To unravel the climatic and environmental dynamics in the borderlands of the Aegean Sea during the early and middle Holocene, and notably for the interval of sapropel S1 (S1) formation, we have analysed terrestrial palynomorphs from a marine core in the northern Aegean Sea. The qualitative results were complemented by quantitative pollen-based climate reconstructions. A land-sea correlation was established based on pollen data and sediment lightness measurements from the same core, and previously published benthic foraminifer data from a nearby core. The borderlands of the Aegean Sea underwent a transition from an open vegetation to oak-dominated woodlands between ~10.4 and ~9.5 ka cal BP. A coeval increase in winter precipitation suggests that moisture availability was the main factor controlling Holocene reforestation. The ~50% higher winter precipitation during S1 formation relative to "pre-sapropelic" conditions suggests a strong contribution from the borderlands of the Aegean Sea to the freshwater surplus during S1 formation. The humid and mild winter conditions during S1 formation were repeatedly punctuated by short-term climatic events that caused a partial deforestation and a reorganisation within the broad-leaved arboreal vegetation. In the marine realm, these events are documented by improved benthic oxygenation. The strongest event represents the regional expression of the 8.2 ka cold event and led to an interruption in S1 formation. Except for the interval of S1 formation, the pollen-derived winter temperatures correlate with the smoothed GISP2 K+ series. They support the previously published, marine-based concept that the intensity of the Siberian High strongly controlled the winter climate in the Aegean region. During S1 formation in the Aegean Sea, however, climate conditions in the borderlands were more strongly affected by the monsoonally influenced climate system of the lower latitudes.

The distribution of diatoms, coccolithophores and planktic foraminifers mirrored the hydrographic and trophic conditions of the surface ocean (0-100 m) across the upwelling area off the Oman coast to the central Arabian Sea during May/June 1997 and July/August 1995. The number of diatoms was increased in waters with local temperature minimum and enhanced nutrient concentration (nitrate, phosphate, silicate) caused by upwelling. Vegetative cells of Chaetoceros dominated the diatom assemblage in the coastal upwelling area. Towards the more nutrient depleted and stratified surface waters to the southeast, the number of diatoms decreased, coccolithophore and planktic foraminiferal numbers increased, and floral and faunal composition changed. In particular, the transition between the eutrophic upwelling region off Oman and the oligotrophic central Arabian Sea was marked by moderate nutrient concentration, and high coccolithophore and foraminifer numbers. Florisphaera profunda, previously often referred as a 'lower-photic-zone-species', was frequent in water depths as shallow as 20 m, and at high nutrient concentration up to 14 µmol NO3/l and 1.2 µmol PO4/. To the oligotrophic southeast of the divergence, cell densities of coccolithophores declined and Umbellosphaera irregularis prevailed throughout the water column down to 100 m depth. In general, total coccolithophore numbers were limited by nutrient threshold concentration, with low numbers (<10*10**3 cells/l) at high [NO3] and [PO4], and high numbers (>70*10**3 cells/l) at low [NO3] and [PO4]. The components of the complex microplankton succession, diatoms, coccoliths and planktic foraminifers (and possibly others), should ideally be used as a combined paleoceanographic proxy. Consequently, models on plankton ecology should be resolved at least for the seasonality, to account for the bias of paleoceanographic transfer calculations.

The fruit is one of the most complex and important structures produced by flowering plants, and understanding the development and maturation process of fruits in different angiosperm species with diverse fruit structures is of immense interest. In the work presented here, molecular genetics and genomic analysis are used to explore the processes that form the fruit in two species: The model organism Arabidopsis and the diploid strawberry Fragaria vesca. One important basic question concerns the molecular genetic basis of fruit patterning. A long-standing model of Arabidopsis fruit (the gynoecium) patterning holds that auxin produced at the apex diffuses downward, forming a gradient that provides apical-basal positional information to specify different tissue types along the gynoecium’s length. The proposed gradient, however, has never been observed and the model appears inconsistent with a number of observations. I present a new, alternative model, wherein auxin acts to establish the adaxial-abaxial domains of the carpel primordia, which then ensures proper development of the final gynoecium. A second project utilizes genomics to identify genes that regulate fruit color by analyzing the genome sequences of Fragaria vesca, a species of wild strawberry. Shared and distinct SNPs among three F. vesca accessions were identified, providing a foundation for locating candidate mutations underlying phenotypic variations among different F. vesca accessions. Through systematic analysis of relevant SNP variants, a candidate SNP in FveMYB10 was identified that may underlie the fruit color in the yellow-fruited accessions, which was subsequently confirmed by functional assays. Our lab has previously generated extensive RNA-sequencing data that depict genome-scale gene expression profiles in F. vesca fruit and flower tissues at different developmental stages. To enhance the accessibility of this dataset, the web-based eFP software was adapted for this dataset, allowing visualization of gene expression in any tissues by user-initiated queries. Together, this thesis work proposes a well-supported new model of fruit patterning in Arabidopsis and provides further resources for F. vesca, including genome-wide variant lists and the ability to visualize gene expression. This work will facilitate future work linking traits of economic importance to specific genes and gaining novel insights into fruit patterning and development.

Strawberry fruits are highly appreciated worldwide due to their pleasant flavor and aroma and to the health benefits associated to their consumption. An important part of these properties is due to their content in secondary metabolites, especially phenolic compounds, of which flavonoids are the most abundant in the strawberry fruit. Although the flavonoid biosynthesis pathway is uncovered, little is known about its regulation. The strawberry Fra a (Fra) genes constitute a large family of homologs of the major birch pollen allergen Bet v 1 and for which no equivalents exist in Arabidopsis. Our group has shown that Fra proteins are involved in the formation of colored compounds in strawberries (Muñoz et al., 2010), which mainly depends on the production of certain flavonoids; that they are structurally homologs to the PYR/PYL/RCAR Arabidopsis ABA receptor, and that they are able to bind flavonoids (Casañal et al., 2013). With these previous results, our working hypothesis is that the Fra proteins are involved in the regulation of the flavonoids pathway. They would mechanistically act as the ABA receptor, binding a protein interactor and a ligand to regulate a signaling cascade and/or act as molecular carriers. The main objective of this research is to characterize the Fra family in strawberry and gain insight into their role in the flavonoid metabolism. By RNAseq expression analysis in ripening fruits we have identified transcripts for 10 members of the Fra family. Although expressed in all tissues analyzed, each family member presents a unique pattern of expression, which suggests functional specialization for each Fra protein. Then, our next approach was to identify the proteins that interact with Fras and their ligands to gain knowledge on the role that these proteins play in the flavonoids pathway. To identify the interacting partners of Fras we have performed a yeast two hybrid (Y2H) screening against cDNA libraries of strawberry fruits at the green and red stages. A protein that shares a 95% homology to the Heat stress transcription factor A-4-C like of Fragaria vesca (HSA4C) interacts specifically with Fra1 and not with other family members, which suggests functional diversification of Fra proteins in specific signaling pathways. The Y2H screening is not yet saturated, so characterization of other interacting proteins with other members of the Fra family will shed light on the functional diversity within this gene family. This research will contribute to gain knowledge on how the flavonoid pathway, and hence, the fruit ripening, is regulated in strawberry; an economically important crop but for which basic research is still very limited. O ácaro rajado, Tetranychus urticae Koch, é considerado uma das principais pragas de hortaliças e varias outras culturas no Brasil, em áreas onde um considerável volume de acaricidas e anualmente empregado no seu controle. O objetivo deste trabalho foi testar a viabilidade técnica do empregado dos fitoseideos Amblyseius idaeus (Denmark & Muma) e Phytoseiulus macropilis (Banks), comumente encontrados no Brasil, no controle de T. urticae em pepino e morangueiro em Jaguariúna, SP. Utilizou-se o delineamento experimental em blocos casualizados, com 4 repetições e 4 tratamentos (T1-T4) para o pepino e 5 tratamentos para o morangueiro (T1-T5): T1, testemunha; T2 pulverização semanais de Malation para a exclusão de predadores nativos; T3, liberação periodicos de A. idaeus; T4, liberações periódicas de P.macropilis; T5, pulverizações semanais de avermectina para a exclusão de ácaros fitófagos e predadores nativos. Apenas A. idaeus se estabeleceu na cultura do pepino, reduzindo significativamente a população de T. urticae. Ambas espécies de predadores se estabeleceram na cultura do morango e reduziram significativamente a população de T. urticae.
