Objetivou-se estudar o comportamento de embriões zigóticos e nucelares aos 120, 130, 140 e 150 dias após serem efetuadas hibridações controladas entre a laranjeira 'Natal' (Citrus sinensis Osb.) e o parental masculino Poncirus trifoliata (L.) Raf. Em cada data, as sementes foram removidas, e os embriões excisados foram caracterizados em estádios de desenvolvimento (globular, cordiforme, torpedo e cotiledonar); coloração (clorofilado ou não); e localização na semente (próximo à micrópila ou mais interiormente). A partir dessas características, foram construídas tabelas de contingência para testar hipóteses de independência entre elas, mediante o teste exato de Fisher e chi² (qui-quadrado). Relações de dependência foram verificadas entre as características: estádios de desenvolvimento embrionário com a localização na semente; estádios de desenvolvimento embrionário com a natureza da plântula (zigótica ou nucelar); e entre natureza da plântula com a localização do embrião na semente. Verificou-se que os embriões zigóticos excisados de frutos com 130 a 150 dias da hibridação controlada, localizam-se, em grande maioria, próximos à região micropilar da semente, em estádio globular e cordiforme de desenvolvimento.

The number of existing protein sequences spans a very small fraction of sequence space. Natural proteins have overcome a strong negative selective pressure to avoid the formation of insoluble aggregates. Stably folded globular proteins and intrinsically disordered proteins (IDP) use alternative solutions to the aggregation problem. While in globular proteins folding minimizes the access to aggregation prone regions IDPs on average display large exposed contact areas. Here, we introduce the concept of average meta-structure correlation map to analyze sequence space. Using this novel conceptual view we show that representative ensembles of folded and ID proteins show distinct characteristics and responds differently to sequence randomization. By studying the way evolutionary constraints act on IDPs to disable a negative function (aggregation) we might gain insight into the mechanisms by which function - enabling information is encoded in IDPs.

Alcoholic liver disease is mediated via activation of TLR4 signaling; MyD88-dependent and -independent signals are important contributors to injury in mouse models. Adiponectin, an anti-inflammatory adipokine, suppresses TLR4/MyD88-dependent responses via induction of heme oxygenase-1 (HO-1). Here we investigated the interactions between chronic ethanol, adiponectin, and HO-1 in regulation of TLR4/MyD88-independent signaling in macrophages and an in vivo mouse model. After chronic ethanol feeding, LPS-stimulated expression of IFN-β and CXCL10 mRNA was increased in primary cultures of Kupffer cells compared with pair-fed control mice. Treatment of Kupffer cells with globular adiponectin (gAcrp) normalized this response. LPS-stimulated IFN-β/CXCL10 mRNA and CXCL10 protein was also reduced in RAW 264.7 macrophages treated with gAcrp or full-length adiponectin. gAcrp and full-length adiponectin acted via adiponectin receptors 1 and 2, respectively. gAcrp decreased TLR4 expression in both Kupffer cells and RAW 264.7 macrophages. Small interfering RNA knockdown of HO-1 or inhibition of HO-1 activity with zinc protoporphyrin blocked these effects of gAcrp. C57BL/6 mice were exposed to chronic ethanol feeding, with or without treatment with cobalt protoporphyrin, to induce HO-1. After chronic ethanol feeding, mice were sensitized to in vivo challenge with LPS, expressing increased IFN-β/CXCL10 mRNA and CXCL10 protein in liver compared with control mice. Pretreatment with cobalt protoporphyrin 24 h before LPS challenge normalized this effect of ethanol. Adiponectin and induction of HO-1 potently suppressed TLR4-dependent/MyD88-independent cytokine expression in primary Kupffer cells from rats and in mouse liver after chronic ethanol exposure. These data suggest that induction of HO-1 may be a useful therapeutic strategy in alcoholic liver disease.

The deduced amino acid sequence of Leishmania major sw3 cDNA reveals the presence of characteristic histone H1 amino acid motifs. However, the open reading frame is of an unusually small size for histone H1 (105 amino acids) because it lacks the coding potential for the central hydrophobic globular domain of linker histones present in other eukaryotes. Here, we provide biochemical evidence that the SW3 protein is indeed a L. major nuclear histone H1, and that it is differentially expressed during the life cycle of the parasite. Due to its high lysine content, the SW3 protein can be purified to a high degree from L. major nuclear lysates with 5% perchloric acid, a histone H1 preparative method. Using an anti-SW3 antibody, this protein is detected as a 17 kDa or as a 17/19 kDa doublet in the nuclear subfraction in different L. major strains. The nuclear localization of the SW3 protein is further supported by immunofluorescence studies. During in vitro promastigote growth, both the sw3 cytoplasmic mRNA and its protein progressively accumulate within parasites from early log phase to stationary phase. Within amastigotes, the high level of H1 expression is maintained but decreases when amastigotes differentiate into promastigotes. Together, these observations suggest that the different levels of this histone H1 protein could influence the varying degrees of chromatin condensation during the life-cycle of the parasite, and provide us with tools to study this mechanism.

L’objectiu d’aquest estudi és plantejar un estat de la qüestió sobre la producció vinària i d’àmfores en època romana a l’àrea de l’ager Tarraconensis i les terres de l’Ebre (possiblement corresponents al territorium de Dertosa), intentant posar en relació el testimoni arqueològic sobre els elements de transformació agrícola (premses, dipòsits) i la producció i distribució de les àmfores. S’ha pogut documentar una producció primerenca d’àmfores de la forma Dressel 1 en època tardorepublicana, reduïda pel que ara sabem a la zona de l’Alt Camp, i potser també present a la zona de Dertosa (forn del Mas d’Aragó). Tanmateix, la producció més important correspon a l’època altimperial. En època d’August es documenta l’aparició dels tallers de l’ager Dertosanus, així com a la part oriental de l’ager Tarraconensis (Darró, el Vilarenc) i al taller de la Canaleta (Vila-seca), on es produïren àmfores de la forma Oberaden 74. Tanmateix, fou en plena època julioclàudia quan les figlinae de l’àrea del Baix Camp es posaren en marxa, amb la producció d’àmfores de les formes Dressel 2-4 i 7-11. Aquesta producció es relaciona amb el vi de Tarraco al qual fan referència les fonts escrites llatines, i arqueològicament sembla que es pot perllongar almenys fins a final del segle II i inici del III.

Pesquisou-se o efeito do estádio de desenvolvimento de embriões resgatados na obtenção de plantas em cruzamentos entre genitores apirenos de videira, utilizando dois métodos de cultivo in vitro de sementes-traço. No método 1 (M1), as sementes-traço foram cultivadas 60 dias em meio-de-cultura ER e no método 2 (M2) 60 dias em ER, mais 30 dias em meio-de-cultura MS. O estádio de desenvolvimento do embrião foi identificado no final destes períodos, quando do resgate e transferência para meio-de-cultura WP. Quatro tipos de embriões foram identificados: globular, cordiforme, torpedo e indefinido. O estádio globular foi encontrado com maior freqüência, levando à obtenção de maior número de plantas, mesmo tendo apresentado menor capacidade de gerar plantas. A maior eficiência em gerar plantas foi obtida com o estádio torpedo. No M2, ocorreu maior número de embriões germinados e de plantas desenvolvidas. Como o estádio de desenvolvimento do embrião influencia na obtenção de plantas, o resgate de embriões em estádio mais avançado do seu desenvolvimento leva a tem maior capacidade de gerar plantas. Portanto, uso de técnicas que favoreçam o resgate de embriões mais desenvolvidos pode aumentar o número de plantas obtidas.

Ja fa uns anys des que J. Miró va publicar el seu llibre sobre les àmfores de la Tarraconense; des d’aleshores noves proves han canviat el panorama d’aquestes àmfores romanes catalanes. El present article tracta d’actualitzar el nostre coneixement actual sobre la pauta de consum i de distribució d’aquests envasos. Nous mètodes i dades quantificades i ben datades (per exemple, segells i fragments d’àmfora) ens permeten reconsiderar la distribució d’aquesta àmfora per l’occident de l’Imperi romà.

In this thesis different parameters influencing critical flux in protein ultrafiltration and membrane foul-ing were studied. Short reviews of proteins, cross-flow ultrafiltration, flux decline and criticalflux and the basic theory of Partial Least Square analysis (PLS) are given at the beginning. The experiments were mainly performed using dilute solutions of globular proteins, commercial polymeric membranes and laboratory scale apparatuses. Fouling was studied by flux, streaming potential and FTIR-ATR measurements. Critical flux was evaluated by different kinds of stepwise procedures and by both con-stant pressure and constant flux methods. The critical flux was affected by transmembrane pressure, flow velocity, protein concentration, mem-brane hydrophobicity and protein and membrane charges. Generally, the lowest critical fluxes were obtained at the isoelectric points of the protein and the highest in the presence of electrostatic repulsion between the membrane surface and the protein molecules. In the laminar flow regime the critical flux increased with flow velocity, but not any more above this region. An increase in concentration de-creased the critical flux. Hydrophobic membranes showed fouling in all charge conditionsand, furthermore, especially at the beginning of the experiment even at very low transmembrane pressures. Fouling of these membranes was thought to be due to protein adsorption by hydrophobic interactions. The hydrophilic membranes used suffered more from reversible fouling and concentration polarisation than from irreversible foul-ing. They became fouled at higher transmembrane pressures becauseof pore blocking. In this thesis some new aspects on critical flux are presented that are important for ultrafiltration and fractionation of proteins.

O objetivo deste trabalho foi o estabelecimento de um protocolo para a embriogênese somática do caquizeiro. Como explantes, foram utilizados embriões zigóticos em diversos estádios de desenvolvimento, retirados de frutos coletados de plantas adultas a campo, a partir de quatro semanas após o pleno florescimento até 22 semanas. O meio básico para os experimentos foi o MS½NO3. O meio inicial de indução foi suplementado com 20µM de 2,4-D e 2µM de cinetina. Os calos escuros obtidos foram repicados para outro meio de indução, com concentrações 10 ou 20 µM de 2,4-D e 2 µM de cinetina. Os calos com massas pró-embriogênicas obtidos foram transferidos para meio de manutenção e multiplicação com 2 µM de cinetina e 2,4-D nas concentrações 2,5; 5,0 e 10 µM. As massas embriogênicas formadas foram transferidas para meio de maturação suplementado com 0,5 µM de AIB e as concentrações 5; 10 e 20 µM de 2-iP. Os embriões formados foram isolados em dois meios de conversão, sendo o primeiro com 5 µM de 2-iP, 5 µM de AG3 e 0,5 µM de AIB e o segundo com 0,5 µM de AG3 e BAP, nas concentrações 0; 0,25; 0,5 e 1,0 µM. Como resultados, obteve-se o padrão indireto de embriogênese somática a partir de embriões zigóticos maduros, com mais de 22 semanas de formação, quando cultivados em meio de cultura com 10 µM de 2,4-D combinado com 2 µM de cinetina. A manutenção e a multiplicação das culturas embriogênicas foram mais eficientes com 5 µM de 2,4-D, na qual os pró-embriões avançaram para o estádio globular. Na fase de maturação, as concentrações de 2-iP testadas atuaram promovendo os embriões globulares a estádios mais avançados da ontogenia como cordiforme, torpedo e cotiledonar. A suplementação do meio de cultura com 1 µM de BAP e 0,5 µM de AG3 gerou a formação de plantas mais desenvolvidas, com maior número e tamanho de folhas.

The scolex of the bothriocephalidean cestode Clestobothrium crassiceps was studied by means of scanning electron microscopy (SEM). The comparative results of various fixation procedures and techniques are presented. The scolex of C. crassiceps is oval to globular and exhibits two deep bothria which appear in the form of two lobes separated by a longitudinal groove. At the apex of the scolex, resembling a beret, an apical disc is present (oval, flattened and with a sinuous edge). Our results are compared with those previously reported in other species of Clestobothrium. This study represents the first report which highlights the presence of an apical disc in the scolex of C. crassiceps. It describes the effects of different procedures applied to our material during preparation and a comparative analysis results obtained using these various methods.

Measles, caused by measles virus (MV), is a highly contagious viral disease causing severe respiratory infection and a typical rash. Despite the availability of a protective vaccine, measles is still the leading vaccine-preventable cause of childhood mortality worldwide. The high mortality associated with the disease is mainly due to an increased susceptibility to secondary infections during the period of immunosuppression that continues for several weeks after recovery. The present study was undertaken to elucidate the role of cytoskeletal components in the regulation of MV infection. The most interesting finding was that MV replication was activated in unstimulated peripheral blood mononuclear cells (PBMC) when globular actin was converted into the filamentous form with jasplakinolide. This provides a new aspect in our understanding of MV infection in PBMC. In the second part of the thesis we investigated MV-induced structural changes of cellular nuclear matrix, which is a proteinaceous framework of the nucleus similar to the cytoskeleton in the cytoplasm. We showed that cleavage of nuclear markers was virusspecific and a general caspase inhibitor rescued MV-infected cells from cell death. Furthermore, we studied MV-induced innate immune mechanisms in lung epithelial and endothelial cells. Our results showed that MV infection resulted in activation of the double stranded RNA (dsRNA) binding molecules melanoma differentiation-associated gene 5 (mda-5), retinoic acid inducible gene I (RIG-I), and toll-like receptor 3 (TLR3) gene expression, followed by high expression of antiviral cytokine mRNA.

The CO2-laser-MAG hybrid welding process has been shown to be a productive choice for the welding industry, being used in e.g. the shipbuilding, pipe and beam manufacturing, and automotive industries. It provides an opportunity to increase the productivity of welding of joints containing air gaps compared with autogenous laser beam welding, with associated reductions in distortion and marked increases in welding speeds and penetration in comparison with both arc and autogenous laser welding. The literature study indicated that the phenomena of laser hybrid welding are mostly being studied using bead-on-plate welding or zero air gap configurations. This study shows it very clearly that the CO2 laser-MAG hybrid welding process is completely different, when there is a groove with an air gap. As in case of industrial use it is excepted that welding is performed for non-zero grooves, this study is of great importance for industrial applications. The results of this study indicate that by using a 6 kW CO2 laser-MAG hybrid welding process, the welding speed may also be increased if an air gap is present in the joint. Experimental trials indicated that the welding speed may be increased by 30-82% when compared with bead-on-plate welding, or welding of a joint with no air gap i.e. a joint prepared as optimum for autogenous laser welding. This study demonstrates very clearly, that the separation of the different processes, as well as the relative configurations of the processes (arc leading or trailing) affect welding performance significantly. These matters influence the droplet size and therefore the metal transfer mode, which in turn determined the resulting weld quality and the ability to bridge air gaps. Welding in bead-onplate mode, or of an I butt joint containing no air gap joint is facilitated by using a leading torch. This is due to the preheating effect of the arc, which increases the absorptivity of the work piece to the laser beam, enabling greater penetration and the use of higher welding speeds. With an air gap present, air gap bridging is more effectively achieved by using a trailing torch because of the lower arc power needed, the wider arc, and the movement of droplets predominantly towards the joint edges. The experiments showed, that the mode of metal transfer has a marked effect on gap bridgeability. Transfer of a single droplet per arc pulse may not be desirable if an air gap is present, because most of the droplets are directed towards the middle of the joint where no base material is present. In such cases, undercut is observed. Pulsed globular and rotational metal transfer modes enable molten metal to also be transferred to the joint edges, and are therefore superior metal transfer modes when bridging air gaps. It was also found very obvious, that process separation is an important factor in gap bridgeability. If process separation is too large, the resulting weld often exhibits sagging, or no weld may be formed at all as a result of the reduced interaction between the component processes. In contrast, if the processes are too close to one another, the processing region contains excess molten metal that may create difficulties for the keyhole to remain open. When the distance is optimised - i.e. a separation of 0-4 mm in this study, depending on the welding speed and beam-arc configuration - the processes act together, creating beneficial synergistic effects. The optimum process separation when using a trailing torch was found to be shorter (0-2 mm) than when a leading torch is used (2-4 mm); a result of the facilitation of weld pool motion when the latter configuration is adopted. This study demonstrates, that the MAG process used has a strong effect on the CO2-laser-MAG hybrid welding process. The laser beam welding component is relatively stable and easy to manage, with only two principal processing parameters (power and welding speed) needing to be adjusted. In contrast, the MAG process has a large number of processing parameters to optimise, all of which play an important role in the interaction between the laser beam and the arc. The parameters used for traditional MAG welding are often not optimal in achieving the most appropriate mode of metal transfer, and weld quality in laser hybrid welding, and must be optimised if the full range of benefits provided by hybrid welding are to be realised.

[cat] Es donen a conèixer diversos objectes singulars localitzats en superfície en els jaciments ibèric i romà de la Fogonussa, que romanien inèdits en la població de Sant Martí de Maldà, municipi de Sant Martí de Riucorb. La selecció està formada per una vora d'àmfora grecoitàlica tardana, que conté una inscripció ibèrica, dues nanses de vasos contenidors ibèrics amb grafits i tres nanses d'àmfora ibèrica estampillades, una d'elles amb la figura d'un griu.

Herein, we report the formation of organized mesoporous silica materials prepared from a novel nonionic gemini surfactant, myristoyl-end capped Jeffamine, synthesized from a polyoxyalkyleneamine (ED900). The behavior of the modified Jeffamine in water was first investigated. A direct micellar phase (L1) and a hexagonal (H1) liquid crystal were found. The structure of the micelles was investigated from the SAXS and the analysis by Generalized Indirect Fourier Transformation (GIFT), which show that the particles are globular of coreshell type. The myristoyl chains, located at the ends of the amphiphile molecule are assembled to form the core of the micelles and, as a consequence, the molecules are folded over on themselves. Mesoporous materials were then synthesized from the self-assembly mechanism. The recovered materials were characterized by SAXS measurements, nitrogen adsorptiondesorption analysis, transmission and scanning electron microscopy. The results clearly evidence that by modifying the synthesis parameters, such as the surfactant/silica precursor molar ratio and the hydrothermal conditions, one can control the size and the nanostructuring of the resulting material. It was observed that, the lower the temperature of the hydrothermal treatment, the better the mesopore ordering.
