Diseñar, aplicar y comprobar el grado de validez de 3 programas dirigidos al desarrollo y mejora de las estrategias necesarias para una adecuada comprensión y producción de textos. La investigación se propone los siguientes objetivos : 1.- Revisar las estrategias de comprensión y expresión escrita empleadas por alumnos del segundo ciclo de secundaria (3 ESO), con anterioridad a la aplicación de los 3 tratamientos. 2.- Analizar la influencia de una instrucción directa en estrategias dirigidas a construir la representación textual sobre la comprensión lectora de los alumnos. 3.- Estudiar la influencia de una instrucción directa en estrategias dirigidas a construir la representación textual sobre la calidad de la producción escrita de los alumnos. 4.- Examinar el efecto de una instrucción directa en los procesos y estrategias de composición sobre la calidad de la producción escrita de los alumnos. 5.- Estudiar el efecto que puede tener una instrucción directa en los procesos y estrategias de composición escrita sobre la construcción de la representación textual implicada en la comprensión lectora. 6.- Comprobar los efectos que puede tener una instrucción conjunta en ambos tipos de estrategias (comprensión lectora y composición escrita) sobre la calidad de las producciones escritas (elementos creativos, mecanismos de cohesión, y coherencia textual) y sobre el modo de elaborar la representación textual implicada en la comprensión lectora. 7.- Contrastar la influencia que puede tener la instrucción directa en estrategias de comprensión lectora sobre el rendimiento académico de los alumnos. 8.- Analizar la influencia que puede tener la instrucción directa en estrategias de composición escrita sobre el rendimiento académico de los alumnos. 9.- Analizar la influencia que puede tener una instrucción integrada por estrategias de comprensión lectora y composición escrita sobre el rendimiento académico de los alumnos participantes en el programa.. La muestra está compuesta por 86 alumnos de tercero de ESO, del IES Rodríguez Fabrés.. Diseño de grupos de control no equivalentes con medidas pretratamiento y postratamiento; parte de grupos formados de manera natural en el IES; aplica una metodología cuasiexperimental. Se eligen aleatoriamente los grupos experimentales y control. Se determina el nivel de comprensión lectora y expresión escrita que poseen los alumnos, realizando para ellos en todos los grupos una estimación inicial (fase pretest). Se realiza una estimación del rendimiento académico de los alumnos durante la primera evaluación a través de las calificaciones obtenidas en 4 áreas: Lengua y Literatura, Matemáticas, Ciencias Sociales y Ciencias Naturales; mediante la puntuación otorgada por el profesorado en cuatro capacidades concretas de los alumnos: Interés, comprensión, expresión y autonomía. Realizada la medición inicial, procede a la aplicación de los diferentes programas en cada uno de los grupos experimentales Aplicados los programas, realiza las mediones postest de las variables anteriormente mencionadas, para comprobar los efectos del tratamiento en los grupos experiementales. Grupo 1: Control. No intervienen en ningún programa, se les aplican pruebas pretest y postest, constituido por 28 sujetos. Grupo 2: Grupo experimental que trabajó el programa 'Mejorar nuestra forma de expresarnos cuando escribimos'. 13 sujetos. Grupo 3: Grupo experimental que trabajó el programa 'Mejorar nuestra comprensión de los textos expositivos', integrado por 19 sujetos. Grupo 4: Grupo experimental donde se desarrolló el programa: 'Mejorar nuestra comprensión y expresión de textos', integrado por 26 sujetos. Clasificación de las variables : A.-Variables dependientes: Pueden haberse modificado como consecuencia de la aplicación de las variables dependientes : A.1.- Rendimiento en comprensión lectora. A.2.- Rendimiento en expresión escrita. A.3.- Rendimiento escala de estrategias cognitivas y actitudes hacia la expresión escrita. A.4.- Rendimiento académico general. B.-Variables independientes : Manipuladas deliberadamente : B.1.- Programa: 'Mejorar nuestra forma de expresarnos cuando escribimos'. B.2.- Programa: 'Mejorar nuestra comprensión de los textos expositivos'. B.3.- Programa: : 'Mejorar nuestra comprensión y expresión de textos'.. Informes del Instituto, Test del factor G de Cattel (TEA), Test de actitudes escolares (TEA 2), 4 pruebas de medición de comprensión lectora (ad hoc), 4 pruebas de medición en expresión escrita (ad hoc), escala de estrategias cognitivas y actitudes hacia la exposición escrita (ad hoc), actas de calificaciones y capacidades del IES y cuestionario de estudio y trabajo intelectual (C.E.T.I.).. Análisis estadístico con el paquete SPSS; análisis de varianza.. Analiza los efectos diferenciales que la implementación de los 3 programas tienen en la mejora de las capacidades comprensivas y expresivas de los alumnos de 3õ ESO en su rendimiento escolar, analizando las diferentes variables. La investigación subraya la validez que cada uno de los programas ha tenido a la hora de mejorar aquellos aspectos en los que se pretendía incidir en el diseño e implementación de la investigación. verificado a través del contraste de hipótesis. Respecto a los programas empleados, la investigación ha demostrado que el programa 'mejor comprensión de textos expositivos' como el programa más potente para mejorar las competencias de los alumnos, verificándose en la mejora de rendimiento logrado por los alumnos.. Una intervención específica orientada al desarrollo y empleo de los procesos y estrategias implicados en la composición de los textos variados potencian la calidad de la expresión escrita de los alumnos. Una intervención específica orientada al desarrollo y empleo de las estrategias necesarias para construir una representación coherente del contenido del texto, contribuye a mejorar la comprensión lectora. Una intervención específica y conjunta en estrategias de comprensión lectora y expresión escrita, mejora el desempeño de ambas competencias , aunque no de forma generalizada para todos los alumnos.

Objective: Peroxynitrite (ONOO-) is formed in the inflamed and degenerating human joint. Peroxynitrite-modified collagen-II (PMC-II) was recently discovered in the serum of patients with osteoarthritis (OA) and rheumatoid arthritis (RA). Therefore we investigated the cellular effects of PMC-II on human mesenchymal progenitor cells (MPCs) as a model of cartilage and cartilage repair cells in the inflamed and degenerating joint. Design: MPCs were isolated from the trabecular bone of patients undergoing reconstructive surgery and were differentiated into a chondrogenic lineage. Cells were exposed to PMC-II and levels of the proinflammatory mediators nitric oxide (NO) and prostaglandin E-2 (PGE(2)) measured. Levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), phosphorylated mitogen activated protein kinases (MAPKs) and nuclear factor kappa B (NF-kappa B) activation were measured by enzyme linked immunosorbent assay (ELISA) together with specific MAPK and NF-kappa B inhibitors. Results: PMC-II induced NO and PGE(2) synthesis through upregulation of iNOS and COX-2 proteins. PMC-II also lead to the phosphorylation of MAPKs, extracellularly regulated kinase 1/2 (ERK1/2) and p38 [but not c-Jun NH2-terminal kinase (JNK1/2)] and the activation of proinflammatory transcription factor NF-kappa B. Inhibitors of p38, ERK1/2 and NF-kappa B prevented PMC-II induced NO and PGE(2) synthesis, NOS and COX-2 protein expression and NF-kappa B activation. Conclusion: iNOS, COX-2, NF-KB and MAPK are known to be activated in the joints of patients with OA and RA. PMC-II induced iNOS and COX-2 synthesis through p38, ERK1/2 and NF-KB dependent pathways suggesting a previously unidentified pathway for the synthesis of the proinflammatory mediators, NO and PGE(2), further suggesting that inhibitors of these pathways may be therapeutic in the inflamed and degenerating human joint. Dietary antioxidants can affect cellular processes relevant to chronic inflammatory diseases such as atherosclerosis. We have used non- standard techniques to quantify effects of the antioxidant soy isoflavones genistein and daidzein on translocation of Nuclear Factor-KB (NF-KB) and nitric oxide (NO) production, which are important in these diseases. Translocation was quantified using confocal immunofluoresecence microscopy and ratiometric image analysis. NO was quantified by an electrochemical method after reduction of its oxidation products in cell culture supernatants. Activation of the RAW 264.7 murine monocyte/macrophage cell line increased the ratio of nuclear to cytoplasmic immunostaining for NF-kB. The increase was exacerbated by pre-treatment with genistein or daidzein. To show that decreases could also be detected, pre-treatment with the pine bark extract Pycnogenol (R) r was examined, and found to reduce translocation. NO production was also increased by activation, but was reduced by pre-treatment with genistein or daidzein. In the EA. hy926 human endothelial cell line, constitutive production was detectable and was increased by thrombin. The confocal and electrochemical methods gave data that agreed with results obtained using the established electromobility shift and Griess assays, but were more sensitive, more convenient, gave more detailed information and avoided the use of radioisotopes.

Pre-eclampsia (PE) is a complication of human pregnancy characterized by hypertension and proteinuria after 20 weeks of gestation. Its incidence varies from 5% to 7% of pregnancies and is a major cause of morbidity and maternal and fetal mortality. This is a multisystemic disease, with focus on vascular dysfunction and is closely related to the exacerbated activation of the immune system. In addition to increased activation of monocytes and granulocytes, there is an elevated production of proinflammatory cytokines in pregnant women with PE. The nuclear transcription factor-kB (NF-kB) is present in the cells of the immune system and is responsible for transcription of genes related to inflammation. Whereas the PE is associated with intense inflammatory response, the use of substances modulating the activity of NF-kB factor could be useful in alleviating the inflammation present in these patients. Silibinin is the main component of silymarin, a polyphenolic extract obtained from fruits and seeds of Sylibum marianum with potent hepatoprotective, anti-inflammatory and anti-fibrotic activities. The silibinin mechanism of action includes the ability to inhibit NF-kB activation and, consequently, its migration to the nucleus. The objective of this study was to assess whether silibinin modulates the activity of NF-kB and the production of inflammatory cytokines in mononuclear cells of patients with PE. We evaluated 34 pregnant women with PE, 20 normotensive pregnant women (GN) and 15 non-pregnant women (NG). Peripheral blood mononuclear cells (PBMC) were obtained from those groups of women and cultured in the presence or absence of silibinin (5 uM or 50 uM) and stimulated or not with lipopolysaccharide (LPS) for 18 h to obtain supernatant for determination of tumor necrosis factor-alpha (TNF-α) and interleukin-1 (IL-1β) by enzyme immunoassay (ELISA). The pineal gland, a circumventricular organ, plays an integrative role in defense responses. The injury-induced suppression of the pineal gland hormone, melatonin, which is triggered by darkness, allows the mounting of innate immune responses. We have previously shown that cultured pineal glands, which express toll-like receptor 4 (TLR4) and tumor necrosis factor receptor 1 (TNFR1), produce TNF when challenged with lipopolysaccharide (LPS). Here our aim was to evaluate which cells present in the pineal gland, astrocytes, microglia or pinealocytes produced TNF, in order to understand the interaction between pineal activity, melatonin production and immune function. Cultured pineal glands or pinealocytes were stimulated with LPS. TNF content was measured using an enzyme-linked immunosorbent assay. TLR4 and TNFR1 expression were analyzed by confocal microscopy. Microglial morphology was analyzed by immunohistochemistry. In the present study, we show that although the main cell types of the pineal gland (pinealocytes, astrocytes and microglia) express TLR4, the production of TNF induced by LPS is mediated by microglia. This effect is due to activation of the nuclear factor kappa B (NF-kB) pathway. In addition, we observed that LPS activates microglia and modulates the expression of TNFR1 in pinealocytes. As TNF has been shown to amplify and prolong inflammatory responses, its production by pineal microglia suggests a glia-pinealocyte network that regulates melatonin output. The current study demonstrates the molecular and cellular basis for understanding how melatonin synthesis is regulated during an innate immune response, thus our results reinforce the role of the pineal gland as sensor of immune status.

Anhidrotic Ectodermal Dysplasia (EDA), is the most frequent form among Ectodermal Dysplasias, hereditary genetic disorders causing ectodermal appendages defective development. Indeed, EDA is characterized by defective formation of hair follicles, sweat glands and teeth both in human patients and animals. EDA, the gene mutated in Anhidrotic Ectodermal Dysplasia, encodes Ectodysplasin, a TNF family member that activates NF-kB mediated transcription. This disease can occur with mutations in other EDA-NF-kB pathway members, as EDA receptor, EDAR and its adapter, EDARADD. Moreover, mutations in TRAF6, NEMO, IKB and NF-kBs genes are responsible for Immunodeficiency associated EDA (EDA-ID). Several molecules, as SHH, WNT/DKK, BMP and LTβ, have already been reported to be EDA pathway regulators or effectors although the knowledge of the full spectrum of EDA targets remains incomplete. During the first part of the research project a gene expression analysis was performed in primary keratinocytes from Wild-type and Tabby (EDA model mouse) mice to identify novel EDA target genes. Earlier expression profiling at various developmental time points in Tabby and Wild-type mouse skin reported genes differentially expressed in the two samples and, to increase the resolution to find genes whose expression may be restricted to epidermal cells, the study was extended to primary keratinocyte cultures established from E19 Wild-type and Tabby skin. Using microarrays bearing 44,000 gene probes, we found 385 “preliminary candidate” genes whose expression was significantly affected by Eda defect. By comparing expression profiles to those from Eda-A1 (where Eda-A1 is highly expressed) transgenic skin, we restricted the list to 38 “candidate EDA targets”, 14 of which were already known to be expressed in hair follicles or epidermis. This work confirmed expression changes for 3 selected genes, Tbx1, Bmp7, and Jag1, both in primary keratinocytes and in Wild-type and Tabby whole skin, by Q-PCR and Western blotting analyses. Thus, this study detected novel candidate pathways downstream of EDA. In the second part of the research project, plasmid constructs were produced and analyzed to create a transgenic mouse model for Immunodeficiency associated EDA disease (XL-EDA-ID). In particular, plasmids containing mouse Wild-type and mutated Nemo cDNA under K-17 epidermis-specific promoter control and a Flag tag, were prepared, on the way to confine transgene expression to mice epidermis and to determine EDA phenotype without immunodeficiency for a comparison to Tabby model phenotype. EDA-ID mutations reported in patients and selected for this study are: C417R (C409R in mouse), causing Zinc Finger protein domain destabilization and A288G (A282G in mouse) affecting oligomerization of the protein. Moreover, the ex-novo mutation, ZnF, C-terminal Zinc Finger domain deletion, was tested. Thus, the constructs were analyzed by transient transfection, Western blotting and luciferase assays techniques, detecting Nemo Wild-type and mutant protein products and residue NF-kB activity in presence of mutants, after TNF stimulation. In particular, MEF_Nemo-/- cell line was used to monitor NF-kB activity without endogenous Nemo gene. Results show reduced NF-kB activity in presence of mutated Nemo forms compared to Wild-type: 81% for A282G (A288G in human); 24% for C409R (C417R in human); 15% for ZnF. C409R mutation (C417R in human), reported in 6 EDA-ID human patients, was selected to prepare transgenic model mouse. Mice (white, FVP) born following K17-promoter-Flag-Nemo_C409R plasmid region pronuclear injection, were analyzed for the transgene presence in the genotype and a preliminar examination of their phenotype was performed. In particular, one mouse showed considerable coat defects if compared to Wild-type mice. This preliminar analysis suggests a possible influence of Nemo mutant over-expression in epidermis without immunodeficiency. Still, more microscopic studies to analyze hair subtypes, Guard, Awl and Zigzag (usually alterated inTabby mouse model), Immunohistochemistry experiments to detect epidermis restricted Nemo expression and sweat glands analysis, will follow. This and other transgene positive mice will be crossed with black mice C57BL6 to obtain at least two indipendent agouti lines to analyze. Theses mice will be used in EDA target genes detection through microarrays. Following, plasmid constructs containing other Nemo mutant forms (A282G and ZnF) might be studied by the same experimental approaches to prepare more transgenic model mice to compare to Nemo_C409R and Tabby mouse models.

Las respuestas que se generan a partir de cuestionamientos del origen del hombre nos permitirán especular hacia dónde evolucionará como especie. Los grandes saltos evolutivos que diferencian a los primates humanos de los no humanos, se podrían describir entre otras características por una eficiente memoria para el uso de herramientas, la dominancia para el uso de la mano junto al desarrollo de la oposición del pulgar y el lenguaje. Se ha descrito un gen, el HSR que expresa para la dominancia para el uso de la mano derecha, habilidades cognitivas relacionadas con el lenguaje y asimetría cerebral en humanos. Este es un gen imprintado, es decir, que se hereda su expresión según el origen parental y cuya expresión está regulada por factores epigenéticos. Estos factores, modifican la expresión del gen sin afectar la estructura primaria del ADN. Se ha estudiado la expresión fenotípica del gen HSR en una población de niños escolarizados de La Rioja, dividida en dos regiones (Región 1 y Región 2). Los resultados obtenidos, que muestran una alteración de las proporciones fenotípicas del gen en la Región 2, apoyan fuertemente la posibilidad de que un factor ambiental estaría condicionando el epigenotipo del gen HSR. Se piensa que el estudio de estos mecanismos regulatorios en estos genes recientemente adquiridos por la evolución y blanco de funciones también recientemente adquiridas, podría dar información de hacia dónde la evolución del hombre podría proyectarse en el futuro.

A nivel mundial, el cáncer mamario representa el 23% de las enfermedades oncológicas que afectan a las mujeres. En el año 2008 fue responsable de 458.400 muertes. Las probabilidades de curar a una paciente dependen del avance de la enfermedad en el momento del diagnóstico. La estimación del pronóstico y predicción del tratamiento de las pacientes es fundamental para el manejo clínico de la enfermedad. La metástasis en ganglios linfáticos, el tamaño y el grado tumoral son los factores pronóstico más importantes, mientras que la expresión de los receptores de estrógeno, progesterona y del factor de crecimiento epidérmico 2 son marcadores para la predicción de la respuesta a distintos tratamientos. Las células tumorales presentan un genoma y un epigenoma distinto a las células normales. La alteración epigenética mejor entendida y más estudiada es la metilación aberrante de genes reguladores del ciclo celular. Esta tesis doctoral tiene como objetivo identificar las alteraciones epigenéticas relacionadas con el cáncer de mama humano. Para ello el trabajo ha sido dividido en tres bloques. El primero, centrado en el estudio de las alteraciones epigenéticas de la carcinogénesis mamaria. El segundo, enfocado en establecer relaciones entre las alteraciones epigenéticas y los aspectos clínicos de la enfermedad. El tercero, enfocado en la detección no invasiva de procesos oncológicos en las pacientes. Esta tesis muestra los resultados del análisis de la metilación aberrante de 49 regiones del genoma involucradas en el desarrollo de enfermedades oncológicas. El estudio ha permitido determinar el perfil de metilación tumoral de 92 tumores mamarios invasores, 17 melanomas, 8 hepatoblastomas y 6 tumores de próstata. Las alteraciones en la metilación de estos genes es específica para cada paciente y la información epigenética permite diferenciar el proceso tumoral mamario del resto de los tumores. La cantidad y el tipo de genes afectados correlacionan con factores de mal pronóstico y factores que predicen respuesta a tratamientos. Las células que escapan a los ganglios mantienen la metilación aberrante del tumor primario, rasgo que ha servido para detectar ADN tumoral circulando en la sangre de las pacientes con la enfermedad. Los resultados aportan a un mejor entendimiento del proceso tumoral mamario y al futuro diseño de estudios para la detección temprana y no invasiva de la enfermedad.

The transcription factor NF-κB is a pivotal regulator of inflammatory responses. While the activation of NF-κB in the arthritic joint has been associated with rheumatoid arthritis (RA), its significance is poorly understood. Here, we examine the role of NF-κB in animal models of RA. We demonstrate that in vitro, NF-κB controlled expression of numerous inflammatory molecules in synoviocytes and protected cells against tumor necrosis factor α (TNFα) and Fas ligand (FasL) cytotoxicity. Similar to that observed in human RA, NF-κB was found to be activated in the synovium of rats with streptococcal cell wall (SCW)-induced arthritis. In vivo suppression of NF-κB by either proteasomal inhibitors or intraarticular adenoviral gene transfer of super-repressor IκBα profoundly enhanced apoptosis in the synovium of rats with SCW- and pristane-induced arthritis. This indicated that the activation of NF-κB protected the cells in the synovium against apoptosis and thus provided the potential link between inflammation and hyperplasia. Intraarticular administration of NF-kB decoys prevented the recurrence of SCW arthritis in treated joints. Unexpectedly, the severity of arthritis also was inhibited significantly in the contralateral, untreated joints, indicating beneficial systemic effects of local suppression of NF-κB. These results establish a mechanism regulating apoptosis in the arthritic joint and indicate the feasibility of therapeutic approaches to RA based on the specific suppression of NF-κB.

The human inducible nitric oxide synthase (hiNOS) gene is expressed in several disease states and is also important in the normal immune response. Previously, we described a cytokine-responsive enhancer between −5.2 and −6.1 kb in the 5′-flanking hiNOS promoter DNA, which contains multiple nuclear factor κβ (NF-κB) elements. Here, we describe the role of the IFN-Jak kinase-Stat (signal transducer and activator of transcription) 1 pathway for regulation of hiNOS gene transcription. In A549 human lung epithelial cells, a combination of cytokines tumor necrosis factor-α, interleukin-1β, and IFN-γ (TNF-α, IL-1β, and IFN-γ) function synergistically for induction of hiNOS transcription. Pharmacological inhibitors of Jak2 kinase inhibit cytokine-induced Stat 1 DNA-binding and hiNOS gene expression. Expression of a dominant-negative mutant Stat 1 inhibits cytokine-induced hiNOS reporter expression. Site-directed mutagenesis of a cis-acting DNA element at −5.8 kb in the hiNOS promoter identifies a bifunctional NF-κB/Stat 1 motif. In contrast, gel shift assays indicate that only Stat 1 binds to the DNA element at −5.2 kb in the hiNOS promoter. Interestingly, Stat 1 is repressive to basal and stimulated iNOS mRNA expression in 2fTGH human fibroblasts, which are refractory to iNOS induction. Overexpression of NF-κB activates hiNOS promoter–reporter expression in Stat 1 mutant fibroblasts, but not in the wild type, suggesting that Stat 1 inhibits NF-κB function in these cells. These results indicate that both Stat 1 and NF-κB are important in the regulation of hiNOS transcription by cytokines in a complex and cell type-specific manner.

TRAF1 and TRAF2 form an oligomeric complex that associates with the cytoplasmic domains of various members of the tumor necrosis factor (TNF) receptor superfamily. TRAF2 action is required for activation of the transcription factor NF-kappaB triggered by TNF and the CD40 ligand. Here we show that TRAF1 and TRAF2 interact with A20, a zinc finger protein, whose expression is induced by agents that activate NF-kappaB. Mutational analysis revealed that the N-terminal half of A20 interacts with the conserved C-terminal TRAF domain of TRAF1 and TRAF2. In cotransfection experiments, A20 blocked TRAF2-mediated NF-kappaB activation. A20 also inhibited TNF and IL-1-induced NF-kappaB activation, suggesting that it may inhibit NF-kappaB activation signaled by diverse stimuli. The ability of A20 to block NF-kappaB activation was mapped to its C-terminal zinc finger domain. Thus, A20 is composed of two functionally distinct domains, an N-terminal TRAF binding domain that recruits A20 to the TRAF2-TRAF1 complex and a C-terminal domain that mediates inhibition of NF-kappaB activation. Our findings suggest a possible molecular mechanism that could explain A20's ability to negatively regulate its own TNF-inducible expression.

Clusterina (CLU) è una proteina ubiquitaria, presente nella maggior parte dei fluidi corporei e implicata in svariati processi fisiologici. Dalla sua scoperta fino ad oggi, CLU è risultata essere una proteina enigmatica, la cui funzione non è ancora stata compresa appieno. Il gene codifica per 3 varianti trascrizionali identificate nel database NCBI con i codici: NM_001831 (CLU 1 in questo lavoro di tesi), NR_038335 (CLU 2 in questo lavoro di tesi) e NR_045494 (CLU 3 in questo lavoro di tesi). Tutte le varianti sono trascritte come pre-mRNA contenenti 9 esoni e 8 introni e si differenziano per l’esone 1, la cui sequenza è unica e caratteristica di ogni variante. Sebbene in NCBI sia annotato che le varianti CLU 2 e CLU 3 non sono codificanti, tramite analisi bioinformatica è stato predetto che da tutti e tre i trascritti possono generarsi proteine di differente lunghezza e localizzazione cellulare. Tra tutte le forme proteiche ipotizzate, l’unica a essere stata isolata e sequenziata è quella tradotta dall’AUG presente sull’esone 2 che dà origine a una proteina di 449 aminoacidi. Il processo di maturazione prevede la formazione di un precursore citoplasmatico (psCLU) che subisce modificazioni post-traduzionali tra cui formazione di ponti disolfuro, glicosilazioni, taglio in due catene denominate β e α prima di essere secreta come eterodimero βα (sCLU) nell’ambiente extracellulare, dove esercita la sua funzione di chaperone ATP-indipendente. Oltre alla forma extracellulare, è possibile osservare una forma intracellulare con localizzazione citosolica la cui funzione non è stata ancora completamente chiarita. Questo lavoro di tesi si è prefissato lo scopo di incrementare le conoscenze in merito ai trascritti CLU 1 e CLU 2 e alla loro regolazione, oltre ad approfondire il ruolo della forma citosolica della proteina in relazione al signaling di NF-kB che svolge un ruolo importante nel processo di sviluppo e metastatizzazione del tumore. Nella prima parte, uno screening di differenti linee cellulari, quali cellule epiteliali di prostata e di mammella, sia normali sia tumorali, fibroblasti di origine polmonare e linfociti di tumore non-Hodgkin, ha permesso di caratterizzare i trascritti CLU 1 e CLU 2. Dall’analisi è emerso che la sequenza di CLU 1 è più corta al 5’ rispetto a quella depositata in NCBI con l’identificativo NM_001831 e il primo AUG disponibile per l’inizio della traduzione è localizzato sull’esone 2. È stato dimostrato che CLU 2, al contrario di quanto riportato in NCBI, è tradotto in proteina a partire dall’AUG presente sull’esone 2, allo stesso modo in cui viene tradotto CLU 1. Inoltre, è stato osservato che i livelli d’espressione dei trascritti variano notevolmente tra le diverse linee cellulari e nelle cellule epiteliali CLU 2 è espressa sempre a bassi livelli. In queste cellule, l’espressione di CLU 2 è silenziata per via epigenetica e la somministrazione di farmaci capaci di rendere la cromatina più accessibile, quali tricostatina A e 5-aza-2’-deossicitidina, è in grado di incrementarne l’espressione. Nella seconda parte, un’analisi bioinformatica seguita da saggi di attività in vitro in cellule epiteliali prostatiche trattate con farmaci epigenetici, hanno permesso di identificare, per la prima volta in uomo, una seconda regione regolatrice denominata P2, capace di controllare l’espressione di CLU 2. Rispetto a P1, il classico promotore di CLU già ampiamente studiato da altri gruppi di ricerca, P2 è un promotore debole, privo di TATA box, che nelle cellule epiteliali prostatiche è silente in condizioni basali e la cui attività incrementa in seguito alla somministrazione di farmaci epigenetici capaci di alterare le modificazioni post-traduzionali delle code istoniche nell’intorno di P2. Ne consegue un rilassamento della cromatina e un successivo aumento di trascrizione di CLU 2. La presenza di un’isola CpG differentemente metilata nell’intorno di P1 spiegherebbe, almeno in parte, i differenti livelli di espressione di CLU che si osservano tra le diverse linee cellulari. Nella terza parte, l’analisi del pathway di NF-kB in un modello sperimentale di tumore prostatico in cui CLU è stata silenziata o sovraespressa, ha permesso di capire come la forma citosolica di CLU abbia un ruolo inibitorio nei confronti dell’attività del fattore trascrizionale NF-kB. CLU inibisce la fosforilazione e l’attivazione di p65, il membro più rappresentativo della famiglia NF-kB, con conseguente riduzione della trascrizione di alcuni geni da esso regolati e coinvolti nel rimodellamento della matrice extracellulare, quali l’urochinasi attivatrice del plasminogeno, la catepsina B e la metallo proteinasi 9. È stato dimostrato che tale inibizione non è dovuta a un’interazione fisica diretta tra CLU e p65, per cui si suppone che CLU interagisca con uno dei componenti più a monte della via di segnalazione responsabile della fosforilazione ed attivazione di p65.

O sistema imunológico preserva a integridade do organismo perante o ambiente que ele está inserido. As reações imunológicas são essenciais para controle e eliminação da infecção, no entanto se os mecanismos contra regulatórios da resposta imunológica forem superados, a homeostasia pode falhar levando a um desequilíbrio no processo de reparo do organismo, podendo causar danos, insuficiência de órgão e até a morte. Muitos estudos têm demonstrado a interação entre o sistema imunológico e os aminoácidos. Visto que a glutamina é utilizada como substrato energético para enterócitos, além de fornecer nitrogênio para síntese de purinas e pirimidinas para proliferação celular e a taurina participa da hemostasia, estabilização de membranas, mobilização de cálcio, além de ser importante agente antioxidante, nos propusemos nesse trabalho investigar os efeitos da glutamina e taurina sobre aspectos relacionados a resposta imunológica de células da linhagem Raw 264.7. Para tanto foram avaliadas: viabilidade celular; a capacidade proliferativa e o ciclo celular; a capacidade de síntese de citocinas: IL-1 α, IL-1β, IL-6, IL-10 e TNF-α e a expressão do fator de transcrição NFκB, bem como de seu inibidor IκBα. Foi possível observar aumento da viabilidade e proliferação celular para células tratadas com glutamina, entretanto não foi observado o mesmo efeito para taurina. Quando ambos aminoácidos foram associados, houve prevalência dos efeitos da glutamina. Observamos neste trabalho, que houve uma tendência da diminuição de expressão da relação de p-NFκB/NFκB quando se aumentou a concentração de glutamina. Paralelamente, o mesmo foi encontrado para relação p-IκB/IκB. Esses resultados corroboram com os resultados encontrados na produção das citocinas pró-inflamatórias IL-1α, Il-1β e TNF-α, uma vez que ao aumentarmos a concentração de glutamina bem como glutamina e taurina, observamos menor produção das mesmas. Complementarmente, encontramos resultados opostos para a citocina anti-inflamatória IL-10, a qual teve maior síntese em resposta ao aumento da concentração dos aminoácidos. Portanto concluímos que tanto a glutamina quanto a taurina possuem capacidade de modular aspectos da resposta imunológica de células Raw 264.7.
