Four aspects of horizontal genetic transfer during heterokaryon formation were examined in the asexual pathogen Fusarium oxysporum f.sp. cubense (Foc): (1) variability based on method of heterokaryon formation; (2) differences in nuclear and mitochondrial inheritance; (3) the occurrence of recombination without nuclear fusion; (4) the occurrence of horizontal genetic transfer between distantly related isolates. The use of non-pathogenic strains of Fusarium oxysporum as biocontrol agents warrants a closer examination at the reproductive life cycle of this fungus, particularly if drug resistance or pathogenicity genes can be transmitted horizontally. Experiments were divided into three phases. Phase I looked at heterokaryon formation by hyphal anastomosis and protoplast fusion. Phase II was a time course of heterokaryon formation to look at patterns of nuclear and mitochondrial inheritance. Phase III examined the genetic relatedness of the different vegetative compatibility groups using a multilocus analysis approach. Heterokaryon formation was evident within and between vegetative compatibility groups. Observation of non-parental genotypes after heterokaryon formation confirmed that, although a rare event, horizontal genetic transfer occurred during heterokaryon formation. Uniparental mitochondria inheritance was observed in heterokaryons formed either by hyphal anastomosis or protoplast fusion. Drug resistance was expressed during heterokaryon formation, even across greater genetic distances than those distances imposed by vegetative compatibility. Phylogenies inferred from different molecular markers were incongruent at a significant level, challenging the clonal origins of Foc. Mating type genes were identified in this asexual pathogen Polymorphisms were detected within a Vegetative Compatibility Group (VCG) suggesting non-clonal inheritance and/or sexual recombination in Foc. Pahayokolides A-D are cytotoxic cyclic polypeptides produced by the freshwater cyanobacterium Lyngbya sp. strain 15-2 that possess an unusual β-amino acid, 3-amino-2,5,7,8-tetrahydroxy-10-methylundecanoic acid (Athmu). The absolute configuration of pahayokolides A-D was determined using advanced Marfey’s method. It was also confirmed that a pendant N-acetyl- N-methyl leucine moiety in pahayokolide A was absent in pahayokolides B and pahayokolides C-D were conformers of pahayokolide A. Feeding experiments indicated that the biosynthesis of the Athmu sidechain arises from leucine or α-ketoisovalerate, however could not be further extended by three rounds of condensation with malonate units. Putative four peptide and one unique polyketide synthetases in Lyngbya sp. strain 15-2 were identified by using a PCR method and degenerate primers derived from conserved core sequences of known NRPSs and PKSs. Identification of one unique KS domain conflicted with the logic rule that the long side chain of Athmu was assembled by three rounds of ketide extensions if PKSs were involved. A gene cluster (pah) encoding a peptide synthetase putatively producing pahayokolide was cloned, partially sequenced and characterized. Seven modules of the non-ribosomal peptide synthetase (NRPS) were identified. Ten additional opening reading frames (ORFs) were found, responsible for peptide resistance, transport and degradation. Although the predicted substrate specificities of NRPS agreed with the structure of pahayokolide A partially, the disagreement could be explained. However, no PKS gene was found in the pah gene cluster.

[EN] Sea turtle nests are exposed to different environmental risks that may affect their hatching success. Human exploitation, predation by wild or domestic animals, nest flooding or severe beach erosion or accession are common causes of egg mortality. However, there is very little information about the impact of microorganisms on turtle eggs. We analyzed loggerhead turtle eggs from Boavista Island (Republic of Cabo Verde) which were incubated under different environmental conditions in order to evaluate the presence and impact of fungus. We have isolated Fusarium oxysporum from dead and live eggs after three days of incubation.

The title compound, C(16)H(18)O(2)P(2), possesses two stereogenic P atoms and shows a distorted s-cis conformation of each O=P-C=C moiety. This has been suggested on the basis of the stereochemical result of 1,3-dipolar cyclo-additions with nitro-nes and is now confirmed by the present crystal structure analysis. There are two crystallographically independent molecules in the asymmetric unit.

A murcha de Fusarium spp. em crisântemo é responsável por sérios prejuízos à cultura no Brasil. Uma alternativa para o seu controle é o uso de substrato supressivo, o qual pode ser obtido pela adição de fontes de matérias orgânicas. Dessa forma, o presente trabalho teve por objetivo desenvolver um substrato supressivo à murcha do Fusarium em crisântemo com a introdução de matéria orgânica aos substratos padrões. Para tanto, lodo de esgoto e lodo de esgoto compostado; torta de mamona; esterco suíno; cama aviária; compostos comerciais LanziC); casca de camarão, biofertilizante e hidrolisado de peixe foram incorporados a substratos à base de casca de Pinus e de turfa em diferentes concentrações e combinações. Os experimentos foram realizados em propriedade produtora de crisântemo Bola-belga com problemas de Fusarium. Em todos os experimentos o número mínimo de repetições foi de 20 vasos por tratamento. Transcorridas 8, 12, 15 e 20 semanas do transplantio foi avaliada a severidade da doença por uma escala de notas de O para planta sadia a 5 para planta morta. Com os dados foram calculadas as áreas abaixo da curva de progresso da severidade da murcha de Fusarium. Além disso, foram realizadas análises dos atributos químicos e da atividade microbiana dos substratos bem como do desenvolvimento das plantas. O lodo de esgoto, lodo de esgoto compostado e a cama aviária induziram a supressividade do substrato à base de casca se Pinus e de turfa, controlando a murcha de Fusarium. O Lanzi®, também foi supressivo ao patógeno. A casca de camarão e o composto Lanzi® também induziram a supressividade dos substratos. Por outro lado, esterco suíno, torta de mamona, hidrolisado de peixe, quitosana e Trichoderma asperellum não interferiu na supressividade à doença. Substratos obtidos com lodo de esgoto e cama aviária, em mistura ou não, nas concentrações de 10, 20 e 30% (v/v) foram os mais adequados do ponto de vista de indução de supressividade e qualidade do produto, sendo os substratos recomendados para uso pelo agricultor. A supressividade observada nos substratos foi devido à união de características químicas e biológicas obtidas com a introdução das matérias orgânicas.

Con el propósito de evaluar la efectividad "in vitro'' de algunos fungicidas usados para el manejo de patógenos causantes de enfermedades del cafeto y reducir sus dosis, se condujo el presente trabajo que se llevó a cabo en el Laboratorio de Sanidad Vegetal del Centro Experimental de la Comisión Nacional del Café (CONCAFE) ubicado en el Km. 7, empalme San Francisco Carretera a San Ramón-Matagalpa y en el Laboratorio de Micología del Centro Nacional de Protección Vegetal (CENAPROVE) que está ubicado en el Km. 12.5 Carretera Sur, San José de la Cañada, Managua, a partir de Mayo a Diciembre de 1991. Se probaron seis fungicidas: Benomyl, Fermate, Propiconazol, Cupravit, Propineb y Captafol, evaluándose en cada uno cuatro dosis que se escogieron partiendo de la dosis comercial recomendada por las casas distribuidoras, la que tomamos como dosis alta, de la cual se derivaron las subsiguientes dosis, para él manejo de: Colletotrichum coffeanum Noack., Rhizoctonia solani Kuhn y Fusarium oxysporum f. sp. Se usó un Diseño Completo al Azar (DCA) con cuatro repeticiones, 24 tratamientos y 1 testigo absoluto sin tratamiento. Se midió el crecimiento del hongo en medio de cultivo PDA, (Papa-Dextrosa-Agar} con los fungicidas, encontrándose que el Propiconazol fue el que mayor efectividad tuvo aún en la dosis más baja, en cualquiera de los patógenos. El Fermate en su dosis media a baja y el Captafol en su dosis media son los productos que lograron en los 3 patógenos ejercer un buen control. Cupravit no mostró ninguna efectividad sobre R. solani y F oxysporum, pero si sobre C.coffeanum. Benomyl ejercio un buen efecto para R. solani y f. oxysporum pero no para C. coffeanum. Propineb mostro su efectividad en R. solani y C. coffeanum pero no ejerció ningun efecto en F. oxysporum.

De marzo 1999 a Mayo 2000 se realizó el presente estudio, en Managua, el cual se basó en la recopilación de información sobre las plagas asociadas a las semillas de cucurbitáceas. El objetivo del estudio fue proporcionar elementos técnicos a Cuarentena Vegetal para la toma de decisiones y aplicación de medidas fitosanitarias en la importación de semillas de cucurbitáceas para siembra procedente de Estados Unidos. La información fue obtenida de Bases de Datos Internacionales de Plagas, Centros de Documentación, Organismos internacionales, consultas a especialistas en foto protección, listado de plagas presentes en los cultivos de Nicaragua y búsqueda en Internet. Para el ordenamiento de la información se realizaron fichas técnicas para cada plaga. De un listado inicial de 1O plagas, solamente 8 plagas fueron sujetas a evaluación y análisis para el manejo del riesgo, después de pasar por las tres etapas de Evaluación de un Análisis de Riesgo de Plagas según la Norma Centroamericana del OIRSA. A las plagas consideradas como cuarentenarias para Nicaragua y que pueden causar grandes daños al país si se llegan a introducir, se les evaluó el riesgo de introducción, establecimiento y dispersión, además se determinaron las medidas de manejo del riesgo de plagas. De las plagas analizadas el hongo Fusarium oxysporum f. sp niveum, el virus Cucumber Green Mottle Mosaic Virus, el virus Melón Necrotic Spot Carmovirus, la bacteria Acidovorax avenae subsp.citrulli y el virus Cucumber Mosaic Cucumovirus, son las especies que presentan mayor riesgo fitosanitario.

杉木是我国重要的速生丰产树种,分布在北纬21°41′到33°41′,东经102°到122°的广大地区,杉木人工林面积约占我国人工林总面积的1/4,随着连栽代数的增加,土壤中毒和生产力下降程度日趋严重。 本论文以分离自与红树林、珍珠贝、海兔子、海绵、软珊瑚等与海洋动、植物共栖或共生存的106株海洋微生物(54株放线菌,52株细菌)为资源,以杉木连栽致害真菌尖孢镰刀菌萎蔫专化型(Fusarium oxysporum f.sp.vasinfectum)菌株SF2为靶菌,通过平板对峙试验和土壤原位定殖试验,筛选到一株分离自红树林木榄(Bruguiera gymnorrhizo)根际土壤的海洋细菌3728菌株;该菌对SF2具有很强抑菌活性,能够高密度在杉木根际土壤中定殖,对杉木幼苗的生长有一定的促进作用。采用传统的细菌学和分子生物学的鉴定方法对其进行了菌种鉴定,为枯草芽孢杆菌(Bacillus subtilis)。 通过对抗菌谱的研究,发现海洋细菌3728除了对杉木连栽主要致害真菌尖孢镰刀菌萎蔫专化型菌株SF2有很强的抑菌活性外,对大豆连作致害菌 (Penicillium purpurogenum),棉花枯萎病菌(Fusarium oxysporum),棉花立枯病菌(Rhizoctonia solani),大豆根腐病菌(Fusarium solani)以及小麦赤霉病菌(Fusarium graminerum)等也有较强的抑制作用。室内模拟试验还表明,在土壤中接种海洋细菌3728后,能够明显增加土壤中氨化细菌和氨化真菌的数量,能够增加土壤中功能性微生物——纤维素分解细菌和纤维素分解真菌的数量和种类,增强了纤维素分解能力。再添加C/N比较低的白三叶草凋落物,土壤中氨化细菌、氨化真菌的数量继续增加,土壤纤维素分解能力更显著提高。这为进一步开展对杉木连栽障碍的生物调控试验,提供了一定的科学依据。

自南海柳珊瑚(Pacifigorgia sp.)中分离得到一株对黄瓜枯萎病菌尖孢镰刀菌黄瓜专化型(Fusarium oxysporum f.sp.cucumer-inum)有较强抑制作用的海洋枯草芽孢杆菌(Bacillus subtilis)3512A。经室内模拟试验表明,该菌株能够在灭菌土和有菌土中高密度定殖,促进黄瓜幼苗的生长。为探讨该菌株在室外对黄瓜枯萎病的生防作用及对黄瓜生长和产量的影响,进行了盆栽试验,结果表明,海洋细菌3512A能够有效地防治黄瓜枯萎病,促进黄瓜的生长,提高叶绿素含量,增加黄瓜的产量。
