This paper presents the design and implementation of a novel optical fiber temperature compensated relative humidity (RH) sensor device, based on fiber Bragg gratings (FBGs) and developed specifically for monitoring water ingress leading to the deterioration of building stone. The performance of the sensor thus created, together with that of conventional sensors, was first assessed in the laboratory where they were characterized under experimental conditions of controlled wetting and drying cycles of limestone blocks, before being employed “in-the-field” to monitor actual building stone in a specially built wall. Although a new construction, this was built specifically using conservation methods similar to those employed in past centuries, to allow an accurate simulation of processes occurring with wetting and drying in the historic walls in the University of Oxford.

The sweet cherry ‘Sunburst’ is highly appreciated by consumers due to its organoleptic traits. Regional producers tend to harvest cherries sooner in order to increase their profits. With the aim of understanding the consequences of this we have tested the effect of different ripeness stages at the moment of harvesting on fruit quality. Quality parameters tested included external colour (L*, a*, b*), fruit texture, total soluble solids (TSS), and titratable acidity (TA). To evaluate nutritional quality total antioxidant activity was measured too. Once again, and in agreement with results obtained in previous studies, we conclude that there is no advantage in picking less ripe cherries.

Nos últimos anos, a Optoelectrónica tem sido estabelecida como um campo de investigação capaz de conduzir a novas soluções tecnológicas. As conquistas abundantes no campo da óptica e lasers, bem como em comunicações ópticas têm sido de grande importância e desencadearam uma série de inovações. Entre o grande número de componentes ópticos existentes, os componentes baseados em fibra óptica são principalmente relevantes devido à sua simplicidade e à elevada de transporte de dados da fibra óptica. Neste trabalho foi focado um destes componentes ópticos: as redes de difracção em fibra óptica, as quais têm propriedades ópticas de processamento únicas. Esta classe de componentes ópticos é extremamente atraente para o desenvolvimento de dispositivos de comunicações ópticas e sensores. O trabalho começou com uma análise teórica aplicada a redes em fibra e foram focados os métodos de fabricação de redes em fibra mais utilizados. A inscrição de redes em fibra também foi abordado neste trabalho, onde um sistema de inscrição automatizada foi implementada para a fibra óptica de sílica, e os resultados experimentais mostraram uma boa aproximação ao estudo de simulação. Também foi desenvolvido um sistema de inscrição de redes de Bragg em fibra óptica de plástico. Foi apresentado um estudo detalhado da modulação acústico-óptica em redes em fibra óptica de sílica e de plástico. Por meio de uma análise detalhada dos modos de excitação mecânica aplicadas ao modulador acústico-óptico, destacou-se que dois modos predominantes de excitação acústica pode ser estabelecidos na fibra óptica, dependendo da frequência acústica aplicada. Através dessa caracterização, foi possível desenvolver novas aplicações para comunicações ópticas. Estudos e implementação de diferentes dispositivos baseados em redes em fibra foram realizados, usando o efeito acústico-óptico e o processo de regeneração em fibra óptica para várias aplicações tais como rápido multiplexador óptico add-drop, atraso de grupo sintonizável de redes de Bragg, redes de Bragg com descolamento de fase sintonizáveis, método para a inscrição de redes de Bragg com perfis complexos, filtro sintonizável para equalização de ganho e filtros ópticos notch ajustáveis.

Este trabalho focou-se no estudo do impacte das condições ambientais, de instalação e de utilização na degradação da fibra ótica, que frequentemente resultam na redução do desempenho das fibras óticas. Entre este fatores, foram estudados os efeitos de ambientes agressivos para o revestimento da fibra, nomeadamente no tempo de vida e resistência. Foi também estudado o efeito da propagação de sinais óticos de elevadas potências em curvaturas apertadas e a sua influência na degradação do desempenho da fibra ótica. Ainda neste âmbito, foi também estudado o desempenho de fibras óticas insensíveis a curvtura e fibras dopadas com Érbio, sendo analisada a dinâmica do efeito rastilho nestas fibras. Como parte integrante das redes óticas, os conetores óticos são de extrema importância na sua estrutura. O seu desempenho será refletido na qualidade do serviço da rede, e por isso é determinante estudar os fatores que contribuem para a sua degradação e mau funcionamento. Assim, este trabalho apresenta um estudo do comportamento de conetores óticos perante situações de mau manuseamento (como são limpeza insuficiente e degradação física da face final). Em adição foi também dado ênfase à reutilização de fibra danificada pelo efeito rastilho no desenvolvimento de sensores, passíveis de serem utilizados na monitorização de índice de refração, pressão hidrostática, tensão ou alta temperatura. Este procedimento surge como uma solução de baixo custo para o desenvolvimento de sensores em fibra ótica a partir de fibra danificada e inutilizável para as suas habituais aplicações em transmissão e/ou reflexão de sinais óticos.

We present a new method for lysis of single cells in continuous flow, where cells are sequentially trapped, lysed and released in an automatic process. Using optimized frequencies, dielectrophoretic trapping allows exposing cells in a reproducible way to high electrical fields for long durations, thereby giving good control on the lysis parameters. In situ evaluation of cytosol extraction on single cells has been studied for Chinese hamster ovary (CHO) cells through out-diffusion of fluorescent molecules for different voltage amplitudes. A diffusion model is proposed to correlate this out-diffusion to the total area of the created pores, which is dependent on the potential drop across the cell membrane and enables evaluation of the total pore area in the membrane. The dielectrophoretic trapping is no longer effective after lysis because of the reduced conductivity inside the cells, leading to cell release. The trapping time is linked to the time required for cytosol extraction and can thus provide additional validation of the effective cytosol extraction for non-fluorescent cells. Furthermore, the application of one single voltage for both trapping and lysis provides a fully automatic process including cell trapping, lysis, and release, allowing operating the device in continuous flow without human intervention.

A simple, effective and inexpensive fiber optic sensor for investigating the setting characteristics of various grades of cement is described. A finite length of unsheathed multimode optical fiber laid inside the cement mix, is subjected to stress during the setting process. The microbends created on the fiber due to this stress directly influence the intensity of light propagating through the fiber. Continuous monitoring of such variations in the light output transmitted through the fiber gives a clear measure of the setting characteristics of the cement mix, thus providing a simple and elegant technique of great practical importance in the field of civil engineering. The smart fiber optic sensor described above can be incorporated into a building during the construction process itself so that continuous monitoring of the deterioration process for the entire life time of the building can be carried out.

A simple fiber optic concentration sensor based on the coupling of light f rom one fiber to another through a solution is discussed. The operational characteristics of the sensor are illustrated by taking the solutions of potassium permanganate and fast green dye as samples.The extrinsic type sensor described here shows linearity at lower concentrations.

Unprocessed seafood harbor high number of bacteria, hence are more prone to spoilage. In this circumstance, the use of spice in fish for reduction of microorganism can play an important role in seafood processing. Many essential oils from herbs and spices are used widely in the food, health and personal care industries and are classified as GRAS (Generally regarded as safe) substances or are permitted food additives. A large number of these compounds have been the subject of extensive toxicological scrutiny. However, their principal function is to impart desirable flavours and aromas and not necessarily to act as antimicrobial agents. Given the high flavour and aroma impact to plant essential oils, the future for using these compound as food preservatives lies in the careful selection and evaluation of their efficacy at low concentrations but in combination with other chemical preservatives or preservation processes. For this reason they are worth of study alone or in combination with processing methods in order to establish if they could extend the shelf-life of foods. In this study, the effect of the spices, clove, turmeric, cardamom, oregano, rosemary and garlic in controlling the spoilage and pathogenic bacteria is investigated. Their effect on biogenic amine formation in tuna especially, histamine, as a result of bacterial control is also studied in detail. The contribution of spice oleoresin in the sensory and textural parameters is investigated using textural profile analysis and sensory panel. Finally, the potential of spices in quality stabilization and in increasing the shelf–life of tuna during frozen storage is analysed

A study was carried out to determine the influence of fibrolytic enzymes derived from mesophilic or thermophilic fungal sources, added at ensiling, on time-course fermentation characteristics and in vitro rumen degradation of maize silage. The mesophilic enzyme was a commercial product derived from Trichodenna reesei (L), whereas the thermophilic enzyme was a crude extract produced from Thermoascus aurantiacus (Ta) in this laboratory. The fungus was cultured using maize cobs as a carbon source. The resulting fermentation extract was deionised to remove sugars and characterised for its protein concentration, main and side enzymic activities, optimal pH, protein molecular mass and isoelectric point. In an additional study, both enzymes were added to maize forage (333.5 g DM/kg, 70.0, 469.8, 227.1 and 307.5 g/kg DM of CP, NDF, ADF and starch, respectively) at two levels each, normalized according to xylanase activity, and ensiled in 0.5 kg capacity laboratory minisilos. Duplicate silos were opened at 2, 4, 8, 15, and 60 days after ensiling, and analysed for chemical characteristics. Silages from 60 days were bulked and in vitro gas production (GP) and organic matter degradability (OMD) profiles evaluated using the Reading Pressure Technique (RPT), in a completely randomised design. The crude enzyme extract contained mainly xylanase and endoglucanase activities, with very low levels of exoglucanase, which probably limited hydrolysis of filter paper. The extract contained three major protein bands of between 29 and 55 kDa, with mainly acidic isoelectric points. Ensiling maize with enzymes lowered (P < 0.05) the final silage pH, with this effect being observed throughout the ensiling process. All enzyme treatments reduced (P < 0.05) ADF contents. Treatments including Ta produced more gas (P < 0.05) than the controls after 24 h incubation in vitro, whereas end point gas production at 96 h was not affected. Addition of Ta increased (P < 0.01) OMD after 12 h (410 and 416 g/kg versus 373 g/kg), whereas both L and Ta increased (P < 0.05) OMD after 24 h. Addition of enzymes from mesophilic or thermophilic sources to maize forage at ensiling increased the rate of acidification of the silages and improved in vitro degradation kinetics, suggesting an improvement in the nutritive quality. A reliable and fast sensor for in vitro evaluation of solar protection factors (SPFs) of cosmetic products, based on the photobleaching kinetics of a nanocrystalline TiO(2)/dye UV-dosimeter, has been devised. The accuracy, robustness and suitability of the new device was demonstrated by the excellent matching of the predicted and the in vivo results up to SPF 70, for four standard samples analyzed in blind. These results strongly suggest that our device can be useful for routine SPF evaluation in laboratories devoted to the development or production of cosmetic formulations, since the conventional in vitro methods tend to exhibit unacceptably high errors above SPF similar to 30 and the conventional in vivo methods tend to be expensive and exceedingly time consuming.
