OBJECTIVE: The factors that induce remission of RA during pregnancy and the relapse occurring after delivery remain an enigma. In a previous study, we investigated gene-expression profiles of peripheral blood mononuclear cells (PBMC) in patients with RA and healthy women in late pregnancy and postpartum. Profiles of samples from both groups were similar in late pregnancy with elevated monocyte and decreased lymphocyte signatures. Postpartum, in RA PBMC the high level of monocyte transcripts persisted. Further increase was observed in adhesion, migration and signalling processes related to monocytes but also in lymphocytes despite similar clinical activity due to intensified drug treatment. This prompted us to investigate correlations between clinical parameters of disease activity and gene profiles. METHODS: Transcriptome data were correlated with RADAI, CRP, monocyte and lymphocyte counts. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway annotations, monocytes and lymphocytes signatures were used as reference information. RESULTS: Comparative analysis of PBMC expression profiles from RA patients during and after pregnancy with RADAI and CRP revealed a correlation of these disease activity parameters predominantly with monocyte transcripts. Genes related to cellular programs of adhesion, migration and response to infections were upregulated. Comparing clinically active and not-active RA patients postpartum revealed a cluster of 19 genes that could also identify active disease during pregnancy. CONCLUSION: The data suggest that an increase of the RADAI and an elevation of CRP is a consequence of molecular activation of monocytes. Furthermore, they indicate that molecular activation of T lymphocytes may remain clinically unrecognized postpartum. It is conceivable that a set of 19 genes may qualify as molecular disease activity marker.

There is a tremendous amount of mystery that surrounds the instruments of Antonio Stradivari. There have been many studies done in the past, but no one completely understands exactly how he made his instruments, or why they are still considered the best in the world. This project is designed to develop an engineering model of one of Stradivari's violins that will accurately simulate the structural and acoustic behavior of the instrument. It also hopes to shine some light on what makes the instruments of Stradivari unique when compared to other violins. It will focus on geometry and material properties, utilizing several modern engineering tools, including CT scanning, experimental modal analysis, finite element analysis, correlation techniques, and acoustic synthesis.

Divergence of anterior-posterior (AP) limb pattern and differences in vertebral column morphology are the two main examples of mammalian evolution. The Hox genes (homeobox containing gene) have been implicated in driving evolution of these structures. However, regarding Hox genes, how they contribute to the generation of mammalian morphological diversities, is still unclear. Implementing comparative gene expression and phenotypic rescue studies for different mammalian Hox genes could aid in unraveling this mystery. In the first part of this thesis, the expression pattern of Hoxd13 gene, a key Hox gene in the establishment of the limb AP pattern, was examined in developing limbs of bats and mice. Bat forelimbs exhibit a pronounced asymmetric AP pattern and offer a good model to study the molecular mechanisms that contribute to the variety of mammalian limbs. The data showed that the expression domain of bat Hoxd13 was shifted prior to the asymmetric limb plate expansion, whereas its domain in mice was much more symmetric. This finding reveals a correlation between the divergence of Hoxd13 expression and the AP patterning difference in limb development. The second part of this thesis details a phenotypic rescue approach by human HOXB1-9 transgenes in mice with Hoxb1-9 deletion, The mouse mutants displayed homeosis in cervical and anterior thoracic vertebrae. The human transgenes entirely rescued the mouse mutants, suggesting that these human HOX genes have similar functions to their mouse orthologues in anterior axial skeletal patterning. The anterior expressing human HOXB transgenes such as HOXB1-3 were expressed in the mouse embryonic trunk in a similar manner as their murine orthologues. However, the anterior boundary of human HOXB9 expression domain was more posterior than that of the mouse Hoxb9 by 2-3 somites. These data provide the molecular support for the hypothesis that Hox genes are responsible for maintaining similar anterior axial skeletal architectures cervical and anterior thoracic regions, but different architectures in lumbar and posterior thoracic regions between humans and mice. ^

This study analyzed the relationship of family support systems and adolescent pregnancy outcomes. The population for the study was 390 adolescents who had attended the Marion County Health Department Adolescent Family Life Project in Indianapolis, Indiana during a two-year period.^ The study is unique in that it afforded the opportunity to compare adolescent pregnancy-related characteristics, of white and non-white adolescents in the same study.^ The pregnancy outcomes studied were: Infant birthweight, school attendance, and pregnancy recidivism.^ Significant results were found in the analysis that supported other research in regard to factors that are associated with school attendance when family support, adolescent's age, and ethnicity were controlled. Infant birthweight and repeat pregnancy outcome relationships were not found to have any consistently significant relationship with independent variables anticipated to be associated. However, the comparisons of infant birthweight among the adolescents with, and without, family support, by ethnicity resulted in some interesting findings. Repeat pregnancy proved an enigma, in that there seemed to be almost no variables in this study that were associated with the adolescent having a repeat pregnancy.^ Familial support in this study seemed to be of less importance as a factor in adolescent pregnancy outcomes than was ethnicity. The non-white adolescents in this study had a better record for remaining in school, both those non-white adolescents who lived with parents, and those who did not live with parents. More low birthweight occurred in the non-white adolescent, both those adolescents who lived with parents, and those who did not live with parents. Repeat pregnancy occurred more in the non-white adolescent whether she lived with parents, or did not live with parents. ^

Ice core records demonstrate a glacial-interglacial atmospheric CO2 increase by ~100 ppm, while 14C calibration efforts document a strong decrease in atmospheric 14C concentration during this period. A calculated transfer of ~530 Gt of 14C depleted carbon is required to produce the deglacial coeval rise of carbon in the atmosphere and terrestrial biosphere. This amount is usually ascribed to oceanic carbon release, although the actual mechanisms remained elusive, since an adequately old and carbon-enriched deep-ocean reservoir seemed unlikely. Here we present a new, though still fragmentary, ocean-wide d14C dataset showing that during the Last Glacial Maximum (LGM) and Heinrich Stadial 1 (HS-1) the maximum 14C age difference between ocean deep waters and the atmosphere exceeded the modern values by up to 1500 14C yr, in the extreme reaching 5100 14C yr. Below 2000 m depth the 14C ventilation age of modern ocean waters is directly linked to the concentration of dissolved inorganic carbon (DIC). We propose as working hypothesis that the modern regression of DIC vs d14C also applies for LGM times, which implies that a mean LGM aging by ~600 14C yr corresponded to a global rise of ~85-115 µmol DIC/kg in the deep ocean. Thus, the prolonged residence time of ocean deep waters may indeed have made it possible to absorb an additional ~730-980 Gt DIC, one third of which possibly originated from intermediate waters. We also infer that LGM deep-water O2 dropped to suboxic values of <10µmol/kg in the Atlantic sector of the Southern Ocean, possibly also in the subpolar North Pacific. The outlined deglacial transfer of the extra aged, deep-ocean carbon to the atmosphere via the dynamic ocean-atmosphere carbon exchange would be sufficient to account for two trends observed, (1) for the increase in atmospheric CO2 and (2) for the 190-permil drop in atmospheric d14C during the so-called HS-1 'Mystery Interval', when atmospheric 14C production rates were largely constant.

Small biserial foraminifera were abundant in the early Miocene (ca. 18.9-17.2 Ma) in the eastern Atlantic and western Indian Oceans, but absent in the western equatorial Atlantic Ocean, Weddell Sea, eastern Indian Ocean, and equatorial Pacific Ocean. They have been assigned to the benthic genus Bolivina, but their high abundances in sediments without evidence for dysoxia could not be explained. Apertural morphology, accumulation rates, and isotopic composition show that they were planktic (genus Streptochilus). Living Streptochilus are common in productive waters with intermittent upwelling. The widespread early Miocene high Streptochilus abundances may reflect vigorous but intermittent upwelling, inducing high phytoplankton growth rates. However, export production (estimated from benthic foraminiferal accumulation rates) was low, possibly due to high regeneration rates in a deep thermocline. The upwelled waters may have been an analog to Subantarctic Mode Waters, carrying nutrients into the eastern Atlantic and western Indian Oceans as the result of the initiation of a deep-reaching Antarctic Circumpolar Current, active Agulhas Leakage, and vigorous vertical mixing in the Southern Oceans.

Only about half of all the CO_2 that has been produced by the burning of fossil fuels now remains in the atmosphere. The CO_2 "missing" from the atmosphere is the subject of an important debate. It was thought that the great majority of the missing CO_2 has invaded the ocean, for this system naturally acts as a giant chemical regulator of the atmosphere. Although it is clear that ocean processes have a major role in the regulation of the carbon dioxide content of the atmosphere through air-sea exchange processes, recent studies of the oceanic carbon cycle and air-sea interaction indicate that oceanic carbon is in a quasi-steady state via the system of biological and physical processes in the ocean interior. It is difficult to determine whether the ocean has the capacity to take up the increasing air-born CO_2 released by human activities over the past five or six decades. To understand this enigma, we need a better understanding of the natural variability of the oceanic carbon cycle.

The Ontong Java Plateau in the western Pacific is anomalous compared to other oceanic large igneous provinces in that it appears to have never formed a large subaerial plateau. Paleoeruption depths (at 122 Ma) estimated from dissolved H2O and CO2 in submarine basaltic glass pillow rims vary from ~1100 m below sea level (mbsl) on the central part of the plateau to 2200-3000 mbsl on the northeastern edge. Our results suggest maximum initial uplift for the plateau of 2500-3600 m above the surrounding seafloor and 1500+/-400 m of postemplacement subsidence since 122 Ma. Our estimates of uplift and subsidence for the plateau are significantly less than predictions from thermal models of oceanic lithosphere, and thus our results are inconsistent with formation of the plateau by a high-temperature mantle plume. Two controversial possibilities to explain the anomalous uplift and subsidence are that the plateau (1) formed as a result of a giant bolide impact, or (2) formed from a mantle plume but has a lower crust of dense garnet granulite and/or eclogite; neither of these possibilities is fully consistent with all available geological, geophysical, and geochemical data. The origin of the largest magmatic event on Earth in the past 200 m.y. thus remains an enigma.

The main motivation for Integrated Ocean Drilling Program Expedition 310 to the Tahitian Archipelago was the assumption that the last deglacial sea-level rise is precisely recorded in the coral reefs of this far-field site. The Tahitian deglacial succession typically consists of coral framework subsequently encrusted by coralline algae and microbialites. The high abundance of microbialites is uncommon for shallow-water coral reefs, and the environmental conditions favouring their development are still poorly understood. Microbioerosion patterns in the three principal framework components (corals, coralline algae, microbialites) are studied with respect to relative light availability during coral growth and subsequent encrustation, in order to constrain the palaeobathymetry and the relative timing of the encrustation. Unexpectedly for a tropical, light-flooded setting, ichnotaxa typical for the deep-euphotic to dysphotic zone dominate. The key ichnotaxa for the shallow euphotic zone are scarce in the analysed sample set, and are restricted tothe baseof thedeglacial succession, thus reflecting thedeglacial sea-level rise. At the base of the deglacial reef succession, the ichnocoenoses present in the corals indicate shallower bathymetries than those in the encrusting microbialites. This is in agreement with radiocarbon data that indicate a time gap of more than 600 years between coral death and microbialite formation. At the top of the deglacial reef succession, in contrast, the microbioerosion patterns in the three framework components indicate a uniform palaeobathymetry, and radiocarbon ages imply that encrustation took place shortly after coral demise. An enigma arises from the fact that the ichnocoenoses imply photic conditions that appear very deep for zooxanthellate coral growth. During the deglacial sea-level rise increased nutrients and fluvial influx may have led to (seasonal?) eutrophication, condensing the photic zonation. This would have exerted stress on the coral ecosystem and played a significant role in initiating microbialite development.

A man of humble beginnings, Gaines had dreams and goals for which he was willing to overcome outdated legal, social and educational systems do not only better himself but make a stand for his fellow man. The story of Lloyd Gaines is one of struggle, hope, success and injury. It is a life and legacy to be recognized, admired and celebrated.

Cpefat mice carry a mutation in the carboxypeptidase E/H gene which encodes an exopeptidase that removes C-terminal basic residues from endoproteolytically cleaved hormone intermediates. These mice have endocrine disorders including obesity, infertility, and hyperproinsulinemia–diabetes syndrome, but the etiology remains an enigma. Because studies have identified membrane carboxypeptidase E as a sorting receptor for targeting prohormones to the regulated secretory pathway for processing and secretion, the intracellular routing and secretion of pro-opiomelanocortin/adrenocorticotropin and growth hormone from anterior pituitary cells were investigated in Cpefat mice. In Cpefat mice, pro-opiomelanocortin was accumulated 24-fold above normal animals in the pituitary and it was poorly processed to adrenocorticotropin. Furthermore, pro-opiomelanocortin was secreted constitutively at high levels, showing no response to stimulation by corticotropin-releasing hormone. Similarly, growth hormone release was constitutive and did not respond to high K+ stimulation. Both pro-opiomelanocortin and growth hormone levels were elevated in the circulation of Cpefat mice versus normal mice. These data provide evidence that the lack of carboxypeptidase E, the sorting receptor, results in the intracellular misrouting and secretion of pro-opiomelanocortin and growth hormone via the constitutive pathway in the pituitary of Cpefat mice.
