3 addition rate and reaction temperature, poly-3-octylthiophenes with different the regioregularities can be obtained, reaching about 80% of heat-to-tail coupling. Different molar mass distributions and polydispersivities were obtained. The preliminary results suggest that the oxidative polymerization process could be improved to yield polythiophenes with higher regioregularity degree and narrower molar mass distributions by just setting some reaction conditions. We also verified that it is possible to solvent extract part of the lower regioregular fraction of the polymer further improving the regioregularity degree. (C) 2011 Wiley Periodicals, Inc. Copper complexes with fluorinated beta-diketones were synthesized and characterized in terms of lipophilicity and peroxide-assisted oxidation of dihydrorhodamine as an indicator of redox activity. The biological activity of the complexes was tested against promastigotes of Leishmania amazonensis. Inhibition of trypanosomatid-specific trypanothione reductase was also tested. It was found that the highly lipophilic and redox-active bis(trifluoroacetylacetonate) derivative had increased toxicity towards promastigotes. These results indicate that it is possible to modulate the activity of metallodrugs based on redox-active metals through the appropriate choice of lipophilic chelators in order to design new antileishmanials. Further work will be necessary to improve selectivity of these compounds against the parasite.

A comparative study using different proportions of CeO2/C (4%, 9% and 13% CeO2) was performed to produce H2O2, a reagent used in the oxidation of organic pollutants and in electro-Fenton reactions for the production of the hydroxyl radical (OH center dot), a strong oxidant agent used in the electrochemical treatment of aqueous wastewater. The CeO2/C materials were prepared by a modified polymeric precursor method (PPM). X-ray diffraction analysis of the CeO2/C prepared by the PPM identified two phases. CeO2 and CeO2. The average size of the crystallites in these materials was close to 7 nm. The kinetics of the oxygen reduction reaction (ORR) were evaluated by the rotating ring-disk electrode technique. The results showed that the 4% CeO2/C prepared by the PPM was the best composite for the production of H2O2 in a 1 mol L-1 NaOH electrolyte solution. For this material, the number of electrons transferred and the H2O2 percentage efficiency were 3.1 and 44%, respectively. The ring-current of the 4% CeO2/C was higher than that of Vulcan carbon, the reference material for H2O2 production, which produced 41% H2O2 and transferred 3.1 electrons per molecule of oxygen. The overpotential for this reaction on the ceria-based catalyst was substantially lower (approximately 200 mV), demonstrating the higher catalytic performance of this material. Gas diffusion electrodes (GDE) containing the catalyst were used to evaluate the real amount of H2O2 produced during exhaustive electrolysis. The 4% CeO2/C GDE produced 871 mg L-1 of H2O2, whereas the Vulcan carbon GDE produced a maximum amount of only 407 mg L-1. Thus, the 4% CeO2/C electrocatalyst prepared by the PPM is a promising material for H2O2 electrogeneration in alkaline media. (C) 2011 Elsevier B.V. All rights reserved.

The putrescine analogue 1,4-diamino-2-butanone (DAB) is highly toxic to various microorganisms, including Trypanosoma cruzi. Similar to other a-aminocarbonyl metabolites. DAB exhibits pro-oxidant properties. DAB undergoes metal-catalyzed oxidation yielding H2O2, NH4+ ion, and a highly toxic alpha-oxoaldehyde. In vitro. DAB decreases mammalian cell viability associated with changes in redox balance. Here, we aim to clarify the DAB pro-oxidant effects on trypomastigotes and on intracellular T. cruzi amastigotes. DAB (0.05-5 mM) exposure in trypomastigotes, the infective stage of T. cruzi, leads to a decline in parasite viability (IC50 c.a. 0.2 mM DAB; 4 h incubation), changes in morphology, thiol redox imbalance, and increased TcSOD activity. Medium supplementation with catalase (2.5 mu M) protects trypomastigotes against DAB toxicity, while host cell invasion by trypomastigotes is hampered by DAB. Additionally, intracellular amastigotes are susceptible to DAB toxicity. Furthermore, pre-treatment with 100-500 mu M buthionine sulfoximine (BSO) of LLC-MK2 potentiates DAB cytotoxicity, whereas 5 mM N-acetyl-cysteine (NAC) protects cells from oxidative stress. Together, these data support the hypothesis that redox imbalance contributes to DAB cytotoxicity in both T. cruzi and mammalian host cells. (C) 2012 Elsevier Inc. All rights reserved.

This work reports the development of GDE for electrogeneration of H2O2 and their application in the degradation process of Reactive Blue 19 dye. GDE produced by carbon black with 20% polytetrafluoroethylene generated up to 500 mg L-1 of H2O2 through the electrolysis of acidic medium at -0.8 V vs Ag/AgCl. Reactive Blue 19 dye was degraded most efficiently with H2O2 electrogenerated in the presence of Fe(II) ions, leading to removal of 95% of the original color and 39% of TOC at -0.8 V vs Ag/AgCl.

The aim of the present thesis, carried out at the Analytical Group of the Faculty of Industrial Chemistry in Bologna, is to develop a new electrochemical method for the determination of the Antioxidant Capacity (AOC). The approach is based on the deposition of a non-conducting polymeric film on the working electrode surface and the following exposition to the radicals OH· produced by H2O2 photolysis. The strongly oxidant action of hydroxyl radicals degrades, causing an increase of the Faradic current, relevant to the redox couple [Ru(NH3)6]2+/3+ monitored by cyclic voltammetry(CV); the presence of an antioxidant compound in solution slows down the radical action, thus protecting the polymeric film and blocking the charge transfer. The parameter adopted for the quantification of the AOC, was the induction time, called also lag phase, which is the time when the degradation of the film starts. Five pure compounds, among most commonly antioxidant, were investigated : Trolox®(an analogue water-soluble of vitamin E), (L)-ascorbic acid, gallic acid, pyrogallol and (-)- epicatechin. The AOC of each antioxidant was expressed by TEAC index (Trolox® Equivalent Antioxidant Capacity), calculated from the ratio between the slope of the calibration curve of the target compound and the slope of the calibration curve of Trolox®. The results from the electrochemical method, have been compared with those obtained from some other standardized methods, widely employed. The assays used for the comparison, have been: ORAC, a spectrofluorimetric method based on the decrease of fluorescein emission after the attack of alkylperoxide radicals, ABTS and DPPH that exploit the decoloration of stable nitrogen radicals when they are reduced in presence of an antioxidant compound and, finally, a potentiometric method based on the response of the redox couple [Fe(CN)6]3-/ [Fe(CN)6]4-. From the results obtained from pure compounds, it has been found that ORAC is the methodology showing the best correlation with the developed electrochemical method, maybe since similar radical species are involved. The comparison between the considered assays, was also extended to the analysis of a real sample of fruit juice. In such a case the TEAC value resulting from the electrochemical method is higher than those from standardized assays.

Hydrogen peroxide (H2O2) is a powerful oxidant which is commonly used in a wide range of applications in the industrial field. Several methods for the quantification of H2O2 have been developed. Among them, electrochemical methods exploit the ability of some hexacyanoferrates (such as Prussian Blue) to detect H2O2 at potentials close to 0.0 V (vs. SCE) avoiding the occurrence of secondary reactions, which are likely to run at large overpotentials. This electrocatalytic behaviour makes hexacyanoferrates excellent redox mediators. When deposited in the form of thin films on the electrode surfaces, they can be employed in the fabrication of sensors and biosensors, normally operated in solutions at pH values close to physiological ones. As hexacyanoferrates show limited stability in not strongly acidic solutions, it is necessary to improve the configuration of the modified electrodes to increase the stability of the films. In this thesis work, organic conducting polymers were used to fabricate composite films with Prussian Blue (PB) to be electro-deposited on Pt surfaces, in order to increase their pH stability. Different electrode configurations and different methods of synthesis of both components were tested, and for each one the achievement of a possible increase in the operational stability of Prussian Blue was verified. Good results were obtained for the polymer 3,3''-didodecyl-2,2':5',2''-terthiophene (poly(3,3''-DDTT)), whose presence created a favourable microenvironment for the electrodeposition of Prussian Blue. The electrochemical behaviour of the modified electrodes was studied in both aqueous and organic solutions. Poly(3,3''-DDTT) showed no response in aqueous solution in the potential range where PB is electroactive, thus in buffered aqueous solution is was possible to characterize the composite material, focusing only on the redox behaviour of PB. A combined effect of anion and cation of the supporting electrolyte was noticed. The response of Pt electrodes modified with films of the PB /poly(3,3''-DDTT) composite was evaluated for the determination of H2O2. The performance of such films was found better than that of the PB alone. It can be concluded that poly(3,3''-DDTT) plays a key role in the stabilization of Prussian Blue causing also a wider linearity range for the electrocatalytic response to H2O2.

Ozon (O3) ist in der Atmosphäre ein wichtiges Oxidanz und Treibhausgas. Während die höchsten Konzentrationen in der Stratosphäre beobachtet werden und die vor der gefährlichen UV-Strahlung schützende Ozonschicht bilden, können sich signifikante Änderungen der Ozon-Konzentration in der Region der Tropopause auf das Klima der Erde auswirken. Des Weiteren ist Ozon eine der Hauptquellen für das Hydroxylradikal (OH) und nimmt damit entscheidend Einfluss auf die Oxidationskraft der Atmosphäre. Der konvektive Transport von Ozon und seinen Vorläufergasen aus Regionen nahe der Erdoberfläche in die freie Troposphäre beeinflusst das Budget dieser Spezies in der Tropopausenregion.rnDie Datengrundlage der Studie in der vorliegenden Arbeit basiert auf den flugzeuggetragenen Messkampagnen GABRIEL 2005 (Suriname, Südamerika) sowie HOOVER I 2006 und HOOVER II 2007 (beide in Europa). Mit dem zur Verfügung stehenden Datensatz wird das Ozonbudget in der freien, unbelasteten Hintergrundatmosphäre und in der durch hochreichende Konvektion gestörten, oberen Troposphäre untersucht. Anhand der auf in-situ Messungen von O3, NO, OH, HO2 und dem aktinischen Strahlungsfluss basierten Berechnung der Netto-Ozonproduktionsrate (NOPR) werden für das Messgebiet Ozontendenzen in der unbelasteten Troposphäre abgeleitet und mit Simulationen des globalen Chemie-Transport-Modells MATCH-MPIC verglichen. Mit Hilfe zweier Fallstudien in den Tropen in Südamerika und den mittleren Breiten in Europa werden die Auswirkungen von hochreichender Konvektion auf die obere Troposphäre quantifiziert.rnDie Ergebnisse zeigen für die Grenzschicht in niedrigen und mittleren Breiten eine eindeutige Tendenz zur Produktion von Ozon, was für den tropischen Regenwald in der Messregion nicht der allgemeinen Erwartung entsprach, nach der diese Region durch die Zerstörung von Ozon charakterisiert sein sollte. In der oberen Troposphäre ab etwa 7 km wird für die beiden Regionen eine leichte Tendenz zur Ozonproduktion beobachtet. Signifikante Unterschiede zeigen die Ergebnisse für die mittlere Troposphäre. Während die Tropen in dieser Region durch eine eindeutige Tendenz zur Zerstörung von Ozon charakterisiert sind, lässt sich über den mittleren Breiten zwar eine hohe photochemische Aktivität aber keine derart klare Tendenz feststellen. Die hohen Breiten zeichnen sich durch eine neutrale Troposphäre in Bezug auf die Ozontendenz aus und weisen kaum photochemische Aktivität auf. Der Vergleich dieser Ergebnisse mit dem MATCH-MPIC Modell zeigt in weiten Teilen der Messregionen eine grundlegende Übereinstimmung in der Tendenz zur Produktion oder Zerstörung von Ozon. Die absoluten Werte werden vom Modell aber generell unterschätzt. Signifikante Unterschiede zwischen in-situ Daten und Modellsimulationen werden in der Grenzschicht über dem tropischen Regenwald identifiziert.rnDer Einfluss der Konvektion ist durch eine signifikant erhöhte NOPR gekennzeichnet. In dieser Arbeit wird in den Tropen mit einem Median-Wert von 0.20 ppbv h−1 eine um den Faktor 3.6 erhöhte NOPR im Vergleich zur ungestörten oberen Troposphäre abgeschätzt. In den mittleren Breiten führt die um eine Größenordnung höhere NO-Konzentration zu einem Wert von 1.89 ppbv h−1, was einer Überhöhung um einen Faktor 6.5 im Vergleich zum ungestörten Zustand entspricht. Diese Ergebnisse zeigen für beide Regionen in der oberen Troposphäre eine erhöhte Ozonproduktion als Folge konvektiver Aktivität. rnrnHochreichende Konvektion ist zudem ein sehr effektiver Mechanismus für den Vertikaltransport aus der Grenzschicht in die obere Troposphäre. Die schnelle Hebung in konvektiven Wolken führt bei Spurengasen mit Quellen an der Erdoberfläche zu einer Erhöhung ihrer Konzentration in der oberen Troposphäre. Die hochgradig löslichen Spurenstoffe Formaldehyd (HCHO) und Wasserstoffperoxid (H2O2) sind wichtige Vorläufergase der HOx-Radikale. Es wird angenommen, dass sie aufgrund ihrer Löslichkeit in Gewitterwolken effektiv ausgewaschen werden.rnIn der vorliegenden Arbeit wird eine Fallstudie von hochreichender Konvektion im Rahmen des HOOVER II Projekts im Sommer 2007 analysiert. Am 19.07.2007 entwickelten sich am Nachmittag am Südostrand eines in nordöstlicher Richtung ziehenden mesoskaligen konvektiven Systems drei zunächst isolierte konvektive Zellen. Flugzeuggetragene Messungen in der Aus- und der Einströmregion einer dieser Gewitterzellen stellen einen exzellenten Datensatz bereit, um die Auswirkungen von hochreichender Konvektion auf die Verteilung verschiedener Spurengase in der oberen Troposphäre zu untersuchen. Der Vergleich der Konzentrationen von Kohlenstoffmonoxid (CO) und Methan (CH4) zwischen der oberen Troposphäre und der Grenzschicht deutet auf einen nahezu unverdünnten Transport dieser langlebigen Spezies in der konvektiven Zelle hin. Die Verhältnisse betragen (0.94±0.04) für CO und (0.99±0.01) für CH4. Für die löslichen Spezies HCHO und H2O2 beträgt dieses Verhältnis in der Ausströmregion (0.55±0.09) bzw. (0.61±0.08). Dies ist ein Indiz dafür, dass diese Spezies nicht so effektiv ausgewaschen werden wie angenommen. Zum besseren Verständnis des Einflusses der Konvektion auf die Budgets dieser Spezies in der oberen Troposphäre wurden im Rahmen dieser Arbeit Boxmodell-Studien für den Beitrag der photochemischen Produktion in der Ausströmregion durchgeführt, wobei die gemessenen Spezies und Photolysefrequenzen als Randbedingungen dienten. Aus den Budgetbetrachtungen für HCHO und H2O2 wird eine Auswascheffizienz von (67±24) % für HCHO und (41±18) % für H2O2 abgeschätzt. Das für H2O2 überraschende Ergebnis lässt darauf schließen, dass dieses Molekül in einer Gewitterwolke deutlich effektiver transportiert werden kann, als aufgrund seiner hohen Löslichkeit aus der Henry-Konstanten zu erwarten wäre. Das Ausgasen von gelöstem H2O2 beim Gefrieren eines Wolkentropfens, d.h. ein Retentionskoeffizient kleiner 1, ist ein möglicher Mechanismus, der zum beobachteten Mischungsverhältnis dieser löslichen Spezies in der Ausströmregion beitragen kann.

CD40 and its ligand regulate pleiotropic biological responses, including cell proliferation, differentiation, and apoptosis. In many inflammatory lung diseases, tissue damage by environmental or endogenous oxidants plays a major role in disease pathogenesis. As the epithelial barrier is a major target for these oxidants, we postulated that CD40, the expression of which is increased in asthma, plays a role in the regulation of apoptosis of bronchial epithelial cells exposed to oxidants. Using 16HBE 14o- cells exposed to oxidant stress, we found that ligation of CD40 (induced by G28-5 monoclonal antibodies) enhanced cell survival and increased the number of cells in G2/M (interphase between DNA synthesis and mitosis) of the cell cycle. This was associated with NF-kappaB and activator protein-1 activation and increased expression of the inhibitor of apoptosis, c-IAP1. However, oxidant stress-induced apoptosis was found to be caspase- and calpain-independent implicating CD40 ligation as a regulator of caspase-independent cell death. This was confirmed by the demonstration that CD40 ligation prevented mitochondrial release and nuclear translocation of apoptosis inducing factor. In conclusion, we demonstrate a novel role for CD40 as a regulator of epithelial cell survival against oxidant stress. Furthermore, we have identified, for the first time, an endogenous inhibitory pathway of caspase-independent cell death.

BACKGROUND: Rapamycines, sirolimus (SRL) and everolimus (ERL), are proliferation signal inhibitors (PSIs). PSI therapy often leads to edema. We hypothesized that increased oxidative stress in response to PSIs may modulate the expression of vascular endothelial (VE)-cadherin on endothelial cells (ECs) and, subsequently, vascular permeability, which in turn may be involved in the development of edema. METHODS: Experiments were performed on human umbilical vein ECs (HUVECs). Oxidative stress was measured by dichlorofluorescein-diacetate. Expression of VE-cadherin was evaluated by immunofluorescent staining and western blot analysis. Endothelial "permeability" was assessed using a transwell model. RESULTS: SRL and ERL, at concentrations of 1, 10 and 100 nmol/liter, enhanced oxidative stress (SRL: 24 +/- 12%, 29 +/- 9%, 41 +/- 13% [p < 0.05, in all three cases]; ERL: 13 +/- 10%, 27 +/- 2%, 40 +/- 12% [p < 0.05, in the latter two cases], respectively) on HUVECs, which was inhibited by the anti-oxidant, N-acetyl-cysteine (NAC) and, to a lesser extent, by the specific inhibitor of nitric oxide synthase, N-Omega-nitro-L-arginine methylester. By the use of NAC, VE-cadherin expression remained comparable with control, according to both immunocytochemistry and western blot analysis. Permeability was significantly increased by SRL and ERL at 100 nmol/liter (29.5 +/- 6.4% and 33.8 +/- 4.2%, respectively); however, co-treatment with NAC abrogated the increased permeability. CONCLUSIONS: EC homeostasis, as indicated by VE-cadherin expression, may be damaged by SRL and ERL, but resolved by the anti-oxidant NAC.

Ice core measurements (H2O2 and CH4/HCHO) and modeling studies indicate a change in the oxidation capacity of the atmosphere since the onset of the Industrial Revolution due to increases in fossil fuel burning emissions [e. g., Lelieveld et al., 2002; Hauglustaine and Brasseur, 2001; Wang and Jacob, 1998; Staffelbach et al., 1991]. The mass-independent fractionation (MIF) in the oxygen isotopes of sulfate and nitrate from a Greenland ice core reveal that biomass-burning events in North America just prior to the Industrial Revolution significantly impacted the oxidation pathways of sulfur and nitrogen species deposited in Greenland ice. This finding highlights the importance of biomass-burning emissions for atmospheric chemistry in preindustrial North America and warrants the inclusion of this impact in modeling studies estimating changes in atmospheric oxidant chemistry since the Industrial Revolution, particularly when using paleo-oxidant data as a reference for model evaluation.

The γ-aminobutyric acid benzodiazepine (GABAA /BZDR) ionophore complex has been widely studied in the central nervous system (CNS) and it regulates Cl− ion movement across the plasma membrane. The complex has been found in the distal tubule and the thick ascending limb of the kidney. The goal of this study was to see if modulation of this complex by agonists or antagonists could affect the way Madin-Darby Canine Kidney (MDCK) cells responded to an oxidant stress induced by menadione. When compared to cells incubated with menadione alone, preincubation with lindane, a nonspecific GABAA antagonist, coincubation with bicuculline, a specific GABAA antagonist, and coincubation with FG7142, an inverse agonist for the BZDR, protected cells from menadione cytotoxicity. Preincubation of cells in media containing PK11195 had no effect on menadione cytotoxicity. Coincubation with flurazepam, a BZDR agonist, exacerbated menadione cytotoxicity. This suggests that modulation of the GABAA/BZDR ionophore complex within MDCK cells with agonists and antagonists can alter the cellular responsiveness to an oxidant-induced injury. These responses via agonists and antagonists may be due to alterations of Cl− ion influx during late stage necrotic cell death. ^

Studies have demonstrated a variable response to ozone among individuals and animal species and strains. For instance, C57BL/6J mice have a greater inflammatory response to ozone exposure than C3H/HeJ mice. In these studies, I utilized these strain differences in an effort to derive a mechanistic explanation to the variable strain sensitivity to ozone exposure. Therefore, alveolar macrophages (AM) from C57BL/6J and C3H/HeJ mice were exposed in vitro to hydrogen peroxide ($\rm H\sb2O\sb2$), heat and acetyl ceramide or in vivo to ozone. Necrosis and DNA fragmentation in macrophages from the two murine strains were determined to assess cytotoxicity following these treatments. In addition, synthesis and expression of the stress proteins, stress protein 72 (SP72) and heme oxygenase (HO-1), were examined following treatments. The in vitro experiments were conducted to eliminate the possibility of in vivo confounders (i.e., differences in breathing rates in the two strains) and thus directly implicate some inherent difference between cells from the two murine strains. $\rm H\sb2O\sb2$ and heat caused greater cytotoxicity in AM from C57BL/6J than C3H/HeJ mice and DNA fragmentation was a particularly sensitive indicator of cell injury. Similarly, AM from C57BL/6J mice were more sensitive to ozone exposure than cells from C3H/HeJ mice. Exposure to either 1 or 0.4 ppm ozone caused greater cytotoxicity in macrophages from C57BL/6J mice compared to macrophages from C3H/HeJ mice. The increased sensitivity of AM to injury was associated with decreased synthesis and expression of stress proteins. AM from C57BL/6J mice synthesized and expressed significantly less stress proteins in response to heat and ozone than AM from C3H/HeJ mice. Heat treatment resulted in greater synthesis and expression of SP72. In addition, macrophages from C57BL/6J mice expressed lower amounts of HO-1 than macrophages from C3H/HeJ mice following 0.4 ppm ozone exposure. Therefore, AM from C57BL/6J mice are more susceptible to oxidative injury than AM from C3H/HeJ mice which might be due to differential expression of stress proteins in these cells. ^
