Apresenta o resultado de um estudo sobre a contribuição das estruturas de Tema para a organização da informação em dissertações. Para essa pesquisa, foram utilizados 44 textos produzidos por alunos do Convênio Especial (CE) do extinto sistema de Ensino Elite. Os textos foram divididos em dois grupos: o primeiro grupo (01 – 22) traz textos com média maior que 6,0, e o segundo (23 – 44) textos com média igual ou menor que 6,0. A base teórica assenta-se nos estudos de Halliday (1994), Thompson (1996), Eggins (1994), Neves (1997, 2002), Bakhtin (1992), Marcuschi (1999, 2002), Travaglia (2002), Rojo (2005), Koch (2002, 2006), Souza (2003), Bronckart (2003), Pillar (2002) e Fries (1994). Nos textos analisados, identifico as estruturas de Tema presentes em cada parte do texto (introdução, desenvolvimento e conclusão) e os elementos que formam essas estruturas. Os resultados apontam para o fato de que não há diferença entre os tipos de Temas escolhidos nos dois grupos, mas os elementos temáticos e o método de desenvolvimento diferem. No que diz respeito aos padrões de desenvolvimento, nos textos do primeiro grupo, a maior incidência foi do padrão zig-zag e, nos do segundo grupo, do tema reiterado. A análise dos textos mostra que não é a escolha de determinada estrutura de Tema que garante uma boa organização do texto, mas sim a escolha de qual elemento é escolhido para ocupar a posição temática.

Vitamin A is important to the proper development and functioning of the body, appearing in several metabolic functions. This experiment was conducted with aim evaluate to the vitamin A supplementation in diets for Goldfish Carassius auratus. The experimental was completely randomized with six treatments (0, 2.000, 4.000, 8.000, 16.000 and 32.000 IU of vitamin A kg(-1) diet) with four repetitions into 24 experimental cages, with capacity for 150 liters useful volume, made of mesh shading, installed inside a masonry circular tank with a capacity of 25 m(3) of water. 192 fish with an average initial weight of 6.66 +/- 0.57 g and initial length of 7.30 +/- 0.33 cm, feed at 8h, 11h, 14h and 17h at satiety were used. Evaluated the effects on productive performance and intensity of yellow-red coloring of the skin of the fish were. The Linear Response Plateau analysis of weight gain and length total, feed conversion and daily growth rate versus vitamin A requirement estimate showed 2,624, 2,385 and 2,358, 4,381 and 2,866 IU of vitamin A per kg diet, respectively. The data indicated that this vitamin has little or no influence on the intensity of the skin color of the Goldfish.

This action research study of approximately 90 high school algebra students investigates how frequent quizzing benefits them during the course of a semester. The intent of the research was to see how well students kept up with the material and if frequent quizzing helped them on the chapter tests. It was also designed to help me gain a better understanding of what students know and how I need to adjust daily routines so that all students stay caught up. I discovered that although frequent quizzes are not the students’ favorite activity to take part in, they learn to accept the quizzes and benefit greatly because of the amount of information students learn from them. Holding students accountable with frequent quizzes forces students to stay caught up and pushes them to excel as many found the tests to be much easier because of the practice they received. My research revealed many advantages to holding students accountable through frequent quizzes and although it can be somewhat time consuming, it is definitely a practice that will be continued in my classroom for years to come.

This PhD thesis describes set up of technological models for obtaining high health value foods and ingredients that preserve the final product characteristics as well as enrich with nutritional components. In particular, the main object of my research has been Virgin Olive Oil (VOO) and its important antioxidant compounds which differentiate it from all other vegetables oils. It is well known how the qualitative and quantitative presence of phenolic molecules extracted from olives during oil production is fundamental for its oxidative and nutritional quality. For this purpose, agronomic and technological conditions of its production have been investigated. It has also been examined how this fraction can be better preserved during storage. Moreover, its relation with VOO sensorial characteristics and its interaction with a protein in emulsion foods have also been studied. Finally, an experimental work was carried out to determine the antioxidative and heat resistance properties of a new antioxidant (EVS-OL) when used for high temperature frying such as is typically employed for the preparation of french fries. Results of the scientific research have been submitted for a publication and some data has already been published in national and international scientific journals.

Mixed tethered bilayer lipid membranes (tBLMs) are described based on the self-assembly of a monolayer on template stripped gold, of an archea analogue thiolipid, 2,3-di-o-phytanyl-sn-glycerol-1-tetraethylene glycol-D,L--lipoic acid ester lipid (DPTL), and a newly designed dilution molecule, tetraethylene glycol-D,L--lipoic acid ester (TEGL). The usage of spacer and addition of extra dilution molecules between the substrate and the bilayer is that this architecture provides an ionic reservoir underneath the membrane, avoiding direct contact of the embedded membrane proteins with the gold electrodes and increasing the lateral diffusion of the bilayer, thus allowing for the incorporation of complex channels proteins which are failed in non-diluted systems. The tBLM is completed by fusion of liposomes made from a mixture of 1,2-diphythanolyl-sn-glycero-3-phosphocholine (DPhyPC), cholesterol, and 1,2-diphytanoyl-sn-Glycero-3-phosphate (DPhyPG) in a molar ratio of 6:3:1. Varying the mixing ratio, the optimum mixing ratio was obtained at a dilution factor of DPTL and TEGL at 90%:10%. Only under these conditions, the mixed tBLM showed electrical properties, as shown by EIS, which are comparable to a BLM. With higher dilution factors, a defect-free lipid bilayer was not formed. Formation of bilayers have been characterized by different techniques, such as surface plasmon resonance (SPR), electrochemical impedance spectroscopy (EIS), atomic force microscopy (AFM), and quartz crystal microbalance (QCM). Different proteins such as hemolysin, melittin, gramicidin, M2, Maxi-K, nAChR and bacteriohodopsin are incorporated into these tBLMs as shown by SPR and EIS studies. Ionic conductivity at 0 V vs. Ag|AgCl, 3M KCl were measured by EIS measurements. Our results indicate that these proteins have been successfully incorporated into a very stable tBLM environment in a functionally active form. Therefore, we conclude that the mixed tBLMs have been successfully designed as a general platform for biosensing and screening purposes of membrane proteins.

Melittin, Hauptbestandteil des Bienengifts, ist ein kationisches Peptid, welches in der Lage ist, die biophysikalischen Eigenschaften der Zellmembran zu beeinflussen. Melittin werden unter anderem auch entzündungshemmende, schmerzlindernde, anti-rheumatische und anti-arthritische Wirkungen zugeschrieben. rnIn dieser Arbeit wurde nachgewiesen, dass Melittin die Proteolyse von ADAM10- und ADAM17-Substraten in verschiedenen Zellen stimuliert. Durch das Sheddingvon TGF-α wurde in HaCaT-Keratinozyten eine Transaktivierung des EGF-Rezeptors und eine daraus resultierende Phosphorylierung der Kinase ERK1/2 beobachtet. Die durch Melittin gesteigerte Aktivität der ADAMs ist calciumunabhängig und wird nicht durch Änderungen in der Membranfluidität verursacht. Eine Beteiligung der P2-Rezeptoren an der Melittin-induzierten ADAM-Aktivierung konnte sowohl durch Inhibition der Rezeptoren als auch durch Transfektion von HEK-Zellen mit dem P2X7-Rezeptor nachgewiesen werden. In diesen wurde nach der Behandlung mit Melittin eine Phosphorylierung von ERK1/2 beobachtet, welche durch ATPasen und P2-Rezeptor-Inhibitoren unterdrückt werden konnte. rnMit Hilfe des Kaninchenerythrozyten-Modells wurde nachgewiesen, dass eine Translokation von Phosphatidylserin von der Innen- zur Außenseite der Membran unmittelbar mit einer erhöhten ADAM-Aktivität korreliert. Sowohl durch Aktivierung des P2X7-Rezeptors als auch durch die Behandlung der Zellen mit dem Ionophor A23187 konnte ein Phosphatidylserin-Flip induziert werden. Dieser Flip führte zu einer erhöhten Aktivität von ADAM10, die durch eine gesteigerte Hämolyse und Spaltung von pVCC nachgewiesen werden konnte. Wurde der Phosphatidylserin-Flip durch Inhibitoren des P2X7-Rezeptors bzw. die Chelation von Ca2+ und Hemmung der Ionenfluxe unterdrückt, blieb auch die erhöhte ADAM-Aktivität aus. Wurde dagegen der Phosphatidylserin-Flip erst induziert und nachträglich die Inhibition des P2X7-Rezeptors bzw. die Chelation von Ca2+ und Hemmung der Ionenfluxe durchgeführt, zeigte dies keine Inhibition der ADAM-Aktivität.rnZusammenfassend zeigen diese Ergebnisse, dass eine Exposition von Phosphatidylserin auf der Außenseite der Membran in einem kausalen Zusammenhang mit einer gesteigerten ADAM-Aktivität steht.rn

Infection of canine footpads with the canine distemper virus (CDV) can cause massive epidermal thickening (hard pad disease), as a consequence of increased proliferation of keratinocytes and hyperkeratosis. Keratinocytes of canine footpad epidermis containing detectable CDV nucleoprotein antigen and CDV mRNA were shown previously to have increased proliferation indices. Because various proteins that play a role in the proliferation of epidermal cells are viral targets, the potential participation of such proteins in CDV-associated keratinocyte proliferation was investigated. Transforming growth factor-alpha (TGF-alpha), cell cycle regulatory proteins p21, p27 and p53, and nuclear factor (NF)-kappaB transcription factor components p50 and p65 were studied in the footpad epidermis from the following groups of dogs inoculated with CDV: group 1, consisting of seven dogs with clinical distemper and CDV in the footpad epidermis; group 2, consisting of four dogs with clinical distemper but no CDV in the footpad epidermis; group 3, consisting of eight dogs with neither clinical distemper nor CDV in the footpad epithelium. Group 4 consisted of two uninoculated control dogs. The expression of TGF-alpha, p21, p27 and p53, and p50 in the basal layer, lower and upper spinous layers, and in the granular layer did not differ statistically between CDV-positive (group 1) and CDV-negative (groups 2-4) footpad epidermis. However, there were differences in the levels of nuclear and cytoplasmic p65 expression between group 1 dogs and the other three groups. Thus, footpads from group 1 dogs had more keratinocytes containing p65 in the cytoplasm and, conversely, fewer nuclei that were positive for p65. These findings indicate that p65 translocation into the nucleus is reduced in CDV-infected footpad epidermis. Such decreased translocation of p65 may help to explain increased keratinocyte proliferation in hard pad disease and suggests interference of CDV with the NF-kappaB pathway.

The gene for agouti signaling protein (ASIP) is centrally involved in the expression of coat color traits in animals. The Mangalitza pig breed is characterized by a black-and-tan phenotype with black dorsal pigmentation and yellow or white ventral pigmentation. We investigated a Mangalitza x Piétrain cross and observed a coat color segregation pattern in the F2 generation that can be explained by virtue of two alleles at the MC1R locus and two alleles at the ASIP locus. Complete linkage of the black-and-tan phenotype to microsatellite alleles at the ASIP locus on SSC 17q21 was observed. Corroborated by the knowledge of similar mouse coat color mutants, it seems therefore conceivable that the black-and-tan pigmentation of Mangalitza pigs is caused by an ASIP allele a(t), which is recessive to the wild-type allele A. Toward positional cloning of the a(t) mutation, a 200-kb genomic BAC/PAC contig of this chromosomal region has been constructed and subsequently sequenced. Full-length ASIP cDNAs obtained by RACE differed in their 5' untranslated regions, whereas they shared a common open reading frame. Comparative sequencing of all ASIP exons and ASIP cDNAs between Mangalitza and Piétrain pigs did not reveal any differences associated with the coat color phenotype. Relative qRT-PCR analyses showed different dorsoventral skin expression intensities of the five ASIP transcripts in black-and-tan Mangalitza. The a(t) mutation is therefore probably a regulatory ASIP mutation that alters its dorsoventral expression pattern.

Consistency within the psychological processes is accepted as a precondition for efficient functioning and good health. Inconsistency in contrast impairs meeting the requirements of environment and therefore impairs the satisfaction of human needs. It is also seen as a stressor and having the potential to increase the vulnerability for mental disorders (Grawe, 1998, 2004). Incongruence is a form of Inconsistency, describing the divergency between the perception of reality and the goals of a person. Discordance as a second form of Inconsistency is the amount of conflict between goals, wishes and motives. According to the Consistency Theory of Grawe, these two forms of Inconsistency together with avoidance goals and satisfaction of human needs play an important role at the emergence and the maintenance of mental disorders, as well as for the wellbeing of humans. This study includes a short overview of the conceptions of Inconsistency in the psychological literature and a metaanalysis about the interrelations between forms of Inconsistency and characteristics of health and disease. The results support mostly the assumptions of Consistency Theory. Almost all forms of Inconsistency are associated with characteristics of subjective wellbeing, health and disease. One has to take into consideration, that most of the results come from correlational studies with only one measuring time. It is therefore not possible to distinguish between cause and effect.

BACKGROUND: Volume resuscitation is one of the primary therapeutic goals in hemorrhagic shock, but data on microcirculatory effects of different colloidal fluid resuscitation regimen are sparse. We investigated sublingual mucosal microcirculatory parameters during hemorrhage and after fluid resuscitation with gelatin, hydroxyethyl starch, or hypertonic saline and hydroxyethyl starch in pigs. METHODS: To induce hemorrhagic shock, 60% of calculated blood volume was withdrawn. Microvascular blood flow was assessed by laser Doppler velocimetry. Microcirculatory hemoglobin oxygen saturation was measured with a tissue reflectance spectrophotometry, and side darkfield imaging was used to visualize the microcirculation and to quantify the flow quality. Systemic hemodynamic variables, systemic acid base and blood gas variables, and lactate measurements were recorded. Measurements were performed at baseline, after hemorrhage, and after fluid resuscitation with a fixed volume regimen. RESULTS: Systemic hemodynamic parameters returned or even exceeded to baseline values in all three groups after fluid resuscitation, but showed significantly higher filling pressures and cardiac output values in animals treated with isotonic colloids. Microcirculatory parameters determined in gelatin and hydroxyethyl starch resuscitated animals, and almost all parameters except microvascular hemoglobin oxygen saturation in animals treated with hypertonic saline and hydroxyethyl starch, were restored after treatment. DISCUSSION: Hemorrhaged pigs can be hemodynamically stabilized with either isotonic or hypertonic colloidal fluids. The main finding is an adequate restoration of sublingual microcirculatory blood flow and flow quality in all three study groups, but only gelatin and hydroxyethyl starch improved microvascular hemoglobin oxygen saturation, indicating some inadequate oxygen supply/demand ratio maybe due to a better restoration of systemic hemodynamics in isotonic colloidal resuscitated animals.

The natural regulation of the water cycle by tropical montane forests is an important ecosystem service. Within this chapter we focus on water balance and regulation of the water cycle. Differences of rainfall-runoff generation across scales change from a near-surface event water driven system in pristine rainforest-covered micro-catchments to a more groundwater pre-event water dominated one on the mesoscale. The highly dynamic discharges are often correlated with total suspended sediment loads. However, we also observed total suspended sediment peaks at times of low flow, indicating a decoupling of erosion and stream transport and a triggering of landslides not directly related to hydrological processes. We also summarize likely future trends of water-related ecosystem services and expect an increase in human use and benefits of fresh water use whereas changes in water regulation and water purification services remain unchanged on a high level.
