Squamous cell carcinoma of oral tongue shows high rates of morbidity and mortality in the population, therefore, great efforts are being made to classify morphological changes and identify biomarkers that have prognostic value and that are able to group patients in individualized therapeutic options. From this perspective, there is the heat shock factor 1 (HSF1), which is a heat shock factor transcription protein (HSPs) that allows the cancer to deal with stressors associated with malignancy, acting differently in tumor progression. This research aimed to perform a clinico-pathological analysis of 70 cases of oral tongue squamous cell carcinoma (OTSCC) and immunohistochemical study of the expression of HSF1 protein in OTSCC, comparing it with 30 specimens of normal oral mucosa (NOM), and correlating this immunostaining with clinico-pathological aspects of OTSCC. To analyze the association between immunoexpression of HSF1 and clinicophatoloical aspects, the cases were categorized in minor and major overexpression, based in the median immunostaining score. Regarding the cases of OTSCC, 57.1% showed clinical stage III or IV, 82.9% were graded as high grade according to Bryne (1998) and 47.1% as high risk of malignancy according to Brandwein-Gensler et al., (2005). A disease free survival rate of 47.84% and overall survival rate of 68.20% was observed in the analyzed cases, and the high degree of malignancy according to Bryne’s system (1998) (p=0.05), tumor size T3 or T4 (p=0.04), local recurrence (p=0.02), and perineural invasion (p=0.02) determined negative impacts in survival time. We observed also a statistically significant result (p<0.01) when comparing the immunoreactivity of HSF1 between NOM and OTSCC. This significantly increased expression of HSF1 in cases of OTSCC suggests that this protein acts, indeed, in the pathogenesis of this disease. However, there were no statistically significant associations between this overexpression and the clinico-pathological parameters analyzed. This finding may reflect the influence of epigenetic events on HSF1 gene or a possible stability of this protein expression throughout disease progression.

Squamous cell carcinoma (SCC ) is the most common malignancy of the oral cavity (OSCC), with a high mortality rate. Due to this, the discovery of biomarkers that facilitate the understanding of the biological behavior of the tumor and improve treatment is necessary. Urokinase type plasminogen activator (uPA) and its receptor, uPAR, are responsible for the proteolysis of structures of the basement membrana and extracellular matrix, facilitating tumor invasion. This study aims to assess the immuno expression of these proteins in 46 cases of squamous cell carcinoma of the oral tongue (OTSCC). These results were related to the presence of metastasis, clinical TNM staging, locoregional recurrence, outcome of the lesion and histological grading. Immunostaining of each case was evaluated semiquantitatively, in the front of invasion and center of the tumor, in which scores were assigned: 0 (0% of positive cells), 1 (1-10% of positive cells), 2 (11 -50% positive cells) and 3 (more than 50% positive cells). The expression of uPA was observed in 93.5% (n=43) of the cases in the front of invasion, with predominance of score 2 (n=16; 34.8%) and in 67.9% (n=31) of the cases in the center of the tumor, with predominance of score 1 (n=15; 32.6%). Overall, the immunoexpression of uPA was not associated with clinical parameters. Regarding the malignant histological grading, a higher expression of uPA was observed in cases of high-grade malignancy comp ared to low-grade malignancy (p=0.05). Regarding the morphological parameters, increased expression of uPA was observed in the worst mode of invasion (p=0.03 ). The expression of uPAR was observed in 73.9% of cases in the front of invasion, with a predominance of score 1 (n=21; 45.6 %), and in 47.5% (n=21) of the cases in the center of the tumor, with a predominance of score 0 (n=25; 54.4%). Although no statistical differences were observed in relation to lymph node metastasis, clinical TNM staging, outcome, and histological grading, there was a higher expression of uPAR in cases with locoregional recurrence (p=0.04). Regarding the tumor intra -localization, it was observed an increased expression of uPA and uPAR at the front of invasion in relation to the center of the tumor (p<0.001). Regarding the correlation between uPA and uPAR, there was no statistical sign ificance. Based on these results, it is suggested that uPA and uPAR are involved in the progression of CELO, mainly in the deeper region of the tumor.

DNA repair systems, genes and proteins are essential for genome integrity maintenance, avoiding serious diseases such as cancer. Deregulation in the expression of those proteins has been associated with both the risk of development and evolution of various human cancers, including oral squamous cell carcinoma. The purpose of this study was to analyze the immunoreactivity of the DNA repair proteins XRCC1, THIIF and XPF in oral tongue squamous cell carcinoma (OTSCC) and to investigate its association with clinical and histopathological parameters, outcome and 5-year survival rate. Seventy-four cases of OTSCC were analyzed semi-quantitatively through immunohistochemistry. We observed that DNA repair proteins were highly expressed in parenchymal cells; however, we only observed a significant association between XRCC1 high expression and better clinical staging (p=0,02). Cox regression showed that tumor size (p<0,01), lymph node involvement (p=0,04), tumor stage (p=0,02) and depth of invasion> 4mm (p=0,05) were prognostic factors. The results of this experiment suggest that XRCC1, TFIIH and XPF participate in the tumorigenic process, however, their immunoexpression may not be used as an independent prognostic indicator for OTSCC.

Introduction: Gastric cancer is currently the fourth higher cancer mortality rate among men in the world and the fifth among women, despite the progressive advances in oncology. The identification of tumor receptors and the development of target-drugs to block them has contributed to increased survival and quality of life of patients, but it becomes important to know the tumor profile of the population being treated, avoiding burdening treatment with examinations and treatments that are not cost-effective. Objective: To evaluate the profile of the population with gastric cancer treated in five years at the Clinical Hospital of the Federal University of Uberlândia and verify the correlation between overexpression of HER-2 receptor with an unfavorable prognosis. Methods: 203 records with gastric cancer were selected through the system database, attending a five-year period, of which 117 paraffin blocks were available for immunohistochemical assessment of HER2 receptor. Results: 2.6% of tumors showed overexpression of HER2, considering for this study two crosses as positive. There was no statistically significant difference in correlation between expression of the HER2 receptor with age, gender, tumor grade, local involvement, Lauren classification, Borrmann classification or staging. Conclusion: For this studied population, we can conclude that there is no need to employ HER2 blockers with high cost as a target-therapy in patients with gastric cancer, since no clinical benefit probably will be obtained due to a low percentage of these patients that demonstrated superexpression of this receptor or even there is no patients with gastric cancer with superexpression of HER2 with more than three crosses of positivity in immunochemistry

Lung cancer is the most common of malignant tumors, with 1.59 million new cases worldwide in 2012. Early detection is the main factor to determine the survival of patients affected by this disease. Furthermore, the correct classification is important to define the most appropriate therapeutic approach as well as suggest the prognosis and the clinical disease evolution. Among the exams used to detect lung cancer, computed tomography have been the most indicated. However, CT images are naturally complex and even experts medical are subject to fault detection or classification. In order to assist the detection of malignant tumors, computer-aided diagnosis systems have been developed to aid reduce the amount of false positives biopsies. In this work it was developed an automatic classification system of pulmonary nodules on CT images by using Artificial Neural Networks. Morphological, texture and intensity attributes were extracted from lung nodules cut tomographic images using elliptical regions of interest that they were subsequently segmented by Otsu method. These features were selected through statistical tests that compare populations (T test of Student and U test of Mann-Whitney); from which it originated a ranking. The features after selected, were inserted in Artificial Neural Networks (backpropagation) to compose two types of classification; one to classify nodules in malignant and benign (network 1); and another to classify two types of malignancies (network 2); featuring a cascade classifier. The best networks were associated and its performance was measured by the area under the ROC curve, where the network 1 and network 2 achieved performance equal to 0.901 and 0.892 respectively.

Para o diagnóstico radiológico das patologias ginecológicas é essencial conhecer e compreender as indicações dos vários exames disponíveis e, para cada achado radiológico, integrar a idade, o contexto clínico e a história pregressa da doente. A Ressonância Magnética (RM) tem hoje um papel crucial no diagnóstico das doenças ginecológicas. Contudo, para maximizar o potencial desta técnica é imprescindível adequar o protocolo utilizado a cada caso e a cada doente e ter em conta algumas regras imprescindíveis à interpretação dos exames, que descreveremos neste artigo. A RM ginecológica é principalmente útil no estadiamento do carcinoma do colo do útero e do endométrio, podendo por vezes ser também útil na sua detecção, na avaliação da resposta ao tratamento, detecção da recidiva ou complicações e na avaliação de lesões anexiais de natureza indeterminada na ecografia. Nas doenças benignas é frequentemente usada na avaliação pré-terapêutica de leiomiomas uterinos, bem como na adenomiose e na endometriose. Em muitas destas situações há potenciais erros e pitfalls, para os quais o médico radiologista deve estar alerta, de forma a minimizar eventuais falhas diagnósticas ou erros de estadiamento.

O carcinoma do endométrio apresenta uma taxa de incidência em Portugal de cerca de 7.2%, sendo a 5ª neoplasia mais comum na mulher. Apesar de apresentar uma prevalência relativamente elevada, o seu prognóstico global é favorável, uma vez que 75% dos casos são diagnosticados em estádio precoce. O estudo por ressonância magnética é geralmente efectuado após a realização de uma ecografia para avaliação de uma hemorragia uterina anormal e após o diagnóstico histológico por histeroscopia ou ressecção. Contudo, a ressonância magnética pode apresentar um papel determinante no diagnóstico em casos de impossibilidade de biópsia e nos quais a biópsia é inconclusiva. Além do mais, apesar de esta técnica não ser contemplada na classificação para o estadiamento do carcinoma do endométrio da International Federation of Gynecology and Obstetrics de 2009, apresenta uma função fundamental no estadiamento pré-operatório destas doentes, sendo crucial para definir a abordagem cirúrgica e terapêutica. No presente artigo, as autoras descrevem o estado da arte da ressonância magnética funcional no diagnóstico e no estadiamento do carcinoma do endométrio, chamando a atenção para o papel do estudo dinâmico após administração de contraste endovenoso e do estudo ponderado em difusão nestes cenários através da revisão da literatura mais recente sobre este tópico.

O carcinoma do colo do útero é um dos tumores malignos mais frequentes a nível mundial. Para garantir a uniformização de critérios entre países com diferentes recursos, o estadiamento deste tumor permanece clínico, segundo as orientações da Federação Internacional de Ginecologia e Obstetrícia, e tem por principal objectivo a identificação das doentes que são candidatas à cirurgia. A avaliação radiológica é amplamente recomendada, quando disponível, com o objectivo de aumentar a acuidade do diagnóstico, assegurando a optimização terapêutica, sendo também recomendada no seguimento. Importa, assim, que o radiologista tenha presente não só o protocolo técnico adequado na suspeita de carcinoma do colo do útero e o respectivo espectro de apresentação radiológica, mas também algumas características da própria doença e possíveis abordagens terapêuticas, de forma a incluir no seu relatório toda a informação relevante. A ressonância magnética permanece o principal pilar na avaliação radiológica destas doentes, embora recentemente o papel da tomografia computorizada por emissão de positrões tenha vindo a ganhar relevo, sobretudo no que respeita à avaliação ganglionar e ao despiste de recidiva. Neste artigo as autoras dão uma perspectiva aprofundada da avaliação radiológica do carcinoma do colo do útero, deste o diagnóstico ao seguimento pós-terapêutico, à luz dos estudos mais recentes.

A ressonância magnética é um método excelente e cada vez é mais indicado na avaliação de alterações da vagina, em virtude da sua capacidade de avaliação multiplanar e alto contraste tecidual. A RM é usada no estadiamento de tumores da vagina e detecção de recidivas. Também é útil na caracterização de malformações congénitas. Neste artigo descrevemos as características em ressonância magnética das diferentes patologias congénitas e adquiridas da vagina.

As neoplasias malignas do pâncreas englobam vários tipos histologicos com características ima giológicas e comportamentos que permitem distingui-los entre si numa boa percentagem de situações. Contudo, nem sempre a sua diferenciação se toma possível sem o recurso às técnicas anátomo-patoló gicas, constituindo um grande desafio à imagiologia o diagnóstico diferencial com lesões benignas, com referencia particular às massas inflamatórias. O adenocarcinoma constitui cerca de 95% das neo plasias pancreáticas e é uma das grandes causas de morte por cancro nos países desenvolvidos. As técnicas de imagem aplicadas no diagnóstico e no estadiamento dos principais tumores do pâncreas são muito diversificadas, englobando meios inócuos, como a ecografia, métodos invasivos utilizando técnicas angiográficas ou recurso aos meios endoscópicos para acesso endocavitário. A utilidade de cada uma destas variedades técnicas depende essencialmente do tipo de tumor a estudar, tomando-se fundamental o correcto e o completo conhecimento das possibilidades e limitações de cada uma, com vista à aplicação racional dos meios na imagiologia do pâncreas.

A clínica veterinária de pequenos animais é um mundo vasto e complexo que necessita de um estudo contínuo. Este relatório de estágio está dividido em duas partes. A primeira parte contém a descrição das atividades realizadas durante o estágio curricular realizado na clínica AlcabidecheVet. A segunda parte corresponde a uma monografia cujo tema é doença renal crónica em animais de companhia. A doença renal crónica acomete tanto cães como gatos em idade avançada, tendo uma progressão irreversível. Vários estudos científicos têm sido realizados para melhorar a sua prevenção e diagnóstico, como o estudo de biomarcadores da função renal. A utitilização de biomarcadores no diagnóstico e controlo da doença renal é essencial para o seu estadiamento e para demarcar o avanço dos danos renais. É também necessário conhecer os mecanismos patológicos em ação nesta doença para que o seu tratamento e monitorização providenciem o máximo bem-estar e o prolongamento da vida do animal; Abstract: (Small Animal Practice and Surgery – Kidney Chronic Disease) The world of small animal veterinary practice is vast and complex and needs a continuous study. This report is divided in two parts. The first describes the activities performed during the curricular internship at the clinic AlcabidecheVet. The second consists of a monograph which theme is chronic kidney disease. Chronic kidney disease affects both dogs and cats of declining age, and has an irreversible progression. There’s been scientific advances in its prevention and diagnosis with the study of biomarkers of kidney function. It’s also necessary to understand the pathological mechanisms in action in this disease, so its treatment and control can aim towards the welfare and the life extension of pets.

O presente relatório apresenta as atividades desenvolvidas no estágio decorrido no Hospital Ars Veterinaria. A primeira parte do relatório refere-se ao relatório de casuística com apresentação de casos e procedimentos acompanhados. A segunda refere-se à monografia com o tema “Opções terapêuticas em hemangiossarcoma canino” e encontra-se ilustrada por três casos clínicos. O hemangiossarcoma é uma neoplasia descrita com origem endotelial, devido à aparência histológica, no entanto descobertas recentes sugerem que provém de células progenitoras hematopoiéticas. Os sinais clínicos são inespecíficos e variáveis. A localização mais comum é o baço, embora ocorra noutras localizações. O diagnóstico requer o estadiamento tumoral e para ser completo deve incluir hematologia e bioquímica sérica, provas de coagulação, exames imagiológicos e confirmação histopatológica. O tratamento base é cirúrgico associado a protocolos quimioterápicos baseados em doxorrubicina. Novas terapias começam a ser investigadas de forma a melhorar os tempos de sobrevivência desta neoplasia de prognóstico reservado; Abstract: Small animal medicine This report was elaborated following a traineeship at the Ars Veterinaria Hospital. The first part concerns about cases report and includes presentation of cases and followed procedures. The second part is the monography under the theme “therapeutical options in canine hemangiosarcoma” and it presents three clinical cases seen during the internship. The hemangiossarcoma is a cancer described to have endothelial origin, because of its histological appearance, but recent discoveries suggest that it originates from hematopoietic progenitor cells. The clinical signs are variable and unspecific. The most common location is the spleen, but others might exist. The diagnosis demands tumoral staging, which to be complete must include hematology and biochemical analysis, coagulation tests, imagiologic tests and histopathologic confirmation. The basic treatment is surgical, associated with chemotherapy based on doxorubicin. New therapies are beginning to be investigated, aiming improve survival times of this cancer with reserved prognosis.
