Rising anthropogenic carbon dioxide (CO2) dissolving into coastal waters is decreasing the pH and carbonate ion concentration, thereby lowering the saturation state of calcium carbonate (CaCO3) minerals through a process named ocean acidification (OA). The unprecedented threats posed by such low pH on calcifying larvae of several edible oyster species have not yet been fully explored. Effects of low pH (7.9, 7.6, 7.4) on the early growth phase of Portuguese oyster (Crassostrea angulata) veliger larvae was examined at ambient salinity (34 ppt) and the low-salinity (27 ppt) treatment. Additionally, the combined effect of pH (8.1, 7.6), salinity (24 and 34 ppt) and temperature (24 °C and 30 °C) was examined using factorial experimental design. Surprisingly, the early growth phase from hatching to 5-day-old veliger stage showed high tolerance to pH 7.9 and pH 7.6 at both 34 ppt and 27 ppt. Larval shell area was significantly smaller at pH 7.4 only in low-salinity. In the 3-factor experiment, shell area was affected by salinity and the interaction between salinity and temperature but not by other combinations. Larvae produced the largest shell at the elevated temperature in low-salinity, regardless of pH. Thus the growth of the Portuguese oyster larvae appears to be robust to near-future pH level (> 7.6) when combined with projected elevated temperature and low-salinity in the coastal aquaculture zones of South China Sea.

Project of modular multifunctional structure in Colombia Guadua (Guadua angustifolia Kunth) is one of the forest species more characteristics of Colombia. Pertaining to the family of the bamboo, this one native plant of Colombia used in the construction allows a reduction of costs, guaranteed, among others advantages, by its physic mechanical properties of flexibility, resistance, hardness, climatic effect and earthquake resistance. The project initially considers a study of the Guadua as structural material. We will study its anatomy, its physical and mechanical properties as well as the existing norm that governs its use in construction and finally some examples will be provided in which its actual use can be verified. Later on, we will proceed with the development of the project which purpose is the design of a multifunctional modular structure required to in a project of cooperation located in Neiva (Colombia). The project will include all the necessary documents for a work of these characteristics: memory, technical specifications, budget, drawings, as well as the Security and Labor Health Study. In order to satisfy the needs raised in the cooperation project, we will undertake the design of two modular structures with the purpose to be used as office and warehouse. The construction normative CTE will be applied for its calculation, but some parameters will be based in the NSR-10, establishing a comparison between both norms. Due to the importance of the seismic action in Colombia, the seismic resistance of the module will be verified according to the “Manual of Construction, evaluation and rehabilitation resistant earthquake of rubblework houses” created by the Colombian Association of Seismic Engineering,. We will also use this Manual to define the types of unions more favorable from seismic point of view, designing the required auxiliary segments according to the type of union.

Under-deck cable-stayed bridges are very effective structural systems for which the strong contribution of the stay cables under live loading allows for the design of very slender decks for persistent and transient loading scenarios. Their behaviour when subjected to seismic excitation is investigated herein and a set of design criteria are presented that relate to the type and arrangement of bearings, the number and configuration of struts, and the transverse distribution of stay cables. The nonlinear behaviour of these bridges when subject to both near-field and far-field accelerograms has been thoroughly investigated through the use of incremental dynamic analyses. An intensity measure that reflects the pertinent contributions to response when several vibration modes are activated was proposed and is shown to be effective for the analysis of this structural type. The under-deck cable-stay system contributes in a very positive manner to reducing the response when the bridges are subject to very strong seismic excitation. For such scenarios, the reduction in the stiffness of the deck because of crack formation, when prestressed concrete decks are used, mobilises the cable system and enhances the overall performance of the system. Sets of natural accelerograms that are compliant with the prescriptions of Eurocode 8 were also applied to propose a set of design criteria for this bridge type in areas prone to earthquakes. Particular attention is given to outlining the optimal strategies for the deployment of bearings

Bridges with deck supported on either sliding or elastomeric bearings are very common in mid-seismicity regions. Their main seismic vulnerabilities are related to the pounding of the deck against abutments or between the different deck elements. A simplified model of the longitudinal behavior of those bridges will allow to characterize the reaction forces developed during pounding using the Pacific Earthquake Engineering Research Center framework formula. In order to ensure the general applicability of the results obtained, a large number of system parameter combinations will be considered. The heart of the formula is the identification of suitable intermediate variables. First, the pseudo acceleration spectral value for the fundamental period of the system (Sa(Ts)) will be used as an intensity measure (IM). This IM will result in a very large non-explained variability of the engineering demand parameter. A portion of this variability will be proved to be related to the relative content of high-frequency energy in the input motion. Two vector-valued IMs including a second parameter taking this energy content into account will then be considered. For both of them, a suitable form for the conditional intensity dependence of the response will be obtained. The question of which one to choose will also be analyzed. Finally, additional issues related to the IM will be studied: its applicability to pulse-type records, the validity of scaling records and the sufficiency of the IM.

This work proposes design energy spectra in terms of velocity, derived through linear dynamic analyses on Turkish registers and intended for regions with design peak acceleration 0.3 g or higher. In the long and mid period ranges the analyses are linear, taking profit of the rather insensitivity of the spectra to the structural parameters other than the fundamental period; in the short period range, the spectra are more sensitive to the structural parameters and nonlinear analyses would be required. The selected records are classified in eight groups according to the design input acceleration, the soil type, the earthquake magnitude and the near-source effects. For each of these groups, median and characteristic spectra are proposed (50% and 95% percentiles). These spectra have an initial linear growing branch in the short period range, a horizontal branch in the mid period range and a descending branch in the long period range.

An evaluation of the seismic hazard in La Hispaniola Island has been carried out, as part of the cooperative project SISMO-HAITI, supported by the Technical University of Madrid (UPM) and developed by several Spanish Universities, the National Observatory of Environment and Vulnerability) ONEV of Haiti, and with contributions from the Puerto Rico Seismic Network (PRSN) and University Seismological Institute of Dominican Republic (ISU). The study was aimed at obtaining results suitable for seismic design purposes. It started with the elaboration of a seismic catalogue for the Hispaniola Island, requiring an exhaustive revision of data reported by more than 20 seismic agencies, apart from these from the PRSN and ISU. The final catalogue contains 96 historical earthquakes and 1690 instrumental events, and it was homogenized to moment magnitude, Mw. Seismotectonic models proposed for the region were revised and a new regional zonation was proposed, taking into account geological andtectonic data, seismicity, focal mechanisms, and GPS observations. In parallel, attenuation models for subduction and crustal zones were revised in previous projects and the most suitable for the Caribbean plate were selected. Then, a seismic hazard analysis was developed in terms of peak ground acceleration, PGA, and spectral accelerations, SA (T), for periods of 0.1, 0.2, 0.5, 1 and 2s, using the Probabilistic Seismic Hazard Assessment (PSHA) methodology. As a result, different hazard maps were obtained for the quoted parameters, together with Uniform Hazard Spectra for Port au Prince and the main cities in the country. Hazard deaggregation was also carried out in these towns, for the target motion given by the PGA and SA (1s) obtained for return periods of 475, 975 and 2475 years. Therefore, the controlling earthquakes for short- and long-period target motions were derived. This study was started a few months after the 2010 earthquake, as a response to an aid request from the Haitian government to the UPM, and the results are available for the definition of the first building code in Haiti.

This work proposes design energy spectra in terms of an equivalent velocity, intended for regions with design peak acceleration 0.3 g or higher. These spectra were derived through linear and nonlinear dynamic analyses on a number of selected Turkish strong ground motion records. In the long and mid period ranges the analyses are linear, given the relative insensitivity of the spectra to structural parameters other than the fundamental period; conversely, in the short period range, the spectra are more sensitive to the structural parameters and, hence, nonlinear analyses are required. The selected records are classified in eight groups with respect to soil type (stiff or soft soil), the severity of the earthquake in terms of surface magnitude Ms(Ms≤ 5.5 and Ms> 5.5) and the relevance of the near-source effects (impulsive or vibratory). For each of these groups, median and characteristic spectra are proposed; such levels would respectively correspond to 50 and 95 % percentiles. These spectra have an initial linear growing branch in the short period range, a horizontal branch in the mid period range and a descending branch in the long period range. Empirical criteria for estimating the hysteretic energy from the input energy are suggested. The proposed design spectra are compared with those obtained from other studies.

On Wednesday 11th May 2011 at 6:47 pm (local time) a magnitude 5.1 Mw earthquake occurred 6 km northeast of Lorca with a depth of around 5 km. As a consequence of the shallow depth and the small epicentral distance, important damage was produced in several masonry constructions and even led to the collapse of one of them. Pieces of the facades of several buildings fell down onto the sidewalk, being one of the reasons for the killing of a total of 9 people. The objective of this paper is to describe and analyze the failure patterns observed in reinforced concrete frame buildings with masonry infill walls ranging from 3 to 8 floors in height. Structural as well as non-structural masonry walls suffered important damage that led to redistributions of forces causing in some cases the failure of columns. The importance of the interaction between the structural frames and the infill panels is analyzed by means of non-linear Finite Element Models. The resulting load levels are compared with the member capacities and the changes of the mechanical properties during the seismic event are described and discussed. In the light of the results obtained the observed failure patterns are explained. Some comments are stated concerning the adequacy of the numerical models that are usually used during the design phase for the seismic analysis.

The design of containment walls suffering seismic loads traditionally has been realized with methods based on pseudoanalitic procedures such as Mononobe-Okabe's method, which it has led in certain occasions to insecure designs, that they have produced the ruin of many containment walls suffering the action of an earthquake. The recommendations gathered in Mononobe-Okabe's theory have been included in numerous Codes of Seismic Design. It is clear that a revision of these recommendations must be done. At present there is taking place an important review of the design methods of anti-seismic structures such as containment walls placed in an area of numerous earthquakes, by means of the introduction at the beginning of the decade of 1990 the Displacement Response Spectrum (DRS) and the Capacity Demand Diagram (CDD) that suppose an important change in the way of presenting the Elastic Response Spectrum (ERS). On the other hand in case of action of an earthquake, the dynamic characteristics of a soil have been referred traditionally to the speed of the shear waves that can be generated in a site, together with the characteristics of plasticity and damping of the soil. The Principle of the energy conservation explains why a shear upward propagating seismic wave can be amplified when travelling from a medium with high shear wave velocity (rock) to other medium with lower velocity (soil deposit), as it happened in the earthquake of Mexico of 1985. This amplification is a function of the speed gradient or of the contrast of impedances in the border of both types of mediums. A method is proposed in this paper for the design of containment walls in different soils, suffering to the action of an earthquake, based on the Performance-Based Seismic Design.

ABSTRACT In the nuclear fusion field running in parallel to ITER (International Thermonuclear Experimental Reactor) as one of the complementary activities headed towards solving the technological barriers, IFMIF (International Fusion Material Irradiation Facility) project aims to provide an irradiation facility to qualify advanced materials resistant to extreme conditions like the ones expected in future fusion reactors like DEMO (DEMOnstration Power Plant). IFMIF consists of two constant wave deuteron accelerators delivering a 125 mA and 40 MeV beam each that will collide on a lithium target producing an intense neutron fluence (1017 neutrons/s) with a similar spectra to that of fusion neutrons [1], [2]. This neutron flux is employed to irradiate the different material candidates to be employed in the future fusion reactors, and the samples examined after irradiation at the so called post-irradiative facilities. As a first step in such an ambitious project, an engineering validation and engineering design activity phase called IFMIF-EVEDA (Engineering Validation and Engineering Design Activities) is presently going on. One of the activities consists on the construction and operation of an accelerator prototype named LIPAc (Linear IFMIF Prototype Accelerator). It is a high intensity deuteron accelerator identical to the low energy part of the IFMIF accelerators. The LIPAc components, which will be installed in Japan, are delivered by different european countries. The accelerator supplies a 9 MeV constant wave beam of deuterons with a power of 1.125 MW, which after being characterized by different instruments has to be stopped safely. For such task a beam dump to absorb the beam energy and take it to a heat sink is needed. Spain has the compromise of delivering such device and CIEMAT (Centro de Investigaciones Energéticas Medioambientales y Tecnológicas) is responsible for such task. The central piece of the beam dump, where the ion beam is stopped, is a copper cone with an angle of 3.5o, 2.5 m long and 5 mm width. This part is cooled by water flowing on its external surface through the channel formed between the copper cone and a concentric piece with the latter. The thesis is developed in this realm, and its objective is designing the LIPAc beam dump cooling system. The design has been performed employing a simplified one dimensional model. The water parameters (pressure, flow, pressure loss) and the required annular channel geometry (width, rugoisty) have been obtained guaranteeing the correct cooling of the beam dump. It has been checked that the cooling design allows variations of the the beam with respect to the nominal position, being the CHF (Critical Heat Flux) at least twice times higher than the nominal deposited heat flux. 3D fluid dynamic simulations employing ANSYS-CFX code in the beam dump cooling channel sections which require a more thorough study have also been performed. The beam dump will activateasaconsequenceofthe deuteron beam interaction, making impossible any change or maintenance task once the accelerator operation has started. Hence the design has to be very robust and all the hypotheses employed in the design mustbecarefully checked. Most of the work in the thesis is concentrated in estimating the heat transfer coefficient which is decisive in the obtained results, and is also employed as boundary condition in the mechanical analysis. For such task, correlations which applicability range is the adequate for the beam dump conditions (annular channel, water-surface temperature differences of tens of degrees) have been compiled. In a second step the heat transfer coefficients obtained from the selected correlation (Petukhov- Gnielinski) have been compared with the ones deduced from the 3D fluid dynamic simulations, obtaining satisfactory results. Finally an experimental validation has been performed employing a prototype and a hydraulic circuit that supplies a flow with the requested parameters in the beam dump. After several tries and improvements in the experiment, the heat transfer coefficients for different flows and heating powers have been obtained. Considering the uncertainty in the measurements the experimental values agree reasonably well (in the order of 15%) with the ones obtained from the correlations. Due to radiological reasons the quality of the cooling water must be controlled, hence minimizing the copper corrosion. After performing a bibligraphic study the most adequate water parameters were identified (conductivity, pH and dissolved oxygen concentration). As part of this thesis a corrosion study of the beam dump cooling circuit has been performed with the double aim of determining if corrosion can pose a risk for the copper beam dump , and obtaining an estimation of the corrosion velocitytodimension the water purification system. TRACT code(TRansport and ACTivation) has been employed for such study adapting the code for the beam dump case. For such study a collaboration with the code responsible (Panos Karditsas) at Culham (UKAEA) was established. The work developed in this thesis has supposed the publication of three articles in JCR journals (”Journal of Nuclear Materials” y ”Fusion Engineering and Design), as well as presentations in more than four conferences and relevant meetings.

After the 2010 Haiti earthquake, that hits the city of Port-au-Prince, capital city of Haiti, a multidisciplinary working group of specialists (seismologist, geologists, engineers and architects) from different Spanish Universities and also from Haiti, joined effort under the SISMO-HAITI project (financed by the Universidad Politecnica de Madrid), with an objective: Evaluation of seismic hazard and risk in Haiti and its application to the seismic design, urban planning, emergency and resource management. In this paper, as a first step for a structural damage estimation of future earthquakes in the country, a calibration of damage functions has been carried out by means of a two-stage procedure. After compiling a database with observed damage in the city after the earthquake, the exposure model (building stock) has been classified and through an iteratively two-step calibration process, a specific set of damage functions for the country has been proposed. Additionally, Next Generation Attenuation Models (NGA) and Vs30 models have been analysed to choose the most appropriate for the seismic risk estimation in the city. Finally in a next paper, these functions will be used to estimate a seismic risk scenario for a future earthquake.

The paper proposes a new application of non-parametric statistical processing of signals recorded from vibration tests for damage detection and evaluation on I-section steel segments. The steel segments investigated constitute the energy dissipating part of a new type of hysteretic damper that is used for passive control of buildings and civil engineering structures subjected to earthquake-type dynamic loadings. Two I-section steel segments with different levels of damage were instrumented with piezoceramic sensors and subjected to controlled white noise random vibrations. The signals recorded during the tests were processed using two non-parametric methods (the power spectral density method and the frequency response function method) that had never previously been applied to hysteretic dampers. The appropriateness of these methods for quantifying the level of damage on the I-shape steel segments is validated experimentally. Based on the results of the random vibrations, the paper proposes a new index that predicts the level of damage and the proximity of failure of the hysteretic damper

Individuals with hemophilia A require frequent infusion of preparations of coagulation factor VIII. The activity of factor VIII (FVIII) as a cofactor for factor IXa in the coagulation cascade is limited by its instability after activation by thrombin. Activation of FVIII occurs through proteolytic cleavage and generates an unstable FVIII heterotrimer that is subject to rapid dissociation of its subunits. In addition, further proteolytic cleavage by thrombin, factor Xa, factor IXa, and activated protein C can lead to inactivation. We have engineered and characterized a FVIII protein, IR8, that has enhanced in vitro stability of FVIII activity due to resistance to subunit dissociation and proteolytic inactivation. FVIII was genetically engineered by deletion of residues 794-1689 so that the A2 domain is covalently attached to the light chain. Missense mutations at thrombin and activated protein C inactivation cleavage sites provided resistance to proteolysis, resulting in a single-chain protein that has maximal activity after a single cleavage after arginine-372. The specific activity of partially purified protein produced in transfected COS-1 monkey cells was 5-fold higher than wild-type (WT) FVIII. Whereas WT FVIII was inactivated by thrombin after 10 min in vitro, IR8 still retained 38% of peak activity after 4 hr. Whereas binding of IR8 to von Willebrand factor (vWF) was reduced 10-fold compared with WT FVIII, in the presence of an anti-light chain antibody, ESH8, binding of IR8 to vWF increased 5-fold. These results demonstrate that residues 1690–2332 of FVIII are sufficient to support high-affinity vWF binding. Whereas ESH8 inhibited WT factor VIII activity, IR8 retained its activity in the presence of ESH8. We propose that resistance to A2 subunit dissociation abrogates inhibition by the ESH8 antibody. The stable FVIIIa described here provides the opportunity to study the activated form of this critical coagulation factor and demonstrates that proteins can be improved by rationale design through genetic engineering technology.
